        WDRP, a DWD protein component of CUL4-based E3 ligases, acts as a receptor of CDPK-related protein kinase 5 to mediate kinase degradation in Arabidopsis

        CRK5 is a member of the Arabidopsis thaliana Ca2+-dependent protein kinase-related kinase family. Here, a yeast two-hybrid screen was performed with a truncated form of AtCRK5 as bait to identify interacting proteins and determine its physiological roles. One gene encoding the DWD protein WDRP was isolated. Furthermore, in vitro and in vivo co-immunoprecipitation results strongly supported that these two proteins interact with each other. Using a cell-free degradation assay, we also established that CRK5 was an unstable protein that was degraded through the proteasome pathway. The rate of CRK5 degradation was delayed in a WDRP knockout line. On the other hand, the degradation of CRK5 mediated by WDRP might not affect the phosphorylation of PIN2 by CRK5. Overall, we demonstrated that AtCRK5 interacted with a DWD protein, AtWDRP; the protein AtWDRP targets the kinase for ubiquitin-dependent degradation. Therefore, this report describes a new kinase regulation pathway for CRK family proteins in Arabidopsis.

      • RTN4 3'-UTR Insertion/Deletion Polymorphism and Susceptibility to Non-Small Cell Lung Cancer in Chinese Han Population

        Nogo protein, encoded by gene reticulon-4 (RTN4), includes three major isoforms by different splicing, named Nogo-A Nogo-B and Nogo-C. Nogo proteins play an important role in the apoptosis of cells, especially in tumor cells. RTN4 single nucleotide polymorphisms (SNPs) can influence the efficiency of transcription and translation thus being related with an individual's predisposition to cancer. The CAA insertion/deletion polymorphism (rs34917480) within RTN4 3'-UTR has been reported to be associated with many cancer types. In order to investigate the relationship between this polymorphism and susceptibility to non-small cell lung cancer (NSCLC) in the Chinese population, we conducted the present case-control study including 411 NSCLC patients and 471 unrelated healthy controls. The genotype distributions were significantly different between cases and controls (p=0.014). We found that the del allele could significantly increase NSCLC risk (ins/ins vs ins/del: p=0.007, OR 1.46, 95%CI=1.11-1.93; dominant model: p=0.004, OR 1.47, 95%CI=1.13-1.92 and allele model: p=0.008, OR 1.35, 95%CI=1.08-1.67). This association was stronger in participants over 60 years old, males and smokers. We therefore conclude that the CAA insertion/deletion polymorphism (rs34917480) contributes to non-small cell lung cancer risk in Chinese population. Age, sex and environmental exposure are also related to carcinogenic effects of rs34917480.

        Evaluation of Anterior Ethmoidal Artery by 320-Slice CT Angiography with Comparison to Three-Dimensional Spin Digital Subtraction Angiography: Initial Experiences

        Objective: To explore the usefulness of 320-slice CT angiography (CTA) for evaluating the course of the anterior ethmoidal artery (AEA) and its relationship with adjacent structures by using three-dimensional (3D) spin digital subtraction angiography (DSA) as standard reference. Materials and Methods: From December 2008 to December 2010, 32 patients with cerebrovascular disease, who underwent both cranial 3D spin DSA and 320-slice CTA within a 30 day period from each other, were retrospectively reviewed. AEA course in ethmoid was analyzed in DSA and CTA. In addition, adjacent bony landmarks (bony notch in medial orbital wall, anterior ethmoidal canal, and anterior ethmoidal sulcus) were evaluated with CTA using the MPR technique oriented along the axial, coronal and oblique coronal planes in all patients. The dose length product (DLP) for CTA and the dose-area product (DAP) for 3D spin DSA were recorded. Effective dose (ED) was calculated. Results: The entire course of the AEA was seen in all 32 cases (100%) with 3D spine DSA and in 29 of 32 cases (90.1%) with 320-slice CTA, with no significant difference (p = 0.24). In three cases where AEA was not visualized on 320-slice CTA, two were due to the dominant posterior ethmoidal artery, while the remaining case was due to diminutive AEA. On MPR images of 320-slice CT, a bony notch in the orbital medial walls was detected in all cases (100%, 64 of 64); anterior ethmoidal canal was seen in 28 of 64 cases (43.8%), and the anterior ethmoidal sulcus was seen in 63 of 64 cases (98.4%). The mean effective dose in CTA was 0.6 ± 0.25 mSv, which was significantly lower than for 3D spin DSA (1.3 ± 0.01 mSv) (p < 0.001). Conclusion: 320-slice CTA has a similar detection rate for AEA to that of 3D spin DSA; however, it is noninvasive, and may be preferentially used for the evaluation of AEA and its adjacent bony variations and pathologic changes in preoperative patients with paranasal sinus diseases.       • Lung Cancer in a Rural Area of China: Rapid Rise in Incidence and Poor Improvement in Survival

        Background: Lung cancer has been a major health problem in developed countries for several decades, and has emerged recently as the leading cause of cancer death in many developing countries. The incidence of lung cancer appears to be increasing more rapidly in rural than in urban areas of China. This paper presents the trends of lung cancer incidence and survival derived from a 40-year population-based cancer monitoring program in a rural area, Qidong, China. Materials and Methods: The Qidong cancer registration data of 1972-2011 were used to calculate the crude rate, age-standardized rate by Chinese population (CASR) and by world population (WASR), birth cohort rates, and other descriptive features. Active and passive methods were used to construct the data set, with a deadline of the latest follow-up of April 30, 2012. Results: The total number of lung cancer cases was 15,340, accounting for 16.5% of all sites combined. The crude incidence rate, CASR and WASR of this cancer were 34.1, 15.7 and 25.4 per 100,000, respectively. Males had higher crude rates than females (49.7 vs 19.0). Rapidly increasing trends were found in annual percent change resulting in lung cancer being a number one cancer site after year 2010 in Qidong. Birth cohort analysis showed incidence rates have increased for all age groups over 24 years old. The 5 year observed survival rates were 3.55% in 1973-1977, 3.92 in 1983-1987, 3.69% in 1993-1997, and 6.32% in 2003-2007. Males experienced poorer survival than did females. Conclusions: Lung cancer has become a major cancer-related health problem in this rural area. The rapid increases in incidence likely result from an increased cigarette smoking rate and evolving environmental risk factors. Lung cancer survival, while showing some improvement in prognosis, still remains well below that observed in the developed areas of the world.

      • A Multi-attribute Keyword Retrieval Mechanism for Encrypted Cloud Data

        With the development of cloud computing technology, more and more users have outsourced their data to cloud platform. To protect the safety of these data, some encryption methods are usually used. But it is very difficult question that how to effective retrieval and to use the encrypted data, we propose a multi-attribute keyword retrieval mechanism for encrypted cloud data. In this mechanism, we first define the local feature attributes of the keywords and the global feature attributes of the document. Then, we construct the mapping relationship between keywords and document attributes according to the basic principle of inverted index algorithm and establish the security index. Based on the above steps, we improve the weight of the key words and calculate the similarity score of the document by combining the multi-attribute score function. Experiments show that this mechanism not only can effectively improve the accuracy of the data retrieval, but also can greatly reduce the bandwidth consumption of system.

        Cathodic Reduction of Cu2+ and Electric Power Generation Using a Microbial Fuel Cell

        When Cu2+ was used as an electron acceptor, removal of Cu2+ was achieved from the synthesized wastewater (SW)in the cathode compartment of a microbial fuel cell (MFC). By addition of KNO3, the different initial pH of the SW showed no effect on the removal efficiency of Cu2+. For Cu2+ concentration of 50 mg/L the removal efficiencies were found to be 99.82%, 99.95%, 99.58%, and 99.97% for the KNO3 concentrations of 0, 50, 100 and 200 mM, and to be 99.4%, 99.9%, 99.7%, and 99.7% for pH values of 2, 3, 4, and 5, respectively. More than 99% Cu2+ was removed for the Cu2+ concentrations of 10, 50, and 100 mg/L, while only 60.1% of Cu2+ was removed for the initial concentration of 200 mg/L (pH 3). The maximum power density was affected by both KNO3 concentration and initial concentration of Cu2+. It was increased by a factor of 1.5 (from 96.2 to 143.6 mW/m2) when the KNO3 concentration was increased from 0 to 200 mM (50 mg/L Cu2+), and by a factor of 2.7 (from 118 to 319 mW/m2) when Cu2+concentration was increased from 10 to 200 mg/L (pH 3).

        Ginsenoside Rh2 reduces depression in offspring of mice with maternal toxoplasma infection during pregnancy by inhibiting microglial activation via the HMGB1/TLR4/NF-kB signaling pathway

        Background: Maternal Toxoplasma gondii (T. gondii) infection during pregnancy has been associated with various mental illnesses in the offspring. Ginsenoside Rh2 (GRh2) is a major bioactive compound obtained from ginseng that has an anti-T. gondii effect and attenuates microglial activation through toll-like receptor 4 (TLR4)/nuclear factor-kappa B (NF-kB) signaling pathway. GRh2 also alleviated tumor-associated or lipopolysaccharide-induced depression. However, the effects and potential mechanisms of GRh2 on depression-like behavior in mouse offspring caused by maternal T. gondii infection during pregnancy have not been investigated. Methods: We examined GRh2 effects on the depression-like behavior in mouse offspring, caused by maternal T. gondii infection during pregnancy, by measuring depression-like behaviors and assaying parameters at the neuronal and molecular level. Results: We showed that GRh2 significantly improved behavioral measures: sucrose consumption, forced swim time and tail suspended immobility time of their offspring. These corresponded with increased tissue concentrations of 5-hydroxytryptamine and dopamine, and attenuated indoleamine 2,3-dioxygenase or enhanced tyrosine hydroxylase expression in the prefrontal cortex. GRh2 ameliorated neuronal damage in the prefrontal cortex. Molecular docking results revealed that GRh2 binds strongly to both TLR4 and high mobility group box 1 (HMGB1). Conclusion: This study demonstrated that GRh2 ameliorated the depression-like behavior in mouse offspring of maternal T. gondii infection during pregnancy by attenuating the excessive activation of microglia and neuroinflammation through the HMGB1/TLR4/NF-kB signaling pathway. It suggests that GRh2 could be considered a potential therapy in preventing and treating psychiatric disorders in the offspring mice of mothers with prenatal exposure to T. gondii infection.

        Four isostructural amino-acid-based polyoxomolybdates, {$M(H_2O)_3(pro)Mo_4O_{13}$}$_2{\cdot}2H_2O$ (pro = proline, M = Co (1), Ni (2), Cu (3), Zn (4)), have been synthesized and structurally characterized by single crystal X-ray diffraction, elemental analysis, IR spectrum, TG analysis. The structures of 1-4 are layered networks built up from {$Mo_8O_{26}(pro)_2$}$^{4-}$ units and {$M(H_2O)_3O_3$} octahedra, the uncoordinated water molecules occupying the interlayer regions.
