        The objective of this article is to carry on the comparative study of the two books, Dongyisusebowon and Myungsunrok. Je-Ma Lee showed, in fact, the intellectual achievement in his book, Dongyisusebowon. And Lee showed more close relationships with Suk-Ji Han's idea. The common ideas from this comparative study of both scholars are as follows. 1. Lee understands the real character of human being according to Chu-Ztu(朱子)'s doctrine, with the supplement of Han's. 2. Lee explains the Sung-Jung(性情) in Cosmology and the theory of Human Nature, but he concentrates upon the Sung-Jung in the theory of Human Nature, especially in constitutional medicine. 3. Lee states the Tai-Geuk(太極) in the meaning of heart from the medical viewpoint, he agrees with the Han's idea. 4. I try to comprehend Myungsunrok in the Wang-Yang-Ming(王陽明)'s concept. 5. Han explains that Sung-Jung-I-Ki(性情理氣) is one concept with two aspects, Che-Yong(體用). In addition, Lee has formed his 'Chun-In-Sung-Myung(天人性命)' thought in harmonization with Han's idea and Chu-Tzu's idea.

      • Construction of Stably Transformed Bm 5 Cells by Using Autographa californica Nuclear Polyhedrosis Virus IE 0 Gene

        To construct transfurmed Bm5 cells, Autographa californica nuclear polyhedrosis virus(AcNPV) IE1 gene, an immediate early viral gene was firstly used in this study. AcNPV IE1 gene, which shares on 95.3% nucleotide sequence homology with Bombyx mori nuclear polyhedrosis virus (BmNPV) IE1 gene, was isolated and cloned into pBluescript. Neomycin gene from pKO-neo was inserted under the control of the IE1 promoter to yield pAcIE1-neo. The plasmid pAcIE1-neo was transfected into Bm5 or Sf9 cells, and neomycin-resistant cells were selected in TC100 medium containing 10% fetal bovine serum (FBS) and 1 mg/ml G4l8 for two weeks. Individual clones were picked and each was amplified for further characterization. The genomic DNA from neomycin-resistant cells was isolated and characterized by PCR using AcNPV IEI gene-specific primers and by Southern blot analysis using neomycin gene probe. We concluded that AcNPV IE1 gene was functional in B. mori-derived Bm5 cells as well as Spodoptera fugjprrda-derived Sf9 cells to produce stably-transformed insect cells

      • Endogenous Cushing`s syndrome in a patient with systemic lupus erythematosus

        Introduction: Systemic lupus erythematosus is one of the autoimmune disease which glucocorticoids are mainstay of treatment. Cushing’s syndrome caused by glucocorticoid excess which could be exogenous or endogenous. Although iatrogenic Cushing’s syndrome is most common form especially in the patient with glucocorticoid treatment, endogenous glucocorticoid excess should be under consideration because of different treatment strategy. Case Report: We describe a 51-year old woman had had a longstanding history of SLE. She was treated with steroid and cytoxan pulse therapy and plasmapheresis. Her lupus activity had remained stable for 7 years with low-dose glucocorticoid treatment. She showed excessive weight gain, easy bruising, moon face, truncal obesity, acne, and menstrual disorder. With history of a steroid therapy, iatrogenic Cushing’s syndrome was considered by priority. However endogenous Cushing’s syndrome was suspected in respect that the feature of Cushing’s syndrome had been aggravated with the long term use of minimal dose glucocorticoid. After evaluation under the impression of Cushing’s syndrome we diagnosed her as concomitant endogenous Cushing’s syndrome due to a left adrenal adenoma.(table1)The patient underwent laparoscopic left adrenalectomy. SLE was controlled with transient apply of low-dose glucocorticoid treatment and her lupus activity remains stable without glucocorticoid treatment until now. Discussions: This is the first reported case of concomitant endogenous Cushing’s syndrome onset with preexisting SLE in Korea. This case shows the importance of differential diagnosis of exogenous Cushing’s syndrome and endogenous Cushing’s syndrome in autoimmune disease patients with glucocorticoid therapy.

      • Environmental enrichment enhances synaptic plasticity by internalization of striatal dopamine transporters

        <P> Environmental enrichment (EE) with a complex combination of physical, cognitive and social stimulations enhances synaptic plasticity and behavioral function. However, the mechanism remains to be elucidated in detail. We aimed to investigate dopamine-related synaptic plasticity underlying functional improvement after EE. For this, six-week-old CD-1 mice were randomly allocated to EE or standard conditions for two months. EE significantly enhanced behavioral functions such as rotarod and ladder walking tests. In a [<SUP>18</SUP>F]FPCIT positron emission tomography scan, binding values of striatal DAT were significantly decreased approximately 18% in the EE mice relative to the control mice. DAT inhibitor administrated to establish the relationship of the DAT down-regulation to the treatment effects also improved rotarod performances, suggesting that DAT inhibition recapitulated EE-mediated treatment benefits. Next, EE-induced internalization of DAT was confirmed using a surface biotinylation assay. In situ proximity ligation assay and immunoprecipitation demonstrated that EE significantly increased the phosphorylation of striatal DAT as well as the levels of DAT bound with protein kinase C (PKC). In conclusion, we suggest that EE enables phosphorylation of striatal DAT via a PKC-mediated pathway and causes DAT internalization. This is the first report to suggest an EE-mediated mechanism of synaptic plasticity by internalization of striatal DAT. </P>

        Background : This study was conducted to determine the effects of mulching, an environmentally friendly organic cultivation method, on antioxidant compounds and growth in Codonopsis lanceolata. Methods and Results : C. lanceolata was treated by mulching with a non-woven fabric, biodegradable film, or rice husks, as well as hand weeding, and non-treatment. The growth and levels of weed control in C. lanceolata were better in plants cultivated under mulching treatments (non-woven fabric, biodegradable film, and rice husks) than under non-mulching treatments (hand weeding, non-treatment). The contents of antioxidant compounds, such as total flavonoids, phenolics, and anthocyanin, were highest under biodegradable film treatment, followed by non-woven fabric treatment, rice husks treatment, hand weeding, and non-treatment. DPPH and ABTS activity were able to identify to have differences their activity in comparison to antioxidant compounds content by solvent fractions. Mulching treatments produced a higher activity in water and ethyl ether fractions for DPPH scavenging activity and in n-butanol fractions for ABTS scavenging activity compared to other fractions, although total activity of DPPH and ABTS were not increased with mulching treatments. Conclusion : Mulching C. lanceolata with biodegradable film and non-woven fabric is an effective method for improving plant growth and inhibiting the occurrence of weeds as well as increasing antioxidant compound content and changing of antioxidant activity.

      • Teneligliptin versus sitagliptin in Korean patients with type 2 diabetes inadequately controlled with metformin and glimepiride: A randomized, double‐blind, non‐inferiority trial

        <P><B>Aim</B></P><P>To assess the efficacy and safety of add‐on therapy with the dipeptidyl peptidase‐4 inhibitor teneligliptin compared with sitagliptin in patients with type 2 diabetes (T2DM) inadequately controlled with metformin and glimepiride.</P><P><B>Materials and Methods</B></P><P>This was a phase 3, randomized, double‐blind, non‐inferiority study of adult Korean subjects with T2DM (n = 201), with HbA1c ranging from 7.0% to 11.0%, on stable doses of metformin plus glimepiride. Subjects were randomized in a 1:1 fashion to receive either oral teneligliptin 20 mg or sitagliptin 100 mg for 24 weeks. The primary endpoint was change from baseline in HbA1c.</P><P><B>Results</B></P><P>At baseline, mean age was 60.56 ± 9.41 years, body mass index was 25.23 ± 2.85 kg/m<SUP>2</SUP> and HbA1c was 8.11% ± 0.79%. At 24 weeks, both groups achieved significant reductions from baseline in HbA1c (teneligliptin, −1.03% ± 0.10% [<I>P</I> < 0.0001]; sitagliptin, −1.02% ± 0.10% [<I>P</I> < 0.0001]). The inter‐group difference was −0.01% (95% confidence interval [CI]: −0.28, 0.26; <I>P</I> = 0.9497); the upper limit of the 95% CI was within the preset limit for non‐inferiority (0.4%). There were no significant differences between groups in the proportion of patients achieving HbA1c targets, or changes from baseline in fasting plasma glucose, body weight or lipid levels at 24 weeks. Rates of adverse events (teneligliptin, n = 63 [61.76%]; sitagliptin, n = 61 [62.24%]; <I>P</I> = 0.9442) and hypoglycaemia (teneligliptin, n = 32 [31.37%]; sitagliptin, n = 28 [28.57%]; <I>P</I> = 0.6656) were similar.</P><P><B>Conclusion</B></P><P>Teneligliptin was non‐inferior to sitagliptin in the context of triple therapy for T2DM and is an important option in this setting.</P>

        Spermatozoa motility, viability, acrosome integrity, mitochondrial membrane potential and plasma membrane integrity in 0.25 mL and 0.5 mL straw after frozen-thawing in Hanwoo bull

        In the present study, we examined the effect of straw size on spermatozoa motility, viability, acrosome integrity, mitochondrial membrane potential, and plasma membrane integrity after freezing-thawing. Hanwoo semen was collected from three bulls and diluted with an animal protein-free extender, divided into two groups, namely, 10 million spermatozoa in 0.25 mL and 20 million spermatozoa in 0.5 mL straw, and cryopreserved. In Experiment 1, the motility and motility parameters of the frozenthawed spermatozoa were evaluated. After freezing-thawing, the spermatozoa motility parameters fast progressive, straight line velocity, and average path velocity were compared between the 0.25 mL straw and 0.5 mL straw groups. They were 35.2 ± 1.0 and 32.3 ± 0.7%, 34.6 ± 0.7 and 31.8 ± 0.5 μm/s, 51.4 ± 1.3 and 47.1 ± 1.1 μm/s, 0.25 mL straw and 0.5 mL straw groups, respectively. In Experiment 2, the viability, acrosome membrane integrity, and mitochondrial membrane potential of the frozen-thawed spermatozoa were assessed. After freezing-thawing, the percentages of spermatozoa with live, intact acrosomes and high mitochondrial membrane potential were compared between the in 0.25 mL straw and 0.5 mL straw groups. They were 48.0 ± 2.6% and 35.6 ± 2.8% between the 0.25 mL straw and 0.5 mL straw groups. In Experiment 3, the plasma membrane integrity of frozen-thawed spermatozoa was compared. After freezingthawing, the plasma membrane integrity was higher for the in 0.25 mL straw group than the 0.5 mL straw group. They were 62.0 ± 2.2 and 54.1 ± 1.3% between the 0.25 mL straw and 0.5 mL straw groups. In conclusion, our results suggest that freezing semen in 0.25 mL straw improves the relative motility, viability, and acrosomal, mitochondrial membrane potential, and plasma membrane integrity of Hanwoo bull spermatozoa.

        A mechanistic study on the hydrogen trapping property and the subsequent electrochemical corrosion behavior of quenched and tempered steel

        <P><B>Abstract</B></P> <P>The hydrogen-facilitated anodic dissolution of steel is an interesting experimental phenomenon, but the persistent gaps in this knowledge area are great. The changes in the Tafel slopes and the reaction rates of steel that has been cathodically charged with hydrogen are interpreted mainly in the context of hydrogen trapping and de-trapping behaviors of steel using a variety of electrochemical methods. This study reveals that the increase in the anodic current density and the decrease in the polarization resistance are attributed primarily to the hydrogen-induced lattice expansion. Based on the Tafel-slope change, the oxidation of hydrogen cation partly contributed to the increase in the total anodic current density together with the dominant anodic reaction of the steel dissolution. The electrochemical permeation measurements showed much slower effusion kinetics of the hydrogen that has been trapped at the ε-carbide particles, and the trapping and de-trapping behavior at the fine particles are one of the controlling factors of the hydrogen-enhanced anodic dissolution of steel. From an engineering aspect, it is believed that the current study will provide an important insight into future perspectives on stress corrosion cracking failure occurring in various high-strength steels.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Hydrogen-facilitated anodic reactions of steel are demonstrated experimentally. </LI> <LI> Effect of fine precipitates (ε-carbide) on hydrogen effusion kinetics is discussed. </LI> <LI> The presence of atomic hydrogen in the steel lattice contributes to lattice expansion. </LI> <LI> Anodic and cathodic reaction rates depend on hydrogen charging and subsequent aging. </LI> <LI> Hydrogen trapping at ε-carbide particles leads to slower hydrogen effusing kinetics. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
