        FACILE SOLVOTHERMAL SYNTHESIS OF MESOSTRUCTURED CHITOSAN-COATED Fe 3 O 4 NANOPARTICLES AND ITS FURTHER MODIFICATION WITH FOLIC ACID FOR IMPROVING TARGETED DRUG DELIVERY

        Mesostructured chitosan-coated Fe 3 O 4 nanoparticles (CS-coated Fe 3 O 4 NPs) were synthesizedby a facile one-step solvothermal method via using chitosan as a surface-modi¯cation agent. Subsequently, the surfaces of CS-coated Fe 3 O 4 NPs were successfully conjugated with folic acid(FA) molecules to obtain FA – CS-coated Fe 3 O 4 NPs for improving targeted drug delivery. Themorphology, chemical component and magnetic property of as-prepared composite nanoparticleswere characterized by Fourier transform infrared spectroscopy (FTIR), X-ray di®raction (XRD),dynamic light scattering (DLS), scanning transmission electron microscopy (SEM), transmissionelectron microscopy (TEM), thermal gravimetric analysis (TGA) and vibrating sample magne-tometer (VSM). Furthermore, doxorubicin hydrochloride (DOX) as a model drug was encap-sulated for investigating drug release pattern in vitro. The results show that the magnetizationsaturation value of FA – CS-coated Fe 3 O 4 NPs was about 28.5 emu/g, exhibiting super-paramagnetic properties and mesostructure. DOX could be loaded to FA – CS-coated Fe 3 O 4 NPswith high capacity about 27.9%, and the release rate of DOX could be adjusted by the pH value. This work demonstrates that the prepared magnetic nanoparticles have potential applications inthe treatment of cancer as targeting drug delivery carriers.

        <P>A general synthetic method capable of producing a kind of water-soluble gold nanoparticles possessing single-phase synthetic route, high concentration, small size under control, easy store, and excellent thermal, pH and salt stability, was demonstrated. The combined studies with transmission electron microscopy (TEM), Energy dispersive X-ray spectroscopy (EDS), X-ray photoelectron spectroscopy (XPS) and UV-visible spectra clearly show that the gold nanoparticles modified by thioethyl-pendant ligand exist.</P>

      • Mechanism of HN-3 cell apoptosis induced by carboplatin: Combination of mitochondrial pathway associated with Ca <sup>2+</sup> and the nucleus pathways

        <P>Laryngeal carcinomas have been recognized as a serious health threat worldwide. In the present study, the mechanism of apoptosis in HN-3 cells induced by carboplatin (CBCDA), a widely used anti-cancer drug, was investigated. The pro-apoptotic effect of CBCDA in HN-3 cells was demonstrated to be time- and dose-dependent. Therefore, the stages of apoptosis were investigated in chronological order. The results demonstrated that excessive generation of cytosolic Ca<SUP>2+</SUP> in HN-3 cells was initially triggered when cells were exposed to CBCDA, followed by the appearance of mitochondrial depolarization and oxidative stress, leading to the release of apoptosis-inducing factor. At later stages, expression of caspase-8 was increased due to the apoptotic signals originating from CBCDA-induced DNA damage, as well as caspase-9 and poly ADP ribose polymerase (PARP) expression upregulation. Glutathione decreased the available CBDCA concentration, decreased apoptosis and alleviating oxidative stress, thus reducing the actual effective concentration. Mechanistic research may benefit the rational design of more efficient therapeutic strategies as well as development of novel platinum-based agents.</P>

        This study analyzed for the present situation and characteristics of the community space of overseas cohousing. Based on this, this study aims to inquire into the necessary fundamental needed when designing a community space of future cohousing. As a result, this study could learn that cohousing is an intentional community of private homes clustered around shared space. Households have independent incomes and private lives, but neighbors collaboratively plan and manage community activities and shared spaces. In addition to personal space, the community has shared space, including children"s paly space, rest space, green space, sports space,learning space and public living space. Residents can hold various activities in the community. So these spaces give residents more opportunities to communicate and enrich their community life.

        Structural Equation Modeling to Analyze the Critical Driving Factors and Paths for Off-site Construction in China

        Off-site Construction (OSC) is an alternative to conventional construction. However, China lags behind developed countries interms of its OSC development. The chief goal of this study is to demonstrate the critical driving forces and solutions that promoteOSC development. A total of 21 hypotheses are established to present the interrelations among eight driving forces. Based on 176valid responses from developers in mainland China, a structural equation model is built to explore the key solutions and the criticaldriving forces. Most driving forces, such as Pursuit of Sustainable Competitiveness (PSC), Governmental Policies and Regulations(GPR), Technological Innovation (TI), Corporate Social Responsibility (CSR), and Construction Market Demand (CMD), can affecta Corporation’s Willingness and Behavior (CWB) in adopting OSC through two mediating factors, namely, Economic Benefits (EB)and social and environmental benefits. The findings highlight five main driving solutions to promote OSC, namely, “PSC → EB →CWB,” “PSC → TI → EB → CWB,” “GPR → TI → EB → CWB,” “GPR → CSR → CWB,” and “GPR → CMD → CWB.”Governments and project clients are also identified as important players in the development of OSC in China. These results provide avaluable reference for the government to understand the underlying interrelationships among the driving forces and key solutions tothe development of OSC in China.

        Overexpression of Uridine-Cytidine Kinase 2 Correlates with Breast Cancer Progression and Poor Prognosis

        Purpose: Uridine-cytidine kinase (UCK) 2 is a rate-limiting enzyme involved in the salvage pathway of pyrimidine-nucleotide biosynthesis. Recent studies have shown that UCK2 is overexpressed in many types of cancer and may play a crucial role in activating antitumor prodrugs in human cancer cells. In the current study, we evaluated the potential prognostic value of UCK2 in breast cancer. Methods: We searched public databases to explore associations between UCK2 gene expression and clinical parameters in patients with breast cancer. Gene set enrichment analysis (GSEA) was performed to identify biological pathways associated with UCK2 gene expression levels. Survival analyses were performed using 10 independent large-scale breast cancer microarray datasets. Results: We found that UCK2 mRNA expression was elevated in breast cancer tissue compared with adjacent nontumorous tissue or breast tissue from healthy controls. High UCK2 levels were correlated with estrogen receptor negativity (p<0.001), advanced tumor grade (p<0.001), and poor tumor differentiation (p<0.001). GSEA revealed that UCK2-high breast cancers were enriched for gene sets associated with metastasis, progenitor-like phenotypes, and poor prognosis. Multivariable Cox proportional hazards regression analyses of microarray datasets verified that high UCK2 gene expression was associated with poor overall survival in a dose-response manner. The prognostic power of UCK2 was superior to that of TNM staging and comparable to that of multiple gene signatures. Conclusion: These findings suggest that UCK2 may be a promising prognostic biomarker for patients with breast cancer.

        The space environment of kindergarten is an important space for children to learn, play, live, grow and develop. It is the external condition of children`s spiritual growth and plays a potential education function. With the development of society and culture, the space design of kindergarten should also strive to meet the emotional and ideological needs of children. The purpose of this dissertation is to solve the problem of backward teaching environment in most Chinese kindergartens in China, to create a suitable space for children to grow, to meet the needs of children`s emotional and ideological needs. Research on the performance characteristics of emotional design and other content and the existing national kindergarten spatial design case analysis for the future design of kindergartens in China to bring help. The case study of this thesis is about three new public demonstration kindergartens in Shanghai. Analyze the performance characteristics of perceptual design in the study space and the activity space and make a qestrionnaire survey and analysis of the emotional design of kindergarten.

        Circular RNA expression profiles in the porcine liver of two distinct phenotype pig breeds

        Objective: An experiment was conducted to identify and characterize the circular RNA expression and metabolic characteristics in the liver of Jinhua pigs and Landrace pigs. Methods: Three Jinhua pigs and three Landrace pigs respectively at 70-day were slaughtered to collect the liver tissue samples. Immediately after slaughter, blood samples were taken to detect serum biochemical indicators. Total RNA extracted from liver tissue samples were used to prepare the library and then sequence on HiSeq 2500. Bioinformatic methods were employed to analyze sequence data to identify the circRNAs and predict the potential roles of differentially expressed circRNAs between the two breeds. Results: Significant differences in physiological and biochemical traits were observed between growing Jinhua and Landrace pigs. We identified 84,864 circRNA candidates in two breeds and 366 circRNAs were detected as significantly differentially expressed. Their host genes are involved in lipid biosynthetic and metabolic processes according to the gene ontology analysis and associated with metabolic pathways. Conclusion: Our research represents the first description of circRNA profiles in the porcine liver from two divergent phenotype pigs. The predicted miRNA-circRNA interaction provides important basis for miRNA-circRNA relationships in the porcine liver. These data expand the repertories of porcine circRNA and are conducive to understanding the possible molecular mechanisms involved in miRNA and circRNA. Our study provides basic data for further research of the biological functions of circRNAs in the porcine liver.

        A putative pathogen-resistant regulatory pathway between MicroRNAs and candidate target genes in maize

        MicroRNAs (miRNAs) are a family of small non-coding RNAs that, in most cases, negatively regulate gene expression at the post-transcriptional level. Plant miRNAs have been implicated in developmental processes and adaptation to environmental stress including biotic and abiotic stresses. Here, we report a comprehensive analysis of miRNAs and associated target genes under banded leaf and sheath blight (BLSB) stress caused by R. solani in maize. Eight differentially expressed miRNAs were randomly selected from deep sequencing results and validated by qRT-PCR together with their putative target genes, most of which are transcription factors as well as metabolic genes involved in auxin signaling. The results revealed that majorities of the analyzed miRNAs show an inverse correlation with their corresponding predicted target genes. In addition, a putative regulatory network of miRNAs-mRNAs responsive to R. solani was constructed. This study provides insight into the regulatory functions of miRNAs, thereby expanding our knowledge of the molecular mechanisms of pathogen resistance.
