        Mangiferin ameliorates cardiac fibrosis in D-galactose-induced aging rats by inhibiting TGF-β/p38/MK2 signaling pathway

        Aging is the process spontaneously occurred in living organisms. Cardiac fibrosis is a pathophysiological process of cardiac aging. Mangiferin is a well-known C-glucoside xanthone in mango leaves with lots of beneficial properties. In this study, rat model of cardiac fibrosis was induced by injected with 150 mg/kg/d D-galactose for 8 weeks. The age-related cardiac decline was estimated by detecting the relative weight of heart, the serum levels of cardiac injury indicators and the expression of hypertrophic biomakers. Cardiac oxidative stress and local inflammation were measured by detecting the levels of malondialdehyde, enzymatic antioxidant status and proinflammatory cytokines. Cardiac fibrosis was evaluated by observing collagen deposition via masson and sirius red staining, as well as by examining the expression of extracellular matrix proteins via Western blot analysis. The cardiac activity of profibrotic TGF-β1/p38/MK2 signaling pathway was assessed by measuring the expression of TGF-β1 and the phosphorylation levels of p38 and MK2. It was observed that mangiferin ameliorated D-galactose-induced cardiac aging, attenuated cardiac oxidative stress, inflammation and fibrosis, as well as inhibited the activation of TGF-β1/p38/MK2 signaling pathway. These results showed that mangiferin could ameliorate cardiac fibrosis in D-galactose-induced aging rats possibly via inhibiting TGF-β/p38/MK2 signaling pathway.

        Zearalenone regulates key factors of the Kelch-like erythroid cell-derived protein with CNC homology-associated protein 1-nuclear factor erythroid 2-related factor 2 signaling pathway in duodenum of post-weaning gilts

        Objective: This study explored the mechanism of the Kelch-like erythroid cell-derived protein with CNC homology-associated protein 1 (Keap1)-nuclear factor erythroid 2-related factor 2 (Nrf2) signaling pathway under conditions of zearalenone (ZEA)-induced oxidative stress in the duodenum of post-weaning gilts. Methods: Forty post-weaning gilts were randomly allocated to four groups and fed diets supplemented with 0, 0.5, 1.0, or 1.5 mg/kg ZEA. Results: The results showed significant reductions in the activity of the antioxidant enzymes total superoxide dismutase and glutathione peroxidase and increases the malondialdehyde content with increasing concentrations of dietary ZEA. Immunohistochemical analysis supported these findings by showing a significantly increased expression of Nrf2 and glutathione peroxidase 1 (GPX1) with increasing concentrations of ZEA. The relative mRNA and protein expression of Nrf2, GPX1 increased linearly (p<0.05) and quadratically (p<0.05), which was consistent with the immunohistochemical results. The relative mRNA expression of Keap1 decreased linearly (p<0.05) and quadratically (p<0.05) in the duodenum as the ZEA concentration increased in the diet. The relative mRNA expression of modifier subunit of glutamate-cysteine ligase (GCLM) increased quadratically (p<0.05) in all ZEA treatment groups and the relative mRNA expression of quinone oxidoreductase 1 (NQO1) catalytic subunit of glutamate-cysteine ligase decreased linearly (p<0.05) and quadratically (p<0.05) in the ZEA1.0 group and ZEA1.5 group. The relative protein expression of Keap1 and GCLM decreased quadratically (p<0.05) in the duodenum as the ZEA concentration increased in the diet, respectively. The relative protein expression of NQO1 increased linearly (p<0.05) and quadratically (p<0.05) in all ZEA treatment groups in the duodenum. Conclusion: These findings suggest that ZEA regulates the expression of key factors of the Keap1-Nrf2 signaling pathway in the duodenum, which enables resistance to ZEA-induced oxidative stress. Further studies are needed to examine the effects of ZEA induced oxidative stress on other tissues and organs in post-weaning gilts.

        Traffic Engineering and Manageability for Multicast Traffic in Hybrid SDN

        Multicast communication can effectively reduce network resources consumption in contrast with unicast. With the advent of SDN, current researches on multicast traffic are mainly conducted in the SDN scenario, thus to mitigate the problems of IP multicast such as the unavoidable difficulty in traffic engineering and high security risk. However, migration to SDN cannot be achieved in one step, hybrid SDN emerges as a transitional networking form for ISP network. In hybrid SDN, for acquiring similar TE and security performance as in SDN multicast, we redirect every multicast traffic to an appropriate SDN node before reaching the destinations of the multicast group, thus to build up a core-based multicast tree substantially which is first introduced in CBT. Based on the core SDN node, it is possible to realize dynamic control over the routing paths to benefit traffic engineering (TE), while multicast traffic manageability can also be obtained, e.g., access control and middlebox-supported network services. On top of that, multiple core-based multicast trees are constructed for each multicast group by fully taking advantage of the routing flexibility of SDN nodes, in order to further enhance the TE performance. The multicast routing and splitting (MRS) algorithm is proposed whereby we jointly and efficiently determine an appropriate core SDN node for each group, as well as optimizing the traffic splitting fractions for the corresponding multiple core-based trees to minimize the maximum link utilization. We conduct simulations with different SDN deployment rate in real network topologies. The results indicate that, when 40% of the SDN switches are deployed in HSDN as well as calculating 2 trees for each group, HSDN multicast adopting MRS algorithm can obtain a comparable TE performance to SDN multicast.

      • Relationship Between PTEN and Livin Expression and Malignancy of Renal Cell Carcinomas

        Objectives: This study focused on PTEN and Livin expression and associations with malignancy in human renal clear cell carcinomas (RCCC). Methods: PTEN and Livin expression was assessed in 100 RCCC tissue samples, 50 paracarcinoma cases, and 20 normal renal tissue samples using the immunohistochemical Streptavidin proxidase (SP) method. The relationships between binding and corresponding biological characteristics, such as histological grade, lymph node metastases, and clinical stages were analyzed. Results: Positive PTEN expression in RCCC was significantly lower than in renal tissue adjacent to carcinoma tissue and normal renal tissue (P<0.01). Livin expression in the renal tissue adjacent to the carcinoma and normal renal tissues exhibited only low levels, whereas overall Livin expression in RCCC was statistically significant (P<0.01). In RCCC, PTEN expression rate gradually decreased with an increase in clinical stage, whereas that of Livin increased to statistically significant levels (P<0.01), PTEN and Livin levels being negatively correlated (r=-0.395, P<0.01). Conclusions: PTEN and Livin are important in RCCC development. The two factors combined are expected to provide indices for estimating RCCC malignancy and progression levels, as well as references for RCCC diagnosis and treatment.

      • Application of nanofiltration membrane in the recovery of aluminum from alkaline sludge solutions

        Large amounts of aluminum hydroxide ($Al(OH)_3$) exist in water purification sludge (WPS) because of the added aluminum coagulant in water treatment process. Notably, $Al(OH)_3$ is an amphoteric compound, can be dissolved in its basic condition using sodium hydroxide to form aluminate ions ($Al(OH)_4{^-}$). However, in a process in which pH is increasing, the humid acid can be dissolved easily from WPS and will inhibit the recovery and reuse of the dissolved aluminate ions. This study attempts to fix this problem by a novel approach to separate $Al(OH)_4{^-}$ ions using nanofiltration (NF) technology. Sludge impurity in a alkaline solution is retained by the NF membrane, such that the process recovers $Al(OH)_4{^-}$ ions, and significantly decreases the organic matter or heavy metal impurities in the permeate solution. The $Al(OH)_4{^-}$ ion is an alkaline substance. Experimental results confirm that a recovered coagulant of $Al(OH)_4{^-}$ ion can effectively remove kaolin particles from slightly acidic synthetic raw water.

      • The Modeling Methods of Heavy NC Machine Tool Bolt Joint Based on Virtual Media

        The Department of Heavy NC machine tool has a great influence on the performance of machine tools. The 30%-50% static stiffness, more than 50% of the dynamic stiffness depends on the combining part, and the influence on damping reached 90%.Machine tool with the Department of static and dynamic performance directly determines the accuracy and reliability of Heavy NC machine tool. Therefore, in conjunction with the Department of research and design of Heavy NC machine tool is very important. In this paper, based on Hertz contact theory, the contact at the micro level, considering the combination form of machine tool, and the machine joints as a virtual media simulation, a combination of virtual media framework, and calculation of characteristic parameters of the virtual material, then use the ANSYS Workbench to solve the combination of the natural frequency of the whole, the first five order modal extraction. Using modal test technology, get the specimens of natural frequency and damping of the true value, natural frequency are compared with theory, prove the formula of virtual media is reliable, can be used in the modeling of heavy machine tool.

        An Importance Sampling Method to Evaluate Value-at-Risk for Assets with Jump Risks

        Risk management is an important issue when there is a catastrophic event that affects asset price in the market such as a sub-prime financial crisis or other financial crisis. By adding a jump term in the geometric Brownian motion, the jump diffusion model can be used to describe abnormal changes in asset prices when there is a seri-ous event in the market. In this paper, we propose an importance sampling algorithm to compute the Value-at-Risk for linear and nonlinear assets under a multi-variate jump diffusion model. To be more precise, an efficient computational procedure is developed for estimating the portfolio loss probability for linear and nonlinear assets with jump risks. And the titling measure can be separated for the diffusion and the jump part under the assumption of independence. The simulation results show that the efficiency of importance sampling improves over the naive Monte Carlo simulation from 7 times to 285 times under various situations. We also show the robustness of the importance sampling algorithm by comparing it with the EVT-Copula method proposed by Oh and Moon (2006).

        Pure Ultraviolet Emission from ZnO Quantum Dots-Based/GaN Heterojunction Diodes by MgO Interlayer

        We demonstrate the fabrication and characterization of ZnO/GaNbasedheterojunction light-emitting diodes (LEDs) by using air-stableand solution-processable ZnO quantum dots (QDs) with a thin MgOinterlayer acting as an electron blocking layer (EBL). The ZnO QDs/MgO/p-GaN heterojunction can only display electroluminescence(EL) characteristic in reverse bias regime. Under sufficient reversebias, a fairly pure ultraviolet EL emission located at 370 nm derivingfrom near band edge of ZnO with a full width at half maximum(FWHM) of 8.3 nm had been obtained, while the deep-level emissionhad been almost totally suppressed. The EL origination andcorresponding carrier transport mechanisms were investigatedqualitatively in terms of photoluminescence (PL) results and energyband diagram.
