        Upregulated lncRNA Gm2044 inhibits male germ cell development by acting as miR-202 host gene

        Long non-coding RNAs (lncRNAs) have been found to participate in the regulation of human spermatogenic cell development. However, little is known about the abnormal expression of lncRNAs associated with spermatogenic failure and their molecular mechanisms. Using lncRNA microarray of testicular tissue for male infertility and bioinformatics methods, we identified the relatively conserved lncRNA Gm2044 which may play important roles in non-obstructive azoospermia. The UCSC Genome Browser showed that lncRNA Gm2044 is the miR-202 host gene. This study revealed that lncRNA Gm2044 and miR-202 were significantly increased in nonobstructive azoospermia of spermatogonial arrest. The mRNA and protein levels of Rbfox2, a known direct target gene of miR-202, were regulated by lncRNA Gm2044. Furthermore, the miR- 202-Rbfox2 signalling pathway was shown to mediate the suppressive effects of lncRNA Gm2044 on the proliferation of human testicular embryonic carcinoma cells. Understanding of the molecular signalling pathways for lncRNA-regulated spermatogenesis will provide new clues into the pathogenesis and treatment of patients with male infertility.

        Involvement of lncRNA-HOTTIP in the Repair of Ultraviolet Light-Induced DNA Damage in Spermatogenic Cells

        Ultraviolet light (UV)-induced cellular response has been studied by numerous investigators for many years. Long noncoding RNAs (lncRNAs) are emerging as new regulators of diverse cellular process; however, little is known about the role of lncRNAs in the cellular response to UV treatment. Here, we demonstrate that levels of lncRNA-HOTTIP significantly increases after UV stimulation and regulates the UV-mediated cellular response to UV through the coordinate activation of its neighboring gene Hoxa13 in GC-1 cells (spermatogonia germ cell line). UV-induced, G2/M-phase arrest and early apoptosis can be regulated by lncRNA-HOTTIP and Hoxa13. Furthermore, lncRNA-HOTTIP can up-regulate ${\gamma}-H_2AX$ and p53 expression via Hoxa13 in UV-irradiated GC-1 cells. In addition, p53 has the ability to regulate the expression of both lncRNA-HOTTIP and Hoxa13 in vitro and in vivo. Our results provide new data regarding the role lncRNAs play in the UV response in spermatogenic cells.

        Complex roles of the stroma in the intrinsic resistance to gemcitabine in pancreatic cancer: where we are and where we are going

        Pancreatic ductal adenocarcinoma (PDAC) is among the most devastating human malignancies. The poor clinical outcome in PDAC is partly attributed to a growth-permissive tumor microenvironment. In the PDAC microenvironment, the stroma is characterized by the development of extensive fibrosis, with stromal components outnumbering pancreatic cancer cells. Each of the components within the stroma has a distinct role in conferring chemoresistance to PDAC, and intrinsic chemoresistance has further worsened this pessimistic prognosis. The nucleoside analog gemcitabine (GEM) is usually the recommended first-line chemotherapeutic agent for PDAC patients and is given alone or in combination with other agents. The mechanisms of intrinsic resistance to GEM are an active area of ongoing research. This review highlights the important role the complex structure of stroma in PDAC plays in the intrinsic resistance to GEM and discusses whether antistroma therapy improves the efficacy of GEM. The addition of antistroma therapy combined with GEM is expected to be a novel therapeutic strategy with significant survival benefits for PDAC patients.

      • Trusted Quantum Botnet Research based on Dual-Chord Protocol

        The controller in the traditional chord-based botnets needs to maintain the periodic communication among all the adjacent peerbots so that every peerbot could efficiently master the new routing information. Once some peerbot is captured by the anti-virus software and is treated as a honeypot, the security personnel can capture more peerbots by tracking the deployed honeypot when it updates the routing table so that the robustness of botnet would be affected. To solve the above-mentioned problem, the trusted chord-based quantum botnet is proposed that utilizes the quantum secure channel to achieve the updating of routing table, and introduces the anti-tracking network for capturing the deployed honeypots. The anlysis result showed that the robustness and stability of trusted chord-based quantum botnet could efficiently be improved.

        Prenylated flavonoids are a special kind of flavonoid derivative possessing one or more prenyl groups in the parent nucleus of the flavonoid. The presence of the prenyl side chain enriched the structural diversity of flavonoids and increased their bioactivity and bioavailability. Prenylated flavonoids show a wide range of biological activities, such as anti-cancer, anti-inflammatory, neuroprotective, anti-diabetic, anti-obesity, cardioprotective effects, and anti-osteoclastogenic activities. In recent years, many compounds with significant activity have been discovered with the continuous excavation of the medicinal value of prenylated flavonoids, and have attracted the extensive attention of pharmacologists. This review summarizes recent progress on research into natural active prenylated flavonoids to promote new discoveries of their medicinal value.

        Superfast Preparation of SWNT Sponge by Flame Burning Method and Its Adsorptive, Elastic and Electrochemical Properties

        A compressible single-walled carbon nanotube (SWNT) sponge was developed by a superfast flame burning method in less than 20 s by moving polyurethane (PU) sponge template coated with SWNTs through an ethanol flame. By adjusting the geometries of the templates, the arbitrary shapes of the SWNT sponges composed of a unique network structure could be prepared as required. The SWNT sponges possessing good hydrophobicity and outstanding organic solvent adsorption capacity could adsorb various organic solvents and oils with high adsorption rate and good adsorption–volatilization and adsorption–combustion recycling performance. The SWNT sponges present good elasticity and compression stability even after a compressive strain of 80% and the 1000th loading/unloading cycle due to the stable skeleton structures. The SWNT sponges as flexible electrodes could also achieve high-specific capacitance of 126.8 F g -1 at 1Ag -1 and 95% capacitance retention after 10 000 charge/discharge cycles. Owing to the availability of the flame, easy decomposition of the PU sponge and flame resistance of SWNTs, this facile flame burning method was demonstrated to be a practical approach to prepare the SWNT sponges on a large scale with controllable shape and density, moderate organic liquid adsorption capability, good elasticity and decent electromechanical properties.

        Determination and prediction of the digestible and metabolizable energy contents of corn germ meal in growing pigs

        Objective: This experiment was conducted to determine the chemical composition, digestible energy (DE) and metabolizable energy (ME) contents of corn germ meals (CGM) and to develop equations to predict the corresponding energy contents based on the chemical characteristics of individual CGM. Methods: Sixty-six barrows (initial body weight = 51.3±4.6 kg) were allotted to 11 diets including a basal diet and 10 CGM test diets in a completely randomized design. In the test diets, CGM was included in replacement of 30% of the energy-providing ingredients in the basal diet, resulting in a final inclusion rate of 29.1%. Each diet was fed to 6 barrows housed in individual metabolism crates for a 7-d acclimation period followed by a 5-d total but separate collection of feces and urine. Results: Considerable variation was observed in acid-hydrolyzed ether extract, ether extract, ash, calcium (Ca) and total phosphorus contents among the CGM samples. On dry matter (DM) basis, the DE and ME contents of the CGM ranged from 10.22 to 15.83 MJ/kg and from 9.94 to 15.43 MJ/kg, respectively. The acid detergent fiber (ADF) contents were negatively correlated with the DE and ME contents of CGM samples. The best-fit prediction equations for the DE and ME values (MJ/kg DM) of the 10 CGM were: DE = 26.85–0.28 insoluble dietary fiber (%)–17.79 Ca (%); ME = 21.05–0.43 ADF (%)–11.40 Ca (%). Conclusion: The chemical compositions of CGM vary depending on sources, particularly in ether extract and Ca. The DE and ME values of CGM can be predicted based on their chemical composition in growing pigs.

        Effects of spherical adsorbent fluidization and self-rotation on removal of VOCs in a cyclonic fluidized bed

        Afluidized bed has the advantages of treating largeflows, intensifying mass and heat transfer, andlowering costs. This study proposed a cyclonicfluidized bed packed with spherical activated carbonadsorbents for volatile organic compounds adsorption. Thefluidization and self-rotation of the ACparticles in a 25 mm cyclonicfluidized bed were studied with high-speed camera testing technology. Theeffects of the particle movement on the adsorption efficiency of toluene were also tested. The resultsshow that most of the particles at the inlet side of the cyclonicfluidized bed were moving up when theinlet airflow rate was greater than 2.0 m3/h. The particles began to move in clusters when the relativepacking height increased to a critical value of 0.57. Increasing the gasflow rate and the diameter andheight of the core column will increase the self-rotation speed of the total particles. The maximum selfrotationspeed of spherical adsorbents reached 1700 rad/s at the inletflow rate of 2.5 m3/h. In the case ofthe same axial velocity of the gas phase in the upper space of the core column, increasing the particle selfrotationspeed can slightly improve the adsorption efficiency. The maximum adsorption efficiencyreached 99% when the inletflow rate is 1.0 m3/h with relative packing height 0.65.

        Synthesis and Structural Studies of an Organic Complex and its Association with BSA

        The self-assembly of one novel organic complex based on chlorogenic acid (HCA) and 2,2'-bipyridine (2,2'-bipy) has been synthesized and characterized. The complex achieved by hydrogen-bonding interactions, adopted a 1:1 stoichiometry in a solid state. The proton transfer occurred from the carboxyl oxygen to the aromatic nitrogen atom to form salts CA${\cdot}$(2,2'-Hbipy), the 2,2'-Hbipy molecule individually occupies the pseudo-tetragonum that is formed with CA. In this paper, the interactions of CA${\cdot}$(2,2'-Hbipy) with bovine serum albumin (BSA) were studied by fluorescence spectrometry. For CA${\cdot}$(2,2'-Hbipy), HCA and 2,2'-bipy, the average quenching constants for BSA were $2.4384{\times}10^4$, $4.653{\times}10^3$, and $3.059{\times}10^3\;L{\cdot}mol^{-1}$, respectively. The mechanism for protein fluorescence quenching is apparently governed by a static quenching process. The Stern-Volmer quenching constants and corresponding thermodynamic parameters ${\Delta}$H, ${\Delta}$G and ${\Delta}$S were calculated. The binding constants and the number of binding sites were also investigated. The conformational changes of BSA were observed from synchronous fluorescence spectra.
