Effect of Highly Pressurized Hydrogen Gas Charging on the Hydrogen Embrittlement of API X70 Steel

During the use of API X70 steel as a pipeline structural material for the transportation of natural gas,hydrogen embrittlement can occur due to the hydrogen contained in natural gas. The aim of this study is toinvestigate the effects of the hydrogen content under high-pressure hydrogen gas conditions on the hydrogenembrittlement of air-cooled API X70 steel. The air-cooled API X70 steel was manufactured by hot rollingand was then air-cooled to room temperature. Tensile test specimens were held for 0 h, 1000 h, and 2000 hwithin a pressure vessel filled with 100% hydrogen gas at a gas pressure of 10 MPa, with the tensile teststhen performed at room temperature. The microstructure of the API X70 steel consists of coarse polygonalferrite, coarse pearlite, and fine acicular ferrite. The yield and tensile strength increased and elongationdecreased considerably after a holding time of 2000 h compared to those of 0 h and 1000 h within the pressurevessel. The morphology of the fracture surface changed from ductile to brittle upon hydrogen gas charging. Secondary cracks were observed in both of the hydrogen-gas-charged specimens. No external cracks wereformed on the surface of the tensile-tested specimen with a 0 h holding time; however, many external crackswere observed on the specimen surface subjected to hydrogen gas charging.

The column chromatographic separation of the MeOH extract from the aerial parts of Prunella vulgaris var. lilacina Nakai led to the isolation of fifteen triterpenoic acids (2-6, 9-13, 16-20), four flavonoids (14,21-23), four phenolics (7, 8, 15, 24), and a diterpene (1). Their structures were determined by spectroscopic methods to be trans-phytol (1), oleanic acid (2) ursolic acid (3), $2{\alpha}$,$3{\alpha}$,$19{\alpha}$-trihydroxyurs-12en-28oic acid (4), $2{\alpha}$,$3{\alpha}$-dihydroxyurs-12en-28oic acid (5), maslinic acid (6), caffeic acid (7), phydroxy cinnamic acid (8), $2{\alpha}$,$3{\alpha}$,$19{\alpha}$,23-tetrahydroxyurs-12en-28oic acid (9), $2{\alpha}$,$3{\alpha}$,23-trihydroxyurs-12en-28oic acid (10), $2{\alpha}$,$3{\beta}$-dihydroxyurs-12en-28oic acid (11), $2{\alpha}$,$3{\beta}$,24-trihydroxyolea-12en-28oic acid (12), (12R, 13S)-$2{\alpha}$,$3{\beta}$,24,trihydroxy-12,13-cyclo-taraxer-14-en-28oic acid (13), quercertin 3-O-$\beta$-D-glucopyranoside (14), rosmarinic acid (15), $2{\alpha}$,$3{\alpha}$,24-trihydroxyurs-12,20(30)-dien-28oic acid (16), $2{\alpha}$,$3{\alpha}$,24-trihydroxyolea-12en-28oic acid (17), $2{\alpha}$,$3{\beta}$,$19{\alpha}$,24-tetrahydroxyurs-12en-28oic acid 28-O-D-glucopyranoside (18), $2{\alpha}$,$3{\alpha}$,$19{\alpha}$,24-tetrahydroxyurs-12en-28oic acid 28-O-D-glucopyranoside (19), prunvuloside A (20), kaempferol 3-O-$\alpha$-L-rhamnopyranosyl($1{\rightarrow}6$)-$\beta$-D-glucopranoside (21), kaempferol 3-O-$\beta$-D-glucopyranoside (22), quercertin 3-O-$\alpha$-L-rhamnopyranosyl($1{\rightarrow}6$)-$\beta$-D-glucopyranoside (23), and 2-hydroxy-3-(3',4'-dihydroxyphenly)propanoic acid (24). Compounds 1, 8-12, 17, 21, 23, and 24 were isolated from this plant source for the first time. The isolated compounds were evaluated for their cytotoxicity against A549, SK-OV-3, SK-MEL-2, and HCTl5 cells in vitro using the sulforhodamin B bioassay (SRB) method. Compound 3 exhibited moderate cytotoxic activity against A549, SK-OV-3, SK-MEL-2, and HCTl5 cells, with $ED_{50}$ values of 3.71, 3.65, 13.62, and $5.44{\mu}M$, respectively.

<P>X-linked deafness type 3 (DFN3), the most prevalent X-linked form of hereditary deafness, is caused by mutations in the POU3F4 locus, which encodes a member of the POU family of transcription factors. Despite numerous reports on clinical evaluations and genetic analyses describing novel POU3F4 mutations, little is known about how such mutations affect normal functions of the POU3F4 protein and cause inner ear malformations and deafness. Here we describe three novel mutations of the POU3F4 gene and their clinical characterizations in three Korean families carrying deafness segregating at the DFN3 locus. The three mutations cause a substitution (p.Arg329Pro) or a deletion (p.Ser310del) of highly conserved amino acid residues in the POU homeodomain or a truncation that eliminates both DNA-binding domains (p.Ala116fs). In an attempt to better understand the molecular mechanisms underlying their inner ear defects, we examined the behavior of the normal and mutant forms of the POU3F4 protein in C3H/10T1/2 mesodermal cells. Protein modeling as well as in vitro assays demonstrated that these mutations are detrimental to the tertiary structure of the POU3F4 protein and severely affect its ability to bind DNA. All three mutated POU3F4 proteins failed to transactivate expression of a reporter gene. In addition, all three failed to inhibit the transcriptional activity of wild-type proteins when both wild-type and mutant proteins were coexpressed. Since most of the mutations reported for DFN3 thus far are associated with regions that encode the DNA binding domains of POU3F4, our results strongly suggest that the deafness in DFN3 patients is largely due to the null function of POU3F4.</P>

<P>Two new oxepin-containing (<B>1</B> and <B>2</B>) and two diketopiperazine-type alkaloids (<B>3</B> and <B>4</B>) have been isolated from an EtOAc extract of the marine-derived fungus <I>Aspergillus</I> sp. SF-5044. The structures of these metabolites were determined through analysis of NMR and MS data, along with Marfey’s method. Objectives: Rectal cancer bleeding occurs approximately 20% of overall rectal cancer patients. The current standard treatments cannot cover minor bleeding enough. Failures of the local or systemic approaches to stop bleeding are frequently reported. Methods: we reported a case of male rectal cancer patient who was treated with herbal decoction and significantly improved on symptoms, such as, loose stools and rectal bleeding. Previous to first visiting to our clinic, he was prescribed to use oral tranexamic acids, which failed to relieve his symptoms. He was administered with an herbal decoction, Inhyungtang, clinically known to have hemostastic effect in hematochezia. Results & Conclusion: The prescription showed significant improvement on his symptom, in particular, his bleeding. Thus, we report his case and we expect further experiences and experimental studies to support efficacy of Inhyungtang.
