        Bifidobacterium longum and Galactooligosaccharide Improve Skin Barrier Dysfunction and Atopic Dermatitis-like Skin

        Purpose: The beneficial effects of a combination therapy using Bifidobacterium longum and galactooligosaccharide (GOS) for the treatment of atopic dermatitis (AD) have not been elucidated. Methods: Gene expressions of interleukin (IL)-4 and IL-13 from peripheral blood mononuclear cells and fecal abundance of B. longum from 12-month-old infants were evaluated. Human primary epidermal keratinocytes (HEKs) and hairless mice were treated with B. longum, GOS, B. longum-derived extracellular vesicles (BLEVs), dinitrochlorobenzene (DNCB), or a synbiotic mixture of B. longum and GOS. Expression of epidermal barrier proteins and cytokines as well as serum immunoglobulin E (IgE) levels were analyzed in HEKs and mice. Dermatitis scores, transepidermal water loss (TEWL), epidermal thickness, and fecal B. longum abundance were evaluated in mice. Results: Fecal abundance of B. longum was negatively correlated with blood IL-13 expression in infants. B. longum or BLEVs increased expression of filaggrin (FLG) and loricrin (LOR) in HEKs. B. longum increased the efficacy of GOS to upregulate FLG and LOR expressions in HEKs. Oral administration of GOS increased fecal abundance of B. longum in mice. Oral administration of B. longum attenuated DNCB-induced skin inflammation, abnormal TEWL, AD-like skin, and deficiency of epidermal barrier proteins. Moreover, the combination of B. longum and GOS showed greater effects to improve DNCB-induced skin inflammation, abnormal TEWL, AD-like skin, serum IgE levels, IL-4 over-expression, and the deficiency of epidermal barrier proteins than the administration of B. longum alone. Conclusions: B. longum and GOS improve DNCB-induced skin barrier dysfunction and AD-like skin.

        Alterations of Epidermal Lipid Profiles and Skin Microbiome in Children With Atopic Dermatitis

        Purpose: We aimed to investigate epidermal lipid profiles and their association with skin microbiome compositions in children with atopic dermatitis (AD). Methods: Specimens were obtained by skin tape stripping from 27 children with AD and 18 healthy subjects matched for age and sex. Proteins and lipids of stratum corneum samples from nonlesional and lesional skin of AD patients and normal subjects were quantified by liquid chromatography tandem mass spectrometry. Skin microbiome profiles were analyzed using bacterial 16S rRNA sequencing. Results: Ceramides with nonhydroxy fatty acids (FAs) and C18 sphingosine as their sphingoid base (C18-NS-CERs) N-acylated with C16, C18 and C22 FAs, sphingomyelin (SM) N-acylated with C18 FAs, and lysophosphatidylcholine (LPC) with C16 FAs were increased in AD lesional skin compared to those in AD nonlesional skin and that of control subjects (all P < 0.01). SMs N-acylated with C16 FAs were increased in AD lesional skin compared to control subjects (P < 0.05). The ratio of NS-CERs with long-chain fatty acids (LCFAs) to short-chain fatty acids (SCFAs) (C24-32:C14-22), the ratio of LPC with LCFAs to SCFAs (C24-30:C16-22) as well as the ratio of total esterified omega-hydroxy ceramides to total NS-CERs were negatively correlated with transepidermal water loss (rho coefficients = −0.738, −0.528, and −0.489, respectively; all P < 0.001). The proportions of Firmicutes and Staphylococcus were positively correlated to SCFAs including NS ceramides (C14-22), SMs (C17-18), and LPCs (C16), while the proportions of Actinobacteria, Proteobacteria, Bacteroidetes, Corynebacterium, Enhydrobacteria, and Micrococcus were negatively correlated to these SCFAs. Conclusions: Our results suggest that pediatric AD skin shows aberrant lipid profiles, and these alterations are associated with skin microbial dysbiosis and cutaneous barrier dysfunction.

      • Overexpression of <i>PYL5</i> in rice enhances drought tolerance, inhibits growth, and modulates gene expression

        <P>Abscisic acid (ABA) is a phytohormone that plays important roles in the regulation of seed dormancy and adaptation to abiotic stresses. Previous work identified OsPYL/RCARs as functional ABA receptors regulating ABA-dependent gene expression in <I>Oryza sativa</I>. <I>OsPYL</I>/<I>RCAR</I>s thus are considered to be good candidate genes for improvement of abiotic stress tolerance in crops. This work demonstrates that the cytosolic ABA receptor OsPYL/RCAR5 in <I>O. sativa</I> functions as a positive regulator of abiotic stress-responsive gene expression. The constitutive expression of <I>OsPYL/RCAR5</I> in rice driven by the <I>Zea mays ubiquitin</I> promoter induced the expression of many stress-responsive genes even under normal growth conditions and resulted in improved drought and salt stress tolerance in rice. However, it slightly reduced plant height under paddy field conditions and severely reduced total seed yield. This suggests that, although exogenous expression of OsPYL/RCAR5 is able to improve abiotic stress tolerance in rice, fine regulation of its expression will be required to avoid deleterious effects on agricultural traits.</P>

        In Vitro Metabolism of a New Cardioprotective Agent, KR-33028 in the Human Liver Microsomes and Cryopreserved Human Hepatocytes

        KR-33028 (N-[4-cyano-benzo[b]thiophene-2-carbonyl]guanidine) is a new cardioprotective agent for preventing ischemia-reperfusion injury. This study was performed to identify the metabolic pathway of KR-33028 in human liver microsomes and to compare its metabolism with that of cryopreserved human hepatocytes. Human liver microsomal incubation of KR-33028 in the presence of NADPH and UDPGA resulted in the formation of four metabolites, M1, M2, M3, and M4. M1 and M2 were identified as 5-hydroxy-KR-33028 and 7-hydroxy-KR-33028, respectively, on the basis of LC/MS/MS analysis with the synthesized authentic standard. M3 and M4 were suggested to be dihydroxy-KR-33028 and hydroxy-KR-33028-glucuronide, respectively. Metabolism of KR-33028 in cryopreserved human hepatocytes resulted in the formation of M1, M2, and M4. These data show a good correlation between major metabolites formed in human liver microsomes and cryopreserved human hepatocytes. In addition, KR­33028 was found to inhibit moderately the metabolism of CYP1A2 substrates. Based on the results obtained metabolic pathway of KR-33028 is proposed.

        Effects of woohwangcheongsimwon suspension on the pharmacokinetics of bupropion and its active metabolite, 4-hydroxybupropion, in healthy subjects

        <P><B>WHAT IS ALREADY KNOWN ABOUT THIS SUBJECT</B></P><P>• Woohwangcheongsimwon suspension has traditionally been used for the treatment and prevention of stroke, hypertension, palpitations, convulsions and unconsciousness in various Asian countries.</P><P>• Woohwangcheongsimwon suspensions showed an inhibitory effect on CYP2B6 activity <I>in vitro</I>. Two terpenoids, borneol and isoborneol, are major constituents of woohwangcheongsimwon suspension, and show a competitive inhibition of CYP2B6 with <I>K</I><SUB>i</SUB> values of 9.5 and 5.9 µ<SMALL>M</SMALL>, respectively.</P><P>• Bupropion undergoes metabolic transformation to the active metabolite, 4-hydroxybupropion, primarily via CYP2B6 both <I>in vivo</I> and <I>in vitro</I>. It is often used as a CYP2B6 substrate for clinical drug–drug interaction studies.</P><P>• Drug interactions may occur between woohwangcheongsimwon suspension and bupropion.</P><P><B>WHAT THIS STUDY ADDS</B></P><P>• Co-administration with woohwangcheongsimwon suspension did not alter the pharmacokinetics of bupropion or its metabolite, 4-hydroxybupropion.</P><P>• Dosage adjustment of bupropion is unnecessary in patients concomitantly administered the highest recommended daily dose of woohwangcheongsimwon suspension.</P><P>AIMS</P><P>To examine the effects of woohwangcheongsimwon suspension on the pharmacokinetics of bupropion and its active metabolite, 4-hydroxybupropion, formed via CYP2B6 <I>in vivo</I>.</P><P>METHODS</P><P>A two-way crossover clinical trial with a 2 week washout period was conducted in 14 healthy volunteers. In phases I and II, subjects received 150 mg bupropion with or without woohwangcheongsimwon suspension four times (at −0.17, 3.5, 23.5 and 47.5 h, with the time of bupropion administration taken as 0 h) in a randomized balanced crossover order. Bupropion and 4-hydroxybupropion plasma concentrations were measured for up to 72 h by LC-MS/MS. Urine was collected up to 24 h to calculate the renal clearance. In addition, the <I>CYP2B6</I>*6 genotype was also analyzed.</P><P>RESULTS</P><P>The geometric mean ratios and 90% confidence interval of bupropion with woohwangcheongsimwon suspension relative to bupropion alone were 0.976 (0.917, 1.04) for AUC(0,∞) and 0.948 (0.830,1.08) for <I>C</I><SUB>max</SUB>, respectively. The corresponding values for 4-hydroxybupropion were 0.856 (0.802, 0.912) and 0.845 (0.782, 0.914), respectively. The <I>t</I><SUB>max</SUB> values of bupropion and 4-hydroxybupropion were not significantly different between the two groups (<I>P</I> > 0.05). The pharmacokinetic parameters of bupropion and 4-hydroxybupropion were unaffected by woohwangcheongsimwon suspension.</P><P>CONCLUSIONS</P><P>These results indicate that woohwangcheongsimwon suspension has a negligible effect on the disposition of a single dose of bupropion <I>in vivo</I>. As a result, temporary co-administration with woohwangcheongsimwon suspension does not seem to require a dosage adjustment of bupropion.</P>

      • <i>In vitro</i> metabolism of a new cardioprotective agent, KR‐32570, in human liver microsomes

        <P><B>Abstract</B></P><P>KR‐32570 (5‐(2‐methoxy‐5‐chlorophenyl)furan‐2‐ylcarbonyl)guanidine) is a new reversible Na<SUP>+</SUP>/H<SUP>+</SUP> exchanger inhibitor for preventing ischemia‐reperfusion injury. This study was performed to identify the metabolic pathway of KR‐32570 in human liver microsomes. Human liver microsomal incubation of KR‐32570 in the presence of NADPH and UDPGA resulted in the formation of six metabolites, M1–M6. M1 was identified as <I>O</I>‐desmethyl‐KR‐32570, on the basis of liquid chromatography/tandem mass spectrometric (LC/MS/MS) analysis with the synthesized authentic standard. M2 and M3 were suggested to be hydroxy‐KR‐32570 and hydroxy‐<I>O</I>‐desmethyl‐KR‐32570, respectively. M1, M2, and M3 were further metabolized to their glucuronide conjugates, M4, M5, and M6, respectively. In addition, the specific P450 isoforms responsible for KR‐32570 oxidation to two major metabolites, <I>O</I>‐desmethyl‐KR‐32570 and hydroxy‐KR‐32570, were identified using a combination of correlation analysis, chemical inhibition in human liver microsomes and metabolism by expressed recombinant P450 isoforms. The inhibitory potency of KR‐32570 on clinically major P450s was investigated in human liver microsomes. The results show that CYP3A4 contributes to the oxidation of KR‐32570 to hydroxy‐KR‐32570, and CYP1A2 play the predominant role in <I>O</I>‐demethylation of KR‐32570. KR‐32570 was found to inhibit moderately the metabolism of CYP2C8 substrates. Copyright © 2006 John Wiley & Sons, Ltd.</P>

        직장 내 소외감형성의 따돌림이 민간경호원의 직무태도에 대한 영향 및 자아 존중감의 조절효과에 관한 연구 : 서울·경기지역을 중심으로

        This study is significant in the sense that it minimize the negative effect of workplace bullying, ultimately it maximize organizational performance and keep private security guards physical and mental health. We tested the hypothesis that workplace bullying negatively impacts private security guard's office attitude, but there will be significant difference in influence with their level of self-esteem scale. First of all, as the result of the correlation analysis of each of variables, a positive correlation was found between self-esteem and job satisfaction but negative correlation between self-esteem they have, the more job satisfaction they get, the lower turnover intention they have. Second, we tried to find out how self-esteem affect them on their job satisfaction. Using regression analysis, we can see that job satisfaction is affected by age, workplace bullying, relational aggression and self-esteem. For example, job satisfaction is increased as they get old and self-esteem is improved, on the other hand, workplace bullying and relational aggression is decreased. Third, to find out how self-esteem affects their turnover intention, we did regression analysis. The result shows that turnover intention is affected by age, working period, workplace bulling, relational aggression and self-esteem. For example, turnover intention if higher as working period, workplace bullying, relational aggression is increased while age and self-esteem is decreased. 이 연구는 민간경호원을 대상으로 직장 내 소외감형성의 따돌림은 부정적 직무태도에 영향을 미치고 있지만 개인의 자아존중감에 따라 차이가 있을 것이라는 연구문제를 설정하여 직장 내 따돌림에 대한 부정적 영향을 최소화하며 궁극적으로는 조직의 응집력을 강화하고 민간경호원의 정신 및 신체적 건강을 지속적으로 유지하는데 그 결과의 의의가 있다고 볼 수 있다. 따라서 이러한 결과를 통하여 얻어진 결론은 다음과 같다. 첫째, 요인들 간의 상관관계를 분석한 결과 업무 따돌림과 인간관계 따돌림, 업무 따돌림과 이직의도, 인간관계 따돌림과 이직의도, 자기존중감과 직무만족은 서로 정적 상관관계를 가지고 있으며, 그 외의 변수들 간에는 서로 부적 상관관계를 가지고 있는 것이 나타났다. 둘째, 집단 내 소외감형성의 따돌림에 따른 자기존중감이 직무만족도에 어떠한 영향을 주는지를 알아보기 위해 회귀분석을 한 결과 연령과 업무 따돌림, 인간관계 따돌림, 자기존중감이 직무만족에 각각 영향을 주며, 연령과 자기존중감이 향상되면 직무만족은 향상되지만 직무 따돌림과 인간관계 따돌림은 감소되는 것이 나타났다. 셋째, 직장 내 집단 따돌림과 자기존중감이 이직의도에 영향을 주는지를 알아보기 위해 회귀분석을 한 결과 연령과 근속 년수, 업무 따돌림, 인간관계 따돌림, 자기존중감이 이직의도에 영향을 주며, 근속 년수와 직무따돌림, 인간관계 따돌림이 증가하게 되면 이직의도도 증가되지만, 연령과 자기존중감은 감소되는 것이 나타났다.
