The objective of this study was to characterise the pattern of p53 mutations in bladder turmor. In this study, 25 bladder transitional cell carcinomas were analyed by immunohistochemistry (IHC) for p53 nuclear overexpression, and the results were compared with those of polymerase chain reaction-single strand conformational polymorphism (PCR-SSCP) analysis in exon 5-8 of the p53 gene and DNA sequencing analysis. 15 out of 20 cases (75%) showed p53 nuclear immunoreactivities on IHC. On PCR-SSCP analysis, 10 out of 25 cases(40%) had abnormal shifts on mobility. 62% of the mutations were in exon 8. Direct DNA sequencing analysis were performed in these 10 cases to confirm the presence of mutated p53 genes and to determine the type of mutations. Sixteen point mutations were detected in 10 cases. Tow specimens had double mutations and another two had triple mutations. G:C→A:T transitions were the most frequent patterns (62.5%). One mutation was a premature stop codon and two were silent mutations. There out of 10 had a point mutation at codon 285 (GAG/Glu→AAG/Lys) and two had at codon 280(GAG/Glu→AAG/Lys). One of 16 mutations was transition at hot spot codon 273 with CpG site. These results suggest that altered expressions and point mutations of p53 occured in all grade of bladder cancer, but are more associated with hight grade bladder tumors. To elucidate the carcinogenesis of bladder cancer, further studies should be carried out.

Purpose: This study was peformed to investigate whether the free to total PSA ratio(F/T PSA ratio) offers a better discriminant for the detection of patient with prostatic cancer(CaP) and those with benign prostatic hyperplasia(BPH) than serum total PSA alone. Materials and Methods: From September 1996 to February 1999, free and total PSA levels were measured on frozen sera samples of 154 men with BPH and 46 with CaP using immunoradiometric assay. Results: In all subjects, the mean total PSA was significantly higher in CaP group(72.57±66.58ng/ml) than BPH group(3.12±4.10ng/ml). The mean PSAD was significantly higher in CaP group(1.64± 1.86) than BPH group(0.08±0.01). The mean F/T PSA ratio was significantly lower in CaP group(0.19± 0.07) than BPH group(0.30±0.13). Among 41 subjects whose total PSA were between 4-20ng/ml, the mean PSAD was significantly higher in CaP group(0.37±7.16) than BPH group(0.17± 0.07), but the mean F/T PSA ratio was not significantly different between CaP(0.19± 0.10) and BPH group(0.25±7.13). By comparing the sensitivity and specificity of total PSA alone, PSAD and F/T PSA ratio on receiver operating characterislic (ROC) Cutie in all subiects and subiects with total PSA between 4-20ng/ml, F/T PSA ratio had no superiority than total PSA alone and PSAD for discrimination between CaP and BPH. Conclusions: We suggest that the F/T PSA ratio is not superior to total PSA alone and FSAD in the detection of prostatic cancer, and further evaluation of the usefulness of F/T PSA ratio should be required.

Diltiazem and nifedipine, have been used throughout the world as an effective antianginal and antihypertensive agent. In order to increase the activity of these drugs, we synthesized the diltiazem-nifedipine coupling compounds. These compounds^(1-8) were tested for antihypertensive effect. Unfortunately, these compounds didn't show any promising activities.
Synthesis of surface-tuned polyacrylonitrile particles and its application to CO2 separation

We synthesized the surface-tuned polyacrylonitrile (PAN) colloid particles as sulfate ions with a size of75.6 nm. The synthesized PAN particles were introduced into a polymeric membrane for CO2/N2 separation. Poly(ethylene oxide) (PEO) was used as the polymer matrix. The PEO/PAN colloid composite membranesshowed an improved ideal CO2 selectivity of 23, whereas the PEO membrane showed a selectivityof 6.7. This improved separation performance is attributable to the difference in solubilities of CO2 and N2due to the presence of polar groups on the colloid surfaces, resulting in the enhanced separationperformance.

Conductive paste inks have received considerable attention as facile conductive materials for theformation of electrode layers. However, conventional paste inks result in fi lms with poor surfacemorphology. In addition, they require a high thermal annealing temperature for achieving high electricalconductivity because of their organic/inorganic composite structure, in which nanosized metalparticles and polymeric organic binders are mixed in solvents. In this work, we prepare solvent-freeand polymeric-binder-free metal nanoparticle (NP) fl uids, which can be used as facile conductivepastes for forming an electrode layer after sintering at a considerably low temperature. We employthiol-terminated imidazolium-type ionic liquid (IL-SH) molecules with a small molecular weight andfl uidic behavior as the surface ligands of Ag NPs. IL-SH-stabilized Ag NPs exhibit fl uidic behaviorand metallic conducting properties at a considerably low sintering temperature of 250 °C.

Next-generation and solution-processed thin-fi lm solar cells have been attracted considerable attention because of theirlow cost, light weight, fl exibility, and aesthetics. However, most of solution-processed thin-fi lm solar cells are now focusedon the use of photovoltaic absorbers containing the toxic element of Pb. In this study, eco-friendly silver-bismuth-iodide(Ag-Bi-I) thin-fi lm photovoltaic devices with high open-circuit voltages ( V OC ) are developed by utilizing polythieno[3,4- b ]-thiophene-co-benzodithiophene (PTB7) as the hole transport layer (HTL). The solution-processed AgBi 2 I 7 semiconductor,which is an Ag-Bi-I ternary compound, exhibit features suitable for photovoltaic layers in thin-fi lm solar cells, including athree-dimensional (3D) crystal structure, good surface morphology, and low optical bandgaps of 1.87 eV. Meanwhile, thesolution-processed AgBi 2 I 7 thin-fi lm solar cell based on the PTB7 HTL exhibit a power conversion effi ciency of 0.94% withan improved V OC value of 0.71 V owing to the deeper highest occupied molecular orbital (HOMO) energy level comparedto that of poly(3-hexylthiophene-2,5-diyl) (P3HT). In other words, the V OC of the PTB7 HTL-based device is 20% higherthan that of the P3HT HTL-based control device. Our results provide a new approach for increasing the V OC of eco-friendlyAg-Bi-I thin-fi lm photovoltaics and indicate that further HTL engineering is necessary to simultaneously improve the V OCand performance of the devices.
Hypoxia Enhances Cell Properties of Human Mesenchymal Stem Cells

<P>Atmospheric (in vitro) oxygen pressure is around 150 mm Hg (20% O-2), whereas physiologic (in vivo) oxygen pressure ranges between 5 and 50 mm Hg (0.7-7% O-2). The normoxic environment in cell culture does not refer to a physiological stem cell niche. The aim of this study is to investigate the effect of oxygen concentration on cell properties of human mesenchymal stem cells (MSCs). We analyzed cell proliferation rate, senescence, immunophenotype, stemness gene expression and differentiation potency with human urine stem cells (USCs), dental pulp stem cells (DPSCs), amniotic fluid stem cells (AFSCs), and bone marrow stromal cells (BMSCs). USCs, DPSCs, AFSCs and BMSCs were cultured under either 5% O-2 hypoxic or 20% O-2 normoxic conditions for 5 days. MSCs cultured under hypoxia showed significantly increased proliferation rate and high percentage of S-phase cells, compared to normoxic condition. In real-time PCR assay, the cells cultured under hypoxia expressed higher level of Oct4, C-Myc, Nanog, Nestin and HIF-1 alpha. In immunophenotype analysis, MSCs cultured under hypoxia maintained higher level of the MSC surface markers, and lower hematopoietic markers. Senescence was inhibited under hypoxia. Hypoxia enhances osteogenic differentiation efficiency compared to normoxia. Hypoxia showed enhanced cell proliferation rate, retention of stem cell properties, inhibition of senescence, and increased differentiation ability compared to normoxia.</P>
