      • Significance of Thrombocytosis in Clinicopathologic Characteristics and Prognosis of Gastric Cancer

        Purpose: We aimed to study the relationship between thrombocytosis and clinical features of gastric cancerfocussing on platelet counts and gastric cancer progression through different TNM stages. Methods: According to the normal range of platelet count in our institution, 1,596 patients were divided to two groups: a thrombocytosis group (120 patients, > $400{\times}1000/{\mu}L$) and a control group (1,476 patients, ${\leq}400{\times}1000/{\mu}L$). Results: The incidence of thrombocytosis was 7.5%. Higher platelet counts were observed in patients with older age, larger tumor size, deeper invasion, lymph node metastasis, distant metastasis and advanced TNM stage. In multivariate logistic regression, tumor size, depth of tumor invasion, lymph node metastasis and TNM stage were independent risk factors for thrombocytosis of gastric cancer patients. On prognostic analysis, age, tumor size, tumor location, histologic type, depth of tumor invasion, lymph node metastasis, distant metastasis and TNM stage and platelet count were important factors. Tumor size, invasion depth, lymph node metastasis, TNM stage and the platelet count were independent prognostic factors. Conclusion: Thrombocytosis is associated with clinical features of gastric cancer patients and correlates with a poor prognosis.

        Fibulin2: a negative regulator of BMSC osteogenic differentiation in infected bone fracture healing

        Bone fracture remains a common occurrence, with a population-weighted incidence of approximately 3.21 per 1000. In addition, approximately 2% to 50% of patients with skeletal fractures will develop an infection, one of the causes of disordered bone healing. Dysfunction of bone marrow mesenchymal stem cells (BMSCs) plays a key role in disordered bone repair. However, the specific mechanisms underlying BMSC dysfunction caused by bone infection are largely unknown. In this study, we discovered that Fibulin2 expression was upregulated in infected bone tissues and that BMSCs were the source of infection-induced Fibulin2. Importantly, Fibulin2 knockout accelerated mineralized bone formation during skeletal development and inhibited inflammatory bone resorption. We demonstrated that Fibulin2 suppressed BMSC osteogenic differentiation by binding to Notch2 and inactivating the Notch2 signaling pathway. Moreover, Fibulin2 knockdown restored Notch2 pathway activation and promoted BMSC osteogenesis; these outcomes were abolished by DAPT, a Notch inhibitor. Furthermore, transplanted Fibulin2 knockdown BMSCs displayed better bone repair potential in vivo. Altogether, Fibulin2 is a negative regulator of BMSC osteogenic differentiation that inhibits osteogenesis by inactivating the Notch2 signaling pathway in infected bone.

        Theoretical and Exprerimental Sutdy on Interactive Local Buckling of Arch-shaped Corrugated Steel Roof

        The arch-shaped corugated steel roof is a new type of cold formed thin-waled structure. Since the width-to-thickness ratiosof the plate asemblies of corrugated sections exceed the limits specified in the current design codes, the load-bearingbehavior, especialy loading modes, initial imperfections and boundary conditions, therefore its ultimate load-carying capacityis determined not only by its global stability, but also by its interactive local buckling. In this paper the finite element methodwas aplied to analyze the local buckling behavior of stel arch panels with corrugated trapezium sections. Some tests on theloads to further investigate the loading mechanism of corrugated steel arch panels and the factors refered above. A comparisonbetween the experimental and theoretical results was made, and good agrement was achieved herein. It is shown that therealso exists interactive local buckling among corugated plate assemblies. The experimental results are lower than theoreticalones for axial loading capacity, vice-versa, the former ones are higher for eccentricaly loaded specimens. It is noted that thealso indicate that the trapezium sections have high local buckling capacities, yet they hardly have post-buckling capacities,which are distinctive from that of the plain ones. All these also give some explanations to some corugated steel arch rofscolapsed suddenly in some areas of China.

        Advances in CRISPR-Cas systems for RNA targeting, tracking and editing

        <P><B>Abstract</B></P> <P>Clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated protein (Cas) systems, especially type II (Cas9) systems, have been widely used in gene/genome targeting. Modifications of Cas9 enable these systems to become platforms for precise DNA manipulations. However, the utilization of CRISPR-Cas systems in RNA targeting remains preliminary. The discovery of type VI CRISPR-Cas systems (Cas13) shed light on RNA-guided RNA targeting. Cas13d, the smallest Cas13 protein, with a length of only ~930 amino acids, is a promising platform for RNA targeting compatible with viral delivery systems. Much effort has also been made to develop Cas9, Cas13a and Cas13b applications for RNA-guided RNA targeting. The discovery of new RNA-targeting CRISPR-Cas systems as well as the development of RNA-targeting platforms with Cas9 and Cas13 will promote RNA-targeting technology substantially. Here, we review new advances in RNA-targeting CRISPR-Cas systems as well as advances in applications of these systems in RNA targeting, tracking and editing. We also compare these Cas protein-based technologies with traditional technologies for RNA targeting, tracking and editing.         Polymorphisms in Epigenetic and Meat Quality Related Genes in Fourteen Cattle Breeds and Association with Beef Quality and Carcass Traits

        Improvement for carcass traits related to beef quality is the key concern in beef production. Recent reports found that epigenetics mediates the interaction of individuals with environment and nutrition. The present study was designed to analyze the genetic effect of single nucleotide polymorphisms (SNPs) in seven epigenetic-related genes (DNMT1, DNMT3a, DNMT3b, DNMT3L, Ago1, Ago2, and HDAC5) and two meat quality candidate genes (CAPN1 and PRKAG3) on fourteen carcass traits related to beef quality in a Snow Dragon beef population, and also to identify SNPs in a total of fourteen cattle populations. Sixteen SNPs were identified and genotyped in 383 individuals sampled from the 14 cattle breeds, which included 147 samples from the Snow Dragon beef population. Data analysis showed significant association of 8 SNPs within 4 genes related to carcass and/or meat quality traits in the beef populations. SNP1 (13154420A>G) in exon 17 of DNMT1 was significantly associated with rib-eye width and lean meat color score (p<0.05). A novel SNP (SNP4, 76198537A>G) of DNMT3a was significantly associated with six beef quality traits. Those individuals with the wild-type genotype AA of DNMT3a showed an increase in carcass weight, chilled carcass weight, flank thicknesses, chuck short rib thickness, chuck short rib score and in chuck flap weight in contrast to the GG genotype. Five out of six SNPs in DNMT3b gene were significantly associated with three beef quality traits. SNP15 (45219258C>T) in CAPN1 was significantly associated with chuck short rib thickness and lean meat color score (p<0.05). The significant effect of SNP15 on lean meat color score individually and in combination with each of other 14 SNPs qualify this SNP to be used as potential marker for improving the trait. In addition, the frequencies of most wild-type alleles were higher than those of the mutant alleles in the native and foreign cattle breeds. Seven SNPs were identified in the epigenetic-related genes. The SNP15 in CAPN1 could be used as a powerful genetic marker in selection programs for beef quality improvement in the Snow Dragon Beef population.

        Ni/ZnO-based Adsorbents Supported on Al2O3, SiO2, TiO2, ZrO2: A Comparison for Desulfurization of Model Gasoline by Reactive Adsorption

        Reactive adsorption desulfurization (RADS) experiments were conducted over a series of commercial metal oxide supports (Al2O3-, SiO2-, TiO2- and ZrO2-) supported Ni/ZnO adsorbents. The adsorbents were characterized by X-ray diffraction (XRD), temperature programmed reduction (TPR), and Fourier transform infrared spectroscopy (FTIR) in order to find out the influence of specific types of surface chemistry and structural characteristics on the sulfur adsorptive capacity. The desulfurization performance of all the studied adsorbents decreased in the following order: Ni/ZnO-TiO2 > Ni/ZnO-ZrO2 > Ni/ZnO-SiO2 > Ni/ZnO-Al2O3. Ni/ZnO-TiO2 shows the best performance and the three hour sulfur capacity can achieve 12.34 mg S/g adsorbent with a WHSV of 4 h−1. Various characterization techniques suggest that weak interaction between active component and support component, high dispersion of NiO and ZnO, high reducibility and large total Lewis acidity of the adsorbents are important factors in achieving better RADS performance.

        The role of layered double hydroxides shell in enhancing the adsorption capacity of NaY zeolite for toluene in wet conditions

        Surface modification of zeolite is known to enhance its adsorption capacity for volatile organic compounds (VOCs). In this study, a hydrophobic adsorbent, NaY@LDHs, with layered double hydroxides (LDHs) as the shell and NaY zeolite as the core, was successfully synthesized. It was found that treating LDHs with ethanol can result in a larger specific surface area and pore volume, enhancing the adsorption performance of toluene under wet conditions. SEM imaging reveals that the LDHs shell grows vertically on the surface of the NaY zeolite. The dynamic adsorption experiment of toluene showed that under humid conditions, NaY@LDHs exhibited a saturated adsorption capacity of 32.42 mg/g, representing a significant 87% increase compared to the parent NaY zeolite. The kinetic results show that the adsorption of toluene on NaY@LDHs is primarily physical adsorption, and other factors influence the adsorption process in addition to intraparticle diffusion. Therefore, the air layer formed by the LDHs shell on the surface of NaY zeolite imparts a hydrophobic effect, making NaY@LDHs a promising material for adsorbing volatile organic compounds in a wet environment.
