이 연구는 교과교육과 교과교육학의관계, 그리고 교과교육학과 교육학의 관계등을 분석하고, 그 결과를 토대로 하여 초등학교 교과교육과 교육대학 교과교육학 교육의 실제를 정확히 이해하고 그 발전 방향을 탐색하기 위한 것이다. 일반적으로, 교과교육학은 교육학에서 제시하고 있는 교육과정의 구성및 교육방법상의 일반 원리를 교과교육이라는 특수한 사태에 적용하는 과정에 관심을 두는 이론으로 간주되고 있다. 이 관점에 의하면, 교과교육학은 교육학의 일반적 원리에 따라 교과교육이라는 특수한 맥락에 처방을 내리기 위한 것으로 파악된다. 이 관점을 교과와 그것을 가르치는 교사의 삶을 사실상 별개의 것으로 분리하는 것이며, 그와 동일한 이유에서 교과교육학과 교과교육학자의 삶 또한 별개의 것임을 전제하는 것이다. 그러나 교과교육과 교과교육학, 그리고 교과교육학과 교육학의 관계는 이론에서 실제로 나아가는 앞의 방향과는 정반대 방향에서도 이해될 수 있다. 이 관점에 의하면, 교과교육학은 교과교육 실제에 처방을 내리기 위한 것이 아니라 그 실제를 기술하고 정당화하기 위한 것으로 파악된다. 교과교육과 교과교육학의 관련이 이렇게 파악될 때, 교과 교육학은 교사나 교과교육학자에게 자신이 지금 하는 일이 자신의 삶과 결코 무관한 것이 아니라는 믿음을 심어 주게 되며, 나아가 그들 자신이 참여하고 있는 초등학교나 교육대학의 교육실제를 기술하거나 정당화하려고 할 때 반드시 필요한 개념적 도구를 제공해 줄 수 있게 된다. 바로 여기에 교과교육학에 관한 종래의 접근과는 구분되는 새로운 접근방식이 요구되는 이유가 있다. The purpose of this study is threefold: The first is to analyse the relations between subject education and subject pedagogy, and between subject pedagogy and general pedagogy. The second is, based on the analysis result, to have the precise understanding of teaching various subjects in the elementary school, and of teaching subject pedagogy in the university of education. The third is to search for the desirable direction of developing the elementary education and the teacher education. Subject pedagogy is usually defined as a theory which is interested in the process of applying pedagogical principles regarding curriculum organization and teaching methods to specific teaching situations, that is, subject education situations. However, this definition has a limitation, because it assumes that a subject and the teacher's life who teaches the subject are separated from each other, and that subject pedagogy and the scholar who studies subject pedagogy are also separated. The relationship between subject teaching and subject pedagogy, and between subject pedagogy and general pedagogy can be understood with quite different viewpoint. This new viewpoint, which is the major discussion matter of this study, assumes that subject pedagogy is never separated from the life of subject teacher/subject pedagogist. This viewpoint will reveal the theoretical features of subject pedagogy better, and give us new meaning on the life of teachers/subject pedagogists. The subject pedagogy in the correct sense should provide teachers/pedagogists with the conceptual instrument based on the new viewpoint. This conceptual instrument will make teachers/pedagogists be able to describe, explain, and validate their own educational practice-such as the content structure of each subject-in the elementary school or the university of education.
Dandy-Walker Malformation Associated with Neurocutaneous Melanosis

Neurocutaneous melanosis associated with Dandy-Walker malformation is a rare dysmorphogenesis that is associated with single or multiple giant pigmented cutaneous nevi and diffuse involvement of the central nervous system. In this article, we present a 2-month-old patient with neurocutaneous melanosis associated with Dandy-Walker malformation. In addition, we reviewed the literature and discussed the pathogenesis based on the preferred hypotheses.

It is not yet understood how the enhanced expression of pancreatic adenocarcinoma up-regulated factor (PAUF; a novel oncogene identified in our recent studies), contributes to the oncogenesis of pancreatic cells. We herein report that PAUF up-regulates the expression and transcriptional activity of ${\beta}$-catenin while the suppression of PAUF by shRNA down-regulates ${\beta}$-catenin. The induction of ${\beta}$-catenin by PAUF is mediated by the activities of Akt and GSK-$3{\beta}$, but inhibition of downstream ERK does not reduce ${\beta}$-catenin expression. To test whether PAUF emulates either the Wnt3a-mediated or the protein kinase A-mediated signaling pathway for the stabilization of ${\beta}$-catenin, we examined the phosphorylation status of ${\beta}$-catenin in the presence of PAUF compared with that of ${\beta}$-catenin during treatment with Wnt3a or dibutyryl cAMP, a cell permeable cyclic AMP analogue. PAUF expression induces phosphorylation at Ser-33/37/Thr-41 and Ser-675 of ${\beta}$-catenin but no phosphorylation at Ser-45, indicating that a unique phosphorylation pattern of ${\beta}$-catenin is caused by PAUF. Finally, the expression of PAUF up-regulates both $cyclin-D1$ and $c-Jun$, target genes of ${\beta}$-catenin, leading to a rapid proliferation of pancreatic cells; conversely decreased PAUF expression (by shRNA) results in the reduced proliferation of pancreatic cells. Treatment with hexachlorophene (an inhibitor of ${\beta}$-catenin) reduces the proliferation of pancreatic cells despite the presence of PAUF. Taken together, we propose that PAUF can up-regulate and stabilize ${\beta}$-catenin $via$ a novel pattern of phosphorylation, thereby contributing to the rapid proliferation of pancreatic cancer cells.

<P><I>Context</I>: Caffeic acid phenethyl ester (CAPE), an active component of honeybee propolis, is known to have antioxidant, anti-inflammatory, and other beneficial medicinal properties. However, the molecular mechanisms underlying its anti-allergic effects in mast cells are unknown.</P><P><I>Objective</I>: The purpose of the present study was to examine whether CAPE modulates the immunoglobulin E (IgE)-mediated local allergic reaction in animals, as well as to elucidate the effects of CAPE on mast cells <I>in vitro</I>.</P><P><I>Materials and methods</I>: To investigate the bioactive potential of CAPE (10 or 20 µM), HMC-1 cells were stimulated with phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI) for 24 h in the presence or absence of CAPE. To study the pharmacological effects of CAPE, enzyme-linked immunosorbent assays (ELISAs), RT-PCR, Western blot analysis, electrophoretic mobility shift assays (EMSAs), and fluorescence assays were used.</P><P><I>Results</I>: CAPE (10 mg/kg) inhibited local IgE-mediated allergic reactions (0.164 versus 0.065 O.D.) in a mouse model. Additionally, CAPE (20 µM) attenuated PMACI-stimulated histamine release (3146.42 versus 2564.83 pg/ml) and the production of inflammatory cytokines, such as interleukin (IL)-1β (4.775 versus 0.713 pg/ml, IC<SUB>50</SUB> = 6.67 µM), IL-6 (4771.5 versus 449.1 pg/ml, IC<SUB>50</SUB> = 5.25 µM), and IL-8 (5991.7 versus 2213.1 pg/ml, IC<SUB>50</SUB> = 9.95 µM) in HMC-1 cells. In activated HMC-1 cells, pretreatment with CAPE decreased the phosphorylation of c-Jun N-terminal kinase. In addition, CAPE inhibited PMACI-induced nuclear factor (NF)-κB activation by suppressing IκBα phosphorylation and its degradation.</P><P><I>Discussion and conclusion</I>: Our results indicated that CAPE can modulate mast cell-mediated allergic disease.</P>

<P>Summary</P><P>Objectives </P><P>We investigated the association between smoking and its additive effects with insulin resistance and &bgr;-cell function on the incidence of type 2 diabetes in a prospective population-based cohort study.</P><P>Design and method </P><P>A total of 10 038 subjects were recruited from rural and urban areas. All subjects underwent 75 g oral glucose tolerance tests and full biochemical assessments at baseline and during 4-year follow-up period. The final analysis was limited to 4041 men due to the low smoking rates in women.</P><P>Results </P><P>The ex- and heavy current smokers had the highest incidence of diabetes of 12·5% and 11·1% respectively, compared with never-smokers (7·9%) during 4 years. After multivariate adjustment by Cox-proportional hazard model, ex- and current smokers reveal a relative risk of 1·60 (95% CI: 1·07–2·39), 2·06 (1·35–3·16, for <20 cigarettes/day) and 2·41 (1·48–3·93, for ≥20 cigarettes/day) respectively compared with never smokers. The risk of new onset diabetes was the highest in those with low homeostasis model assessment for beta cell function (HOMA-&bgr;) and high homeostasis model assessment for insulin resistance (HOMA-IR) group in both smokers and never smokers.</P><P>Conclusions </P><P>Smoking is an independent risk factor for type 2 diabetes mellitus and showed synergistic interaction with the status of low insulin secretion and high insulin resistance for developing diabetes. Given the high rates of smoking and growing burden of diabetes in the world, cessation of smoking should be considered as one of the key factors for diabetes prevention and treatment programmes.</P>

Galectins are generally believed to be potential candidates for use in the development of novel antiinflammatory agents or as selective modulators of the immune response. In particular, galectin-9 exhibits some of the extracellular functions, including cell aggregation, adhesion, chemoattraction, activation, and apoptosis. Tl-galectin (Tl-gal, galectin-9 homologue gene) was isolated from an adult worm of the Toxascaris leonina. The full-length Tl-gal gene, which was incorporated into pET-28a, was overexpressed in E. coli and purified by nickel affinity and gel filtration chromatographies. The purified Tl-gal was crystallized using the hangingdrop vapor-diffusion method. The crystal belonged to the tetragonal space group $P4_1$, with unit-cell parameters of a = b = $75.7\AA$ and c = $248.4\AA$. The crystals were obtained at $20^{\circ}C$ and diffracted to a resolution of $3.0\AA$. The asymmetric unit contained four molecules of Tl-gal, which gave a crystal volume per protein mass (Vm) of $2.8\AA^3Da^{-1}$ and a solvent content of 54.1%.
