      • Influence of the MACC1 Gene on Sensitivity to Chemotherapy in Human U251 Glioblastoma Cells

Shang, Chao,Hong, Yang,Guo, Yan,Liu, Yun-Hui,Xue, Yi-Xue Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.1

Background: This study was conducted to determine the influence of MACC1 expression on chemotherapy sensitivity in human U251 glioblastoma cells. Materials and Methods: Expression of the MACC1 gene in 49 cases of human brain glioma was determined by quantitative real-time PCR. Silencing effects of RNA interference on MACC1 was detected by Western-blotting. Flow cytometry methods and methyl thiazolyl tetrazolium assay (MTT) were used to determine the apoptosis and growth inhibitory rates of the U251 cells with MACC1 silencing. before and after treatment with cisplatin (DDP). Results: MACC1 mRNA in gliomas was up-regulated remarkably, to 158.8% of that in peri-cancerous tissues (P<0.05). The siRNA-MACC1 could inhibit the expression of MACC1 protein significantly (p<0.05), associated with an increase in apoptosis rate from 2.57% to 5.39% in U251 cells and elevation of the growth inhibitory rate from 1.5% to 17.8% (p<0.05 for both). After treatment with DDP at various concentrations (1, 3, $5{\mu}g/ml$), compared with control U251 cells, the apoptosis rate of MACC1-silenced U251 cells rose from 8.41%, 13.2% and 19.5% to 12.8%, 17.8% and 25.8%; the growth inhibitory rate increased from 16.2%, 19.3% and 24.5% to 23.7%, 28.4% and 36.3%. Conclusions: There is a notable relationship between over-expression of MACC1 and the characteristics of glioma cells. Silencing of MACC1 was found to enhance the apoptosis and growth inhibitory rates of U251 glioma cells, and thereby increase their sensitivity to DDP chemotherapy.

Poly(4-vinylphenol-co-methyl methacrylate)/Titanium Dioxide Nanocomposite Gate Insulators for 6,13-Bis(triisopropylsilylethynyl)-Pentacene Thin-film Transistors

        Xue Zhang,Ji-Ho Park,Sungkeun Baang,박재훈,Shang Hao Piao,김소희,최형진 한국물리학회 2014 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.65 No.11

Poly(4-vinylphenol-co-methyl methacrylate)/titanium dioxide (TiO2) nanocomposite insulatorswere fabricated for application in 6,13-bis(triisopropylsilylethynyl) pentacene (TIPS-Pn) thin-filmtransistors (TFTs). The capacitance of the fabricated capacitors with this nanocomposite insulatorincreased with increasing content of the high-dielectric-constant TiO2 nanoparticles. Nonetheless, particle aggregates, which were invariably produced in the insulator at higher TiO2 contents, augmented gate-leakage currents during device operation while the rough surface of the insulatorobstructed charge transport in the conducting channel of the TIPS-Pn TFTs. These results suggesta significant effect of the morphological characteristics of nanocomposite insulators on TFTperformance, as well as on their dielectric properties. Herein, the optimal particle composition wasdetermined to be approximately 1.5 wt%, which contributed to characteristic improvements in thedrain current, field-effect mobility, and threshold voltage of TIPS-Pn TFTs.

      • Regulatory Network of MicroRNAs, Target Genes, Transcription Factors and Host Genes in Endometrial Cancer

Xue, Lu-Chen,Xu, Zhi-Wen,Wang, Kun-Hao,Wang, Ning,Zhang, Xiao-Xu,Wang, Shang Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.2

Genes and microRNAs (miRNAs) have important roles in human oncology. However, most of the biological factors are reported in disperse form which makes it hard to discover the pathology. In this study, genes and miRNAs involved in human endometrial cancer(EC) were collected and formed into regulatory networks following their interactive relations, including miRNAs targeting genes, transcription factors (TFs) regulating miRNAs and miRNAs included in their host genes. Networks are constructed hierarchically at three levels: differentially expressed, related and global. Among the three, the differentially expressed network is the most important and fundamental network that contains the key genes and miRNAs in EC. The target genes, TFs and miRNAs are differentially expressed in EC so that any mutation in them may impact on EC development. Some key pathways in networks were highlighted to analyze how they interactively influence other factors and carcinogenesis. Upstream and downstream pathways of the differentially expressed genes and miRNAs were compared and analyzed. The purpose of this study was to partially reveal the deep regulatory mechanisms in EC using a new method that combines comprehensive genes and miRNAs together with their relationships. It may contribute to cancer prevention and gene therapy of EC.

Highrisk human papillomavirus genotype distribution and attribution to cervical cancer and precancerous lesions in a rural Chinese population

        Xue-Lian Zhao,Shang-Ying Hu,Qian Zhang,Li Dong,Rui-Mei Feng,Ross Han,Fang-Hui Zhao 대한부인종양학회 2017 Journal of Gynecologic Oncology Vol.28 No.4

Objective: To explore the genotype distribution of high-risk human papillomavirus (HR-HPV) and its attribution to different grades of cervical lesions in rural China, which will contribute to type-specific HPV screening tests and the development of new polyvalent HPV vaccines among the Chinese population. Methods: One thousand two hundred ninety-two subjects were followed based on the Shanxi Province Cervical Cancer Screening Study I (SPOCCS-I), and screened by HPV DNA testing (hybrid capture® 2 [HC2]), liquid-based cytology (LBC), and if necessary, directed or random colposcopy-guided quadrant biopsies. HPV genotyping with linear inverse probe hybridization (SPF10-PCR-LiPA) was performed in HC2 positive specimens. Attribution of specific HR-HPV type to different grades of cervical lesions was estimated using a fractional contribution approach. Results: After excluding incomplete data, 1,274 women were included in the final statistical analysis. Fifteen point two percent (194/1,274) of women were HR-HPV positive for any of 13 HR-HPV types (HPV16, 18, 31, 33, 35, 39, 45, 51, 52, 56, 58, 59, and 68) and the most common HR-HPV types were HPV16 (19.1%) and HPV52 (16.5%). The genotypes most frequently detected in HR-HPV-positive cervical intraepithelial neoplasia grade 1 (CIN1) were HPV52 (24.1%), HPV31 (20.7%), HPV16 (13.8%), HPV33 (13.8%), HPV39 (10.3%), and HPV56 (10.3%); in HR-HPV-positive cervical intraepithelial neoplasia grade 2 or worse (CIN2+): HPV16 (53.1%), HPV58 (15.6%), HPV33 (12.5%), HPV51 (9.4%), and HPV52 (6.3%). HPV52, 31, 16, 33, 39, and 56 together contributed to 89.7% of HR-HPV-positive CIN1, and HPV16, 33, 58, 51, and 52 together contributed to 87.5% of CIN2+. Conclusion: In summary, we found substantial differences in prevalence and attribution of CINs between different oncogenic HPV types in a rural Chinese population, especially for HPV16, 31, 33, 52, and 58. These differences may be relevant for both clinical management and the design of preventive strategies.

Cloning, Expression, and Characterization of a Cold-Adapted and Surfactant-Stable Alginate Lyase from Marine Bacterium Agarivorans sp. L11

( Shang Yong Li ),( Xue Mei Yang ),( Lan Zhang ),( Wen Gong Yu ),( Feng Han ) 한국미생물 · 생명공학회 2015 Journal of microbiology and biotechnology Vol.25 No.5

The purpose of this study was to find a cold-adapted and surfactant-stable alginate lyase as a candidate for biotechnological and industrial applications. The gene for a new alginate lyase, AlyL1, from Agarivorans sp. L11 was cloned and expressed in Escherichia coli. The recombinant AlyL1 was most active at 40ºC (1,370 U/mg). It was a cold-adapted alginate lyase, which showed 54.5% and 72.1% of maximum activity at 15ºC and 20ºC, respectively. AlyL1 was an alkaliphilic enzyme and most active at pH 8.6. In addition, it showed high stability in the presence of various surfactants at a high concentration (from 0.1% to 1% (w/v)). AlyL1 was an endo-type alginate lyase that degraded both polyM and polyG blocks, yielding disaccharides and trisaccharides as the main products. This is the first report of the cloning and functional expression of a cold-adapted and surfactant-stable alginate lyase. AlyL1 might be an interesting candidate for biotechnological and industrial applications.

A new converter for improving efficiency of multi-actuators fluid power system

Yong Xue,JianZhong Shang,JunHong Yang,Zhuo Wang 대한기계학회 2016 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.30 No.5

This paper is concerned with the application of energy efficient fluid power in mobile robots system and proposes a new fluid power converter system which is analogous to a boost converter in power electronics. The fluid power converter system is based on the principle of pulse-width modulation. The fluid power converter has an effect akin to an electrical switched inductance transformer, wherein the output pressure or flow rate can be stepped up or down. Using an inductive reactance device (an inertia mass-block), the output flow and pressure can be varied to meet the load by a means that does not rely on dissipation of power (the resistance control). The simulation model based on the mathematics models of the components is built to analyse the performance of the fluid power converter. It is clearly shown that the fluid power converter has higher energy efficiency than conventional resistance control manners.

Entanglement Dynamics of Electron Spins in Quantum Dots Under a Nonuniform Magnetic Field

        Feng-Xue Zhou,Yi-Hong Qi,Yue-Ping Niu,Shang-Qing Gong,Jun Qian,Ting Yu,Franco Nori 한국물리학회 2012 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.60 No.8

We investigate entanglement of two coupled electron spins in quantum dots (QDs) in the presence of an inhomogeneous magnetic field. The great effects of the inhomogeneous field are discussed for the dynamics of entanglement. It is shown that due to the system's symmetry, the inhomogeneity of the field does not affect the evolution of entanglement for Φ-type Bell state, while it plays a key role for Ψ-type Bell state. For the maximal entangled Bell states, the field is positive for the entanglement dynamics. The mean field can increase the entanglement revival for Φ-type Bell state while the inhomogeneous field can promote the entanglement revival for Ψ-type Bell state. For the unentangled initial states, the field is destructive for the entanglement generation induced by the coupling of the two spins.

        Yang Shang-Yang,Li Shu-Cai,Xue Yi-Guo,Zhang Qing-Song 보안공학연구지원센터 2016 International Journal of Hybrid Information Techno Vol.9 No.11

Triaxial compression tests had been performed to determine the properties of the rock mass around unlining underground crude oil a storage cavern which was the first one in China. The execution situation of the tunnel project and the seepage law of groundwater were taken into account. The stress and the seepage field around the tunnel in different working states were simulated by applying Comsol around a underground crude oil storage caverns. According to the test results, it was found that the excavation process may arise the local damage. The extension of the loose zone induced by excavation ranged from 0 to 15.6 m, depending on the buried depth of the caverns. According to numerical simulation results, the crown settlement and stress concentration was depended on the buried depth and the water pressure distribution after excavation of the main cavity. This research results can provide the reference for analysis on the stability of the underground cavities under low stress level and on the water sealed underground petroleum storage rock caverns.
