Background Development of genetically modified crops has rapidly increased in last few years. The most widely grown GM crops express genes that confer herbicide tolerance and insect resistance. Detection system of GM crops is important for safety evaluation before its consumption. Objective The purpose of this research is to detect GM crops, especially PAT, in food-samples. Results The bar gene (PAT protein, herbicide resistant) was cloned in pGEX-4T-1 and expressed by E. coli. The high-affinity PAT-specific single-stranded DNA (ssDNA) aptamers were obtained from a random DNA library. MOE docking study was performed to identify the potential binding region of the selected aptamers on PAT. Aptamer-linked immobilized sorbent assay (ALISA) method was used to detect PAT. Conclusion We screened aptamer against PAT for developing an efficient detection method. The selected PAT specific aptamers, HRPA-05 and HRPA-07, showed the distinct target binding behaviors, and detected PAT protein by aptamer-linked immobilized sorbent assay method with high efficiency and selectivity.

A 3-month-old, 3.85kg female Jin-do dog with chief complaint of bloody diarrhea and depression was referred to the Veterinary Teaching Hospital, Chungnam National University. In fecal examination, trophozoites of giardia were observed and It was diagnosed as canine giardiasis. For treatment, metronidazole was administered orally for 5 days at a dose of 22mg/kg q12h. Diarrhea was not observed on 3 days after medication and most of clinical signs were disappeared on 7 days after medication with metronidazole. General conditions and activitivities became good on 13 days after medication. In addition, cysts or trophozoites of giardia were not detected any more in fecal examination using floating method with zinc sulphate solution. The present patient was a case of canine giardiasis that showed favorable response to metronidazole.

A 4 years old, mixed male dog with a history of traffic accident was presented to the Veterinary Medical Teaching Hospital of Chungnam National University with tachypnea and respiratory distress. Physical examination showed that the dog was auscultated crackles sounds, dysynchronous abdominal breathing pattern, hemoptysis, hematuria, urodynia and right and left scleral congestion. Blood and blood chemical examination revealed that WBC, AST and ALT was highly elevated. Parenchymal pulmonary fluid and closed pneumothorax were observed by thoracic radiography. This case was diagnosed as pulmonary contusion. Intravenous fluid and oxygen was administered and also dexamethasone(1mg/kg, Ⅳ), furosemide(2mg/kg, Ⅳ), (ampicillin, 20mg/kg, Ⅳ) and meloxicam(0.2mg/kg, PO) were intravenously injected for 5 days. Abnormal clinical signs were disappeared after treatment.

3D structural analysis of aptamer and diagnostic platforms for detecting hepatocellular carcinoma

Background Alpha-fetoprotein (AFP) has been used as a biomarker for early diagnosis of hepatocellular carcinoma (HCC) for a long time. Aptamer is a type of material that is receiving attention as a new alternative in the diagnosis and treatment of diseases. Objective We applied aptamer to AFP detection and utilized in silico analysis techniques. Here, we describe the selection of aptamers that can bind to AFP as well as in vitro and in silico validation of the binding site. We also developed an aptamerbased detection platform called hybrid-aptablotting sandwich assay (Hybrid-ABSA) and a point-of-care AFP detection kit. Results To construct the AFP detection platform, we screened a total of 18 candidate AFP-binding aptamers and identifi ed a pair of aptamers without non-overlapping-binding sites using dot blotting. We validated results of dot blotting using in silico 3D binding structure docking simulation analysis. The aptamer-based Hybris-ABSA for AFP detection using this pair of aptamers demonstrated superior detection performance to antibody-based ELISA detection methods. Furthermore, we confi rmed that AFP could be detected using the developed point-of-care lateral fl ow In Vitro Diagnostic (IVD) kit. Conclusion Results of this study demonstrate that aptamer-based disease diagnostic technology has potential of replace conventional diagnostic methods. This study also confi rms that in silico techniques have suffi cient utility in biological assays.

<P>Aptamer-based paper strip sensor for detecting <I>Vibrio fischeri</I> was developed. Our method was based on the aptamer sandwich assay between whole live cells, <I>V. fischeri</I> and DNA aptamer probes. Following 9 rounds of Cell-SELEX and one of the negative-SELEX, <I>V. fischeri</I> Cell Aptamer (VFCA)-02 and -03 were isolated, with the former showing approximately 10-fold greater avidity (in the subnanomolar range) for the target cells when arrayed on a surface. The colorimetric response of a paper sensor based on VFCA-02 was linear in the range of 4 × 10<SUP>1</SUP> to 4 × 10<SUP>5</SUP> CFU/mL of target cell by using scanning reader. The linear regression correlation coefficient (<I>R</I><SUP>2</SUP>) was 0.9809. This system shows promise for use in aptamer-conjugated gold nanoparticle probes in paper strip format for in-field detection of marine bioindicating bacteria.</P> [FIG OMISSION]</BR>
