Correlations between abnormalities of morphological details and DNA fragmentation in human sperm

Nguyen, Hiep Tuyet Thi,Dang, Hong Nhan Thi,Nguyen, Thai Thanh Thi,Nguyen, Trung Van,Dang, Thuan Cong,Nguyen, Quoc Huy Vu,Le, Minh Tam The Korean Society for Reproductive Medicine 2022 Clinical and Experimental Reproductive Medicine Vol.49 No.1

Objective: As the associations of sperm DNA fragmentation with morphology have not been examined in detail, this study aimed to investigate the relationship between abnormalities of morphological details and DNA integrity in human sperm. Methods: In this cross-sectional study, men from infertile couples were enrolled at Hue Center for Reproductive Endocrinology and Infertility, Vietnam. Conventional semen parameters, including morphological details, were analyzed following the World Health Organization 2010 criteria. Sperm DNA fragmentation was evaluated using a sperm chromatin dispersion assay. The relationships and correlations between semen parameters, sperm morphology, and the type of halosperm and the DNA fragmentation index (DFI) were analyzed. Results: Among 130 men in infertile couples, statistically significant differences were not found in the sperm halo type between the normal and abnormal sperm morphology groups. The percentage of round-head spermatozoa was higher in the DFI >15% group (16.98%±12.50%) than in the DFI ≤15% group (13.13% ±8.82%), higher values for amorphous heads were found in the DFI >15% group, and lower values for tapered heads were observed in the DFI ≤15% group; however, these differences were not statistically significant. Small-halo sperm and the DFI were positively correlated with round-head sperm (r=0.243, p=0.005 and r=0.197, p=0.025, respectively). Conclusion: The rate of general sperm morphological abnormalities in semen analysis was not related to sperm DNA integrity. However, round sperm heads were closely associated with sperm DNA fragmentation.

Fabrication of Cross-linked Nano-Fibrous Chitosan Membranes and Their Biocompatibility Evaluation

        Nguyen, Thi-Hiep,Lee, Seong-Jin,Min, Young-Ki,Lee, Byong-Taek Materials Research Society of Korea 2011 한국재료학회지 Vol.21 No.2

Fibrous chitosan membranes were fabricated as a substrate for skin applications using an electro-spinning process with different solvents and varying concentrations. Scanning electron microscopy (SEM) images confirmed that the formation of the chitosan fibrous membrane in trifluoroacetic acid was better than that in acetic acid. Fourier transform infrared spectroscopy showed that the chitosan fibers were cross-linked with glutaraldehyde, and that the cytotoxicity of the aldehyde groups was reduced by glycine and washing by NaOH and DI water. Chitosan cross-linked fibrous membranes were insoluble in water and could be washed thoroughly to wash away glycine and excess NaOH and prevent the infiltration of other water soluble bio-toxic agents using DI water. MTT assay method was employed to test the cytotoxicity of chitosan membranes during fabricating, treating and washing processes. After the dehydration of cell cultured chitosan membranes, cell attachment behavior on the material was evaluated using SEM method. Effect of the treatment processes on the biocompatibility of the chitosan membranes was shown by comparing of filopodium and lamellipodium of fibroblast cells on grown washed and unwashed chitosan fibrous membrane. The MTT assay and SEM morphology confirmed that the washed chitosan fibrous membrane increased cell attachment and cell growth, and decreased toxicity compared to results for the unwashed chitosan fibrous membrane.

Oral cavity: An open horizon for nanopharmaceuticals

Nguyen Oanh Oanh Thi,Tran Khue Dan,Ha Nhan Thi,Doan Sang Minh,Dinh Thi Thanh Hai,Tran Tuan Hiep 한국약제학회 2021 Journal of Pharmaceutical Investigation Vol.51 No.4

Background Oral administration is the primary route employed for drug delivery. In addition to the gastrointestinal tract, the oral cavity has been drawing considerable attention owing to its physiological properties and advances in drug carrier development. Notably, nanotechnology has made immense contributions to enhancing drug bioavailability, stability, and efficacy, both in research and clinical settings. Area covered The present review comprises lessons from physiological characteristics to fundamental material science knowledge, summarizing the pros and cons of nanotechnology applications in therapeutic drug delivery. The oral cavity presents inherent challenges, including oral cancer, dental caries, oral infection, or oral inflammation, that necessitate local solutions. Simultaneously, the vascular system under the tongue and in the buccal region offers a portal for systemic drug absorption. Expert opinion Among available delivery systems, nanoparticle-based drug delivery, with the oral cavity as the action or absorption site, can be developed as a principal dosage formulation. Although convenience and fast onset might be advantageous, additional studies are warranted for preclinical characterization, especially in animal models, and for determining manufacturing competence, prior to human trials to meet standards in clinical settings.

A hybrid electrospun PU/PCL scaffold satisfied the requirements of blood vessel prosthesis in terms of mechanical properties, pore size, and biocompatibility

Nguyen, Thi-Hiep,Padalhin, Andrew R.,Seo, Hyung Seok,Lee, Byong-Taek Informa UK (TaylorFrancis) 2013 Journal of Biomaterials Science. Polymer Edition Vol.24 No.14

In this study, a novel hybrid polyurethane/polycaprolactone (PU/PCL) tubular scaffold was fabricated using the electrospinning process for blood vessel prosthesis applications. The detailed microstructure and material properties such as porosity, tensile and bust strength, contact angle, and biocompatibility were investigated and compared with those of monolithic PU and PCL scaffolds. The mechanical properties of the hybrid PU/PCL scaffold (tensile strength: 18?MPa, pressure strength: 590?mmHg) were found to be within the range needed for artificial blood vessel applications. The pore sizes of the PU/PCL scaffold ranged from 5-150?um in diameter, are sufficient enough to allow nutrient diffusion across the membrane. The reduced hydrophobic property of the PU/PCL scaffold was the result of the addition of relatively less hydrophobic PU compared with monolithic PCL scaffold. The biocompatibility of the PU/PCL scaffold was evaluated through cytotoxicity testing, and morphological observation by scanning electron microscopy and confocal microscopy using cow pulmonary artery endothelial cells and fibroblast like cells (L929).

Effect of nitrogen doping on the catalytic activity of carbon nano-onions for the oxygen reduction reaction in microbial fuel cells

Thi Hiep Han,Debananda Mohapatra,Neelima Mahato,Smrutiranjan Parida,Jun Ho Shim,Anh Thi Nguyet Nguyen,Van Quang Nguyen,조문환,Jae-Jin Shim 한국공업화학회 2020 Journal of Industrial and Engineering Chemistry Vol.81 No.-

In this study, highly graphitic nitrogen-doped carbon nano-onions (N-CNOs) were prepared by a onestep, direct, in situflame synthesis technique and their potential applications as catalysts for oxygenreduction reaction (ORR) in a microbial fuel cell (MFC) were evaluated for thefirst time. The ORR activityof the CNO, N-CNO, and the commercial Pt/C were measured using a rotating ring-disk electrode (RRDE). The reaction mechanism for the N-CNO was found to follow a four-electron transfer pathway and possessa higher onset potential in RRDE measurement than CNOs. The ORR activity of N-CNO was 5.4 timesbetter than that of CNO, which was attributed to the introduction of nitrogen to the carbon faramework. The MFC fabricated with the N-CNO cathode produced a maximum power density of 49.6 mW m–2, whichwas approximately double the performance of the CNO-based MFC. The performance of N-CNO was lowcompared to Pt/C but the cost per power was only 1/310th. These results confirmed that N-CNOs could beused as a low-cost alternative and an energy-efficient metal-free ORR catalyst for practical MFCapplications.
