        Interaction of silver(I) and copper(I) with an O<sub>2</sub>S<sub>2</sub>-macrocycle - A comparative structural study

        The 14-membered O<SUB>2</SUB>S<SUB>2</SUB>-macrocycle L reacts with AgPF<SUB>6</SUB> in acetonitrile/dichloromethane (1:1) to yield the cyclic, double S-Ag-S bridged complex, [Ag<SUB>2</SUB>L<SUB>2</SUB>(CH<SUB>3</SUB>CN)<SUB>4</SUB>](PF<SUB>6</SUB>)<SUB>2</SUB> (2), in which each Ag(I) is bound to two exo-oriented S donors arising from different macrocycles to form a 12-membered, di-Ag metallacycle. Two PF<SUB>6</SUB><SUP>-</SUP> ions form weak intramolecular contacts [Ag...F, 2.925(2)A] between the Ag centres in a bridging bidentate fashion. The coordination sphere of each Ag is completed by two weakly bound acetonitrile molecules. The respective macrocyclic ligands are arranged trans to the mean plane through the metallacycle. When the above synthetic procedure was repeated employing methanol/dichloromethane (1:1) as solvent, a complex of type [Ag<SUB>2</SUB>L<SUB>2</SUB>](PF<SUB>6</SUB>)<SUB>2</SUB> (3) was obtained whose structure incorporates a similar 12-membered metallacycle but with the respective macrocyclic rings now oriented towards the same side of the metallacyclic plane. The reaction of L with AgCF<SUB>3</SUB>SO<SUB>3</SUB> yields [Ag<SUB>2</SUB>L<SUB>2</SUB>(CF<SUB>3</SUB>SO<SUB>3</SUB>)<SUB>2</SUB>].2CH<SUB>3</SUB>CN (4) whose structure resembles that of [Ag<SUB>2</SUB>L<SUB>2</SUB>(CH<SUB>3</SUB>CN)<SUB>4</SUB>](PF<SUB>6</SUB>)<SUB>2</SUB> (2); each complex unit incorporates a di-Ag(I) 12-membered metallacycle, with symmetrical bridging bidentate CF<SUB>3</SUB>SO<SUB>3</SUB><SUP>-</SUP> ions weakly linking Ag centres. Long Ag1...Ag1C contacts are present between pairs of complex units. Reaction of Cu(CH<SUB>3</SUB>CN)<SUB>4</SUB>PF<SUB>6</SUB> (one equiv.) with L in methanol/dichloromethane (1:1) resulted in isolation of [CuL<SUB>2</SUB>]PF<SUB>6</SUB> (5) incorporating a distorted tetrahedral S<SUB>4</SUB>-coordination sphere, with each macrocycle binding as a bidentate ligand via its two (exo-orientated) S atoms. In contrast, reaction of [Cu(CH<SUB>3</SUB>CN)<SUB>4</SUB>]PF<SUB>6</SUB> (two equiv.) with L in either acetonitrile/dichloromethane (1:1) or methanol/dichloromethane (1:1) yielded [Cu<SUB>2</SUB>L<SUB>3</SUB>]PF<SUB>6</SUB> (6) in which the Cu(I) centres are bound exo to the macrocyclic cavity by individual S-donors from one L, with the coordination sphere of each Cu(I) completed by out-of-plane binding of the S<SUB>2</SUB>O donors from a second L. Unsymmetrical F...Cu contacts from a PF<SUB>6</SUB><SUP>-</SUP> anion weakly link the pair of Cu centres in a bridging bidentate fashion.

        The development of a cryogenic detector with CaMoO<sub>4</sub> crystals for neutrinoless double beta decay search

        The search for neutrinoless double beta decay (0νββ) is a key experiment for evaluating the mass of the neutrino and for determining the Majorana/Dirac nature of neutrinos. A scintillating CaMoO<SUB>4</SUB> crystal is a good material for use in investigating 0νββ with cryogenic detectors. The high transition energy (Q=3034keV) and nearly 10% natural abundance of <SUP>100</SUP>Mo, together with the scintillating property of the crystal, provide favorable conditions to search for 0νββ of <SUP>100</SUP>Mo. We report a prototype experiment operating at low temperatures using a CaMoO<SUB>4</SUB> crystal as a particle absorber. Prior to testing of isotopically-enriched <SUP>40</SUP>Ca<SUP>100</SUP>MoO<SUB>4</SUB> crystals, a CaMoO<SUB>4</SUB> crystal with Mo of natural isotopic composition was tested. Using a metallic magnetic calorimeter as a temperature sensor, we achieved high energy resolutions for alpha particles and low-energy gamma rays. This experiment shows the feasibility of scaling up the crystal size to perform a high sensitivity search for 0νββ decay of <SUP>100</SUP>Mo.

      • Side-chain engineering of ladder-structured polysilsesquioxane membranes for gas separations

        A comprehensive and fundamental gas transport study of ladder-structured polysilsesquioxanes (LPSQs) was systematically performed by investigating the effects of various alkyl substituents, different copolymer ratios, and UV-irradiation induced photo-crosslinking on gas separations. Overall, LPSQ membranes are more suitable for CO<SUB>2</SUB>/N<SUB>2</SUB> and CO<SUB>2</SUB>/H<SUB>2</SUB> separations due to a relatively high affinity towards CO<SUB>2</SUB> as well as rubbery polymer properties. The gas transport in LPSQ membranes was well interpreted by two important parameters, the inter-chain distance and the side chain mobility. A combination of larger inter-chain distance and higher side chain rigidity tends to increase the fractional free volume, resulting in higher gas permeability. Also, it was successfully demonstrated that the separation performance of LPSQ membranes can be predicted by using a logarithmic permeability relationship based on the transport characterization for a series of ladder-structured poly(phenyl-co-methacryloxypropyl)silsesquioxanes. Lastly, the UV-curing process reduced the permeability of LPSQ membranes, increasing the selectivity due to the restricted chain mobility.

      • Differential regulation of Akt/protein kinase B isoforms during cell cycle progression

        Phosphatidylinositol 3-kinase pathways play key regulatory roles in cell cycle progression into S phase. In this study, we demonstrated that Akt1/PKBα isoform plays an essential role in G<SUB>1</SUB>/S transition and proliferation. Cells lacking Akt1/PKBα showed an attenuated proliferation as well as G<SUB>1</SUB>/S transition, whereas cells lacking Akt2/PKBβ showed normal proliferation and G<SUB>1</SUB>/S transition. The effect of Akt1/PKBα on cell proliferation and G<SUB>1</SUB>/S transition was completely abolished by swapping pleckstrin homology (PH) domain with that of Akt2/PKBβ. Finally, full activation of Akt/PKB and cyclin D expression was achieved by the Akt1/PKBα or chimeric proteins containing the PH domain of Akt1/PKBα indicating that the PH domain of Akt1/PKBα provides full kinase activity and is necessary for the G<SUB>1</SUB>/S transition.

        Selection for Duration of Fertility and Mule Duck White Plumage Colour in a Synthetic Strain of Ducks (Anas platyrhynchos)

        A synthetic strain of ducks (Anas platyrhynchos) was developed by introducing genes for long duration of fertility to be used as mother of mule ducklings and a seven-generation selection experiment was conducted to increase the number of fertile eggs after a single artificial insemination (AI) with pooled Muscovy semen. Reciprocal crossbreeding between Brown Tsaiya LRI-2 (with long duration of fertility) and Pekin L-201 (with white plumage mule ducklings) ducks produced the G0. Then G1 were intercrossed to produce G2 and so on for the following generations. Each female duck was inseminated 3 times, at 26, 29, and 32 weeks of age. The eggs were collected for 14 days from day 2 after AI. Individual data regarding the number of incubated eggs (Ie), the number of fertile eggs at candling at day 7 of incubation (F), the total number of dead embryos (M), the maximum duration of fertility (Dm) and the number of hatched mule ducklings (H) with plumage colour were recorded. The selection criterion was the breeding values of the best linear unbiased prediction animal model for F. The results show high percentage of exhibited heterosis in G2 for traits to improve (19.1% for F and 12.9% for H); F with a value of 5.92 (vs 3.74 in the Pekin L-201) was improved in the G2. Heritabilities were found to be low for Ie ($h^2=0.07{\pm}0.03$) and M ($h^2=0.07{\pm}0.01$), moderately low for Dm ($h^2=0.13{\pm}0.02$), of medium values for H ($h^2=0.20{\pm}0.03$) and F ($h^2=0.23{\pm}0.03$). High and favourable genetic correlations existed between F and Dm ($r_g=0.93$), between F and H ($r_g=0.97$) and between Dm and H ($r_g=0.90$). The selection experiment showed a positive trend for phenotypic values of F (6.38 fertile eggs in G10 of synthetic strain vs 5.59 eggs in G4, and 3.74 eggs in Pekin L-201), with correlated response for increasing H (5.73 ducklings in G10 vs 4.86 in G4, and 3.09 ducklings in Pekin L-201) and maximum duration of the fertile period without increasing the embryo mortality rate. The average predicted genetic response for F was 40% of genetic standard deviation per generation of selection. The mule ducklings' feather colour also was improved. It was concluded that this study provided results for a better understanding of the genetics of the duration of fertility traits in the common female duck bred for mule and that the selection of a synthetic strain was effective method of improvement.

        Cleaved c-FLIP mediates the antiviral effect of TNF-α against hepatitis B virus by dysregulating hepatocyte nuclear factors

        <P>Background & Aims: Cytokines are key molecules implicated in the defense against virus infection. Tumor necrosis factor-alpha (TNF-alpha) is well known to block the replication of hepatitis B virus (HBV). However, the molecular mechanism and the downstream effector molecules remain largely unknown. Methods: In this study, we investigated the antiviral effect and mechanism of p22-FLIP (FLICE-inhibitory protein) by ectopic expression in vitro and in vivo. In addition, to provide the biological relevance of our study, we examined that the p22-FLIP is involved in TNF-alpha-mediated suppression of HBV in primary human hepatocytes. Results: We found that p22-FLIP, a newly discovered c-FLIP cleavage product, inhibited HBV replication at the transcriptional level in both hepatoma cells and primary human hepatocytes, and that c-FLIP conversion to p22-FLIP was stimulated by the TNF-alpha/NF-kappa B pathway. p22-FLIP inhibited HBV replication through the upregulation of HNF3 beta but downregulation of HNF4 alpha, thus inhibiting both HBV enhancer elements. Finally, p22-FLIP potently inhibited HBV DNA replication in a mouse model of HBV replication. Conclusions: Taken together, these findings suggest that the anti-apoptotic p22-FLIP serves a novel function of inhibiting HBV transcription, and mediates the antiviral effect of TNF-a against HBV replication.         Microsatellite typing for DRB1 alleles: application to the analysis of HLA associations with rheumatoid arthritis

        The current methods for molecular typing of HLA-DR alleles incur a substantial financial burden when performing large population studies. In the current study, we aimed to provide much less expensive typing approach with high predictability for DRB1 genotype. We have used a panel of three microsatellite markers in the class II region (D6S2666, D6S2665 and D6S2446) for genotyping and haplotype reconstruction in a total of 1687 Caucasian (1313 RA patients and 374 controls) and 1364 Korean individuals (744 RA patients and 620 controls), all of whom were previously genotyped for DRB1. We found that a total of 88.4 and 87.4% of all observed three-marker haplotypes could determine the DR type with a positive predictive value >0.8 with high sensitivity and specificity. There was a high degree of haplotype conservation when comparing Caucasian and Asian populations. Interestingly, we found that the majority of DRB1<SUP>*</SUP>09 and DRB1<SUP>*</SUP>10 alleles share a common three-marker haplotype in both Caucasian and Asian populations. This is unexpected, since these two alleles are found on very different haplotype families.         Doxorubicin/heparin composite nanoparticles for caspase-activated prodrug chemotherapy

        <P>Caspase-activated prodrug chemotherapy is introduced and demonstrated using the composite nano particles (NPs), which deliver doxorubicin (DOX) and DEVD-S-DOX together to the tumor tissue. DEVD-S-DOX, DOX linked to a peptide moiety (DEVD), is a prodrug that is cleaved into free DOX by caspase-3 upon apoptosis. DEVD-S-DOX has no therapeutic efficacy, but it changes into free DOX with the expression of caspase-3. With the accumulation of the composite NPs in the tumor tissue by the enhanced permeation and retention (EPR) effect, a small exposure of DOX in the tumor. cells initiated apoptosis in a localized area of the tumor tissue, which induced caspase-3 activation. Cleavage of DEVD-S-DOX into free DOX by caspase-3 continued with repetitive activation of caspase-3 and cleavage of DEVD-S-DOX at the tumor site. The composite NPs were characterized with transmittance electron microscopy (TEM) and particle size analyzer. We then evaluated the nanoparticle drug release, therapeutic efficacy, and in vivo biodistribution for tumor targeting using a non-invasive live animal imaging technology and the quantification of DOX with high performance liquid chromatography. DOX-induced apoptosis-targeted chemotherapy (DIATC) was verified by in vitro/in vivo DEVD-S-DOX response to free DOX and cellular uptake behavior of the composite NPs with flow cytometry analysis. Significant antitumor efficacy with minimal cardiotoxicity was also observed, which supported DIATC for improved chemotherapy. (C) 2016 Elsevier Ltd. All rights reserved.</P>
