      • SEAWEED EXTRACTS AS A POTENTIAL TOOL FOR THE ATTENUATION OF OXIDATIVE DAMAGE IN OBESITY‐RELATED PATHOLOGIES

        <P>Recent studies suggest that seaweed extracts are a significant source of bioactive compounds comparable to the dietary phytochemicals such as onion and tea extracts. The exploration of natural antioxidants that attenuate oxidative damage is important for developing strategies to treat obesity‐related pathologies. The objective of this study was to screen the effects of seaweed extracts of 49 species on adipocyte differentiation and reactive oxygen species (ROS) production during the adipogenesis in 3T3‐L1 adipocytes, and to investigate their total phenol contents and 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) radical scavenging activities. Our results show that high total phenol contents were observed in the extracts of <I>Ecklonia cava</I> (see Table?1 for taxonomic authors) (681.1 ± 16.0 μg gallic acid equivalents [GAE] · g<SUP>−1</SUP>), <I>Dictyopteris undulata</I> (641.3 ± 70.7 μg GAE · g<SUP>−1</SUP>), and <I>Laurencia intermedia</I> (560.9 ± 48.1 μg GAE · g<SUP>−1</SUP>). In addition, DPPH radical scavenging activities were markedly higher in <I>Sargassum macrocarpum</I> (60.2%), <I>Polysiphonia morrowii</I> (55.0%), and <I>Ishige okamurae</I> (52.9%) than those of other seaweed extracts (<I>P </I><<I> </I>0.05). Moreover, treatment with several seaweed extracts including <I>D. undulata</I>, <I>Sargassum micracanthum</I>, <I>Chondrus ocellatus</I>, <I>Gelidium amansii</I>, <I>Gracilaria verrucosa</I>, and <I>Grateloupia lanceolata</I> significantly inhibited adipocyte differentiation and ROS production during differentiation of 3T3‐L1 preadipocytes. Furthermore, the production of ROS was positively correlated with lipid accumulation (<I>R</I><SUP>2</SUP> = 0.8149). According to these preliminary results, some of the seaweed extracts can inhibit ROS generation, which may protect against oxidative stress that is linked to obesity.         Sarcomatoid Carcinoma Arising from Mature Cystic Teratoma

        Malignant transformation of mature cystic teratoma (MCT) is rare. Sarcomatoid carcinoma is a neoplasm comprising malignant mesenchymal cells and a conventional carcinomatous area. Here, we report on a case of sarcomatoid carcinoma arising from an MCT in the left ovary of a 45-year-old female. A unilocular cyst consistent with MCT was observed; however, a nodule within the cyst was confirmed from the resected ovary. Microscopically, the nodule showed both squamous cell carcinoma and pleomorphic sarcomatous components admixing with each other. Lining epithelial cells at the periphery of the main tumor showed squamous metaplasia. When a sarcomatous component is observed in the ovary tumor, it is important to find a squamous cell component, either benign or malignant.

        Development of an ISSR-Derived SCAR Marker in Korean Ginseng Cultivars (Panax ginseng C. A. Meyer)

        Recently, new ginseng cultivars having superior agricultural traits have been developed in Korea. For newly developed plant cultivars, the identification of distinctiveness is very important factors not only in plant cultivar management but also in breeding programs. Thus, eighty-five inter simple sequence repeat (ISSR) primers were applied to detect polymorphisms among six major Korean ginseng cultivars and two foreign ginsengs. A total of 197 polymorphic bands with an average 5.8 polymorphic bands and 2.9 banding patterns per assay unit across six Korean ginseng cultivars and foreign ginsengs from 236 amplified ISSR loci with an average 6.9 loci per assay unit were generated by 34 out of 85 ISSR primers. Three species of Panax ginseng including the Korean ginseng cultivars, P. quinquefolius, and P. notoginseng, could be readily discriminated using most tested primers. UBC-821, UBC-868, and UBC-878 generated polymorphic bands among the six Korean ginseng cultivars, and could distinguish them from foreign ginsengs. Sequence characterized amplified region (SCAR) marker system was introduced in order to increase the reproducibility of the polymorphism. One SCAR marker, PgI821C650, was successfully converted from the randomly amplified polymorphism by UBC-821. It showed the expected dominant polymorphism among ginseng samples. In addition, the specific polymorphism for Sunwon was generated by treating Taq I restriction enzyme to polymerase chain reaction products of PgI821C650. These results will serve as useful DNA markers for identification of Korean ginseng, especially Sunwon cultivar, seed management, and molecular breeding program supplemented with marker-assisted selection.

        Quantum Efficiency in Multi-quantum well InGaN/GaN Light-emitting Diodes with Electroluminescence Characteristics

        The temperature dependence of the electroluminescence spectra of InGaN/GaN multi-quantumwell light-emitting diodes (LEDs) has been investigated over a range of temperatures and currents. The presence of localization effects in the active InGaN layers can be deduced from the observed considerable blue shifts of the electroluminescence emission peak with increasing temperature in the low-temperature region. Exciton-phonon couplings in the active layer are also observed at low temperatures. The electroluminescence intensity was measured to determine the internal quantum efficiency of the LEDs as a function of temperature. When the temperature decreases from room temperature to 170~200 K, the electroluminescence intensity increases due to a reduction in the nonradiative recombination, and the internal quantum efficiency is improved. At T < 170 K, the electroluminescence intensity is reduced significantly. This reduction of the electroluminescence intensity mainly results from the low carrier capture efficiency in the active layer due to the high Mg activation energy, the electron-hole separation in multi quantum wells, and the high internal piezoelectric field. However, in the case of LEDs using a narrow barrier, the electroluminescence intensity is reduced slightly at low temperatures. Therefore, in order to measure the internal quantum efficiency by using the temperature dependence of the electroluminescence spectra in LEDs, one must carefully consider the effects of carrier injection at low temperature.

        Production of Monoclonal Antibody to Avian Infectious Bronchitis Virus

        마사츄셋형 전염성 기관지염 바이러스(IBV)를 SPF 발육란의 뇨막강내에서 증식시켜 Sucrose 밀도구배 초원심분리에 의해 정제한 다음 BALB/c 마우스에 면역시켰다. 면역 마우스에서 채취한 비장세포와 마우스 골수암세포와 여러 차례 융합시험을 실시하였다. 많은 융합세포 중에서 IBV에 특이적으로 작용하는 단클론항체(monoclonal antibody : MCA)를 산생하는 hybridoma클론은 2주밖에 얻지 못했다. 2주의 MCA는 모두 IgG형이었고 IBV중화능이나 혈구응집 억제능이 인정되지 않았다. 간접형광항체법으로 작성된 MCA를 이용하여 인공접종한 닭의 기관도말표본에서 10일간의 시험기간중 계속 IBV를 검출할 수 있었다. Avian infectious bronchitis virus(IBV) was propagated in SPF eggs and purified by sucrose density gradient centrifugation in order to prepare the antigen. Several fusions were made between mouse myeloma cells and spleen cells from BALB/c mouse immunized with IBV antigen and two hybridoma clones producing specific monoclonal antibody(MCA) against the IBV were established. The MCAs were classified as IgG type and revealed no neutralizing and hemagglutination inhibition activity. Using the MCA IBV antigen was detected by IFA method in tracheal smears made from chickens infected with IBV during the experimental period of 10 days.

        Production of Reactive Oxygen Species and Changes in Antioxidant Enzyme Activities during Differentiation of 3T3-L1 Adipocyte

        Obesity, a major public health problem around the world, is a strong risk factor for the development of type 2 diabetes, atherosclerosis, hypertension and cardiovascular diseases. Recent research suggests that increased production of reactive oxygen species (ROS) from accumulated fat in obesity leads to elevated systemic oxidative stress and contributes to the development of obesitylinked chronic disorders. The aim of the current study was to investigate changes of key enzymes associated with antioxidant response and metabolic pathways in 3T3-L1 cell, a preadipocyte cell line that undergoes differentiation into mature adipocytes. The changes in lipid accumulation, ROS production, glucose-6-phosphate dehydrogenase (G6PDH), superoxide dismutase (SOD) and catalase (CAT) during the course of differentiation were determined. The ROS production and G6PDH activity exponentially increased as differentiation progressed. However, CAT activity showed a sharp decrease until day 2, followed by a gradual increase up to day 6 and then leveled off. Given the importance of adipocyte differentiation and ROS production in oxidation-linked diseases, these data provide a biochemical rationale for further studies to identify cellular mechanisms that can counter ROS generation and modulate cellular redox environment in adipocytes.

      • Development of a Pain Measurement Device Using Electrical Stimulation and Pressure: A Pilot Study

        <P>Understanding and precise assessment of pain level are key factors in rehabilitation therapy. Pain is a complex and subjective experience that is affected by an individual’s emotion and health conditions. Various methods have been developed for quantitative evaluation of pain level; however, these methods have several drawbacks. In this work, we developed a pain measurement device for quantitative pain assessment. The system consists of two parts, a component for electrical stimulation and a pressure dolorimeter, for application of two different stresses. Regarding electrical stimulation, the degree of pain is assessed by the applied current. Skin resistance was also analyzed by applying current to remove the effects caused by skin conditions. The electrical stimulation did not induce any histological changes or inflammation in the tissues. Using the pressure dolorimeter, the pain level was assessed according to the degree of inflammation. This system could be used for the quantitative assessment of pain induced by inflammation, wounds, and other factors. Since the described system is the first of its kind, there are many problems that remain to be solved. However, with continuous development, our system could provide more accurate pain assessment by removing skin condition effects and through cross-validation.</P>
