        Fine mapping and candidate gene analysis of the dwarf gene d162(t) in rice (Oryza sativa L.)

        In our previous study, d162(t), a single recessive gene, which caused rice dwarf mutant, had been mapped on the short arm of chromosome 3. In this study, the d162(t) gene was fine mapped to a confined region about 0.82 cM by RM14641 and RM3134, and co-segregated with InDel361-2, InDel361-3,InDel361-5, RM14645, RM1022 and RM14643, where no known gene involved in plant height has been identified. Based on the annotation results of TIGR, dozens of open reading frames (ORFs) were predicted in this region, among them,five ORFs were the most possible genes related to the phenotype. In these ORFs, Os03g13010, related to U-box domain containing protein, had a 62bp segment deletion in the coding region in 162d (mutant type, MT). The results of RT-PCR showed that the transcriptional level of Os03g13010was significantly different between Shuhui162 (wild type, WT)and 162d (MT). Therefore, the gene (Os03g13010) encoding a U-box domain containing protein was considered as the candidate gene of d162(t).

        Fabrication of recyclable UiO-66-NH2/PVDF hybrid fibrous membrane for Cr(VI) removal in wastewater

        Efficient and recyclable water treatment technology plays a vital role in practical application of Cr(Ⅵ)removal. Zr-based metal–organic framework (UiO-66-NH2) is widely used in wastewater treatmentdue to its water stability. Powdery UiO-66-NH2 has disadvantages of easy aggregation, reclamation difficultyand secondary pollution. In this work, UiO-66-NH2 was successfully loaded on electrospunpolyvinylidene fluoride (PVDF) fibers by in-situ growth to achieve efficient adsorption and photoreductionof Cr(Ⅵ). Grooved surface of PVDF fibers and truncated octahedron shape of UiO-66-NH2 ensuredthe loading uniformity and firmness. The loading rate and size of UiO-66-NH2 crystals increased withthe amount of trifluoroacetic acid (TFA) in the preparation process. Meantime, the introduction of TFAin synthesis was conducive to the separation efficiency of photogenerated carries. When the additionamount of TFA was 5 ml, UiO-PVDF-3 with loading rate of 44.8% exhibited best Cr(VI) adsorption andphotocatalysis performance. For the pure adsorption process, the adsorption capacity of UiO-PVDF-3 at90 min was 3.76 mg/cm2, and the corresponding removal rate was 95.8%. When visible light was applied,the Cr(VI) removal rate for UiO-PVDF-3 reached to about 95.0% at 45 min. Furthermore, regenerationexperiments showed UiO-PVDF-3 had excellent adsorption and photocatalysis reusability. Based on thismethod, synthesized UiO-66-NH2/PVDF hybrid membrane as a bifunctional material for adsorption andphotocatalysis realizes the reuse of UiO-66-NH2 crystals conveniently and avoids secondary pollutionduring Cr(VI) wastewater treatment.

        Research on the Accessibility of Urban Green Space Based on Road Network- A Case Study of the Park Green Space in City Proper of Nanjing

        Accessibility of urban green space, as an important index of evaluating the service level of urban green space, not only shows the resident's access to green space, but also reflects the environment quality of urban living. Since the quantity, area and space distribution of the patches in urban green space directly affect the function of urban green space and the quality of urban environment, the research on the accessibility of urban green space holds great significance to improve the service of urban green space and the living environment quality. Based on GIS software, this research uses the road network and the population distribution data of Nanjing's city proper to conduct quantitative analysis of the accessibility of park green space and the convenience for residents to travel under different forms of transportation. After the case analysis, the author tests the application of road network in studying the accessibility of urban park green space and proposes a GIS-based method to study the accessibility and the rational layout of urban green space.

      • Removal of the Glycosylation of Prion Protein Provokes Apoptosis in SF126

        Although the function of cellular prion protein (PrP$^C$) and the pathogenesis of prion diseases have been widely described, the mechanisms are not fully clarified. In this study, increases of the portion of non-glycosylated prion protein deposited in the hamster brains infected with scrapie strain 263K were described. To elucidate the pathological role of glycosylation profile of PrP, wild type human PrP (HuPrP) and two genetic engineering generated non-glycosylated PrP mutants (N181Q/N197Q and T183A/T199A) were transiently expressed in human astrocytoma cell line SF126. The results revealed that expressions of non-glycosylated PrP induced significantly more apoptosis cells than that of wild type PrP. It illustrated that Bcl-2 proteins might be involved in the apoptosis pathway of non-glycosylated PrPs. Our data highlights that removal of glycosylation of prion protein provokes cells apoptosis.

        Study on changes and mechanisms of cytokines for alloxan-induced hepatic injury by Cr3+-treatment in mice

        This study investigated the effects of treatment with trivalent chromium (Cr3+) on the cytokine levels in mice that had alloxan-induced hepatic injury. The results showed that chromium picolinate (CrPic) may have a protective effect against hepatic injury and that these effects might be related to changes in cytokine expression. Blood ALT and AST levels in the alloxan mice were higher than those in the control and CrPic-treated groups (no significant differences between the latter groups). Luminex assays of cytokines from Th1 (TNF-α, IL-2 and IL-9) and Th2 (IL-3, IL- 5, IL-6, IL-10, IL-13) - as well as G-CSF, M-CSF, and GM-CSF - demonstrated increased levels of TNF-α, IL-2, and IL-9 (55.10%, 1.05-fold, and 2.14-fold, respectively) in alloxan-treated mice compared to control mice. Further, compared to the control group, liver IL- 3, -5, -6, -10, and -13 levels were decreased 2.45-fold, 1.54-fold, 1.01-fold, 12.6% and 37.3%, respectively, in the alloxan-treated mice. In comparison to the control mice, the levels of G-CSF, M-CSF and GM-CSF were decreased in the alloxan-treated mice (25.2%, 49.5%, and 52.9%, respectively). These cytokines were increased 7.91-fold, 43.67% and 34.05%, respectively, in the CrPic-treated group. These findings suggest that Th1/Th2 type cytokines are involved in hepatic injury and induced by alloxan, which may provide a new possible therapeutic approach through CrPic-treatment.

      • Association of CYP2C19 Polymorphisms with Survival of Breast Cancer Patients Using Tamoxifen: Results of a Meta-analysis

        Background: Previous studies accessing the association of CYP2C19 with outcomes of patients using tamoxifen for breast cancer have yielded conflicting results. The aim of this meta-analysis is to obtain a more precise estimate of effects of CYP2C19 polymorphisms and to clarify their effects on survival of the breast cancer patients using tamoxifen. Materials and Methods: A systematic search of PubMed and Embase was performed, comparing patients with or without $CYP2C19^*2$ and $CYP2C19^*17$, relevant articles searched for. The following outcomes were included from the eligible studies: disease-free survival (DFS) and overall survival (OS), expressed by hazard ratios (HR) with corresponding 95% confidence interval (CI). Subgroup analysis by genotypes was also performed. Pooled estimates were calculated using random-effect model in accordance to the heterogeneity. Results: Six studies met the inclusion criteria. The integrated OR on the association between CYP2C19 and DFS, calculated by the random-effect model, was 0.54 (95%CI=0.34-0.84, p=0.013). Subgroup analysis showed that both $CYP2C19^*2$ and $CYP2C19^*17$ were associated with increased survival. The pooled results of two studies for OS were OR=0.46 (95%CI=0.21-1.01, p=0.233). Conclusions: This meta-analysis suggests that the $CYP2C19^*2$ and $CYP2C19^*17$ genotypes are associated with increased survival in breast cancer patients using tamoxifen.

        Profiling Gene Expression During Gland Morphogenesis of a Glanded and a Glandless Upland Cotton

        The pigment gland is an important character of the Gossypium plant. With the aim of identifying genes involved in pigment gland morphogenesis in cotton, gene expression during pigment gland morphogenesis in Chuan 2802, which is glanded both in seed and plant, and a glandless line N5 was profiled using Affymetrix Cotton microarray. The results showed that there were 564 differentially expressed genes greater than twofold during gland morphogenesis. About 60.2% of these genes shares similarity with known genes on GenBank and about 39.8% with no functional description in the database. These described genes may play roles in defense response, response to oxidative stress, peroxidase activity, and the other metabolic pathways. The KEGG Orthology-Based Annotation System indicated that these above twofold expressed genes involved seven biochemical pathways on KEGG. These findings suggest that a complicated regulation is associated with pigment gland morphogenesis and the associated defense response including gossypol biosynthesis in cotton.

        STABILITY ANALYSIS UPON HIGH-PRESSURE COMMON RAIL FUEL INJECTION SYSTEM UNDER MULTIPLE INJECTION MODES

        Fuel injection stability of high-pressure common rail fuel injection system (HPCRFIS) under multiple injection modes is a recognized technical problem. To explain it essentially, a numerical model based on bond graph method, which provides a standardized series of symbol system to describe various components in different physical fields as well as interactive effect analysis tool for different energy categories, was used to investigate the problem. As HPCRFIS is a typical nonlinear system with multi-physical coupling, it is very difficult to analyze system stability directly. Therefore, the system was linearized at typical moments. The system matrix for each typical moment was obtained thereby. To investigate the influential mechanism of system stability deeply, the variations of rank and eigenvalues distributions of state matrices were analyzed. The results show that the rank of state matrices changes abruptly when the needle or control valve moves. In terms of system stability, the oscillation characteristics are subject to the movement of needle or control valve. The ending of the former injection causes the system to show damping oscillation characteristics before the latter injection starts.The motion of control valve makes more contribution to system stability than that of needle, which could be concluded by Lyapunov stability criterion.
