      • The Control of Collaboration for Comprehensive Transportation Corridor

        Regional comprehensive corridor takes on the characteristics of heavy transport demand and supply, and consists of many travel modes; meanwhile, competition is severe among different trip modes. In order to avoid vicious competition so that transportation market is disturbed, multi-objective and nonlinear programming model is built up, which is based on the maximization of unity of traveler. The objective of model is minimization of distance between practical capacity coefficient and ideal capacity coefficient. Multi-objective is transferred into single objective by TOPSIS method before model is solved. The transformed objective function is multi-peak function, so niche GA is introduced into solving the problem to obtain all of optimal solution. K-means clustering is adapted to classify the set of optimal solution. The centre of gravity of each cluster is taken as value of decision variables. At last a numeric example demonstrates the validity and feasibility.

        Bioinformatic and integrated analysis identifies an lncRNA–miRNA–mRNA interaction mechanism in gastric adenocarcinoma

        Background lncRNAs–miRNAs–mRNAs networks play an important role in Gastric adenocarcinoma (GA). Identifcation of these networks provide new insight into the role of these RNAs in gastric cancer. Objectives Biological information databases were screened to characterize and examine the regulatory networks and to further investigate the potential prognostic relationship this regulation has in GA. Methods By mining The Cancer Genome Atlas (TCGA) database, we gathered information on GA-related lncRNAs, miRNAs, and mRNAs. We identifed diferentially expressed (DE) lncRNAs, miRNAs, and mRNAs using R software. The lncRNA–miRNA–mRNA interaction network was constructed and subsequent survival examination was performed. Representative genes were selected out using The Biological Networks Gene Ontology plug-in tool on Cytoscape. Additional analysis of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) terms were used to screen representative genes for functional enrichment. Reverse transcription quantitative polymerase chain reaction (RT-qPCR) were used to identify the expression of fve candidate diferential expressed RNAs. Results Information of samples from 375 cases of gastric cancer and 32 healthy cases (normal tissues) were downloaded from the TCGA database. A total of 1632 DE-mRNAs, 1008 DE-lncRNAs and 104 DE-miRNAs were identifed and screened. Among them, 65 DE-lncRNAs, 10 DE-miRNAs, and 10 DE-mRNAs form lncRNAs–miRNAs–mRNAs regulatory network. Additionally, 10 lncRNAs and 2 mRNAs were associated with the prognosis of GA. Multivariable COX analysis revealed that AC018781.1 and VCAN-AS1 were independent risk factors for GA. GO functional enrichment analysis found DE-mRNA was signifcantly enriched TERM (P<0.05). The KEGG signal regulatory network analysis found 11 signifcantly enrichment networks, the most prevailing was for the AGE-RAGE signaling pathway associated with Diabetic complications. Results of RT-qPCR was consistent with the in silico results. Conclusions The results of the present study represent a view of GA from a analysis of lncRNA, miRNA and mRNA. The network of lncRNA–miRNA–mRNA interactions revealed here may potentially further experimental studies and may help biomarker development for GA.

        Study on Anti-Proliferative Activity in Cancer Cells and Preliminary Structure–Activity Relationship of Pseudo-Peptide Chiral Thioureas

        In our previous studies, we have shown that thiourea compounds containing phosphate esters have potent antitumor activity and can be used as a novel strategy for the development of antitumor agents. Herein, a series of novel phosphonate thioureas 5–38 have been synthesized, which were fully characterized by 1H NMR, 13C NMR spectrum, elemental analysis. Three human cancer cell lines (Bcap-37, BGC-823, and PC-3) have been used to investigate these compounds’ antitumor activities. After the summarization of the structure–activity relationships, we found that the variation of R, R1, and R2 in these novel phosphonate thioureas contribute to the antitumor activities. All these SAR-guided efforts may lead to novel antitumor drugs in the market in the near future.

        Functional Magnetic Resonance Imaging in the Diagnosis of Locally Recurrent Prostate Cancer: Are All Pulse Sequences Helpful?

        Objective: To perform a meta-analysis to quantitatively assess functional magnetic resonance imaging (MRI) in the diagnosis of locally recurrent prostate cancer. Materials and Methods: A comprehensive search of the PubMed, Embase, Cochrane Central Register of Controlled Trials, and Cochrane Database of Systematic Reviews was conducted from January 1, 1995 to December 31, 2016. Diagnostic accuracy was quantitatively pooled for all studies by using hierarchical logistic regression modeling, including bivariate modeling and hierarchical summary receiver operating characteristic (HSROC) curves (AUCs). The Z test was used to determine whether adding functional MRI to T2-weighted imaging (T2WI) results in significantly increased diagnostic sensitivity and specificity. Results: Meta-analysis of 13 studies involving 826 patients who underwent radical prostatectomy showed a pooled sensitivity and specificity of 91%, and the AUC was 0.96. Meta-analysis of 7 studies involving 329 patients who underwent radiotherapy showed a pooled sensitivity of 80% and specificity of 81%, and the AUC was 0.88. Meta-analysis of 11 studies reporting 1669 sextant biopsies from patients who underwent radiotherapy showed a pooled sensitivity of 54% and specificity of 91%, and the AUC was 0.85. Sensitivity after radiotherapy was significantly higher when diffusion-weighted MRI data were combined with T2WI than when only T2WI results were used. This was true when meta-analysis was performed on a per-patient basis (p = 0.027) or per sextant biopsy (p = 0.046). A similar result was found when 1H-magnetic resonance spectroscopy (1H-MRS) data were combined with T2WI and sextant biopsy was the unit of analysis (p = 0.036). Conclusion: Functional MRI data may not strengthen the ability of T2WI to detect locally recurrent prostate cancer in patients who have undergone radical prostatectomy. By contrast, diffusion-weight MRI and 1H-MRS data may improve the sensitivity of T2WI for patients who have undergone radiotherapy.

      • Clinical Efficacy of Bevacizumab Concomitant with Pemetrexed in Patients with Advanced Non-small Cell Lung Cancer

        Objective: To observe the clinical efficacy of bevacizumab concomitant with pemetrexed in patients with advanced non-small cell lung cancer (NSCLC). Materials and Methods: A total of 72 patients were randomly divided into a combination group (pemetrexed+bevacizumab, n=36) and a pemetrexed group (n=36) and assessed for disease control (CR+PR+SD) after 4-cycles of first-line GP chemotherapy (gemcitabine+cisplatin). Clinical efficacy, progression-free survival time (PFS), overall survival time (OS), overall response rate (ORR), disease control rate (DCR) and rate of adverse responses between two groups were observed and compared. Results: ORR and DCR were 27.8% and 83.4% in combination group, and 16.7% and 69.5% in the pemetrexed group, respectively, but there were no significant differences (P>0.05). PFS in combination group and pemetrexed group were 4.6 months and 3.9 months respectively (P=0.09), whereas OS in the combination group was 14 months, evidently higher than in the pemetrexed group (11 months, P=0.004). Adverse responses in both groups included high blood pressure, bleeding, thrombocytopenia, anemia, elevated transaminase, diarrhea, vomiting and proteinuria, but there were no significant differences (P>0.05). Conclusions: Bevacizumab concomitant with pemetrexed has better clinical efficacy and safety, giving rise to prolonged survival time in patients with advanced NSCLC.

      • 0346 Occupational Exposure to Lead and Cancer in Two Cohort Studies of Men and Women in Shanghai, China

        <P><B>Objectives</B></P><P>Epidemiologic studies of occupational lead exposure have suggested increased risks of cancers of the brain, kidney, lung, meninges, and stomach; however, the totality of the evidence is inconsistent. To clarify whether lead is a carcinogen, we investigated the relationship between occupational lead exposure and risks of these five cancer sites in two prospective cohort studies in Shanghai, China.</P><P><B>Method</B></P><P>Annual job/industry-specific estimates of lead fume and lead dust exposure were derived from a statistical model that combined expert ratings of lead intensity with inspection measurements collected by the Shanghai Centre for Disease Control and Prevention. The job/industry estimates were applied to the lifetime work histories of subjects from the Shanghai Women’s Health Study (73 363 participants) and the Shanghai Men’s Health Study (61 379 participants) to estimate cumulative exposure to lead dust and lead fume. Cohort-specific relative hazard rate ratios (RRs) and 95% confidence intervals (CI) were estimated using Cox proportional hazards regression models and then pooled using a random effects meta-analysis model.</P><P><B>Results</B></P><P>We observed a statistically significant increased risk of meningioma among individuals with estimated occupational exposure to lead dust or fumes (RR=2.4, 95% CI:1.1–5.0), and in particular among those with an above-median cumulative exposure to dust or fumes (RR=3.1, 95% CI:1.3–7.4). We observed suggestive associations with lead exposure for cancers of the kidney (RR=1.4, 95% CI:0.9–2.3) and brain (RR=1.8, 95% CI:0.7–4.8), and null findings for cancers of the lung and stomach.</P><P><B>Conclusions</B></P><P>Our findings provide additional evidence that occupational lead exposure increases risk of meningioma.</P>

        Dynamic response of aluminum honeycomb sandwich panels subjected to hypervelocity impact by porous volcanic rock projectile

        The dynamic response and damage behavior of aluminum honeycomb sandwich panels (HC/SPs) subjected to hypervelocity impact by volcanic rock projectiles were investigated by hypervelocity impact tests and hydrocode simulations. The experiments were conducted using a two stage light gas gun and the results showed that the failure modes in HC/SPs subjected to hypervelocity impact by volcanic rock projectiles mainly took forms of front-face denting and circular perforation, honeycomb core collapsing and rapture, rear-face petalling and perforation etc. A 3D discrete configuration of the porous volcanic rock projectiles was set up. The hypervelocity impact behavior of the HC/SPs was investigated through hydrocode modeling, within a Lagrange-SPH coupling method in LS-DYNA solver. It was found that the dynamic response and failure modes in the HC/SPs were significantly influenced by the impact location and the impact velocity of the volcanic rock projectile.

        Structural Damage Identification Based on Convolutional Neural Network Group Considering the Sensor Fault

        This article proposes a structural damage identification method based on one-dimensional convolutional neural network group considering sensor faults. The method aims to reduce the damage misjudgment caused by sensor faults. In the proposed method, according to the sensor layout, some convolutional neural network sub-models are established to extract the features from raw vibration data for sensor fault diagnosis and structural damage identification; then two convolutional neural networks groups, namely the sensor fault diagnosis group and the damage identification group are designed on the basis of the functions of each sub-model. The sensor fault diagnosis group determines whether the sensor data is abnormal and truncates the abnormal signal. The remaining normal signal are entered into the damage identification group and the final damage identification results are calculated according to the statistical decision module. The effectiveness of the devised method is verified by the IASC–ASCE benchmark structure and laboratory experiments. The results demonstrate that the sensor fault diagnosis and damage identification accuracy of each sub-model ranges from 98.54% to 99.77% and from 87.21% to 91.74% respectively at different noise levels; the damage identification group can reduce the impact of sub-model misjudgment on the structural damage identification. The accuracy of the final damage identification results is 100%. The identification time of all samples in the test set is 53.09 s and 22.93 s, respectively, for SHM benchmark and Laboratory experiment cases. And the average judgment time of each submodel in the sensor fault diagnosis group was 278 and 94 ms, and that of each submodel in the damage identification group was 294 and 105 ms, respectively, for a single test sample, which fulfills the requirements of online damage identification for structural health monitoring.

        Effect of all-trans retinoic acid on casein and fatty acid synthesis in MAC-T cells

        Objective: Caseins and fatty acids of milk are synthesized and secreted by the epithelial cells of the mammary gland. All-trans retinoic acid (ATRA), an active metabolite of vitamin A, has been shown to promote mammary development. This study was conducted to determine the effect of ATRA on casein synthesis and fatty acid composition in MAC-T cells. Methods: MAC-T cells were allowed to differentiate for 4 d, treated with ATRA (0, 1.0, 1.5, and 2.0 μM), and incubated for 3 d. We analyzed the fatty acid composition, the mRNA expression of casein and fatty acid synthesis-related genes, and the phosphorylation of casein synthesis-related proteins of MAC-T cells by gas chromatography, quantitative polymerase chain reaction, and western blotting, respectively. Results: In MAC-T cells, ATRA increased the mRNA levels of α<sub>S1</sub>-casein and β-casein, janus kinase 2 (JAK2) and E74-like factor 5 of the signal transducer and activator of transcription 5 β (STAT5-β) pathway, ribosomal protein S6 kinase beta-1 (S6K1) and eukaryotic translation initiation factor 4E binding protein 1 of the mammalian target of rapamycin (mTOR) pathway, inhibited the mRNA expression of phosphoinositide 3-kinase and eukaryotic initiation factor 4E of the mTOR pathway, and promoted the phosphorylation of STAT5-β and S6K1 proteins. Additionally, ATRA increased the de novo synthesis of fatty acids, reduced the content of long-chain fatty acids, the ratio of monounsaturated fatty acids to saturated fatty acids (SFA), the ratio of polyunsaturated fatty acids (PUFA) to SFA, and the ratio of ω-6 to ω-3 PUFA. The mRNA levels of acetyl-CoA carboxylase 1, fatty acid synthase, lipoprotein lipase, stearoyl-CoA desaturase, peroxisome proliferator-activated receptor gamma, and sterol regulatory element-binding protein 1 (SREBP1) were enhanced by ATRA. Conclusion: ATRA promotes the synthesis of casein by regulating JAK2/STAT5 pathway and downstream mTOR signaling pathway, and it improves the fatty acid composition of MAC-T cells by regulating SREBP1-related genes.

        In vitro evaluation of 9-(2-phosphonylmethoxyethyl)adenine ester analogues, a series of anti-HBV structures with improved plasma stability and liver release

        Chronic hepatitis B virus (HBV) infection maylead to liver cirrhosis and hepatocellular carcinoma, butfew drugs are available for its treatment. Acyclic nucleosidephosphonates (ANPs) have remarkable antivirusactivities but are not easily absorbed from the gastrointestinaltract and accumulate in the kidneys, resulting innephrotoxicity. Therefore, there is a need to find effectiveliver site-specific prodrugs. The dipivaloyloxymethyl esterof 9-(2-phosphonylmethoxyethyl)adenine (PMEA)—adefovirdipivoxil (ADV)—is a first-line therapy drug forchronic hepatitis B with a low therapeutic index because ofrenal toxicity and low hepatic uptake. In this study, a seriesof PMEA derivatives were synthesized to enhance plasmastability and liver release. The metabolic stability of ADV(Chemical I) and its two analogues (Chemicals II and III)was evaluated in rat plasma and liver homogenate in vitro. An ion-pair reverse-phase HPLC–UV method and a hybridion trap and high-resolution time-of-flight mass spectrometry(LC-IT-TOF-MS) were used to evaluate the degradationrate of the analogues and to identify their intermediatemetabolites, respectively. Chemicals I and II were hydrolyzedby cleavage of the C–O bond to give monoesters. Sufficient enzymatic activation in the liver homogenatethrough a relatively simple metabolic pathway, in additionto a favorable stability profile in rat plasma, made ChemicalII an optimal candidate. Next, six analogues based onthe structure of Chemical II were synthesized and evaluatedin plasma and liver homogenate. Compared toChemical II, these compounds generated less active PMEAlevels in rat liver homogenate. Therefore, chemical modificationof Chemical II may lead to new promising PMEAderivatives with enhanced plasma stability and liveractivation.
