<P>Seaweeds are considered a potential source of antiobesity agents. Because Caulerpa, a seaweed, has been consumed for food in Japan, China, South Korea, and Australia, we hypothesized that Caulerpa okamurae may have antiobesity effects in an animal model of high-fat diet (HFD)-induced obesity in C57BL/6 mice. Herein, we found that the ethanolic extract of C okamurae (COE) significantly inhibited lipid accumulation and reduced the expression of the master regulator of adipogenesis, peroxisome proliferator-activated receptor-gamma, sterol regulatory element binding protein-1c, and CCAAT/enhancer-binding protein-alpha in 3T3-L1 adipocytes. Moreover, COE significantly decreased body weight, fat weight, and liver weight in HFD-fed mice. This effect is comparable to that of positive control Garcinia cambogia extract, which has been approved by the Korean Food and Drug Administration as a weight loss food supplement in South Korea. Similarly, markers of weight gain such as free fatty acids, triglyceride, total cholesterol, glucose, and insulin in the plasma and free fatty acid, triglyceride, total cholesterol, and total lipid in the liver are significantly reduced in COE-treated HFD-fed mice. We found significantly reduced peroxisome proliferator-activated receptor-gamma, CCAAT/enhancer-binding protein-alpha, fatty acid synthase, sterol regulatory element binding protein-1c, cluster of differentiation 36, and acetyl-CoA synthetase in the adipose tissue of COE-treated HFD-fed mice. In conclusion, our results demonstrated that COE is effective in preventing body weight gain and fat accumulation and reduces plasma and hepatic lipid profiles. Together, these findings suggest that C okamurae may be used as a possible treatment option for the management of obesity and associated metabolic disorders. (C) 2017 Elsevier Inc. <P><I>Lespedeza </I>has been used for the management of diabetes in folklore medicine. The purpose of this study is to investigate the protective effects of the methanol extract of<I> Lespedeza davurica</I> (LD) on cytokine-induced <I>β</I>-cell damage and streptozotocin- (STZ-) induced diabetes. RINm5F cells were treated with interleukin- (IL-) 1<I>β</I> and interferon- (IFN-) <I>γ</I> to induce pancreatic <I>β</I>-cell damage. The exposure of LD extract significantly decreased cell death, nitric oxide (NO) production, nitric oxide synthase (iNOS) expression, and nucleus factor-kappa B (NF-<I>κ</I>B) p65 activation. Antidiabetic effects of LD extract were observed by oral glucose tolerance test (OGTT) in normal rats and by checking the biochemical, physiological, and histopathological parameters in STZ-induced diabetic rats. In OGTT, glucose clearance levels improved by oral treatment of LD extract. The water intake, urine volume, blood glucose, and serum TG, TC, TBARS, and DPP-IV levels were significantly decreased, and liver glycogen content was significantly increased by treatment of LD extract (250 mg/kg BW) in STZ-induced diabetic rats. Also, insulin immunoreactivity of the pancreases was increased in LD extract administrated rats compared with diabetic control rats. These results indicate that LD extract may protect pancreatic <I>β</I>-cell damage and regulate the blood glucose in STZ-induced diabetic rats.</P>

The aim of this study was to determine whether Caulerpa lentillifea extract (CLE) can protect pancreatic beta cells and enhance insulin signaling in adipocytes. We measured the protein tyrosine phosphatase (PTP)-1B inhibitory effect of CLE using an in-vitro enzyme assay. Proteins involved in the pancreatic beta-cell death and insulin signaling were measured by western blotting. Oil-red O staining was used to measure the insulin mimetic effect of CLE. CLE strongly inhibited the PTP1B enzyme. In rat insulinoma (RIN)-m5F cells, CLE decreased the activation of extracellular regulated kinase (ERK)-1/2, P38 mitogen activated protein kinase (P38), c-Jun NH2-terminal kinase (JNK), and nuclear factor kappa-light-chain-enhancer of the activated B cells (NF-κB). Furthermore, CLE showed insulin-mimetic effect and enhanced the activation of insulin-signaling molecules including IRS, AKT, PI3K, and GSK-3β in 3T3-L1 adipocytes. Our results suggested that CLE-inhibited PTP1B, protected the pancreatic beta cells, and enhanced insulin sensitization in the adipocytes.

The aim of this study was to evaluate the anti-diabetic effects of the water extract of Lespedeza cuneata (LCW) using rat insulinoma (RIN) m5F cells and streptozotocin (STZ)-induced diabetic rats. The effect of LCW on the protection of pancreatic beta cells was assessed using MTT assay, and nitric oxide production was assessed using Griess reagent. STZ-induced diabetic rats were treated with 100 and 400 mg/kg body weight of LCW for 5 weeks. In results, LCW significantly protected cytokine-induced toxicity and NO production, and increased insulin secretion in RINm5F cells. LCW significantly decreased serum blood glucose, thiobarbituric acid reactive substances (TBARS), blood urea nitrogen (BUN) and advanced glycation end products (AGEs) levels, and renal fibronectin expression in STZ-induced diabetic rats. Also, LCW effectively improved BW loss in STZ-induced diabetic rats. Thus, our results suggest that LCW has a beneficial effect on cytokine-induced pancreatic beta cell damage and biomarkers of diabetic complication in hyperglycemic rats.

Crateva unilocularis Buch-Ham has traditionally been used in Nepal for the treatment and prevention of diabetes. However, scientific verification through studies on the preventive effect of C. unilocularis on diabetes has not been done properly. In this study we investigated the effect of C. unilocularis leaf extract (CULE) on glucose uptake and insulin resistance. The effect of CULE on glucose uptake in an in vivo system was measured using zebrafish. In the cell-free system, enzymes activities related to diabetes were measured. Moreover, in the cell-cultured system, RINm5F pancreatic beta cells, 3T3-L1 adipocytes, and L6 myotubes were used to measure the effect of CULE on insulin secretion and glucose metabolism. CULE effectively enhanced glucose uptake in zebrafish larvae, and inhibited the activity of dipeptidyl peptidase-IV (DPP-IV) and protein tyrosine phosphatase-1B (PTP-1B) enzymes related to insulin secretion and insulin signaling activation. Also, CULE significantly increased insulin secretion with suppression of NO production in RIN-m5F pancreatic beta cells. In L6 myotubes and TNF-α-induced insulin resistance model of 3T3-L1 adipocytes, CULE significantly increased glucose uptake and immunofluorescence staining of glucose transporter (GLUT)4 protein. Furthermore, the regulatory response of glucose metabolism by CULE was a close correlation with the activation of insulin signaling (IRβ, PI3K, AKT) and 5′-AMP-activated protein kinase (AMPK)α and the reduction of p38 mitogen-activated protein kinase (MAPK) in TNF-α-induced insulin resistance model of 3T3-L1 adipocytes. Thus, our results suggest that CULE may act as a potential agent for the prevention and treatment of metabolic syndrome associated with type 2 diabetes.

Obesity is a complex metabolic disorder that increases the risk for type 2 diabetes, hyperlipidemia, hypertension, and atherosclerosis. In this study, we evaluated the anti-obesity effects of Nelumbo nucifera leaf (NL) extract in 3T3-L1 adipocytes and obese db/db mice. NL extract among various parts (leaf, seed, and root) of N. nucifera most effectively reduced adipogenesis via inhibiting CCAAT enhancer binding protein α (C/EBPα) and peroxisome proliferator activated receptor γ (PPARγ) expression in 3T3-L1 adipocytes. The addition of NL extract enhanced the protein expression of uncoupling protein 2 (UCP2) as compared to untreated 3T3-L1 adipocytes. The oral administration of NL extract (100 mg/kg BW) significantly reduced food efficacy ratio, body weight, and face or total cholesterol level in obese db/db mice. Also, administration of NL extract significantly decreased adipocyte size and C/EBPα or PPARγ expression in the adipose tissues as compared with control (obese db/db mice). Therefore, our results suggest that NL extract among various parts of N. nucifera could be used as a functional food ingredient for the prevention and treatment of metabolic diseases including obesity and diabetes.

Purpose: Sexually transmitted infections are a major worldwide concern, and human papillomavirus (HPV) is one of the significant risk factors. Many populations suffer from various diseases caused by HPV, and the overall death toll due to cervical carcinoma is remarkable. Despite vaccine availability, perception about vaccine safety and efficacy, its’ preventive outcome is still inferior among the health professionals and vaccine providers. So, this study aims to assess the knowledge, attitude, and practice level of HPV and its’ vaccination among doctors, dentists, and medical students. Materials and Methods: This cross-sectional survey was carried out between April to August 2021, where 626 participants from all types of medical institutions of Bangladesh were interviewed using a validated and structured questionnaire that consists of four extensive areas; socio-demographic characteristics, HPV knowledge, attitude, and practices regarding vaccination. Results: The knowledge and practice standards showed very poor outcomes where 43.29% of the participants showed good knowledge and only 11.82% conveyed good practices. Nevertheless, the attitude towards HPV vaccination was revealed high (75.88%). Female participants showed more positive attitudes than males. Conclusion: Physicians and dentists play vital roles in raising public knowledge about HPV and awareness regarding HPV vaccination programs. The provision of medical education on HPV must be prioritized, and current training techniques must be re-evaluated.
