Characterization of Lactobacillus acidophilus Isolated from Piglets and Chicken

Lactic acid bacteria were isolated from piglets and chicken and characterized. Lactic acid bacteria showing resistance to low pH and bile, adhesion to intestinal epithelium cells, and the inhibition of Escherichia coli and Salmonella spp. were identified as Lactobacillus acidophilus. L. acidophilus PF01 survived for 2 h in MRS broth adjusted to pH 2. L. acidophilus CF07 was less resistant than L. acidophilus PF01 to pH 2, but survived at pH 2.5 for 2 h. Both of isolates were able to grow in MRS broth containing 0.3% (w/v) bile, with L. acidophilus CF07 being more tolerant to bile than L. acidophilus PF01. L. acidophilus PF01 and CF07 adhered specifically to the duodenal and jejunal epithelium cells of piglet, and the cecal and duodenal epithelium cells of chicken, respectively. Both of isolates did not adhere to the epithelium cells of the various animal intestines from which they were isolated. When L. acidophilus was cultured with E. coli and Salmonella spp. in MRS broth, MRS broth containing 2% skim milk powder or modified tryptic soy broth at $37^{\circ}C$, L. acidophilus PF01 and CF07 inhibited the growths of E. coli K88 and K99, and S. enteritidis and S. typhimurium, respectively. Both of isolates were found to possess the essential characteristics of probiotic lactic acid bacteria for piglet and chicken.

본논문에서는 L- R퍼지수의 집합-이론적 연산의 개념을 정의하고, 이들 개념의 성질들을 조사한다. 이들 연산들의 결과들을 이용하여 제2형 퍼지집합의 기수개념에 관하여 연구한다. In this paper, we define concepts of some set-theoretical operations of L- R fuzzy numbers and discuss some properties of these concepts. Using these results, we discuss a concept of cardinality of type-two fuzzy sets.
Elimination of the cryptic plasmid in Leuconostoc citreum by CRISPR/Cas9 system

<P>Leuconostoc spp. are important lactic acid bacteria for the fermentation of foods. In particular, L. citreum strains isolated from various foods have been used as host strains for genetic and metabolic engineering studies. In order to develop a food-grade genetic engineering system, L. citreum CB2567 was isolated from Kimchi. However, the isolated bacterium contained a cryptic plasmid which was difficult to eliminate. As the existence of the plasmid might hinder strain engineering, we eliminated the plasmid using an RNA-guided DNA endonuclease CRISPR/Cas9 system. We demonstrated that a plasmid-free L. citreum CB2567 host strain could be efficiently constructed through a two-step procedure: 1) transformation of the 'killer' plasmid expressing Cas9 endonuclease and a guide RNA (gRNA) targeting for a specific sequence in the cryptic plasmid, and 2) serial subculture without antibiotics for curing the killer plasmid. When the crude extract of L. citreum expressing Cas9 and the guide RNA was incubated with a PCR fragment containing the specific sequence recognized by the guide RNA, the PCR fragment was cleaved. Also, the cryptic plasmid pCB42 was successfully eliminated from the host strain after transforming the plasmid harboring Cas9 and the guide RNA. The Cas9 and gRNA expression plasmid used in this study can be applied for genome engineering purposes by additionally introducing an editing DNA template to repair the double strand DNA breakage caused by Cas9 in the genome of L. citreum. This study demonstrates the feasibility of developing CRISPR/Cas9-based genetic engineering tools to develop a safe host strain and construct food-grade lactic acid bacteria without residual antibiotic markers.</P>

<P>Sol-gel-derived, crack-free, and condensed TiOx thin films with improved barrier properties were successfully fabricated on polymeric substrates with a simple two-step heat treatment at low temperatures. To assess the barrier properties of the TiOx thin films, Ca corrosion tests were conducted and their water vapor transmission rates (WVTRs) were measured. We found that the two-step heat treatment (at 45 degrees C for 90 min and 110 degrees C for 60 min) produces a close-packed TiOx structure that substantially reduces the WVTRs of the coated polymeric substrates. The WVTRs of 86 nm thick TiOx thin films on polyethylene naphthalate (PEN) substrates at a relative humidity (RH) of 90% were found to be 0.133 g m(-2) day(-1) at 38 degrees C and 0.0387 g m(-2) day(-1) at 25 degrees C. In addition, the WVTR value of the TiOx thin films on PEN substrates are stable with respect to bending: it was found to increase by only similar to 13% after 100 repetitions of bending with a 20 mm radius. <P><B>Abstract</B></P> <P>To identify pathogenic variants in 107 Korean patients with sporadic frontotemporal dementia (FTD), 46 genes related to FTD, amyotrophic lateral sclerosis, and other dementias were screened by next-generation sequencing. Hexanucleotide repeats in <I>C9orf72</I> gene were also tested by repeat-primed polymerase chain reaction. Next-generation sequencing revealed one known pathogenic variant (c.708+1G>A) in the <I>GRN</I> gene in a patient with behavioral variant FTD (bvFTD). In addition, a novel in-frame deletion (c.2675_2683del) in the <I>CSF1R</I> gene was identified in a patient with bvFTD who had severe bifrontal atrophy with frontal subcortical white matter changes. Novel compound heterozygous variants in the <I>AARS2</I> gene, c.1040+1G>A and c.636G>A (p.Met212Ile), were found in a patient with bvFTD. Forty-six variants of uncertain significance were detected in other patients. None of the patients had expanded hexanucleotide repeats in <I>C9orf72</I>. These results show that pathogenic variants of known FTD genes are rare in Korean FTD patients but the <I>CSF1R</I> and <I>AARS2</I> genes should be screened for a genetic diagnosis of FTD or other dementias.</P>

<P><B>Abstract</B></P> <P>Bi<SUB>2</SUB>TeO<SUB>5</SUB> has been obtained in polycrystalline form via solid state reaction and structurally characterized from both synchrotron and neutron powder diffraction data, enabling the precise determination of the atomic positions. The crystal structure is defined in the acentric <I>Amb</I>2 space group, containing three crystallographically unequivalent Bi atoms. The position of the stereochemically active electron lone pairs of both Bi<SUP>3+</SUP> and Te<SUP>4+</SUP> ions is inferred to be opposite to the strongly covalent BiO and TeO chemical bonds; the global electron polarization arises from the non-compensated Bi<SUP>3+</SUP> lone electron pairs. Additionally, from difference Fourier maps between synchrotron and neutron diffraction data (X-N technique) it was possible to observe experimental evidence of the lone electron pair for Bi<SUP>3+</SUP>. This study was tested for the bending stress analysis of hand splint for Hemiplegic Patients. Hemiplegia was represented the stroke and the cerebral palsy. Hand splints of four materials was an Orthoplast hand splint, a Polypropylene hand splint, a Yogips hand splint and an Aluminum hand splint. The Stress Analysis system was made by the electronic oscilloscope, strain gage sensors, amplifier, A/D converter, PC with C program. It will be able to using the important data in splint design. The results of analysis were obtained various different graphes and maximum data in an Orthoplast hand splint, a Polypropylene hand splint, a Yogips hand splint and an Aluminum hand splint.

Amendments of a recirculating nutrient solution with potassium silicate were evaluated as a means to control Phytophthora capsici infections on pepper plant(Capsicum annuum L.). Supplying the solutions with 100 or 200 ppm of silicate significantly reduced motility, root decay, and yield losses attributed to infection of P. capsici. Treating inoculated plants with potassium silicate increased root dry weights and number of fruit, especially high-grade fruit. Results were slightly superior to non-inoculated controls. The two varieties, PBC 137 and PBC 602, responded similarly to the treatments. No significant differences were observed between the 100- and 200 ppm silicate treatments. Results were better when greenhouse conditions favored the spread of P. capsici. Silicon alone did not increase pepper yield, suggesting that it acts as a disease suppression agent rather than as a fertilizer, The phenomena by which silicon confers protection against P. capsici infection and disease development are not fully understood, but our results indicate that mechanisms other than a mechanical barrier to fungal penetration are involved.

Dendritic cells (DCs), the most abundant antigen-presenting cells in the lung, have been drawing attention for their roles in specific allergic responses to aeroallergens with support of Th lymphocytes, and in persistent inflammatory changes in allergic asthma. To identify genetic factors that may be involved in the asthma susceptibility and development of the disease phenotypes, we examined association of DC-specific DCNP1 polymorphisms with the disease risk. The case–control study revealed association of the nucleotide variants with serum immunoglobulin E (IgE) levels specific for Dermatophagoides farinae (Der f 1) and Dermatophagoides pteronyssinus (Der p 1), major aeroallergens of dust mites, among subjects with asthma. In particular, the T-allele-carrying genotype frequencies for one of the variants (c.−1289C>T) located in the promoter region were found increased in the asthmatic group with low levels of the mite-specific IgE (odds ratio (OR)=0.63 (0.48–0.83) for Der p 1). Results from functional analyses indicated that the promoter variant would affect the gene expression by modulating DNA–protein interaction. We propose that the genetic polymorphism of DCNP1 may influence production of specific IgE by altering DC functions in the mite allergen presenting and/or processing.
