In this study, we screened molecules from marine natural products that stimulate osteoblast differentiation. We found that phorbaketal A significantly stimulates osteoblast differentiation in mesenchymal cells. Increased interaction of TAZ and Runx2 stimulated phorbaketal A-induced expression of osteoblastic marker genes. The activation of ERK was important for the stimulation of differentiation because an inhibitor of ERK blocked phorbaketal A-induced osteogenic differentiation.         SRC activates TAZ for intestinal tumorigenesis and regeneration

        <P><B>Abstract</B></P> <P>Proto-oncogene tyrosine-protein kinase Src (cSRC) is involved in colorectal cancer (CRC) development and damage-induced intestinal regeneration, although the cellular mechanisms involved are poorly understood. Here, we report that transcriptional coactivator with PDZ binding domain (TAZ) is activated by cSRC, regulating CRC cell proliferation and tumor formation, where cSRC overexpression increases TAZ expression in CRC cells. In contrast, knockdown of <I>cSRC</I> decreases TAZ expression. Additionally, direct phosphorylation of TAZ at Tyr316 by cSRC stimulates nuclear localization and facilitates transcriptional enhancer factor TEF-3 (TEAD4)-mediated transcription. However, a TAZ phosphorylation mutant significantly decreased cell proliferation, wound healing, colony forming, and tumor formation. In a CRC mouse model, <I>Apc</I> <SUP> <I>Min/+</I> </SUP>, activated SRC expression was associated with increased TAZ expression in polyps and TAZ depletion decreased polyp formation. Moreover, intestinal TAZ knockout mice had intestinal regeneration defects following γ-irradiation. Finally, significant correspondence between SRC activation and TAZ overexpression was observed in CRC patients.         Effect of fermented sarco oyster extract on age induced sarcopenia muscle repair by modulating regulatory T cells

        Sarcopenia is an age-related, progressive skeletal muscle disorder involving the loss of muscle mass and strength. Previous studies have shown that γ-aminobutyric acid (GABA) from fermented oysters aids in regulatory T cells (Tregs) cell expansion and function by enhancing autophagy, and concomitantly mediate muscle regeneration by modulating muscle inflammation and satellite cell function. The fermentation process of oysters not only increases the GABA content but also enhances the content of branched amino acids and free amino acids that aid the level of protein absorption and muscle strength, mass, and repair. In this study, the effect of GABA-enriched fermented sarco oyster extract (FSO) on reduced muscle mass and functions via Treg modulation and enhanced autophagy in aged mice was investigated. Results showed that FSO enhanced the expression of autophagy markers (autophagy-related gene 5 [ATG5] and GABA receptor-associated protein [GABARAP]), forkhead box protein 3 (FoxP3) expression, and levels of anti-inflammatory cytokines (interleukin [IL]-10 and transforming growth factor [TGF]-β) secreted by Tregs while reducing pro-inflammatory cytokine levels (IL-17A and interferon [IFN]-γ). Furthermore, FSO increased the expression of IL-33 and its receptor IL-1 receptor-like 1 (ST2); well-known signaling pathways that increase amphiregulin (Areg) secretion and expression of myogenesis markers (myogenic factor 5, myoblast determination protein 1, and myogenin). Muscle mass and function were also enhanced via FSO. Overall, the current study suggests that FSO increased autophagy, which enhanced Treg accumulation and function, decreased muscle inflammation, and increased satellite cell function for muscle regeneration and therefore could decrease the loss of muscle mass and function with aging.

        This study is designed to evaluate the effects of an oriental herbal medication namely Danggwihwahyeolsan-gamibang and Gami-Jeonssibaekchulsan, on ulcerative colitis. Subject: A patient diagnosed ulcerative colitis who're not improved through western medication. Methods: The Clinical data was analyzed on a pastient with ulcerative clitis due to Changpung(腸風) resulted from Hanyeolchakjab(寒熱錯雜), whose symptoms were combined diarrhea, abdominal pain et al. The patient visited at the internal medicine department of Daegu Haany University affiliated Daegu Oriental Hospital on May 18. 2007 to June 25. 2007. The improvement of symptoms were checked by Seo Colitis Activity Index(AI).

        (−)-Epicatechin Gallate (ECG) Stimulates Osteoblast Differentiation via Runt-related Transcription Factor 2 (RUNX2) and Transcriptional Coactivator with PDZ-binding Motif (TAZ)-mediated Transcriptional Activation

        <P>Osteoporosis is a degenerative bone disease characterized by low bone mass and is caused by an imbalance between osteoblastic bone formation and osteoclastic bone resorption. It is known that the bioactive compounds present in green tea increase osteogenic activity and decrease the risk of fracture by improving bone mineral density. However, the detailed mechanism underlying these beneficial effects has yet to be elucidated. In this study, we investigated the osteogenic effect of (−)-epicatechin gallate (ECG), a major bioactive compound found in green tea. We found that ECG effectively stimulates osteoblast differentiation, indicated by the increased expression of osteoblastic marker genes. Up-regulation of osteoblast marker genes is mediated by increased expression and interaction of the transcriptional coactivator with PDZ-binding motif (TAZ) and Runt-related transcription factor 2 (RUNX2). ECG facilitates nuclear localization of TAZ through PP1A. PP1A is essential for osteoblast differentiation because inhibition of PP1A activity was shown to suppress ECG-mediated osteogenic differentiation. Taken together, the results showed that ECG stimulates osteoblast differentiation through the activation of TAZ and RUNX2, revealing a novel mechanism for green tea-stimulated osteoblast differentiation.</P>

        Cyclosporine 0.05% Ophthalmic Emulsion for Dry Eye in Korea: A Prospective, Multicenter, Open-Label, Surveillance Study

        <P><B>Purpose</B></P><P>To assess the effectiveness and tolerability of cyclosporine ophthalmic emulsion (CsA) 0.05% in patients with moderate to severe dry eye disease in Korea.</P><P><B>Methods</B></P><P>This was a prospective, multicenter, open-label, surveillance study of 392 Korean patients with moderate to severe dry eye disease who were treated with CsA 0.05% for three months. An assessment of effectiveness was performed at baseline, and after 1, 2, and 3 months. The primary effectiveness outcomes were changes in ocular symptoms and Schirmer score. The secondary effectiveness outcomes were a change in conjunctival staining, use of artificial tears, global evaluation of treatment, and patient satisfaction. The primary safety outcome was the incidence and nature of adverse events.</P><P><B>Results</B></P><P>A total of 362 patients completed the study. After three months, all ocular symptom scores were significantly reduced compared to the baseline values, while the Schirmer scores were significantly increased relative to baseline (<I>p</I> < 0.0001). After three months, there were significant reductions from baseline in conjunctival staining (<I>p</I> < 0.01) and use of artificial tears (<I>p</I> < 0.0001). According to clinicians' global evaluations, most patients (>50%) experienced at least a 25% to 50% improvement in symptoms from baseline at each follow-up visit. The majority of patients (72.0%) were satisfied with the treatment results, and 57.2% reported having no or mild symptoms after treatment. The most common adverse events were ocular pain (11.0%).</P><P><B>Conclusions</B></P><P>Our findings indicate that CsA 0.05% is an effective and tolerable treatment for dry eye disease in Korean clinical practice.</P>

        내장을 첨가한 전복죽의 품질 특성

        전통적 기호식품 및 건강식품인 전복죽 제조 시 쌀 형태에 따른 내장 첨가량을 결정하기 위해 1%, 2%, 3%, 4%, 5%의 내장 첨가량에 따른 이화학적 특성, 물성 및 관능적 품질특성을 연구하였다. 이화학적 특성에서, pH에서는 쌀입자 크기와 내장첨가량에 따른 차이는 보이지 않았으나 반쌀죽과 쌀가루죽에서는 내장첨가량이 증가할수록 약간의 감소경향을 보였다(p<0.05). 산화도는 전반적으로 낮은 수치를 보였으며 온쌀죽>반쌀죽>쌀가루죽의 순으로 유의적(p<0.05)으로 산화도가 높게 나타났고 반쌀죽을 제외한 온쌀죽과 쌀가루죽에서는 내장의 첨가량이 증가함에 따라 증가하는 경향을 보였다. 색도에서, 명도 및 적색도는 유의적인 차이는 없었으나 황색도는 내장첨가량이 증가수록 유의적(p<0.05)으로 증가하였으며 온쌀죽>반쌀죽>쌀가루죽 순으로 높게 나타났다. 물성에서, 쌀입자 크기에 따른 물성의 차이는 유의적(p<0.05)으로 나타나 점조성과 견고성은 온쌀죽>반쌀죽>쌀가루죽 순으로 높게 나타났고 점성과 응집성은 쌀가루죽>반쌀죽>온쌀죽 순으로 높게 나타났다. 내장첨가량에 따른 물성을 살펴본 결과, 온쌀죽에서 내장첨가량이 증가할수록 점조성과 견고성이 다소 감소하는 경향을 보였으나 전반적으로 유의적인 차이는 없었다. 쌀입자 크기와 내장첨가량에 따른 관능평가를 실시한 결과, 식전 냄새를 제외한 나머지 관능특성에서 모두 유의적(p<0.05)인 차이를 보였다. 쌀입자 크기에 따른 관능특성에서 온쌀죽과 반쌀죽이 쌀가루죽보다 질감, 맛 및 전체적인 관능점수에서 유의적(p<0.05)으로 높은 점수를 보였으며 내장첨가량에 따른 관능특성에서는 온쌀죽은 내장 2~5% 첨가, 반쌀죽은 내장 3~5% 첨가구에서 유의적(p<0.05)으로 높은 점수를 보였으나 쌀가루죽은 내장 1~2% 첨가구에서 유의적(p<0.05)으로 낮은 점수를 보였다. 이상의 연구 결과를 통해 온쌀은 2%, 반쌀은 3%, 쌀가루는 4%의 내장을 첨가하여 제조한 전복죽이 이화학적, 물성적 및 관능적으로 우수한 것으로 나타났다. To develop an optimal composite recipe for a functional abalone porridge including abalone viscera, this study investigated the effects of added viscera on the physical, textural and sensory characteristics of the porridge. Several kinds of abalone porridge were prepared with 1%, 2%, 3%, 4%, or 5% of the viscera (w/w) and with round rice, half rice, or ground rice. pH of the porridge with half and ground rice decreased with increasing amounts of the viscera. TBARS value increased with increasing size of rice and increased with increasing amount of the viscera in the porridge with round and ground rice. Yellowness of the porridge increased significantly by the addition of the viscra. Among the four textural properties, consistency and firmness increased with increasing size of rice; in contrast, viscosity and cohesivness decreased with increasing size of rice. However, textural properties of the porridge were not significantly different by the addition of the viscera. In the sensory evaluation, sensory scores of the porridge with round and half rice were higher than with round rice for texture, taste and total. In conclusion, concerning overall sensory evaluation, round rice porridge with 2% viscera, half rice porridge with 3% viscera and ground rice porridge with 4% viscera showed the best results, implying that developing functional abalone porridge using the viscera may be worthy.

        Hepatoprotective Effects of GongJin-dan, on Ethanol-mediated Experimental Liver Damage in Rats

        Background : A traditional Oriental medicine, GongJjn-dan (GJD), is one of the most well-known tonic agents in Korea. Among 6 types of GJD components, antler, red ginseng, and Cornus fructus have shown antioxidant effects, while EtOH-induced tissue damage may be a consequence of oxidative stress. Objectives & Methods : The hepatoprotective effects of GJD were evaluated on EtOH-mediated experimental liver damaged rats at 50, 100, 250 and 500mg/kg comparing with 100mg/kg of silymarin as a reference drug in the present study. Test substances were dosed once a day for 60 days with oral administration of 20% ethanol 2.5ml/100g body weight twice a day (equivalent to 7.9g ethanol/kg/day). Each of 8 rats per group was selected using body weight at 10 days after acclimatization. Experimental animals were sacrificed after 60 days of continuous oral treatment of test substances with 20% ethanol treatment, and changes on the body weight, liver weight, and serum AST and ALT were observed. Results : There were dramatic decreases of body weight and increases of liver weight and serum AST and ALT. Similar inhibition effects on the EtOH-induced hepatic damages were detected between equal dosages of GJD and silymarin. Conclusion : Based on these results. it is concluded that GJD showed clear hepatoprotective effects on EtOH-induced hepatic damage.
