      • Inhibitory effect of Au@Pt-NSs on proliferation, migration, and invasion of EJ bladder carcinoma cells: involvement of cell cycle regulators, signaling pathways, and transcription factor-mediated MMP-9 expression

        <P><B>Background</B></P><P>Although the diverse biological properties of nanoparticles have been studied intensively, research into their mechanism of action is relatively rare. In this study, we investigated the molecular mechanisms of the anticancer activity of heterometallic Au@Pt-nanoseeds (NSs) against bladder cancers.</P><P><B>Materials and methods</B></P><P>Mode of action of Au@Pt-NSs was investigated through MTT assay, flow cytometry analysis, Western immunoblots, real-time qPCR, wound-healing migration and invasion assays, zymography, and electrophoretic mobility shift assay (EMSA).</P><P><B>Results</B></P><P>Treatment with Au@Pt-NSs significantly inhibited the proliferation of EJ cells in a dose-dependent manner by inducing G1 phase cell cycle arrest. Among the regulators associated with the G1 cell cycle phase, CDK2, CDK4, cyclin D1, cyclin E, and p21WAF1 were shown to participate in the inhibitory pathways of Au@Pt-NSs. In addition, treatment with Au@Pt-NSs led to upregulation of phospho-p38 MAPK and downregulation of phospho-AKT in EJ cells. Interestingly, Au@Pt-NSs inhibited the migratory and invasive potential of the cells, which was attributed to the suppression of the enzymatic activity of matrix metalloproteinase-9 (MMP-9). Using MMP-9-specific oligonucleotides, we showed that transcription factors such as NF-κB and Sp-1 were responsible for the MMP-9-mediated metastatic potential of EJ cells.</P><P><B>Conclusion</B></P><P>Au@Pt-NSs significantly limited the progression, migration, and invasion of bladder cancer EJ cells. Our data represent a novel insight into developing cisplatin-like chemotherapeutic reagents with fewer side effects and provide useful information on molecular markers to monitor patients under Au@Pt-NSs-based chemotherapy.</P>

        <P><B>ABSTRACT</B></P><P><B>Background</B>: Diallyl trisulfide (DATS), a bioactive sulfur compound in garlic, has been highlighted due to its strong anti-carcinogenic activity.</P><P><B>Objective</B>: The current study investigated the molecular mechanism of garlic-derived DATS in cancer cells. Additionally, we explored possible molecular markers to monitoring clinical responses to DATS-based chemotherapy.</P><P><B>Design</B>: EJ bladder carcinoma cells were treated with different concentration of DATS. Molecular changes including differentially expressed genes in EJ cells were examined using immunoblot, FACS cell cycle analysis, migration and invasion assays, electrophoresis mobility shift assay <I>(EMSA)</I>, microarray, and bioinformatics analysis.</P><P><B>Results</B>: DATS inhibited EJ cell growth via G<SUB>2</SUB>/M-phase cell cycle arrest. ATM-CHK2-Cdc25c-p21WAF1-Cdc2 signaling cascade, MAPKs, and AKT were associated with the DATS-mediated growth inhibition of EJ cells. DATS-induced inhibition of migration and invasion was correlated with down-regulated MMP-9 via reduced activation of AP-1, Sp-1, and NF-κB. Through microarray gene expression analysis, ANGPTL4, PLCXD1, and MMP3 were identified as candidates of molecular targets of DATS. Introduction of each gene to EJ cells revealed that ANGPTL4 was associated with the DATS-induced inhibition of cell growth, migration, and invasion.</P><P><B>Conclusions</B>: ANGPTL4 regulates DATS-mediated inhibition of proliferation, migration, and invasion of EJ cells, and thus, has potential as a prognostic marker for bladder cancer patients.</P>

        해조류 첨가 오메기술의 품질 특성

        본 연구에서는 지역 특색을 살린 전통주를 개발하여 사장되고 있는 제주 전통주 문화를 정립하고자 제주지역의 대표적 민속주인 오메기술의 제조과정에 다양한 생리활성물질이 함유된 톳, 모자반과 같은 해조류를 물 첨가량 대비 5, 10%로 첨가한 후, 해조류의 함량 차이에 따른 오메기술의 발효중 pH, 산도, 알코올 함량, 색도, 미생물 수, 유기산 함량 및 항산화력을 측정하였다. pH는 담금 직후 4.80~5.35에서 발효 2일째 급격하게 감소하였고 발효기간에 따라 점차 낮아져 발효 10일에는 모자반을 10% 첨가한 오메기술이 3.72로 유의적으로 가장 낮았다. 총산은 담금 직후에 비해 발효 2일째 0.158~0.190%로 급격하게 증가하였고 발효기간에 따라 점차적으로 증가하는 경향을 보였다. 알코올 함량은 발효 2일째에 급격하게 상승하였으며 발효 4일째부터 완만하게 증가하여 발효 10일 8.37~9.00%로 나타났다. 색도는 발효 10일 L값과 a값, b값 모두 톳을 10% 첨가한 오메기술에서 높게 나타났다. 총균수와 젖산균 수는 발효 초기 증가하다가 발효 후기는 감소하는 경향을 나타내었는데 발효 10일째에 톳을 10% 첨가한 오메기술에서 총균수는 1.45×10⁸ CFU/mL, 젖산균 수는 2.21×10⁸ CFU/mL로 나타났다. 유기산에서는 lactic acid와 succinic acid가 많이 검출되었으며 lactic acid는 발효 10일째의 모자반 10%를 첨가한 오메기술에서 7.474 ㎎/mL로 가장 높게 나타났고 succinic acid는 발효 10일 0.598~0.924 ㎎/mL로 유의적인 차이가 없었다. 항산화 활성을 확인하기 위해 DPPH 및 ABTS radical 소거 활성을 측정한 결과 DPPH radical 소거 활성에서는 발효가 진행됨에 따라 점차 증가하여 발효 10일째에는 모자반을 10% 첨가한 오메기술이 69.81%로 가장 높게 나타났고, ABTS radical 소거 활성에서는 발효가 진행됨에 따라 증가하여 발효 8일째에 78.25~81.61%로 모자반을 10% 첨가한 오메기술에서 유의적으로 가장 높게 나타났으나 발효 10일째에는 모든 실험군에서 감소하였다. 본 실험결과에서는 전반적인 품질 비교를 통해 모자반을 10% 첨가한 오메기술이 다른 비교구에 비해 탁월한 결과를 보여 기능성 오메기술로서의 가능성을 확인할 수 있었으며, 이후 지역 특성을 살린 전통주로 기능성 오메기술을 상품화하기 위한 기초자료로서 유용하게 활용될 것으로 판단된다. 현재 오메기술의 소비자 기호도 조사를 수행하고 있으며 그 결과 또한 보고할 계획이다. In this study, we developed traditional liquor, Omegisul, with seaweeds to support the regional culture of Jeju Island in manufacturing traditional liquors. Seaweed extracts from Sargassum fulvellum and Hizikia fusiformis were added for manufacturing Omegisul at a final concentration of either 5 or 10% and fermented for 10 days. During fermentation, samples were collected to measure changes in quality of Omegisul including pH, acidity, alcohol content, chromaticity, total microbes, organic acid content, and antioxidant activity. Both pH and acidity of Omegisul were significantly altered in the early stages of fermentation. Alcohol contents increased over the fermentation period up to 9%. Regarding chromaticity of the product, L, a, and b values were highest in Omegisul with 10% Hizikia fusiformis. Both total plate count and lactic acid bacterial count tended to increase in the early stage of fermentation and decrease later, reaching their highest points in Omegisul with 10% Hizikia fusiformis at 10 days: 1.45×10⁸ CFU/mL and 2.21×10⁸ CFU/mL, respectively (P<0.05). Among organic acids, lactic acid contents were significantly higher during fermentation with the highest concentration of 7.474 ㎎/mL in Omegisul containing 10% Sargassum fulvellum. 1,1-Diphenyl-2-picrylhydrazyl (DPPH) and 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging assays showed that antioxidant activity of Omegisul was increased during fermentation period. Among the samples, Omegisul with 10% Sargassum fulvellum showed the highest antioxidant activities of 69.81% and 81.61% in DPPH and ABTS assays, respectively. Taken together, Omegisul with 10% Sargassum fulvellum showed better characteristics in terms of quality than the other groups and has potential as functional Omegisul.

        한국산 고구마 전분의 이화학적 특성에 관한 연구

        Starch granules of sweet potatoes, the Suwon 147 and the Chunmi were observed by photomicroscope and scanning electron microscope were round and polygonal. Granule sizes of the Suwon 147 and the Chunmi were 14∼30㎛, 16∼34㎛, respectively. X-ray diffraction pattern of starch granules resulted weak crystallinity at 2θ 14.8, 17.2, 22.5. Starch granules were of the Ca crystalline type. The blue values of the Suwon 147 and the Chasnmi were determined to be 0.342 and 0.279, amylose contents 27.6% and 23.6%, and water binding capacity 178.7% and 185.5%. Swelling of the starches negligible until 50℃ thereafter it increased rapidly. Swelling power of the Suwon 147 was more slightly than that of the Chunmi. Optical transmittance of 0.2% starch suspensions was increased rapidly from 65% and the gelatinization at 65∼80℃ was of single stage. Amylogram patterns of 4% starch solutions were similar to no peak viscosity. Viscosity of the Suwon 147 was increased, but that of the Chunmi was decreased at 92.5℃.

        가열방법에 따른 고구마의 호화도 측정

        Changes in moisture content and degrees of gelatinization during cooking of sweet potatoes by different cooking methods were studied. The samples were cooked by boiling, steaming, gas oven or microwave oven baking. Changes in the moisture content of sweet potato during cooking were +30.0% by boiling, +4.4% by steaming, -5.8% by gas oven baking, and -46.6% by microwave oven baking. The degrees of gelatinization of sweet potatoes were 83.8%, 75.7%, 73.0% and 67.7%, respectively.

        EFDC모형을 이용한 가로림만의 조력발전 위치 타당성 검토

        해양에너지는 무궁무진한 부존량을 지니고 있으며 2차적인 환경문제를 야기 시키는 위험성이 상대적으로 작기 때문에 더욱 기대가 모아지고 있다. 우리나라의 서해안은 조수간만의 차가 크기 때문에 해양에너지 중에서도 조력발전을 통한 대규모 신재생에너지 창출이 유리한 지역이다. 아울러 조력발전을 위한 발전소를 해상에 설치 할 경우, 댐 건설에 따른 교량효과, 관광자원효과 및 만내 하천범람을 조절효과 등 다목적의 개발 기대효과를 얻을 수 있다. 조력발전을 위해서는 외해 조석과 발전소 운전에 따른 조지수위 변화에 따라 결정되는 가용수두가 크고, 비교적 광역의 조지면적이 필요하다. 본 연구에서는 가용수두가 클 뿐만 아니라, 만입구가 좁고, 만내에 넓은 조지면적을 지닌 서해안에 위치한 가로림만에 조력발전소를 건설할 경우, 조력발전에 따른 효과를 예측하는데 있어서 필수적인 수리특성의 변화를 EFDC모델을 이용한 수치모형실험을 통해 검토하였다. 검토 결과, 조력발전 입지로서의 가로림만의 입지타당성을 확인하였으며, 대상지역은 창조시의 유속보다는 낙조시의 유속이 빠르기 때문에 낙조시 유속이 빠른 서측수로에 수차발전기를 설치하고 동측수로에 수문을 설치해야 하는 등의 설계 지견을 얻을 수 있었다. Fossil fuel energy has become a worldwide environmental issue due to its effect on global warming and depletion in its supply. Therefore, the interest in developing alternative energy source has been rising. Ocean energy, especially, has gained strength as an alternative energy source for its unlimited supply with low secondary risks. Among all the ocean energy, the west coast of Korea holds the field of large-scale energy development because of its distinctive tidal range. Tidal power plant construction at the sea may expedite multi development effects such as bridge roles, tourism resource effects and adjustability of flood inundation at the inner bay. This study introduces the validity of tidal power plant construction at Garilim Bay in west coast of Korea by examining anticipated hydraulic characteristics using EFDC model. Through EFDC numerical simulations, the feasibility of Garolim Bay as a tidal power plant field has been proved. And the most effective tidal power plant construction would be to install hydraulic turbine in the west side of bay entrance where ebb current is stronger, and install water gate in the east side of bay entrance where the flood current is superior.
