Development and Validation of the Non-Alcoholic Fatty Liver Disease Self-Management Questionnaire

Aims: Non-alcoholic fatty liver disease (NAFLD) is one of common causes of chronic liver disease. The prevalence of NAFLD is increasing with other weight related disease such as obesity, diabetes and metabolic diseases. Self-management is known as a crucial key to improve health outcomes of patients with chronic diseases including NAFLD. However, there is no instrument to measure the level of self-management for patients with NAFLD. This study aims to develop and validate NAFLD self-management questionnaires. Methods: A 23-item of Non-Alcoholic Fatty Liver Disease Self-Management Questionnaire (NAFLD-SMQ) was initially developed after theoretical and literature review by three phases: (1) Items generation; (2) Evaluation of the items; and (3) Psychometric evaluation. Data collection was from April-November in 2019. Participants were 155 individuals recruiting from S hospital in South Korea. The questionnaire was tested by the construct validity using for exploratory factor analysis. Results: Items of the questionnaire were generated based on guideline for patients with NAFLD and the Individual and Family Self-Management Theory. A six-factor was extracted from construct validation using exploratory factor analysis: lifestyle management, drinking management, sleep management, health-supplements management, medical treatment compliance, and family support. These factors accounted for 66.2% of total variation, and have an eigenvalue higher than 1 in the scale. The corrected item-total correlation coefficients ranged 0.33-0.59, and all items were significantly correlated with the total score. Cronbach’s alpha of the total items was 0.87. Conclusions: The NAFLD-SMQ was developed through three phases, and the results of EFA were determined to be valid and reliable. Healthcare providers should assess and evaluate the level of self-management to provide tailored interventions based on their care needs. This instrument would provide useful information for healthcare providers who assess self-management level for individuals with NAFLD. In addition, it may be used as the indicator of health outcomes in this population.
Baculovirus–mediated gene delivery into plant cells

Baculovirus has been widely used for production of numerous recombinant proteins in insect cells. Baculovirus expression has the following advantages: proper post-translational modification, a high capacity for expression of multiple genes or a large insert, biosafety, and high yield driven by the strong promoters polyhedrin or p10. Recently, recombinant proteins have been expressed in both insect cell and mammalian cells by baculovirus system. To date, however, there has been no report of recombinant protein expressed by baculovirus infection in plant. In this study, this expression system was exploited to express GFP (green fluorescence protein). To investigate the baculovirus capability to express the recombinant protein in plant cell, the GFP was cloned under the control of polyhedrin (PPH) promoter and 35S (Cauliflower Mosaic Virus) promoter into the pFastBacTM dual vector, respectively. Gene expression cassetees carrying the GFP genes were transposed into a bacmid in Escherichiacoli (DH10Bac). Bacmid DNA containing GFP gene expression cassette was isolated from transformed DH10Bac using Bac-to-Bac system. The existence of GFP gene in bacmid was confirmed by PCR with GFP specific primers. Both bacmid and baculovirus generated from bacmid-infected Sf9 cells was transfected NicotianaBenthamiana plant leaves, respectively. Confocal immunoflouorescence and fluorescence microscopic analyses confirmed expression of GFP in baculovirus-infected plant leaves and protoplasts isolated from baculovirus–infected plant leaves. In additon, reverse transcription-polymerase chain reaction (RT-PCR) confirmed transcription of GFP gene. In this study, taken together, these results suggest that the baculovirus is able to express recombinant protein gene in plant cells.
Effect of coupled graphene oxide on the sensitivity of surface plasmon resonance detection

<P>We investigated graphene-oxide-(GO-) coupled surface plasmon resonance (SPR) detection sensitivity for sandwiched antigen-antibody interaction between human and antihuman immunoglobulin G molecules. GO was prepared in a Langmuir-Blodgett solution on gold and dielectric surfaces. Theoretical and experimental data suggest that an increased dielectric spacer thickness reduces resonance shifts for GO-coupled SPR detection as dielectric properties of GO appear to prevail. In general, a metal-enhanced structure was shown to provide a larger resonance shift by plasmonic field enhancement. The far-field properties were described in terms of near-field overlap. The peak resonance shift that was obtained with GO-coupled SPR detection was enhanced to 113% of the resonance shift obtained by conventional thin-film-based SPR detection and may further be improved by GO stacking.</P>

When the adenoviral protein E1B55K binds death domain-associated protein (Daxx), the proteasome-dependentdegradation of Daxx is initiated, and adenoviral replication is effectively maintained. Here, we show that the cellularlevels of Daxx differ between human and mouse cancer cell lines. Specifically, we observed higher cellular Daxx levelsand the diminished replication of oncolytic adenovirus in mouse cancer cell lines, suggesting that cellular Daxx levelslimit the replication of oncolytic adenoviruses that lack E1B55K in murine cells. Indeed, the replication of oncolyticadenoviruses that lack E1B55K was significantly increased following infection with oncolytic adenovirus expressingDaxx-specific shRNA. Cellular Daxx levels were decreased in mouse cells expressing heat shock protein 25 (HSP25;homolog of human HSP27) following heat shock or stable transfection with HSP25-bearing plasmids. Furthermore,Daxx expression in murine cell lines was primarily regulated at the transcriptional level via HSP25-mediated inhibitionof the nuclear translocation of the signal transducer and activator of transcription 3 (stat3) protein, which typicallyupregulates Daxx transcription. Conversely, human HSP27 enhanced stat3 activity to increase Daxx transcription. Interestingly, human Daxx, but not mouse Daxx, was degraded as normal by ubiquitin-dependent lysosomaldegradation; however, HSP27 downregulation induced the ubiquitin-independent proteasomal degradation of Daxx.

This paper proposes a solution to the problem of finding a subgraph for a given instance of many terminals on a Euclidean plane. The subgraph is a tree, whose nodes represent the chosen terminals from the problem instance, and whose edges are line segments that connect two corresponding terminals. The tree is required to have the maximum number of nodes while the length is limited and is not sufficient to interconnect all the given terminals. The problem is shown to be NP-hard, and therefore a genetic algorithm is designed as an efficient practical approach. The method is suitable to various probable applications in layout optimization in areas such as communication network construction, industrial construction, and a variety of machine and electronics design problems. The proposed heuristic can be used as a general-purpose practical solver to reduce industrial costs by determining feasible interconnections among many types of components over different types of physical planes.
