        Phenolic Content and Antioxidant and Antiacetylcholinesterase Properties of Honeys from Different Floral Origins

        Twenty-three honey samples of Apis mellifera L. forged on plants from northeastern Brazil were analyzed to determine total phenolic content, flavonoid content, antioxidant activity, and antiacetylcholinesterase activity. The total phenol content was determined by using the Folin–Ciocalteu method, and the flavonoid content was analyzed using by the aluminum chloride method. The antioxidant activity was evaluated using the diphenyl-1-picrylhydrazyl–scavenging test. Honey samples from Lippia sidoides Cham. (mean [±standard deviation] 50% inhibitory concentration [IC_50], 4.20±1.07 mg/mL) and Myracrodruon urundeuva Fr. All. (IC_50, 28.27±1.41 mg/mL) showed better antioxidant activity and presented higher total phenol values (108.50±3.52 mg gallic acid equivalents/100 g for L. sidoides and 68.55±1.01 mg gallic acid equivalents/100 g for M. urundeuva). Several honey samples had relevant results on antiacetylcholinesterase assay. The biological activity of honeys is related to their floral origin, and medicinal plants constitute a useful resource for the generation of functional foods.

        Influence of category−heifers, primiparous and multiparous lactating cows−in a large-scale resynchronization fixed-time artificial insemination program

        This study was conducted to evaluate the influence of category (heifers, primiparous or multiparous cows) on pregnancy rates in a large scaleresynchronization ovulation program. Nelore heifers (n = 903), primiparous lactating cows (n = 338) and multiparous lactating cows (n =1,223) were synchronized using a conventional protocol of estradiol/P4-based fixed-time artificial insemination (FTAI). Thirty days afterultrasonography, females who failed the first FTAI were resynchronized with the same hormonal protocol prior to a second FTAI. Thepregnancy status of each cohort was evaluated by ultrasonography 30 days after each FTAI. The average conception rate after the first FTAIand resynchronization was 80.5%. Heifers had a higher conception rate (85%) than primiparous (76%) or multiparous cows (78%; p = 0.0001). The conception rate after the first FTAI was similar among heifers (57%), primiparous cows (51%) and multiparous cows (56%; p = 0.193). After the second FTAI, heifers exhibited a higher conception rate (66%) than primiparous or multiparous cows (51%; p = 0.0001). These resultsdemonstrate the feasibility of resynchronization in large beef herds for providing consistent pregnancy rates in a short period of time. We alsodemonstrated that ovulation resynchronization 30 days after FTAI is particularly effective for heifers, providing a conception rate of up to66%.

        Effects of Feeding Licury (Syagrus coronate) Cake to Growing Goats

        The objectives of this study were to determine the highest inclusion of licury (Syagrus coronate) cake in the diet of growing Boer goats without adverse effects on intake and digestibility and to determine its effects on ingestive behavior and physiological responses. Twenty entire, one year old 3/4 Boer goats, 18.1 kg (DS = 2.2) average body weight (BW), were allocated to dietary treatments in a completely randomized design. Each animal was confined in a $1.0\;m^2$ pen with a suspended floor and given ad libitum access to clean, fresh water. Diets were formulated to meet NRC (2007) requirements and the ingredients were: 50% of Tifton-85 (Cynodon sp.) hay, corn meal, soybean meal, mineral and vitamin premix, and licury cake. The treatments were: i) no addition of licury cake to the diet, ii) 15% (DM basis) addition of licury cake, iii) 30% licury cake and, iv) 45% licury cake. The experiment lasted for 17 days; the first 10 days were used to adapt the animals to the diets and facilities. The inclusion of licury cake increased the fiber concentration of the diets; however, there was no effect on either dry matter (DM) or organic matter (OM) intake. There was a linear increase (p<0.05) in the EE content of the diet as the addition of licury cake increased; however, EE intake did not differ (p>0.05) between treatments. The digestibility of non-fibrous carbohydrates (NFC) decreased with increasing inclusion of licury cake, as did NFC intake. The efficiency of ingestion of DM and NDF presented a negative quadratic effect with the inclusion of licury cake. Results from this study indicate that licury cake can be fed to goats at up to 45% of the diet without adverse effects on either intake or digestibility.

        Effects of surrounding and underlying shades on the color adjustment potential of a single-shade composite used in a thin layer

        Objectives This study aimed to evaluate the surrounding and underlying shades’ effect on the color adjustment potential (CAP) of a single-shade composite used in a thin layer. Materials and Methods Cylinder specimens (1.0 mm thick) were built with the Vittra APS Unique composite, surrounded (dual specimens) or not (simple specimens) by a control composite (shade A1, A2, or A3). Simple specimens were also built only with the control composites. Each specimen’s color was measured against white and black backgrounds or the simple control specimens with a spectrophotometer (CIELAB system). The whiteness index for dentistry (WID) and translucency parameters (TP00) were calculated for simple specimens. Differences (ΔE00) in color between the simple/dual specimens and the controls were calculated. The CAP was calculated based on the ratios between data from simple and dual specimens. Results The Vittra APS Unique composite showed higher WID and TP00 values than the controls. The highest values of ΔE00 were observed among simple specimens. The color measurements of Vittra APS Unique (simple or dual) against the control specimens presented the lowest color differences. Only surrounding the single-shade composite with a shaded composite barely impacted the ΔE00. The highest CAP values were obtained using a shaded composite under simple or dual specimens. Conclusions The CAP of Vittra APS Unique was strongly affected by the underlying shade, while surrounding this composite with a shaded one barely affected its color adjustment.

        Effects of dietary L-glutamine and glutamic acid combination, and whey powder on the performance and nutrient digestion in weaned piglets fed grain-based diets

        Objective: The present study aimed to evaluate the influence of including L-glutamine along with glutamic acid as a supplement in weaned piglets’ diets with and without whey powder. Methods: Two assays were carried out. A total of 40 piglets ([Landrace×Large White]× Pietrain) weaned at 24 days of age with an initial body weight of 6.6±0.6 kg were used in the first assay, and the following parameters were evaluated: growth performance, the incidence of diarrhea, morphometry, intestinal integrity, and hepatic glycogen index. The animals were then blocked into four groups according to different diets: diet all-grain feeding (G); diet all-grain feeding with whey powder (GW); and with vs without 1% supplementation of the commercial product containing L-glutamine and glutamic acid (A or NA). Whey powder was added according to the stage of life, corresponding to 17%, 10%, and 5%, respectively, in order to meet the need for lactose. The animals were evaluated at 24 to 42 days and at 24 to 55 days of age. The nutrient digestibility for the second assay was carried out by using 24 animals with an average weight of 11.49±1.6 kg, and the same diets were tested. Results: The supplementation of L-glutamine + glutamic acid or the addition of whey powder in diets for weaned piglets provided (p<0.05) greater feed intake, greater weight gain and improved feed conversion in the initial period (24 to 42 days age). However, in the whole period (24 to 55 days age) only amino acid supplementation affected (p<0.05) growth performance. There was a positive interaction (p<0.05) between the type of diet and L-glutamine + glutamic acid supplementation on villus height, crypt depth and the villus:crypt ratio in the duodenum. In addition, L-glutamine + glutamic acid supplementation reduced (p<0.05) the crypt depth and improved the villus:crypt ratio in the jejunum. The inclusion of whey powder affected (p<0.05) positively the digestibility coefficients analyzed except mineral matter digestibility coeficients. The supplementation of 1% the commercial product composed of L-glutamine and glutamic acid improved (p<0.05) only the digestibility coefficient of crude protein. Conclusion: These results indicate that supplementation of 1% commercial product containing L-glutamine + glutamic acid in diets for piglets from 24 to 55 days of age, dispenses with the use of whey powder when evaluating growth performance. Amino acid supplementation alone or associated with whey powder affects (p<0.05) positively the indicators of the intestinal integrity.         Ferroelectric switching behavior of pulsed laser deposited Ba0.8Sr0.2TiO3 thin films

        The effect of pulse amplitude on the ferroelectric and switching properties of pulsed laser deposited Ba0.8Sr0.2TiO3 thin films has been studied. The structural and morphological analysis revealed that the films had a well crystallized perovskite phase and grain size of about 30e40 nm. A well saturated PeE hysteresis loop was observed with a remnant polarization (Pr)z4.8 mC/cm2 and a coercive fieldz100 kV/cm at a frequency of 1 kHz. The Pr has been found to be decreased only 4.3% after passing 8.0 × 108 cycles. The analysis of switching response with nucleation limited switching model reveals that characteristic switching time (t0) variance is due to the random distribution of the local electric fields. The peak value of polarization current and t0 exhibits exponential dependence on reciprocal of pulse amplitude.         Proliferative Effect of Food Plant Species from Brazil's Cerrado on Cultured Fibroblasts

        Brazilian Cerrado is the second largest biome in South America and contains many unstudied valuable plant species rich in bioactive substances. In this study we investigated the phenolic content and proliferative effects on cultured fibroblasts of 32 extracts of different polarities prepared from 11 plants found in Cerrado regions. Eight extracts from six species increased cell proliferation and significantly induced ATP production by the cells. Four of these extracts were obtained from plants used as food, specifically from its fruits or seeds. A high phenolic content for these eight extracts, which directly correlated with the induction of cell proliferation, was corroborated by mass spectrometry analysis. We suggest that the bioactive substance content of these species shows an interesting potential use in cosmetic and food industry, which can contribute to the conservation and sustainable development of this region.

      • Identification of Novel Compounds Inhibiting Chikungunya Virus-Induced Cell Death by High Throughput Screening of a Kinase Inhibitor Library

        <▼1><P>Chikungunya virus (CHIKV) is a mosquito-borne arthrogenic alphavirus that causes acute febrile illness in humans accompanied by joint pains and in many cases, persistent arthralgia lasting weeks to years. The re-emergence of CHIKV has resulted in numerous outbreaks in the eastern hemisphere, and threatens to expand in the foreseeable future. Unfortunately, no effective treatment is currently available. The present study reports the use of resazurin in a cell-based high-throughput assay, and an image-based high-content assay to identify and characterize inhibitors of CHIKV-infection in vitro. CHIKV is a highly cytopathic virus that rapidly kills infected cells. Thus, cell viability of HuH-7 cells infected with CHIKV in the presence of compounds was determined by measuring metabolic reduction of resazurin to identify inhibitors of CHIKV-associated cell death. A kinase inhibitor library of 4,000 compounds was screened against CHIKV infection of HuH-7 cells using the resazurin reduction assay, and the cell toxicity was also measured in non-infected cells. Seventy-two compounds showing ≥50% inhibition property against CHIKV at 10 µM were selected as primary hits. Four compounds having a benzofuran core scaffold (CND0335, CND0364, CND0366 and CND0415), one pyrrolopyridine (CND0545) and one thiazol-carboxamide (CND3514) inhibited CHIKV-associated cell death in a dose-dependent manner, with EC<SUB>50</SUB> values between 2.2 µM and 7.1 µM. Based on image analysis, these 6 hit compounds did not inhibit CHIKV replication in the host cell. However, CHIKV-infected cells manifested less prominent apoptotic blebs typical of CHIKV cytopathic effect compared with the control infection. Moreover, treatment with these compounds reduced viral titers in the medium of CHIKV-infected cells by up to 100-fold. In conclusion, this cell-based high-throughput screening assay using resazurin, combined with the image-based high content assay approach identified compounds against CHIKV having a novel antiviral activity - inhibition of virus-induced CPE - likely by targeting kinases involved in apoptosis.</P></▼1><▼2><P><B>Author Summary</B></P><P>Recent outbreaks and expanding global distribution of Chikungunya virus (CHIKV) in different regions of Asia, Africa and Europe necessitates the development of effective therapeutic interventions. At present, only two antiviral compounds (chloroquine and ribavirin) that inhibit viral infection <I>in vitro</I> have been used in clinical cases of chikungunya infections. However, neither of these compounds have shown strong efficacy in vivo. Recent attempts to identify new antiviral candidates for CHIKV using cell-based phenotypic approach have been reported. In this study, we developed a simple cell-based high-throughput assay using resazurin to identify potential anti-CHIKV compounds. This high-throughput assay is based on the metabolic reduction of resazurin to the highly fluorescent resorufin by viable cells as an indicator of activity against CHIKV-induced CPE. We screened 4,000 small molecules belonging to the BioFocus kinase inhibitor chemical library and found a cluster of related molecules with antiviral activity against CHIKV. Finally, we characterized the putative mode of action of these active compounds using an image-based high content assay and conventional virological methods (<I>i.e.</I>, virus yield reduction assay, microneutralization assay).</P></▼2>

        Polyphasic delimitation of a filamentous marine genus, Capillus gen. nov. (Cyanobacteria, Oscillatoriaceae) with the description of two Brazilian species

        Lyngbya C. Agardh ex Gomont is a nonheterocytous cyanobacterial genus whose evolutionary history is still poorly known. The traditionally defined Lyngbya has been demonstrated to be polyphyletic, including at least five distinct clades, some of which have been proposed as new genera. Intraspecific diversity is also clearly underestimated in Lyngbya due to the lack of unique morphological characters to differentiate species. In this study, we describe the new genus Capillus T. A. Caires, C. L. Sant'Anna et J. M. C. Nunes from benthic marine environments, including two new Brazilian species (here described as C. salinus T. A. Caires, C. L. Sant'Anna et J. M. C. Nunes, and C. tropicalis T. A. Caires, C. L. Sant'Anna et J. M. C. Nunes), and two species yet to be described, one of them from India (Capillus sp. 2.1), and the other from United States of America, based on strain PCC 7419. Capillus species presented cross-wise diagonal fragmentation, assisted or not by necridic cells, which has not been previously mentioned for Lyngbya. Ultrastructural analyses showed that C. salinus and C. tropicalis have numerous gas vesicles, which are rarely described for benthic marine species. The new genus formed a well-supported clade, and the D1-D1' and Box B secondary structures of internal transcribed spacer also supported the proposal of its new species. These findings help to clarify the diversity of species in the Lyngbya complex and the taxonomy of the group, and highlight the need of further floristic surveys in tropical coastal environments, which remain poorly studied.
