      • Conjugated Linoleic Acid Changes fatty Acid Composition by Decreasing Monounsaturated fatty Acids in Rabbits and Hep G2 Cells

        Conjugated dienoic derivatives of linoleic acid(CLA) are a mixture of positional and geometric isomers of linoleic acid(LA). We previously found that CLA changes the fatty acid profile in chicken eggs and serum by decreasing monounsaturated fatty acids. Studies were conducted to explore the effects of CLA on fatty acid composition. Rabbits were fed a semisynthetic diet with or without CLA(0.5g CLA/rabbit/day) for 22 weeks. Compared to the control, rabbits fed CLA had significantly lower monounsaturated fatty acid levels(palmitoleic acid Cl6 : 1 by 50% and oleic acid Cl8 : 1, by 20%) in plasma lipids. We found similar differences in fatty acid composition in the liver and the aorta. The inhibitory effect of CLA on $\Delta$9 desaturation was confirmed in a human hepatoma cell line, Hep G2. CLA significantly decreased $\Delta$9 desaturation in 4-5 hours as shown by an increase in the ratio of Cl6 : 0 to C 16 1, This is apparently due to a decrease in $\Delta$9 desaturase(stearoyl-CoA desaturase, SCD) activity ; it was decreased more than 50%. These results, along with our previous findings, indicate that CLA is an inhibitor of $\Delta$9 desaturase in the liver.

        Effect of nutrient composition in a mixed meal on the postprandial glycemic response in healthy people

        BACKGROUND/OBJECTIVES: The glycemic index (GI) is a measure of the postprandial glucose response (PPGR) to food items, and glycemic load (GL) is a measure of the PPGR to the diet. For those who need to maintain a healthy diet, it is beneficial to regulate appropriate levels of blood glucose. In reality, what influences the meal GI or GL depends on the macronutrient composition and the physical chemistry reactions in vivo. Thus, we investigated whether different macronutrients in a meal significantly affect the PPGR and the validity of calculated GI and GL values for mixed meals. SUBJECTS/METHODS: 12 healthy subjects (6 male, 6 female) were recruited at a campus setting, and subjects consumed a total of 6 test meals one by one, each morning between 8:00 and 8:30 am after 12 h of fasting. PPGR was measured after each consumed meal and serial finger pricks were performed at indicated times. Test meals included 1) 68 g oral glucose, 2) 210 g rice, 3) rice plus 170 g egg white (RE), 4) rice plus 200 g bean sprouts (RS), 5) rice plus 10 g oil (RO), and 6) rice plus, egg white, bean sprouts, and oil (RESO). The incremental area under the curve (iAUC) was calculated to assess the PPGR. Mixed meal GI and GL values were calculated based on the nutrients the subjects consumed in each of the test meals. RESULTS: The iAUC for all meals containing two macronutrients (RS, RO, or RE) were not significantly different from the rice iAUC, whereas, the RESO iAUC (2,237.5 ± 264.9) was significantly lower (P < 0.05). The RESO meal’s calculated GI and GL values were different from the actual GI and GL values measured from the study subjects (P < 0.05). CONCLUSIONS: The mixed meal containing three macronutrients (RESO) decreased the PPGR in healthy individuals, leading to significantly lower actual GI and GL values than those derived by nutrient-based calculations. Thus, consuming various macronutrient containing meals is beneficial in regulating PPGR.

        Effect of a Nutritionally Balanced Diet Comprising Whole Grains and Vegetables Alone or in Combination with Probiotic Supplementation on the Gut Microbiota

        Dysbiosis is a microbial imbalance, which often causes diseases and can be triggered by diet. Here, we determined the effect of a nutritionally balanced diet rich in vegetables and whole grains alone and/or in combination with probiotics on the gut microbiota of healthy adults. We conducted a parallel-group randomized trial enrolling 63 healthy participants who were administered either a balanced diet (B-diet group), a probiotic capsule containing Lactobacillus plantarum PMO 08 (probiotics group), or a balanced diet plus probiotic capsule (synbiotics group) once daily for 2 weeks. The gut microbiota of each participant was analyzed via 16S ribosomal RNA MiSeq-based sequencing. Gastrointestinal symptoms and defecation habits were evaluated using questionnaires. The B-diet group showed significantly reduced Firmicutes-to-Bacteroidetes ratio (P<0.05) and abundances of the genera Blautia (P<0.01), Dorea (P<0.05), and Lachnoclostridium (P<0.05). Furthermore, the abundance of Bacteroides increased (P<0.05) compared to baseline levels. In the synbiotics group, Lactobacillus abundance increased significantly (P<0.05) and defecation difficulty decreased (P<0.05), confirming a synergistic effect of combined intake. All groups showed a significant reduction in the abundance of Clostridiaceae (P<0.001) and alleviation of bloating symptoms (P<0.05). Moreover, the relative abundance of Faecalibacterium significantly increased in the probiotics group (P<0.05). Therefore, the individual or combined intake of a nutritionally balanced diet and L. plantarum PMO 08 beneficially modifies the gut microbiota with the potential to alleviate gastrointestinal symptoms and improve defecation habits.

        식단의 당부하량에 따른 20대 성인의 체중 감량 효과 연구

        본 연구에서는 한국인이 평소 섭취하는 일반식사와 유사한 수준의 당부하량을 가진 high GL식사와 당부하량이 낮게 설계된 low GL 식사를 섭취한 후 체중, 체지방, 혈압 및 혈액 내 생활습관병 위험 지표 등의 변화를 분석 및 비교하였다. 20대 성인남녀를 대상으로 체중 감량을 위해 wash-out 기간인 중간 2주를 제외하고 개인 필요 열량의 30%를 줄인 low GL 식사와 high GL 식사를 각 2주간 제공하였다. Low GL 식사와 high GL 식사의 효과를 비교한 결과 low GL식사가 high GL 식사에 비해 체중 (p < 0.001), 체지방량 (p = 0.024), 체질량지수 (p < 0.001)에 서 유의적으로 감소량이 더 컸다. 하지만 혈압, 혈액 내 비만 질환 위험 지표에서는 유의한 차이가 없었다. Low GL 식사의 전·후를 비교한 결과 체중, 체지방량, 체질량지수, 체지방률, 이완기 혈압, 중성지방, 총콜레스테롤, HDL-콜레스테롤, LDL-콜레스테롤, 공복혈당, C-peptide HOMA 지수가, high GL 식사에서는 체중, 체지방량, 체질량지수, 총콜레스테롤, HDL-콜레스테롤, 공복혈당, C-peptide, C-peptide HOMA 지수에서 각 식사 전보다 후에 유의적으로 감소되었다. 또한 실험 식사 섭취 전과 섭취 후를 비교한 결과 체중, 체지방량, 체질량지수, 체지방률, 혈압 (수축기, 이완기), 총콜레스테롤, HDL-콜레스테롤, LDL-콜레스테롤, 공복혈당, 당화혈색소, C-peptide, C-peptide HOMA 지수가 모두 유의적으로 감소되었다. 본 연구 결과 체중감량 및 생활습관병 위험을 줄이기 위해서는 단순히 섭취 열량을 낮추기보다는 열량과 함께 당부하량을 고려하여 섭취하는 것이 체중을 감소시키는 데에 더 효과적일 것으로 사료된다. Purpose: This study compared the effects of a high glycemic load (high GL) diet and low glycemic load (low GL) diet on the body weight, body fat, blood pressure, and blood lipid indicators. Methods: Twenty-one young adults aged between 21 and 28 years who were overweighted or obese (body mass index [BMI] between 23 and 33.5 kg/m²) before the study and after calorie reduction diets with either low GL or high GL for 2 weeks each were examined. The study was a randomized crossover design with a 2-week washout period between the 2 types of diet. The order of the low GL and high GL diet periods was randomized. The body weight, body fat, blood pressure, levels of blood lipids, fasting glucose, insulin, homeostatic model assessment (HOMA) insulin, C-peptide, and HOMA C-peptide were measured at the baseline, as well as 2, 4, and 6 weeks after starting the experiment. Results: When subjects were on the low GL diet, they lost more weight than those eating the high GL diet (mean ± SD, -2.77 ± 1.09 vs. -1.56 ± 0.78 kg; p < 0.001); there were greater decreases in body fat mass (-1.62 ± 1.19 vs. -0.88 ± 0.91 kg; p = 0.024) and BMI (-0.95 ± 0.32 vs. -0.56 ± 1.08 kg/m²; p < 0.001). On the other hand, there were no significant differences in changes in biochemical parameters, such as blood lipids and fasting glucose levels, and blood pressure. The body weight, body fat mass, BMI, percent body fat, blood pressure, cholesterol (total, low-density lipoprotein, and high-density lipoprotein), fasting glucose, C-peptide, HOMA-insulin resistance-C-peptide levels were decreased significantly at 6 weeks. Conclusion: The low GL diet may be more effective in losing body weight, body fat mass, and BMI than the high GL diet for 2 weeks in healthy young overweight or obese adults.

        어린이집 조리종사자 대상의 나트륨 저감화 행동변화단계에 따른 영양지식, 식태도, 식행동, 자아효능감 비교

        본 연구는 서울, 경기도, 충청도 지역에 소재하는 어린이집 조리종사자 333명에게 나트륨관련 영양지식, 식태도, 식행동, 자아효능감 등에 대해 설문조사하고, ‘나트륨 섭취줄이기’ 행동변화단계에 따라 ‘행동전단계 (고려전/고려/준비)’, ‘행동단계 (행동/유지)’ 두 군으로 분류하여 변수간의 차이를 분석하였다. 1) 대상자의 평균 연령은 48.5세이며 대상자의 68.4%는 중년에 해당되었다. 나트륨관련해서 식사관리에 관한 관심도는 대상자의 54.8%가 ‘매우 관심 있다’고 답하였고, 나트륨관련 식생활정보는 주로 TV/라디오 (56.6%)를 통해서 얻고 있었다. 나트륨관련 영양교육을 1회 경험한 대상자가 가장 많았고 (49.7%), 영양교육의 내용으로는 ‘나트륨을 줄이는 조리 방법’, ‘나트륨과 혈압의 관계’, ‘나트륨과 질환의 관계’ 순이었다. 2) 나트륨관련 영양 지식 총점은 평균 9.3점으로 행동단계군 (9.4점)은 행동전단계군 (9.2점)보다 점수가 조금 높았고 두 군 간의 유의적인 차이는 없었다. 문항별로 ‘영유아기의 나트륨 섭취의 중요성’, ‘가공식품과 자연식품의 나트륨 함량 비교’에서 대상자들은 매우 높은 정답률을 보였고, ‘성인의 1일 나트륨 목표섭취량’ (27.0%), ‘영양성분표의 나트륨 함량 산출’ (30.3%)에서 정답률이 상당히 저조하였다. 3) 나트륨 저감화에 관한 식태도 총점은 39.6점으로 행동단계군 (40.3점)은 행동전단계군 (36.6점)보다 식태도가 긍정적이었다 (p < 0.001). 행동단계군은 식태도 항목 ‘음식은 싱겁게 먹어야 함’, ‘짭짤한 맛이 좋음’, ‘식사 시 국이나 찌개를 먹는 것을 좋아함’, ‘식사 시 김치나 장아찌를 먹는 것을 좋아함’, ‘국수, 라면 등 국물류를 좋아함’에서 행동전단계군과 유의적인 차이를 보였고 (p < 0.001), 행동전단계군보다 바람직한 식태도에는 더 동의하고, 바람직하지 않은 식태도에는 덜 동의하였다. 4) 대상자의 식행동 총점은 49.6점으로 행동단계군(49.9점)은 행동전단계군 (48.5점)보다 식행동이 양호하였으나 유의적인 차이는 없었다. 대부분의 식행동 항목에서 행동단계군은 행동전단계군보다 양호하였는데, 일반 식행동 중 ‘잡곡밥 먹기’, ‘기름진 음식 먹기’ (p < 0.05), 나트륨관련 식행동 중 ‘간장 등 양념장을 찍어먹는 음식 먹기’, ‘짠 스낵 먹기’, ‘가공식품류 먹기’, ‘외식, 배달음식 먹기’ (p< 0.01)에서 유의적인 차이가 있었다. 5) 나트륨 저감화 방법에 관한 자아효능감 총점은 평균 57.1점으로 행동단계군 (58.2점)은 행동전단계군 (52.5점)보다 자아효능감이 높았다 (p < 0.001). 행동단계군은 대부분의 방법에서 행동전단계군보다 자아효능감이 높았는데, 특히 ‘가공식품 피하기’, ‘소금, MSG 대신 천연조미료 사용하기’, ‘음식의 간은 조리를 마친 후 하기’, ‘국그릇 크기 줄이기’, ‘저염 양념장 만들어 사용하기’ 방법에서는 두군간에 큰 차이를 보였다 (p < 0.001). 6) 식태도는 식행동, 자아효능감과 양의 상관관계 (p <0.001)를 식행동은 자아효능감과 양의 상관관계 (p <0.001)를 나타내었고, 영양 지식은 식태도, 식행동, 자아효능감과 양의 상관관계를 보였으나 유의적이지 않았다. 7) 연구 결과 어린이집 조리종사자 대상의 나트륨 저감화 영양사업의 확대가 요구되며 영양교육 등 영양사업 시행 시 나트륨 저감화 행동변화단계별로 대상자를 구분하여 접근할 필요가 있다. 즉, 행동전단계 대상자에게는 나트륨 저감화 실천의 필요성을 인식시키고 태도 변화를 위해 동기 부여 요소를 활용하며 행동단계 대상자에게는 실천이 지속되도록 자기조절 방법, 실천 방법 등을 활용하면 좀 더 효과적일 것이다. 또한 가공식품 산업계에서는 나트륨 저감화 제품 개발이 더욱 활성화됨으로써 급식에 적용 가능한 나트륨 저감화 제품이 확대되어야 할 것으로 사료된다. Purpose: The purpose of this study was to examine sodium-related nutrition knowledge, eating attitudes, eating behaviors, and self-efficacy by stages of behavioral change in reducing sodium intake among childcare center foodservice employees. Methods: Subjects (n = 333) were categorized according to two groups based on the stages of change; Pre-action stage (PA group: precontemplation/contemplation/preparation stage), Action stage (A group: action/maintenance stage). Results: A major source of sodium-related nutrition information was TV/radio (56.6%) and only 166 people (49.8%) have experienced nutrition education specific to sodium. Although the A group showed slightly higher scores for nutrition knowledge than the PA group, the difference was not significant. The percentages of correct answers for ‘daily goal of sodium intake for adults (27.0%)’, ‘calculation of sodium content in nutrition labeling (30.3%)’ were low for both groups. The A group (total score: 40.3) had more desirable eating attitudes regarding reducing sodium intake than the PA group (36.6, p < 0.001). The total score for eating behaviors was slightly higher in the A group (49.6) than in the PA group (48.5), but without statistical significance. The A group (total score: 58.2) also received higher scores for self-efficacy regarding reducing sodium intake than the PA group (52.5, p < 0.001). Conclusion: This study suggests that nutrition education for childcare center foodservice employees should be expanded and customized education should be implemented according to the stages in reducing sodium intake. It is also suggested that food companies make efforts to develop low-sodium products.
