        KI-180과 KI-188 칼슘조성물이 어린쥐의 성장발육에 미치는 영향

        본 연구에서는 KI-188 칼슘과 KI-180 조성물이 성장발육에 미치는 효과를 조사하였다. 시험기간중 KI-188 칼슘과 KI-180 식이군이 대조군 보다 성장률과 평균 식이섭취량은 증가하였다. 체장은 KI-188 칼슘과 KI-180 식이군이 대조군에 비해 평균 3.55 mm 더 길었고, 등뼈길이는 KI-180 식이군이 대조군 보다 통계적인 차이는 없었지만 0.5 mm 더 길었다. 대퇴골 무게는 KI-188 칼슘과 KI-180 식이군이 대조군에 비해 통계적으로 무거웠으며, 대퇴골길이는 대조군에 비해 각각 평균 0.89, 1.09 mm 더 길었다. 혈당, 총콜레스테롤 및 중성지방 농도는 차이가 없었고, 칼슘농도는 KI-188 칼슘과 KI-180 식이군 보다 대조군이 유의하게 높았다. 백혈구와 혈소판수는 차이가 없었고, 적혈구, 혈색소 및 헤마토크릿치는 대조군이 KI-188 칼슘과 KI-180 식이군 보다 통계적으로 높았다. ALP활성은 KI-180 식이군이 대조군 보다 통계적으로 높았고, osteocalcin농도는 KI-188 칼슘과 KI-180 식이군이 대조군에 비해 통계적으로 유의하게 높았다. 혈청 testosterone 농도는 KI-188 칼슘과 KI-180 식이군과 대조군 간에 차이가 없었다. IGF-1과 IGFBP-3 농도는 KI-180 조성물이 대조군 보다 각각 20%, 11% 정도 유의하게 더 높았다. 이상의 결과를 검토할 때 KI-188 칼슘과 KI-180 조성물은 어린쥐의 성장과 골격의 발달을 촉진시키고, 성장호르몬의 분비능을 양적으로 유도하여 전반적인 성장발육에 유효한 작용이 있는 것으로 사료된다. Herbs have active components that promote the growth rate of both animals and human. The KI-180 and KI-188 calcium food formulae contain Acanthopanacis cortex, Bombysis corpus and hoelen, seaweed calcium, chlorella extract, spirulina, colostrum powder, and other natural and functional components. We evaluated the growth-promoting effects of these formulae by analyzing the weight, femur and backbone, alkaline phosphatase, osteocalcin, testosterone, insulin-like growth factor (IGF)-1, and IGF binding protein-3 (IGFBP-3) of growing rats. Growing rats administered with KI-180 and KI-188 calcium showed the increase of body weight, body length, and femur weight and length of growing rats. In addition, administration of KI-180 and KI-188 calcium increased the alkaline phosphatase activity, the levels of osteocalcin and the growth hormones IGF-1 and IGFBP-3 of growing rats. The impact of KI-180 and KI-188 calcium on the physical development of growing rats suggests that the incorporation of these food formulae in the diets of growing children may promote the physical development.

        <P>Mesenchymal stem cells (MSC) have emerged as a new therapeutic tool for a number of clinical applications, because they have multipotency and paracrine effects via various factors. In the present study, we investigated the effects of adipose-derived MSC (Ad-MSC) transplantation via intrathecal injection through the cisterna magna on cell proliferation and differentiation of endogenous stem cells in the hippocampal dentate gyrus (DG) using Ki-67 (a marker for proliferating cells), and doublecortin (DCX, a marker for neuroblasts). The transplanted Ad-MSC were detected in the meninges, not in the hippocampal parenchyma. However, the number of Ki-67-immunoreactive cells was significantly increased by 83% in the DG 2 days after single Ad-MSC injection, and by 67% at 23 days after repeated Ad-MSC treatment compared with that in the vehicle-treated group after Ad-MSC transplantation. On the other hand, the number of DCX-immunoreactive cells in the DG was not changed at 2 days after single Ad-MSC injection; however, it was significantly increased by 62% 9 days after single Ad-MSC injection. At 23 days after repeated Ad-MSC application, the number of DCX-immunoreactive cells was much more increased (223% of the vehicle-treated group). At this time point, DCX protein levels were also significantly increased compared with those in the vehicle-treated group. These results suggest that the intrathecal injection of Ad-MSC could enhance endogenous cell proliferation, and the repeated Ad-MSC injection could be more efficient for an enhancement of endogenous cell proliferation and differentiation in the brain.</P>

        이매방 살풀이춤의 ‘여형화(女形化)’에 대한 논의

        우리는 이매방의 춤을 이야기할 때 ‘아름답다’, ‘교태적이다’, ‘농염하다’라고 표현한다. 이러한 수사(修辭)는 그의 춤이 다분히 여성적인 속성을 갖고 있기 때문에 붙여진 것이다. 본 연구는 이매방의 살풀이춤이 여형화의 양식체계로 구축되어 있음을 논의하였다. 춤에서 ‘여형화’란 남성이 여성의 모습으로 춤을 추는 형식, 혹은 양식이라고 할수 있다. 이매방의 춤은 다음과 같은 여형화의 특성을 보인다. 첫째, 이매방의 살풀이춤은 경극의 ‘화단’이나 가부키의 ‘게이세이’와 같이 화려하고 교태스런 배역 특성을 보인다. 둘째, 살풀이춤의 복식은 남성과 여성의 구분을 두고 있으되 상당히 장식적인 여성성을 지향하고 있다. 셋째, 이매방의 양성 지향적인 성 정체성은 탐미적인 미의식으로 확장되어 여형화 된 춤 양식 체계로 환원되었다. 넷째, 이매방 살풀이춤의 기법은 가공된 여성의 신체상을 기반으로 구축되었다. 즉, 응축된 근육의 밀도 높은 작용을 통해 신체를 ‘축소’하거나 ‘왜곡’하여 관능적이고 요염한 여성성을 신체상으로 구현하여 춤에 반영하였다. 이상으로 미루어 이매방 살풀이춤은 남성이 추는 여성의 춤, 혹은 기방춤을 전형 특성의 하나로 구축하고 있다고 하겠다. When we talk about Lee Mae-bang's Salpul-i chum, we use the expressions ‘beautiful’, ‘exciting’, and ‘congested’. This knowledge was attached because his dance has a feminine attribute. This study discussed that Lee Mae-bang's Salpul-i chum is constructed as Yeohyeonghwa's aquaculture system. First, Lee Mae-bang's Salpul-i chum exhibits gorgeous and cunning character, such as Peking Opera's 'Huā dàn' or Kabuki's 'Keisei'. In dance[chum], ‘Yeohyeonghwa’ can be said to be a form or style in which men dance in the form of women. Lee Mae-bang's dance shows the following characteristics of Yeohyeonghwa. Second, Salpul-i chum's costumes differentiate between men and women, but aim for a highly decorative femininity. Third, Lee Mae-bang's bisexual sexual identity has been stylized into a feminine attitude through aesthetic pursuit. Fourth, Lee Mae-bang Salpul-i chum's technique expresses the processed female body image. In other words, Salpul-i chum expresses the body of a sensual and fascinating woman by 'shrinking' or 'distorting' the body through the dense action of condensed muscles. In this regard, Lee Mae-bang is said to be constructing a male dance or gibangchum as one of the characteristics of Jeonhyeong[original form].

      • Protective Effect of <i>Artemisia argyi</i> and Its Flavonoid Constituents against Contrast-Induced Cytotoxicity by Iodixanol in LLC-PK1 Cells

        <P>Preventive effects and corresponding molecular mechanisms of mugwort (<I>Artemisia argyi</I>) extract and its flavonoid constituents on contrast-induced nephrotoxicity were explored in the present study. We treated cultured LLC-PK1 cells with iodixanol to induce contrast-induced nephrotoxicity, and found that <I>A. argyi</I> extracts ameliorated the reduction in cellular viability following iodixanol treatment. The anti-apoptotic effect of <I>A. argyi</I> extracts on contrast-induced nephrotoxicity was mediated by the inhibition of mitogen-activated protein kinase (MAPK) phosphorylation and the activation of caspases. The flavonoid compounds isolated from <I>A. argyi</I> improved the viability of iodixanol-treated cells against contrast-induced nephrotoxicity. Seven compounds (<B>1</B>, <B>2</B>, <B>3</B>, <B>15</B>, <B>16</B>, <B>18</B>, and <B>19</B>) from 19 flavonoids exerted a significant protective effect. Based on the in silico oral-bioavailability and drug-likeness assessment, which evaluate the drug potential of these compounds, compound <B>2</B> (artemetin) showed the highest oral bioavailability (49.55%) and drug-likeness (0.48) values. We further investigated the compound–target–disease network of compound <B>2</B>, and proliferator-activated receptor gamma (PPAR-γ) emerged as a predicted key marker for the treatment of contrast-induced nephrotoxicity. Consequently, compound <B>2</B> was the preferred candidate, and its protective effect was mediated by inhibiting the contrast-induced inflammatory response through activation of PPAR-γ and inhibition of MAPK phosphorylation and activation of caspases.</P>

      • Recessive optic atrophy, sensorimotor neuropathy and cataract associated with novel compound heterozygous mutations in <i>OPA1</i>

        <P>Mutations in the optic atrophy 1 gene (<I>OPA1</I>) are associated with autosomal dominant optic atrophy and 20% of patients demonstrate extra-ocular manifestations. In addition to these autosomal dominant cases, only a few syndromic cases have been reported thus far with compound heterozygous <I>OPA1</I> mutations, suggestive of either recessive or semi-dominant patterns of inheritance. The majority of these patients were diagnosed with Behr syndrome, characterized by optic atrophy, ataxia and peripheral neuropathy. The present study describes a 10-year-old boy with Behr syndrome presenting with early-onset severe optic atrophy, sensorimotor neuropathy, ataxia and congenital cataracts. He had optic atrophy and was declared legally blind at six years old. Electrophysiological, radiological, and histopathological findings were compatible with axonal sensorimotor polyneuropathy. At birth, he presented with a congenital cataract, which has not been previously described in patients with <I>OPA1</I> mutations. Whole exome sequencing indicated a pair of novel compound heterozygous mutations: p.L620fs*13 (c.1857–1858delinsT) and p.R905Q (c.G2714A). Neither mutation was observed in controls (n=300), and thus, they were predicted to be pathogenic by multiple <I>in silico</I> analyses. The mutation sites were highly conserved throughout different vertebrate species. The patients parents did not have any ophthalmic or neurologic symptoms and the results of electrophysiological studies were normal, suggestive of an autosomal recessive pattern of inheritance. The present study identified novel compound heterozygous <I>OPA1</I> mutations in a patient with recessive optic atrophy, sensorimotor neuropathy and congenital cataracts, indicating an expansion of the clinical spectrum of pathologies associated with <I>OPA1</I> mutations. Thus, <I>OPA1</I> gene screening is advisable in the workup of patients with recessive optic atrophy, particularly with Behr syndrome and cataracts.</P>

        Melanogenesis regulatory activity of the ethyl acetate fraction from <i>Arctium lappa</i> L. leaf on α-MSH–induced B16/F10 melanoma cells

        <P><B>Abstract</B></P> <P>Phenolics obtained from plants as natural compounds have been used in cosmetic industry as potential alternatives to synthetic chemicals due to its excellent antioxidant and whitening effects. This study investigated the possibility of using <I>Arctium lappa</I> L. leaf extracts as cosmetic substances by assessing whitening effects through its antioxidant and physiological activity in B16/F10 melanoma cells. The ethyl acetate fraction from <I>Arctium lappa</I> L. leaf showed the highest total phenolics (216.75 mg gallic acid equivalent (GAE)/g) relative to the other fractions (<I>n</I>-hexane, chloroform, and distilled water). The antioxidant activities of EAFA were evaluated based on 2,2′-azino-bis (3-ethyl benzthiazoline-6-sulfonic acid) diammonium salt (ABTS) /α-diphenyl-β-picrylhydrazyl (DPPH) radical scavenging activity, ferric reducing/antioxidant power (FRAP), and malondialdehyde (MDA) inhibitory effects. The whitening effects of EAFA were examined using tyrosinase inhibitory activity and inhibition of α-melanocyte stimulating hormone (α-MSH) -induced melanogenesis in B16/F10 cells. The ethyl acetate fraction from <I>Arctium lappa</I> L. leaf effectively inhibited the tyrosinase activity and decreased melanin contents as following α-MSH stimulation. Based on these results, the inhibitory mechanism of melanogenesis was confirmed by measuring phosphorylated c-Jun N-terminal kinase (<I>p</I>-JNK), microphthalmia-associated transcription factor (MITF), tyrosinase-related protein 1 (TRP-1), and tyrosinase in α-MSH–induced B16/F10 cells. The ethyl acetate fraction from <I>Arctium lappa</I> L. leaf downregulated the levels of <I>p</I>-JNK, MITF, TRP-1, and tyrosinase. Finally, the main compounds of EAFA were analyzed using the ultra-performance liquid chromatography-quadrupole-time-of-flight (UPLC Q-TOF) MS system, and the components were identified as follows: trans-5-caffeoylqunic acid, rutin, kaempferol-3-O-rutinoside, 3,5-di-O-caffeoylqunic acid, and 4,5-di-<I>O</I>-caffeoylqunic acid.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Ethyl acetate fraction from <I>Arctium lappa</I> L. leaf showed an effective antioxidant activity. </LI> <LI> Ethyl acetate fraction from <I>Arctium lappa</I> L. leaf showed a tyrosinase inhibitory effect. </LI> <LI> Ethyl acetate fraction from <I>Arctium lappa</I> L. leaf indicated a regulatory activity on melanogenesis. </LI> <LI> Major compounds were analyzed as rutin and hydroxycinnamic acid. </LI> </UL> </P>
