단미음료제(單味飮料劑)를 통한 소갈(消渴)의 치료(治療)에 관한 고찰(考察)

In Oriental medicine, So-gal(消渴) is similar to Diabetes Mellitus in the symptoms. In the medical treatment of So-gal(消渴), one kind of herb medicine is recorded in ancent medicine literatures. That herb medicine is a juice of vegetable drunken with herb medicine of So-gal(消渴). Milk(牛乳汁), human milk(人乳汁), gam-ja-jup(甘蔗汁), wo-jul-jup(藕節汁), li-jup(梨汁), sang-gang-jup(生薑汁) etc. is that. In that juices, Milk(牛乳汁), human milk(人乳汁), gam-ja-jup(甘蔗汁), wo-jul-jup(藕節汁), li-jup(梨汁) have sweety tastes, and cold nature. So they cure So-gal(消渴) come from dry and hot nature. Sang-gang-jup(生薑汁) is different with others in the taste and nature that sweety and hot taste and hot nature. Sang-gang-jup(生薑汁) cure the sequela of So-gal(消渴) by his hot nature. We think In Diabetes Mellitus, that juice can be used by drink for the treatment of diet because of simility between So-gal(消渴) and DM.
배추의 배축절편으로부터 캘러스와 뿌리 발생을 통한 안정적 형질전환

'정상' 배추의 배축절편을 선발마커로서 paromomycin 저항 성유전자를 갖고 있는 pPTN290으로 각각 형질전환된 EHA101, LBA4404, GV3101균주와 공동배양한 후 갤러스유도배지에서 형질전환캘러스를 얻은 후, 뿌리유도배지에서 부정근을 그리고 신초유도배지에서 신초를 각각 순차적으로 유도하였다. 형질전환캘러스를 얻은 후, 뿌리유도배지에서 부정근을 그리고 신초유도배지에서 신초를 가각 순차적으로 유도하였다. 형질전환캘러스 형성은 Agrobacterium균주에 따라 차이가 있었으며, 특히 EHA101균주에 공동배양된 배축절편으로부터 최대 6.1%까지 얻어졌다. 또한 각각의 형질전환캘러스 클론으로부터 형질전환 부정근과 신초 발생은 EHA101균주에서 60.7%와 38.2%, LBA4404에서 8.3%와 0%, GV3101에서 20.5%와 85.7%까지 각각 얻을 수 있었다. 형질전환식물체는 특별한 형태적 이상 없이 온실에서 정상적으로 자라 $T_{2}$종자를 얻을 수 있었다. GUS방법으로 7개의 후대 유식물체를 분석한 결과 gus유전자가 안정적으로 발현하고 있음을 확인하였고, 배추 genome에 single 또는 multiple copy로 전달되고 있음을 추측할 수 있었다. Hypocotyl explants of Chinese cabbage (cvs. "Jeong Sang") produced transgenic calli on callus induction medium (MS salt, B5 vitamin, 5 mg/L acetosyringone, 1 mg/L 2,4-D, 3% sucrose, 400 mg/L cefotaxime, 100 mg/L paromomycin, pH 5.8) after cocultivation with strains of Agrobacterium tumefaciens (EHA101, LBA4404, GV3101) harboring the pPTN290 containing paromomycin-resistance gene as a selectable marker, and then they transferred to root induction medium (1/2MS salt, MS vitamins, 2% sucrose, 100 mg/L paromomycin, 100 mg/L cefotaxime, pH 5.8) and shoot induction medium (MS salt, B5 vitamin, 4 mg/L $AgNO_3$, 4 mg/L 6-benzyladenine, 3 mg/L alpha-naphthaleneacetic acid, 100 mg/L paromomycin, 100 mg/L cefotaxime, 3% sucrose, pH 5.8) in order. There was a significant difference in the frequency of transgenic calli depending on Agrobacterium strains. In particular, the highest frequency (6.1%) of transgenic calli was obtained from the hypocotyls cocultivated with EHA101 strains. Also, the frequency (%) of transgenic root and plants from each transgenic callus clone were obtained with 60.7% and 38.2% in EHA101, with 8.3% and 0% in LBA4404, with 20.5% and 85.7% in GV3101 strains, respectively. They were grown to maturity in a greenhouse and normally produced $T_2$ seeds. GUS histochemical assay for progeny ($T_2$) revealed that the transgenes was expressed in the plant genome, and progeny analysis from 7 independent transgenic events demonstrated that the transformants transmitted the transgene as a single or multiple functional locus.

Objective : The purpose of this study was to evidence the effect of Dong-Shi Acupuncture by improving degree of forty four patients who have received a Dong-Shi Acupuncture therapy at Dong Eui oriental hospital OPD and chiefly complained for neck pain and related symptoms. Materials and Methods: Forty four patients were diagnosed with physical inspection and radiography, and investigated into the effect of treatment sharing before treatment and after 2 weeks and 4 weeks treatment by Dr. Kim's method. Results and Conclusions: The conclusion of this study was that improving degree of forty four patients who have received a Dong-Shi acupuncture therapy for all items and excellence of the Dong-Shi Acupuncture was evidenced.

Bone marrow stroma-derived growth factor (SF20/IL25) regulates proliferation of lymphoid cells. We previously observed the upregulation of SF20/IL25 in alveolar macrophages treated with fine $TiO_2$ particles and in a $TiO_2$ particle-treated animal model. To identify the mechanism behind $TiO_2$ particle-induced expression of SF20/IL25, we examined the dependence of the induction process on reactive oxygen species and on cytokine-mediated signaling transduction. For in vivo studies, $TiO_2$ particles were intratracheally instilled in Sprague-Dawley rats. For in vitro studies, alveolar macrophages obtained from rat bronchoalveolar lavage cells were cultured in the presence of $TiO_2$ particles. The antioxidant N-acetyl-L-cysteine (NAC) was used to block the formation of reactive oxygen species both in vivo and in vitro, and TPCK, SB203580, and GF10923X were used to inhibit signal transduction mediated by nuclear factor (NF)-${\kappa}B$, p38 MAP kinase, and protein kinase C, respectively, in vitro. In $TiO_2$ particle-treated rats, intraperitoneal administration of NAC significantly decreased lung inflammation and attenuated the production of SF20/IL25 mRNA and protein. In $TiO_2$ particle-stimulated alveolar macrophages, the upregulation of SF20/IL25 mRNA expression was abolished by NAC in a dose-dependent manner. The expression of SF20/IL25 mRNA in the stimulated macrophages was dose-dependently attenuated by TPCK, but not by SB203580 or GF10923X. Fine $TiO_2$ particles stimulate alveolar macrophages to produce SF20/IL25 via the NF-${\kappa}B$ dependent generation of reactive oxygen species.

<P>We investigated the characteristics of MgO surface with MgO nanocrystal powders due to the long-term (500 hours) ion bombardment comparing with the conventional MgO surface in this study. When the MgO nanocrystal powders were coated on the conventional MgO surface, it was observed that the sputtered Mg particles from MgO surface were re-deposited on the MgO nanocrystal powders, which was able to significantly suppress the re-crystallization on the phosphor layers. We confirm that the MgO nanocrystal powders play a significant role in suppressing the degradation of the MgO surface and phosphor layer after long-term severe ion bombardments. Accordingly, when the MgO nanocrystal powers were applied to the conventional MgO surface, the variations of discharge characteristics, such as address discharge delay time, firing voltage of sustain and address discharge, and luminance, were significantly reduced comparing with the conventional MgO surface.</P>

The behavioral phenotypes of out-bred ICR mice were compared with those of in-bred C57BL/6 and BALB/c mice. In particular, this study examined the locomotor activity and two forms of hippocampus-dependent learning paradigms, passive avoidance and object recognition memory. The basal open-field activity of the ICR strain was greater than that of the C57BL/6 and BALB/c strains. In the passive avoidance task, all the mice showed a significant increase in the cross-over latency when tested 24 hours after training. The strength of memory retention in the ICR mice was relatively weak and measurable, as indicated by the shorter cross-over latency than the C57BL/6 and BALB/c mice. In the object recognition memory test, all strains had a significant preference for the novel object during testing. The index for the preference of a novel object was lower for the ICR and BALB/c mice. Nevertheless, the variance and the standard deviation in these strains were comparable. Overall, these results confirm the strain differences on locomotor activity and hippocampus-dependent learning and memory in mice.
Uptake of Mitochondrial DNA fragment into Boar Spermatoza for Sperm-Mediated Gene Transfer

Sperm-mediated gene transfer(SMGT) can be used to transfer exogenous DNA into the oocyte at fertilization. The main objective of this study was to assess efficiency of transferring mitochondrial DNA(mtDNA) fragment into boar spermatozoa in either presence or absence of liposome and quality of transfected spermatozoa. The mtDNA of chicken liver was isolated and purified by phenol and alkaline lysis extraction, and it was inserted to plasmid. The genome of transfected spermatozoa treated with DNase I was purified by alkaline lysis, and then amplified by the PCR analysis. After electrophoresis, DNA quantitation of each well was calculated by comparison of the band intensity with standard. As a result, exogenous DNA was composed of mtDNA fragment(1.2 kb) and plasmid(2.7 kb). On the other hand, efficiency of transfection by liposome($9.0{\pm}0.34ng/{\mu}l$) in SMGT was higher than that by DNA solution($6.9{\pm}0.53ng/{\mu}l$). However, there was no significant difference. Transfering exogenous DNA into spermatozoa was completed within 90 min of incubation. In another experiment, there were significant (p<0.05) differences between transfected spermatozoa using both DNA solution and DNA/liposome completes with unheated spermatozoa for viability ($70.8{\pm}1.80$ and $68.0{\pm}2.16%$ vs. $83.3{\pm}1.69%$, respectively) and motility($78.7{\pm}1.59$ and $79.3{\pm}2.14%$ vs. $86.7{\pm}1.59%$, respectively). This study indicates that exogenous mtDNA can be efficiently transferred into boar spermatozoa regardless of the presence of liposome, and transfected spermatozoa can also use insemination and in vitro fertilization to generate transgenic pig.
HCC : O-048 ; Laparoscopic liver resection for T1 and T2 hepatocellular carcinoma

Background: Laparoscopic liver resection has gained much popularity in recent years, but relatively few centers have performed laparoscopic hepatectomies in hepatocellular carcinoma (HCC) patients due to the technical difficulties. We now present our early experience with laparoscopic liver resection in HCC performed in a single institution. Methods: From January 2008 until February 2011, total 761 patients of hepatectomy was done and we excluded TNM stage 3, tumors over 5cm and location of tumors of segment I, VII, VIII. 84 laparoscopic liver resections were performed and open conversion was 13 cases (15.4%). To reduce selection bias we made a propensity case matching and selected 50 pairs of each operation group. Results: The cause of open conversion were bleeding for 7 cases, inability to find a mass for 3 cases, severe adhesion for 2 cases and tumor margin for 1 case. Preoperative variables of each group were comparable. The length of hospital stay was shorter in laparoscopic resection group than open resection group. (9.5 days vs 13.2 days, p=0.018) Disease free survival was not different with each group and overall survival was also comparable. 3 year disease free survival in laparoscopic resection group was 35.3% and open resection group was 51.6%. (p=0.409) 3 year survival rate in laparoscopic resection group was 81.3% and open resection group was 90.2%. (p=0.590) Conclusions: Laparoscopic liver resection for HCC can be a feasible and safe operative method in early stage HCC. To draw a final conclusion, randomized control study and long term follow up is needed.

<P>Bimetallic Pt-Au catalysts supported on ZnO/Al2O3 were prepared by incipient wetness impregnation (IW-IMP) method with different pretreatment conditions such as flow velocity, calcination temperature, and heating rate under H2 during the calcination procedure, and characterized by X-ray diffraction (XRD), CO chemisorption, and scanning transmission electron microscopy (STEM) equipped energy dispersive spectroscopy (EDS). Furthermore, catalytic activity for complete oxidation of toluene was measured using a flow reactor under atmospheric pressure. Finally, relationship between the particle sizes with pretreatment conditions and catalytic activity for toluene on the bimetallic Pt-Au catalysts was discussed. In these results, nanosized bimetallic Pt-Au particles on ZnO/Al2O3 could be prepared by IW-IMP method. Relationship between the Pt and Au particle size and activity for toluene oxidation was clearly observed.</P>
