        Optimizing bioconversion of ferulic acid to vanillin by Bacillus subtilis in the stirred packed reactor using Box-Behnken design and desirability function

        A stirring bioreactor packed with a carbon fiber textiles (FT) biofilm formed by Bacillus subtilis was used to produce vanillin from ferulic acid. Biofilm formation was characterized by scanning electron microscopy. The interactive effects of three variables on vanillin molar yield (M) and conversion efficiency of ferulic acid (E) were evaluated by response surface methodology (RSM) with a Box-Behnken design (BBD). The optimal conversion conditions with a maximum overall desirability D of 0.983 were obtained by a desirability function. Considering the actual operation, the confirmation tests were performed using the slightly modified optimal conditions (initial ferulic acid concentration 1.55 g/ L, temperature 35oC, stirring speed 220 rpm). The results showed that M and E were 57.42 and 93.53%, respectively. This was only 1.03% and 1.87%, respectively, different from the predicted values, confirming the validity of the predicted models. These revealed that the stirred packed reactor could be successfully used in vanillin bioconversion from ferulic acid.

        H-형태 양친매성 펜타블록 공중합체의 화학효소적 합성과 자기회합거동 평가

        H-shaped amphiphilic pentablock copolymers (PSt)2-b-PCL-b-PEO-b-PCL-b-(PSt)2 was synthesized via chemoenzymatic method by combining enzyme-catalyzed ring-opening polymerization (eROP) of ε-caprolactone (ε-CL) and atom transfer radical polymerization (ATRP) of styrene. By this process, we obtained copolymers with controlled molecular weight and low polydispersity. The structure and composition of the obtained copolymers were characterized by nuclear magnetic resonance (NMR), gel permeation chromatography (GPC) and infrared spectroscopy analysis (IR). The crystallization behavior of the copolymers was analyzed by differential scanning calorimetry (DSC) and X-ray diffraction (XRD). The crystallization behavior of the H-shaped block copolymers demonstrated a PCL dominate crystallization. The self-assembly behavior of the copolymers was investigated in aqueous media. The hydrodynamic diameters of the copolymer micelles in aqueous solution were measured by dynamic light scattering (DLS). The morphology of the copolymer micelles was observed by atomic force microscopy (AFM) and transmission electron microscopy (TEM). The hydrodynamic diameters of spherical micelles declined gradually with the increase of the hydrophobic chain lengths of the copolymers. The critical micelle concentration (CMC) values were determined from fluorescence emission, and it was found that the CMCs decreased with an increase of PSt hydrophobic block lengths.

      • Knockdown of HMGN5 Expression by RNA Interference Induces Cell Cycle Arrest in Human Lung Cancer Cells

        HMGN5 is a typical member of the HMGN (high mobility group nucleosome-binding protein) family which may function as a nucleosomal binding and transcriptional activating protein. Overexpression of HMGN5 has been observed in several human tumors but its role in tumorigenesis has not been fully clarified. To investigate its significance for human lung cancer progression, we successfully constructed a shRNA expression lentiviral vector in which sense and antisense sequences targeting the human HMGN5 were linked with a 9-nucleotide loop. Inhibitory effects of siRNA on endogenous HMGN5 gene expression and protein synthesis were demonstrated via real-time RT-PCR and western blotting. We found HMGN5 silencing to significantly inhibit A549 and H1299 cell proliferation assessed by MTT, BrdU incorporation and colony formation assays. Furthermore, flow cytometry analysis showed that specific knockdown of HMGN5 slowed down the cell cycle at the G0/G1 phase and decreased the populations of A549 and H1299 cells at the S and G2/M phases. Taken together, these results suggest that HMGN5 is directly involved in regulation cell proliferation in A549 and H1299 cells by influencing signaling pathways involved in cell cycle progression. Thus, our finding suggests that targeting HMGN5 may be an effective strategy for human lung cancer treatment.

        Coring Damage Extent of Rock Cores Retrieved from High In-situ Stress Condition: A Case Study

        The distribution of damage in deep rock cores is critical for assessing its influence on rock mechanical characteristics. This paper presents a case study on estimating the coring damage extent induced by the stress redistribution through a combined method of numerical simulation and X-ray Computed Tomography (CT) scanning. Rock cores are taken at 1900 m depth from the test tunnel at Jin-ping Second Stage Hydropower Station (JPII) in China. Firstly, the stress path experienced by the rock core during coring has been examined by the numerical simulation, and the distribution of tensile zone in the cross-section of rock core is also discussed. Then two kinds of samples, collected from the same position under different stress levels, are scanned to obtain CT images, and a special CT value analysis strategy was adopted to evaluate coring damage intensities of these samples. The result indicates that the stress state can be regarded as the principal factor for the distribution of coring damage. During coring in the Test Tunnel of JPII under the quasi-hydrostatic stress condition, high tensile stresses (over 5 MPa) are observed at the exterior edge of rock cores, which may lead to the nucleation of microcracks around the core boundary, and the coring damage then propagates to the core centre. The CT scanning also illustrates that the coring damage zone may cover approximately 70~80% of the entire cross-section from the outside inside (stress level of 50 MPa), and that the central part of the core is less damaged or eventually undisturbed. Thus, intact rock samples can be expected to be obtained by overcoring the original rock cores. However, the applicability of the overcoring method seems to be largely dependent on the state of in-situ stress at the coring site. The whole core may be damaged if the lateral stress coefficient reaches a critical value (e.g. greater than 3.0), in which case a special coring equipment should be adopted to improve the stress state during coring.

        Molecular organization of the B mating type locus of a Lentinula edodes monokaryon strain SUP2

        Lentinula edodes is an important cultivated mushroom in China. The development of Lentinula edodes production promotes more studies on it. In our previous work, degenerate PCR and chromosome walking technologies were used to obtain one pheromone receptor gene and one pheromone precursor gene from Lentinula edodes. In this study, four pairs of specific primers were designed according to the whole genome sequencing of the protoplast monokaryon of Lentinula edodes strain 135, to amplify STE3-like pheromone receptor gene and its flanking conserved genes in the protoplast monokaryon strain SUP2 derived from Lentinula edodes strain Suxiang and 33655bp DNA sequence was obtained. By BlastX search, seven putative genes were identified, and three of them are pheromone receptor encoded genes. Furthermore, near to two pheromone receptor genes, four genes encoding proteins with conserved motifs of pheromone precursors were found. This study firstly reveals the molecular organization of the B mating type locus of Lentinula edodes.

        Risk Factors for Anxiety in Major Depressive Disorder Patients

        Objective: To analyze the sociodemographic and clinical factors related to anxiety in patients with major depressive disorder (MDD). Methods: This study involved a secondary analysis of data obtained from the Diagnostic Assessment Service for People with Bipolar Disorders in China (DASP), which was initiated by the Chinese Society of Psychiatry (CSP) and conducted from September 1, 2010 to February 28, 2011. Based on the presence or absence of anxiety-related characteristics, 1,178 MDD patients were classified as suffering from anxious depression (n=915) or non-anxious depression (n=263), respectively. Results: Compared with the non-anxious group, the anxious-depression group had an older age at onset (t=−4.39, p<0.001), were older (t=−4.69, p<0.001), reported more lifetime depressive episodes (z=−3.24, p=0.001), were more likely to experience seasonal depressive episodes (χ2=6.896, p=0.009) and depressive episodes following stressful life events (χ2=59.350, p <0.001), and were more likely to have a family history of psychiatric disorders (χ2=6.091, p=0.014). Their positive and total scores on the Mood Disorder Questionnaire (MDQ) and the 32-item Hypomania Checklist (HCL-32) (p<0.05) were also lower. The logistic regression analysis indicated that age (odds ratio [OR]=1.03, p<0.001), a lower total MDQ score (OR=0.94, p=0.011), depressive episodes following stressful life events (OR=3.04, p<0.001), and seasonal depressive episodes (OR=1.75, p=0.039) were significantly associated with anxious depression. Conclusion: These findings indicate that older age, fewer subclinical bipolar features, an increased number of depressive episodes following stressful life events, and seasonal depressive episodes may be risk factors for anxiety-related characteristics in patients with MDD.

      • Anti-metastasis Activity of Black Rice Anthocyanins Against Breast Cancer: Analyses Using an ErbB2 Positive Breast Cancer Cell Line and Tumoral Xenograft Model

        Background: Increasing evidence from animal, epidemiological and clinical investigations suggest that dietary anthocyanins have potential to prevent chronic diseases, including cancers. It is also noteworthy that human epidermal growth factor receptor 2 (ErbB2) protein overexpression or ErbB2 gene amplification has been included as an indicator for metastasis and higher risk of recurrence for breast cancer. Materials and Methods: The present experiments investigated the anti-metastasis effects of black rice anthocyanins (BRACs) on ErbB2 positive breast cancer cells in vivo and in vitro. Results: Oral administration of BRACs (150 mg/kg/day) reduced transplanted tumor growth, inhibited pulmonary metastasis, and decreased lung tumor nodules in BALB/c nude mice bearing ErbB2 positive breast cancer cell MDA-MB-453 xenografts. The capacity for migration, adhesion, motility and invasion was also inhibited by BRACs in MDA-MB-453 cells in a concentration dependent manner, accompanied by decreased activity of a transfer promoting factor, urokinase-type plasminogen activator (u-PA). Conclusions: Together, our results indicated that BRACs possess anti-metastasis potential against ErbB2 positive human breast cancer cells in vivo and in vitro through inhibition of metastasis promoting molecules.

        Electrophysiological and behavioral responses of Tenebrio molitor L. to fourteen kinds of plant volatiles

        To utilize plant volatiles in themonitoring and biological control of Tenebrio molitor L., electroantennogramography (EAG), Y-tube olfactometry and wind tunnel bioassays were conducted to test the electrophysiological and behavioral responses of T. molitor to nine individual volatiles and five essential oils (compounded volatiles). The aim of this studywas to select effective plant compounds that could be used in the development of semi-chemical based push– pullmethods for the control of this harmful stored product pest. T. molitor displayed strong EAG responses to cis-3- hexenol, isoeugenol, α-pinene, turpentine oil, eucalyptus oil and peppermint essential oil. Y-tube olfactometry assays indicated that peppermint essential oil showed greater luring effects on adult T. molitor than the control. Cis- 3-hexenol elicited strong repellent effects on adult T. molitor, and wind tunnel assays indicated that cis-3-hexenol showed strong repellent effects on adult T. molitor. Peppermint essential oil showed greater luring effects than the control. These results provide a basis to further the development of T. molitor luring and repellent agents.
