        Ginsenoside Rg1 alleviates A deposition by inhibiting NADPH oxidase 2 activation in APP/PS1 mice

        Background: Ginsenoside Rg1 (Rg1), an active ingredient in ginseng, may be a potential agent for thetreatment of Alzheimer’s disease (AD). However, the protective effect of Rg1 on neurodegeneration in ADand its mechanism of action are still incompletely understood. Methods: Wild type (WT) and APP/PS1 AD mice, from 6 to 9 months old, were used in the experiment. The open field test (OFT) and Morris water maze (MWM) were used to detect behavioral changes. Neuronal damage was assessed by hematoxylin and eosin (H&E) and Nissl staining. Immunofluorescence,western blotting, and quantitative real-time polymerase chain reaction (q-PCR) were used toexamine postsynaptic density 95 (PSD95) expression, amyloid beta (Ab) deposition, Tau and phosphorylatedTau (p-Tau) expression, reactive oxygen species (ROS) production, and NAPDH oxidase 2(NOX2) expression. Results: Rg1 treatment for 12 weeks significantly ameliorated cognitive impairments and neuronaldamage and decreased the p-Tau level, amyloid precursor protein (APP) expression, and Ab generation inAPP/PS1 mice. Meanwhile, Rg1 treatment significantly decreased the ROS level and NOX2 expression inthe hippocampus and cortex of APP/PS1 mice. Conclusions: Rg1 alleviates cognitive impairments, neuronal damage, and reduce Ab deposition byinhibiting NOX2 activation in APP/PS1 mice.

        Preparation, characterization, and in vivo evaluation of a polymorphic form of valnemulin hydrogen tartrate

        We prepared a polymorphic form of valnemulin hydrogen tartrate (Form I) to overcome the instability and irritating odor of valnemulin hydrochloride that affect its use in the production and application of veterinary drugs. The physicochemical properties of Form I were characterized by scanning electron microscopy, X-ray powder diffraction, infraredspectroscopy, differential scanning calorimetry, and thermogravimetric analysis. The results showed the crystal structure and thermal properties of Form I were very different from those of a commercially available form of valnemulin hydrogen tartrate (Form II). Form I and Form II were more stable than valnemulin hydrochloride after storage under irradiation and high humidity conditions, respectively. The solubility of Form I was 2.6 times that of Form II, and Form I was selected for use in pharmaceutical kinetics experiments in vivo. Compared to valnemulin hydrochloride, after oral administration at a dose of 10 mg/kg in pigs, Form I had similar pharmaceutical kinetic behavior but a slightly higher area under the concentration– time curve from time zero to the last measurable concentration. Consequently, Form I should be suitable for the development of simple formulations and be effective in the clinical application of veterinary drugs.

        A double-layer optimization method for forging process parameters of hinge beam structure and size of intermediate billet

        A double-layer optimization method for the forging process parameters of a hinge beam structure and the design of intermediate billet structure is proposed. First, a primary optimization system for the forging process parameters of hinge beam was established based on orthogonal experiments and range analysis method, and the optimal combination of forging process parameters was selected. On this basis, a mathematical model of the secondary optimization of the final forging of the hinge beam was established. Through radial basis function (RBF) agent model and multi-island genetic algorithm, the secondary optimization of intermediate billet structure of hinge beam was carried out, which improved the grain refinement degree of final forging and reduced the forming load required for forging. Through numerical simulation and optimization design of forging process, the quality and performance of final forging of hinge beam were improved, which has certain guiding significance for the actual production of hinge beam.

        An energy-efficient hierarchical algorithm of dynamic obstacle avoidance for unmanned surface vehicle

        Most of the existing studies developed and improved local path planning algorithms independently of global planning, i.e., ignoring the global optimal constrains. To meet the requirements of practical applications, this paper presented an energy-efficient hierarchical collision avoidance algorithm for unmanned surface vehicle operating in clustered dynamic environments. For the global level, genetic algorithm was modified by strategies of greedy-inspired population initialization, penalty-based multiobjective fitness function, and joint crossover. For the local level, velocity obstacle was combined with dynamic window approach to provide the kinematic constraints of the vehicle to its admissible velocities and simplified collision avoidance rules to guide the evasive maneuvers. Simulations showed that the proposed global algorithm was superior to three other algorithms in terms of path length, path smoothness, and convergence speed regardless of the environment size. The performance of the local algorithm was also verified for various encounter scenarios and speed ratios. In addition, the combination of the global and local planning can effectively solve the path optimization and dynamic obstacle avoidance in a designed offshore environment of fish cage culture.

        The Clinical Feature of Behçet’s Disease in Northeastern China

        Purpose: Behçet’s disease (BD) is a chronic, relapsing, multi-system vasculitis of unknown aetiology with complicated and diversified clinical features predominantly involving oral and genital ulcers, and ocular and cutaneous lesions. The clinical features of this disease have been described to be different according to geographical areas and gender. We investigated the specific clinical features of BD patients in Northeastern China. Materials and Methods: 116 patients involved in this study fulfilled the classification criteria of the International Study Group for BD. The clinical manifestations and results of laboratory tests of BD were recorded in each patient. Results: The onset was typically between 20-39 years with a slight female predominance. Oral ulcers were the most common manifestation, followed by skin lesions, positive pathergy reaction/genital ulcers, and ocular lesions. Vascular lesion and epididymitis were rare in patients with BD. The frequency of erythema nodosum-like lesion and articular involvement were significantly higher in females, while gastrointestinal involvement was significantly higher in males. The results of laboratory tests showed that the human leukocyte antigen (HLA)-B*51 alleles were positive in 30.9% of patients and the immunological abnormities were present in some patients. Conclusion: The clinical features of BD showed geographical and gender difference. Genetic and immune factors might participate in aetiopathogenesis of BD.       • Highly sensitive and wearable gel-based sensors with a dynamic physically cross-linked structure for strain-stimulus detection over a wide temperature range

        <P>Traditional hydrogel sensors can only be applicable in a limited temperature range, such as ambient temperature, since water is easily frozen or evaporated under extreme conditions, which seriously affects their practical application. Here, a highly sensitive wearable strain sensor is fabricated from a flexible, self-healing, anti-freezing and anti-drying gel with a physical cross-linking structure, which is composed of polyacrylic acid, chitosan, and graphene oxide in a mixed solvent of water and glycerol. The dynamic cross-linking enables the network structure and ion channels of the gel to be rapidly recovered and reconstituted, thereby allowing the gel-based strain sensor to display excellent stretchability (more than 1000%) and outstanding sensing performance with a rapid response time of 40 ms, and remarkable repeatability and stability. As a result, the gel can be applied as an epidermal strain sensor for real-time detection of human motions, including joint motions, speaking and breathing. Moreover, the gel can maintain excellent flexibility, stretchability and conductivity over a wide temperature range from −20 °C to 70 °C, which effectively improves the practicality and durability of the gel in practical applications. Therefore, such flexible, conductive, anti-freezing and anti-drying gel may have promising applications in the field of wearable devices, soft robot systems and other applications that need to be applied under changeable conditions.</P>

      • Centimeter-Long and Large-Scale Micropatterns of Reduced Graphene Oxide Films: Fabrication and Sensing Applications

        <P>Recently, the field-effect transistors (FETs) with graphene as the conducting channels have been used as a promising chemical and biological sensors. However, the lack of low cost and reliable and large-scale preparation of graphene films limits their applications. In this contribution, we report the fabrication of centimeter-long, ultrathin (1−3 nm), and electrically continuous micropatterns of highly uniform parallel arrays of reduced graphene oxide (rGO) films on various substrates including the flexible polyethylene terephthalate (PET) films by using the micromolding in capillary method. Compared to other methods for the fabrication of graphene patterns, our method is fast, facile, and substrate independent.         Polymorphisms in Epigenetic and Meat Quality Related Genes in Fourteen Cattle Breeds and Association with Beef Quality and Carcass Traits

        Improvement for carcass traits related to beef quality is the key concern in beef production. Recent reports found that epigenetics mediates the interaction of individuals with environment and nutrition. The present study was designed to analyze the genetic effect of single nucleotide polymorphisms (SNPs) in seven epigenetic-related genes (DNMT1, DNMT3a, DNMT3b, DNMT3L, Ago1, Ago2, and HDAC5) and two meat quality candidate genes (CAPN1 and PRKAG3) on fourteen carcass traits related to beef quality in a Snow Dragon beef population, and also to identify SNPs in a total of fourteen cattle populations. Sixteen SNPs were identified and genotyped in 383 individuals sampled from the 14 cattle breeds, which included 147 samples from the Snow Dragon beef population. Data analysis showed significant association of 8 SNPs within 4 genes related to carcass and/or meat quality traits in the beef populations. SNP1 (13154420A>G) in exon 17 of DNMT1 was significantly associated with rib-eye width and lean meat color score (p<0.05). A novel SNP (SNP4, 76198537A>G) of DNMT3a was significantly associated with six beef quality traits. Those individuals with the wild-type genotype AA of DNMT3a showed an increase in carcass weight, chilled carcass weight, flank thicknesses, chuck short rib thickness, chuck short rib score and in chuck flap weight in contrast to the GG genotype. Five out of six SNPs in DNMT3b gene were significantly associated with three beef quality traits. SNP15 (45219258C>T) in CAPN1 was significantly associated with chuck short rib thickness and lean meat color score (p<0.05). The significant effect of SNP15 on lean meat color score individually and in combination with each of other 14 SNPs qualify this SNP to be used as potential marker for improving the trait. In addition, the frequencies of most wild-type alleles were higher than those of the mutant alleles in the native and foreign cattle breeds. Seven SNPs were identified in the epigenetic-related genes. The SNP15 in CAPN1 could be used as a powerful genetic marker in selection programs for beef quality improvement in the Snow Dragon Beef population.
