      • Golgi Phosphoprotein 2 Down-regulates the Th1 Response in Human Gastric Cancer Cells by Suppressing IL-12A

        Golgi phosphoprotein 2 (GOLPH2) is a very important biomarker in a variety of diseases. Its biological function is not clear, particularly in gastric cancer. To investigate the role of GOLPH2 in human gastric cancer, and determine its effect on the Th1 lymphocyte response, its expression and that of IL-12A were measured by real-time PCR and immunohistochemistry. The relationship between GOLPH2 and IL-12A was analysed statistically. The effect of GOLPH2 on the Th1 lymphocyte response was investigated with an in vitro co-culture system. The results showed that in human gastric cancer, the expression of GOLPH2 was significantly higher and the expression of IL-12A was lower than in normal gastric mucosal tissues, and the expression levels of GOLPH2 and IL-12A were negatively correlated. In addition, obvious down-regulation of the Th1 response was observed when lymphocytes were co-cultured with gastric cancer SGC7901 cells over-expressing GOLPH2. GOLPH2 down-regulated the expression of IL-12A, and inhibited the expression of TNF-${\alpha}$ and IFN-${\gamma}$. The results indicated that GOLPH2 down-regulates the Th1 response via suppression of IL-12A in human gastric cancer, and this might provide a target for the prevention and treatment.

        Lysinibacillus tabacifolii sp. nov., a Novel Endophytic Bacterium Isolated from Nicotiana tabacum Leaves

        A Gram-positive, catalase- and oxidase-positive, strictly aerobic, endospore-forming rod bacterium, designated K3514T, was isolated from the leaves of Nicotiana tabacum. The strain was able to grow at temperatures of 8–40°C, pH 5.0–10.0 and NaCl concentrations of 0–7%. The predominant quinones (>30%) of this strain were MK-7(H2) and MK-7. Phylogenetic analysis of 16S rRNA gene sequence showed that strain K3514T was affiliated to the genus Lysinibacillus, with its closest relatives being Lysinibacillus mangiferihumi (98.3% sequence similarity), Lysinibacillus sphaericus (97.9% sequence similarity), Lysinibacillus fusiformis (97.4% sequence similarity), and Lysinibacillus xylanilyticus (97.3% sequence similarity). However, low levels of DNA-DNA relatedness values suggested that the isolate was distinct from the other closest Lysinibacillus species. Additionally, based on analysis of morphological, physiological, and biochemical characteristics, the isolate could be differentiated from the closest known relatives. Therefore, based on polyphasic taxonomic data, the novel isolate likely represents a novel species, for which the name Lysinibacillus tabacifolii sp. nov. and the type strain K3514T (=KCTC 33042T =CCTCC AB 2012050T) are proposed.

      • Influence of the temperature and hydraulic retention time in bioelectrochemical anaerobic digestion of sewage sludge

        <P><B>Abstract</B></P> <P>At ambient temperature (25 °C), bioelectrochemical anaerobic digestion of sewage sludge was investigated with a hydraulic retention time (HRT) of 10 days and compared to that at a mesophilic condition (35 °C). The methane production and methane content in the biogas at ambient temperature were 612.8 mL/L·d and 73.3%, respectively, which were not significantly lower than that of the mesophilic condition. Additionally, the VS removal was 54.5% which was similar for both temperature conditions. However, for a HRT of 20 days, the bioelectrochemical anaerobic digestion at ambient temperature became more stable, and the VS removal improved up to 65.0%. For the HRT of 10 days, the net energy production at ambient temperature was about 168 kJ/L·d, which was similar to the mesophilic condition; however, the apparent energy efficiency at ambient temperature was 249.2% which was significantly higher than 197.7% at the mesophilic condition. The bioelectrochemical anaerobic digestion that can save the thermal energy input at ambient temperature is recommended for the treatment of organic waste including sewage sludge in moderate and cold climate regions.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The bioelectrochemical anaerobic digestion (BEAD) has advantages at ambient temperature. </LI> <LI> CH<SUB>4</SUB> production at 25 °C in BEAD is not significantly reduced compared to 35 °C. </LI> <LI> Apparent energy efficiency (249.2%) at 25 °C in BEAD is higher than at 197.2% at 35 °C. </LI> <LI> Extending HRT in BEAD improves the process stability and VS removal at 25 °C. </LI> <LI> BEAD for organic waste at 25 °C is a feasible process in moderate and cold regions. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Effect of metal oxide partial substitution of V2O5 in V2O5–WO3/TiO2 on selective catalytic reduction of NO with NH3

        After partial substitution of V2O5 in V2O5–WO3/TiO2 by metal oxide (MxOy, M = Fe, Co, Ni, Cu, Sr, La, and Ce), the NO conversions for the MxOy–V2O5–WO3/TiO2 catalysts (M-VW) showed the following sequence: Co–VW > Fe–VW > Sr-VW Ce-VW La-VW > VW > Ni-VW > Cu-VW. The reduction activities for almost all the M-VW catalysts were enhanced at reaction temperature >400 8C, but only the Co-VW catalyst showed an increased activity at temperature <400 8C. Among the M-VW catalysts tested, the Co-VW sample had the highest catalytic activity with a temperature range of 300–550 8C for more than 90% NO removal at a GHSV of 60 000 h1. Moreover, the Co-VW sample exhibited high space- velocity, H2O and SO2 resistance, and low N2O yield. Co partial substitution for V in the VW led to more Lewis acid sites and Brønsted acid sites, and an obvious increase in the ratio of the adsorbed oxygen to the lattice oxygen from 12.74% for VW to 36.73% for Co-VW. The increased adsorbed oxygen and Lewis and Brønsted acid sites contributed to the improved SCR activity of the Co-VW sample.

      • PU.1 Is Identified as a Novel Metastasis Suppressor in Hepatocellular Carcinoma Regulating the miR-615-5p/IGF2 Axis

        Invasion and metastasis is the major cause of tumor recurrence, difficulty for cure and low survival rate. Excavating key transcription factors, which can regulate tumor invasion and metastasis, are crucial to the development of therapeutic strategies for cancers. PU.1 is a master hematopoietic transcription factor and a vital regulator in life. Here, we report that, compared to adjacent non-cancerous tissues, expression of PU.1 mRNA in metastatic hepatocellular carcinoma (HCC), but not primary HCC, was significantly down-regulated. In addition, levels of PU.1 mRNA in metastatic hepatoma cell lines MHCC97L and MHCC97H were much lower than in non-metastatic Hep3B cells. Transwell invasion assays after PU.1 siRNA transfection showed that the invasion of hepatoma cell lines was increased markedly by PU.1 knockdown. Oppositely, overexpression of PU.1 suppressed the invasion of these cells. However, knockdown and overexpression of PU.1 did not influence proliferation. Finally, we tried to explore the potential mechanism of PU.1 suppressing hepatoma cell invasion. ChIP-qPCR analysis showed that PU.1 exhibited a high binding capacity with miR-615-5p promoter sequence. Overexpression of PU.1 caused a dramatic increase of pri-, pre- and mature miR-615-5p, as well as a marked decrease of miR-615-5p target gene IGF2. These data indicate that PU.1 inhibits invasion of human HCC through promoting miR-615-5p and suppressing IGF2. These findings improve our understanding of PU.1 regulatory roles and provided a potential target for metastatic HCC diagnosis and therapy.

        Bioelectrochemical enhancement of direct interspecies electron transfer in upflow anaerobic reactor with effluent recirculation for acidic distillery wastewater

        <P><B>Abstract</B></P> <P>Methane production in the upflow anaerobic bioelectrochemical reactor (UABE) treating acidic distillery wastewater was compared to the upflow anaerobic sludge blanket reactor (UASB), and the electron transfer pathways for methane production were also evaluated in the effluent recirculation. The methane productions from reactors were influenced by the low pH of influent wastewater. However, the methane production rate and yield of the UABE were 2.08L/L.d and 320mL/g COD<SUB>r</SUB>, which were higher than the UASB. The effluent recirculation containing alkalinity neutralized the acidic influent and increased the upflow velocity in both reactors, and improved the direct interspecies electron transfer more in the UABE. When the effluent recirculation ratio was 3.0 in the UABE, the methane production rate and yield were reached up to 3.88L/L.d and 501.0mL/g COD<SUB>r</SUB>, respectively. The UABE requires electrode installation and electrical energy for operation, but the benefits from increased methane production are much higher.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Electroactive bacteria enriched in UABE activate bDIET for methane production. </LI> <LI> bDIET for methane production is less susceptible to low influent pH. </LI> <LI> Effluent recirculation in UABE further improves bDIET for methane production. </LI> <LI> UABE with effluent recirculation is a recommended process for distillery wastewater. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

      • Electroactive microorganisms in bulk solution contribute significantly to methane production in bioelectrochemical anaerobic reactor

        <P><B>Abstract</B></P> <P>The role of anaerobic microorganisms suspended in the bulk solution on methane production was investigated in a bioelectrochemical anaerobic reactor with the electrode polarized at 0.5 V. The electron transfer from substrate to methane and hydrogen were 25% and 7.5%, respectively, in the absence of the anaerobic microorganisms in the bulk solution. As the anaerobic microorganisms increased to 4400 mg/L, the electrons transferred to methane increased to 83.3% but decreased to 0.3% in hydrogen. The electroactive microorganisms (EAM), including exoelectrogens and electrotrophs, enriched in the bulk solution that confirmed by the redox peaks in the cyclic voltammogram was proportional to the anaerobic microorganism. The methane yield based on COD removal was dependent on the anaerobic microorganisms in the bulk solution rather than on the bioelectrode surface.         Patterns of Failure and Survival Trends in 3,808 Patients with Stage II Nasopharyngeal Carcinoma Diagnosed from 1990 to 2012: A Large-Scale Retrospective Cohort Study

        Purpose The purpose of this study was to investigate the survival trends and patterns of failure in patients with stage II nasopharyngeal carcinoma (NPC) treated with radiotherapy (RT) and chemotherapy over the last 20 years. Materials and Methods Thirty-eight hundred and eight patients diagnosed with stage II NPC between January 1990 and December 2012 were involved in this retrospective cohort study. All patients were treated with RT. According to the main imaging techniques and RT technology, we categorized these patients into four calendar periods: 1990-1996, 1997-2002, 2003-2007, and 2008-2012. Overall survival (OS), progression-free survival (PFS), locoregional relapse-free survival (LRFS), and distant metastasis–free survival (DMFS) were served as the clinical outcome. Results After a median follow-up period of 84.7 months, we observed increasing trends in survival and disease control. The 3- and 5-year OS rates increased from 87.1% and 78.7% in the first calendar period to 97.4% and 94.5% in the last calendar period, respectively (p < 0.001). Additionally, significant increasing trends could be seen in the PFS and LRFS during the four calendar periods. In the subgroup analysis, the LRFS in patients older than 50 years at diagnosis showed greater improvement than younger patients. However, the rate of distant metastasis was stable and relatively low, as the 5-year DMFS ranged from 90.5% to 94.7% among the four calendar periods. Conclusion The survival rates in patients with stage II NPC showed increasing trends from 1990 to 2012. The advance of RT provided excellent locoregional control and enhanced OS.
