Allicin Treatment Enhances the Developmental Potential of Porcine Oocytes during IVM

Allicin (AL) regulates the cellular redox, proliferation, viability, and cell cycle of different cells against extracellular-derived stress. This study was to investigate the effect of allicin treatment during in vitro maturation (IVM) on porcine oocyte maturation and developmental competence. Porcine follicular oocytes were cultured in 0 (control), 0.01, 0.1, 1, 10, and 100 μM AL added IVM media. The rate of polar body emission was higher in the 0.1 μM AL-treated group (74.5% ± 2.3%) than in the control (68.0% ± 2.6%). After parthenogenetic activation, the rates of cleavage and blastocyst formation were significantly higher in the 0.1 μM AL-treated group than in the control (p < 0.05). The reactive oxygen species level at metaphase II was not significantly different among all groups. In matured oocytes, the relative mRNA expression of both BAK and CASP3, and BIRC5 were significantly lower and higher, respectively, in the 0.1 AL-treated group than in the control. Also, the mRNA expression of BMP15 and cyclin B, and the activity of phospho-p44/42 MAPK, was significantly increased. These results indicate that supplementation of oocyte maturation medium with allicin during IVM improves the maturation of oocytes and the subsequent developmental competence of porcine oocytes.

Vaccine-induced immune thrombotic thrombocytopenia (VITT) is a rare but life-threatening complication. VITT strongly mimics heparin-induced thrombocytopenia (HIT) and shares clinical features. Heparin is commonly used to prevent coagulation during hemodialysis. Therefore, nephrologists might encounter patients needing dialysis with a history of heparin exposure who developed thrombotic thrombocytopenia after vaccination. A 70-year-old male presented with acute kidney injury and altered mental status due to lithium intoxication. He needed consecutive hemodialysis using heparin. Deep vein thrombosis of left lower extremity and accompanying severe thrombocytopenia of 15,000/µL on 24 days after vaccination and at the same time, nine days after heparin use. Anti-platelet factor 4 antibody test was positive. Anticoagulation with apixaban and intravenous immunoglobulin (IVIG) infusion resolved swelling of his left calf and thrombocytopenia. There were no definitive diagnostic tools capable of differentiating between VITT and HIT in this patient. Although VITT and HIT share treatment with IVIG and non-heparin anticoagulation, distinguishing between VITT and HIT will make it possible to establish a follow-up vaccination plan in a person who has had a thrombocytopenic thrombotic event. Further research is needed to develop the tools to make a clear distinction between the clinical syndromes.

<P><B>Background</B></P><P>This study was designed to compare the quality assurance (QA) results of four dosimetric tools used for intensity modulated radiation therapy (IMRT) and to suggest universal criteria for the passing rate in QA, irrespective of the dosimetric tool used.</P><P><B>Materials and methods.</B></P><P>Thirty fields of IMRT plans from five patients were selected, followed by irradiation onto radiochromic film, a diode array (Mapcheck), an ion chamber array (MatriXX) and an electronic portal imaging device (EPID) for patient-specific QA. The measured doses from the four dosimetric tools were compared with the dose calculated by the treatment planning system. The passing rates of the four dosimetric tools were calculated using the gamma index method, using as criteria a dose difference of 3% and a distance-to-agreement of 3 mm.</P><P><B>Results</B></P><P>The QA results based on Mapcheck, MatriXX and EPID showed good agreement, with average passing rates of 99.61%, 99.04% and 99.29%, respectively. However, the average passing rate based on film measurement was significantly lower, 95.88%. The average uncertainty (1 standard deviation) of passing rates for 6 intensity modulated fields was around 0.31 for film measurement, larger than those of the other three dosimetric tools.</P><P><B>Conclusions</B></P><P>QA results and consistencies depend on the choice of dosimetric tool. Universal passing rates should depend on the normalization or inter-comparisons of dosimetric tools if more than one dosimetric tool is used for patient specific QA.</P>

<P>The cell-surface markers used routinely to define the undifferentiated state and pluripotency of human embryonic stem cells (hESCs) are those used in mouse embryonic stem cells (mESCs) because of a lack of markers directly originated from hESC itself. To identify more hESC-specific cell-surface markers, we generated a panel of monoclonal antibodies (MAbs) by immunizing the irradiated cell clumps of hESC line Miz-hES1, and selected 26 MAbs that were able to bind to Miz-hES1 cells but not to mESCs, mouse embryonic fibroblast cells, and STO cells. Most antibodies did not bind to human neural progenitor cells derived from the Miz-hES1 cells, either. Of these, MAb 20-202S (IgG1, kappa) immunoprecipitated a cell-surface protein of 72-kDa from the lysate of biotin-labeled Miz-hES1 cells, which was identified to be heat shock 70-kDa protein 8 isoform 1 (HSPA8) by quadrupole time-of-flight tandem mass spectrometry. Immunocytochemical analyses proved that the HSPA8 protein was also present on the surface of hESC lines Miz-hES4, Miz-hES6, and HSF6. Two-color flow cytometric analysis of Miz-hES1 and HSF6 showed the coexpression of the HSPA8 protein with other hESC markers such as stage-specific embryonic antigen 3 (SSEA3), SSEA4, TRA-1-60, and TRA-1-81. Flow cytometric and Western blot analyses using various cells showed that MAb 20-202S specifically bound to the HSPA8 protein on the surface of Miz-hES1, contrary to other anti-HSP70 antibodies examined. Furthermore, the surface expression of the HSPA8 protein on Miz-hES1 was markedly downregulated upon differentiation. These data indicate that a novel MAb 20-202S recognizes the HSPA8 protein on the surface of hESCs and suggest that the HSPA8 protein is a putative cell-surface marker for undifferentiated hESCs.</P>
A fast high-resolution vibration measurement method based on vision technology for structures

Various types of sensors are used at industrial sites to measure vibration. With the increase in the diversity of vibration measurement methods, vibration monitoring methods using camera equipment have recently been introduced. However, owing to the physical limitations of the hardware, the measurement resolution is lower than that of conventional sensors, and real-time processing is difficult because of extensive image processing. As a result, most such methods in practice only monitor status trends. To address these disadvantages, a high-resolution vibration measurement method using image analysis of the edge region of the structure has been reported. While this method exhibits higher resolution than the existing vibration measurement technique using a camera, it requires significant amount of computation. In this study, a method is proposed for rapidly processing considerable amount of image data acquired from vision equipment, and measuring the vibration of structures with high resolution. The method is then verified through experiments. It was shown that the proposed method can fast measure vibrations of structures remotely.

Idiopathic Hypereosinophilic Syndrome (IHES) is defined by significant prolonged eosinophilia (>1,500 cells/ul) without a known underlying cause which leads to end-organ damage or dysfunction. Primary organ involvement includes heart, lung, gastrointestinal tract and bone marrow. The initiation of treatment is recommended regardless of the duration of eosinophilia. In this study, we experienced 61-yearold male who complained of right upper quadrant area abdominal pain. There was no evidence of allergic disease or parasitic infestation. Blood tests showed profound peripheral eosinophilia, leukocytosis and elevation of serum IgG4. The abdominopelvic computed tomography showed multiple small low attenuating lesions scattered in the liver. Additional examination by esophagoduodenoscopy and colonoscopy revealed severe eosinophilic infi ltration on esophagus, stomach and colonic mucosa. Eosinophilic infl ammation and fi brosis were subsequently confi rmed by liver and bone marrow biopsy, leading to a diagnosis of IHES with IgG4-related disease overlapped. Treatment of high-dose corticosteroids resulted in a dramatic clinical response. To the best of our knowledge, this is the fi rst case in which IHES involving multiple organs with suspicion of muscle infi ltration and IgG4-related disease being overlapped. As seen in our case, high-dose of glucocorticoid treatment had effectively suppressed hypereosinophilia and corrected the coagulation abnormality. Our case showed a high value of IgG4 which is likely to be an example of IgG4-related disease. IgG4-related disease is characterized by elevated serum IgG4 or tissue infi ltration by IgG4 plasma cells. IgG4-related disease often associated with eosinophilia and responds effectively to glucocorticoid. Our study findings imply that the interaction between HES and IgG4-related is a diffi cult task. Therefore, careful attention should be made to reveal its obscure clinical manifestations and physician should aware to conduct through examination and perform adequate biopsy throughout gastrointestinal tract to rule out the IHES and IgG4-related disease.
