        Comparative transcript profiling and cytological observation of the newly bred recessive genic male sterility non‑heading Chinese cabbage (Brassica rapa ssp. chinensis) line WS24‑3A

        Background WS24-3A is a newly bred non-heading Chinese cabbage genic male-sterile line, in which sterility is controlled by a recessive gene, designated as Bra2ms. WS24-3A has been used for hybrid breeding. Objective To reveal the underlying molecular mechanisms responsible for the sterility of WS24-3A. Methods Cytological observation of the process of sterile/fertile anther development was performed to determine the tissue and stage in which sterility occurs. Phenotyping and transcriptomic analyses were performed to identify differentially expressed genes (DEGs) between sterile and fertile flower buds at different stages. Results Cytological analysis revealed no tetrads at stage 7 or at later stages of anther development, and the degradation of callose was delayed. Abnormal meiocytes were surrounded by sustaining callose that degenerated gradually in WS24-3A. Comparative transcript profiling identified 3282 DEGs during three anther developmental stages, namely, pre-meiotic anther, meiotic anther, and anthers with single-celled pollen stage. The difference in DEG percentage between up-regulated and down-regulated at meiotic anther stage was obviously larger than at the other two stages; further, most DEGs are important for male meiosis, callose synthesis and dissolution, and tapetum development. Ten DEGs were found to be involved in anther and pollen development, which were analyzed by quantitative PCR. Conclusion Bra2ms affected gene expression in meiocytes and associated with callose synthesis, degradation and tapetum development. Our results provide clues to elucidate the molecular mechanism of genic male sterility in non-heading Chinese cabbage.

        A Method for k Nearest Neighbor Query of Line Segment in Obstructed Spaces

        In order to make up the deficiencies of the existing research results which cannot effectively deal with the nearest neighbor query based on the line segments in obstacle space, the k nearest neighbor query method of line segment in obstacle space is proposed and the STA_OLkNN algorithm under the circumstance of static obstacle data set is put forward. The query process is divided into two stages, including the filtering process and refining process. In the filtration process, according to the properties of the line segment Voronoi diagram, the corresponding pruning rules are proposed and the filtering algorithm is presented. In the refining process, according to the relationship of the position between the line segments, the corresponding distance expression method is put forward and the final result is obtained by comparing the distance. Theoretical research and experimental results show that the proposed algorithm can effectively deal with the problem of k nearest neighbor query of the line segment in the obstacle environment.

        Modeling and vibration characteristics analysis of a DMF rotor system

        To analyze the vibration response of a rotor system with circumferential short spring dual mass flywheel (DMF) when the primary flywheel speed changes, the interactions (forces and torques) between the primary flywheel, spring seat, spring, and secondary flywheel are analyzed in detail, and the dynamic analysis model of the DMF rotor system is established considering the influence of clearance and friction between parts in the DMF in this study. The vibration response of the DMF is investigated by numerical method. By analyzing the bifurcation diagram, time history, phase trajectories, Poincaré section, and frequency domain of the relative angular displacement, the variation of vibration form of the system under different excitation frequencies are discussed. Besides, the effects of load, primary flywheel speed amplitude, and spring stiffness on system vibration are also analyzed. Finally, some of the results from the analytical study are verified through the DMF rotor system experiments.

        Fast modulation strategy for open-end winding PMSM with common DC bus

        An open-end winding PMSM drive system with a common DC bus can produce zero-sequence current and high switching loss, which reduce the performance and efficiency of the system. Aiming at this problem, this paper proposes a fast modulation strategy that can effectively suppress the zero-sequence current, reduce the number of switching times in a sampling period to 6, and reduce the average switching loss by 50%. In addition, when compared with the conventional CBPWM and SVPWM algorithms, this strategy does not need to decompose the modulation wave, or synthesize the voltage vector. Furthermore, the calculation burden is small, which makes it very easy to implement. Experimental results verify the effectiveness and feasibility of this strategy.

      • Research on the Dynamic Complex Spatial Network Relations in Spatial Database

        Representation and analysis of the complex spatial network relations are of great significance in fields such as Geographic Information System,spatial database, spatial data mining and intelligent inference. As the existing research achievements cannot be used to deal with the dynamic complex spatial network relations effectively in reality, in order to make up for the deficiency of existing methods, the dynamic complex spatial network relations is studied in detail. The predicate representation methods of the strict and extended complex spatial network relations are proposed respectively. The dynamic logical hierarchical relationship and dynamic migration relationship of the strict spatial network relation are given. Furthermore, this paper lay emphasis on the study of the dynamic conversion and adjacent relevance of the extended complex spatial network, and some instance analyses are provided. The research achievements in this paper is rather suitable for dealing with issues about the representation, error correction and forecast of the dynamic complex spatial network relations, therefore, the ability of spatial database to deal with spatial relations of complex spatial object has been enhanced.

        Model Predictive Control based Discontinuous PWM Algorithm for 3L-NPC Inverter

        This paper proposes a novel model predictive control based discontinuous PWM strategy for 3L-NPC inverter. Through the model predictive control principle and special output level sequence arrangement, the optimal voltage vector and zero sequence voltage are selected, so that the neutral point voltage can be controlled on the basis of discontinuous modulation strategy, thus achieve the purpose of balancing the neutral point voltage and reducing the switching loss. Because there are most three elements in the control set, the calculation of this method is quite simple. Compared with carrier based PWM and space vector PWM, it does not need vector synthesis and complex zero sequence voltage calculation, so it is easy to implement. Experimental results verify the validity of the method.

        RNA Interference-Mediated Knockdown of Astrocyte Elevated Gene-1 Inhibits Growth, Induces Apoptosis, and Increases the Chemosensitivity to 5-Fluorouracil in Renal Cancer Caki-1 Cells

        Astrocyte elevated gene-1 (AEG-1) is a recently discov-ered oncogene that has been reported to be highly expressed in various types of malignant tumors, including renal cell carcinoma. However, the precise role of AEG-1 in renal cancer cell proliferation and apoptosis has not been clarified. In this study, we transfected the renal cancer cell line Caki-1 with a plasmid expressing AEG-1 short hairpin RNA (shRNA) and obtained cell colonies with stable knockdown of AEG-1. We found that AEG-1 down-regulation inhibited cell proliferation and colony formation and arrested cell cycle progression at the sub-G1 and G0/G1 phase. Western blot analysis indicated that the expression of proliferating cell nuclear antigen (PCNA), cyclin D1 and cyclin E were significantly reduced following AEG-1 down-regulation. In addition, AEG-1 knockdown led to the appearance of apop-totic bodies in renal cancer cells, and the ratio of apoptotic cells significantly increased. Expression of the anti-apoptotic factor Bcl-2 was dramatically reduced, whereas the pro-apoptotic factors Bax, caspase-3 and poly (ADP-ribose) polymerase (PARP) were significantly activated. Finally, AEG-1 knockdown in Caki-1 cells remarkably suppressed cell proliferation and enhanced cell apoptosis in response to 5-fluorouracil (5-FU) treatment, suggesting that AEG-1 inhibition sensitizes Caki-1 cells to 5-FU. Taken together, our data suggest that AEG-1 plays an important role in renal cancer formation and development and may be a potential target for future gene therapy for renal cell carcinoma.

        Effect of calcium carbonate in waste office paper on enzymatic hydrolysis efficiency and enhancement procedures

        Hydrolysis of waste office paper (WOP) into fermentable sugars is an important option of WOP utilization. In this work, the effect of major chemicals in WOP on its hydrolysis using industrial cellulase Accellerase 1000 (Genencor,Rochester, NY, USA) was investigated, and calcium carbonate (CaCO_3) was found to be the key parameter affecting the enzymatic hydrolysis efficiency of WOP. The pretreatment methods, acid washing and acid presoaking, were tested for the removal of CaCO_3 from WOP. It was found that the presoaking of sulfuric acid (H_2SO_4) in WOP was an effective way. The pretreating parameters of WOP were studied on maximizing the hydrolysis efficiency. The conversion yield of cellulose to glucose and cellobiose using the pretreated WOP reached 73.3% after 96 hours hydrolysis at the optimal conditions. The results provided the WOP utilization with a practical enzymatic hydrolysis method with industrial application potential.
