5-Lipoxygenase synthesizes leukotrienes from arachidonic acid. We developed three novel 5-LO inhibi-tors having a benzoxazole scaffold as a potential anti-osteoclastogenics. They significantly suppressed RANKL-induced osteoclast formation in mouse bone marrow-derived macrophages. Furthermore, one compound. K7, inhibited the bone resorptive activity of osteoclasts. The anti-osteoclastogenic effect of K7 was mainly attributable to reduction in the expression of NFATc1. an essential transcription factor for osteoclast differentiation. K7 inhibited osteoclast formation via ERK and p38 MAPK. as well as NF-KB signaling pathways. K7 reduced lipopolysaccharide (LPS)-induced osteoclast formation in vivo. corroborating the in vitro data. Thus, IG exerted an inhibitory effect on osteoclast formation in vitro and in vivo, properties that make it a potential candidate for the treatment of bone diseases associated with excessive bone resorption.

Regulation of proper gene expression is important for cellular and organismal survival, main- tenance, and growth. Abnormal gene expression, even for a single critical gene, can thwart cellular integrity and normal physiology to cause diseases, aging, and death. Therefore, gene expression profiling serves as a powerful tool to understand the pathology of diseases and to cure them. In this study, the difference in gene expression in Flammulina velutipes was compared between the wild type (WT) mushroom and the mutant one with clogging phe- nomenon. Differentially expressed transcripts were screened to identify the candidate genes responsible for the mutant phenotype using the DNA microarray analysis. A total of 88 genes including 60 upregulated and 28 downregulated genes were validated using the real- time quantitative PCR analysis. In addition, proteomic differences between the WT and mutant mushroom were analyzed using two–dimensional gel electrophoresis and matrix- assisted laser desorption/ionization-time of flight (MALDI-TOF). Interestingly, the genes iden- tified by these genomic and proteomic analyses were involved in stress response, transla- tion, and energy/sugar metabolism, including HSP70, elongation factor 2, and pyruvate kinase. Together, our data suggest that the aberrant expression of these genes attributes to the mutant clogging phenotype. We propose that these genes can be targeted to foster normal growth in F. velutipes.

<P>Osteoporosis is a disorder in which bone mass decreases and is responsible for many degenerative bone diseases. The excessive formation and activity of osteoclasts results in pathological disorders of the bone. Receptor Activator of Nuclear Factor kappa B Ligand (RANKL) is regarded as a key regulator of osteoclast activity and as a new therapeutic target for treating osteoporosis. Herein, we have synthesized several new small molecules and tested their inhibition activity on RANKL-induced osteoclast formation. The active compounds 2c and 4d showed inhibitory activity against RANKL-induced osteoclast differentiation (IC50 = 1.56 and 2.20 mu M, respectively). The most active compound 2c prevented LPS-induced osteoclastogenesis in vivo. These data imply that the compound may be the potential candidate for a new therapeutic drug for treatment of bone resorption-associated diseases.</P>

N/A Both commercially pure titanium and Ti-6AI-4V alloy have been widely used as biomaterials because of their excellent biocompatibility, corrosion resistance and mechanical properties. However, in recent years, vanadium has been found to cause cytotoxic effects and adverse tissue reactions, while aluminium has been associated with potential neurological disorders. A newly designed α+ß type Ti alloy, Ti-10Ta-10Nb alloy showed superior properties to CP Ti and Ti-6AI-4V alloy in the point of biomaterial, and elucidated the future uses as a biomaterial. Microstructural changes of Ti-10Ta-10Nb alloy after hot-rolling, warm-rolling, solution and aging treatment were investigated. According to TEM results, the microstructures after solution treatment were composed of mostly α phase with a trace of ß phase due to adding ß-phase stabilizer tantalum and niobium. The microstructures after warm-rolling is coarse and elongated a phase and hot rolling resulted in very fine a widmansta¨tten. The highest value of hardness was obtained by aging treatment at 400℃ for 20hr in which microstructure consisted of very fine a phase in β matrix.

This study examined the relationships between parenting stress, parenting sense of competence, and parenting behavior in Korean and Japanese mothers of early school-aged children. A sample of 166 Korean mothers and 112 Japanese mothers completed questionnaires. First, in terms of results, Korean mothers reported higher scores in parenting stress, parenting sense of competence, and warmth and neglect than Japanese mothers did. In contrast, Japanese mothers scored higher on intrusiveness than Korean mothers. Second, for both groups, parenting stress was negatively related to warmth and reasoning, and positively related to intrusiveness, coercion, and neglect. Third, for both groups, parenting sense of competence was positively linked to warmth and reasoning, and negatively linked to intrusiveness, coercion, and neglect. Fourth, parenting stress was negatively associated with parenting sense of competence for both groups. Finally, parenting sense of competence mediated the associations between parenting stress and warmth for both groups. Only in the case of Korean mothers did parenting stress influence reasoning and intrusiveness through parenting sense of competence. These results imply that there may indeed be cross-cultural similarities and differences in three parenting-related variables between Korean and Japanese mothers.

The microstructure, chemical composition, and lead isotope ratio of the Buddhist bell of Yongmoon Mountain Sangwonsa temple, which was selected as one of the three great bells of Korea by Japanese historians, were analyzed in order to estimate the origin of the material and the time of casting. The microstructure of the temple bell was composed of a copper matrix phase with α, a face centered cubit lattice structure, a δ phase with Cu41 (Sn, Ag, Sb)11 as the chemical structural formula, dispersed lead and Cu2S particles, and locally agglomerated fine particles. Through analysis of the chemical composition of the bell, a criterion (Pb: 0-3.0 wt%, Sn: 10-15 wt%) for distinguishing the bells of the Shilla dynasty from the bells of the Koryo Chosun dynasty is proposed, Examining the lead isotope ratio of 207Pb / 206Pb and 208Pb / 206Pb of the Buddhist bell of Sangwonsa temple proved that the bell was fabricated using raw materials in South Korea, which led to the conclusion that the bell was cast in Korea and the top board of the bell has been damaged by an unknown individual. The criteria of distinguishing the bells from the Shilla dynasty from the bells of the Koryo Chosun dynasty presented for the first time in this research is expected to aid in identifying and estimating the previously unclear production years of other bells.
Effect of Liuweidihuang-tang on Alcohol-induced Decreasse in New Cell Formation in Rat Dentate Gyrus

Liuweidihuang-tang has been traditionally used for the treatment of delayed mental and physical development in children, complications of diabetes, and glomerulonephritis. In the present study, the effect of Liuweidihuang-tang on cell proliferation in the dentate gyrus of alcohol-intoxicated Sprague-Dawley rats was investigated via 5-bromo-2’-deoxyuridine (BrdU) immunohistochemistry. Alcohol administration was shown to decrease the number of BrdU-positive cells in the dentate gyrus, while Liuweidihuang-tang treatment increased new cell formation in the dentate gyrus under normal conditions and alcohol intoxication. It was thus demonstrated that aqueous extract of Liuweidihuang-tang exerts a protective effect against alcohol-induced decrease in new formation.

The shear strength and creep properties of Sn-57 wt.%Bi/Cu and Sn-57 wt.%Bi-0.5 wt.%Zn/Cu joints were studied. Modified double lap shear solder joints were aged for 60 days at 80℃ and then loaded to failure in shear. The shear strength of solder/Cu joints was decreased with aging treatment and the rapid decrease in shear strength of the Sn-57 wt.%Bi/Cu joint was appeared in the 60 days aged specimen. The crack path of aged solder/Cu joint were changed from through the solder to through the interface between Cu substrate and reaction layer with Zn addition. But the crack path under creep test was through the solder regardless of Zn addition.
