TGF-${\beta}$ induces IgA class switching by B cells. We previously reported that Smad3 and Smad4, pivotal TGF-${\beta}$ signal-transducing transcription factors, mediate germline (GL) ${\alpha}$ transcription induced by TGF-${\beta}1$, resulting in IgA switching by mouse B cells. Post-translational sumoylation of Smad3 and Smad4 regulates TGF-${\beta}$-induced transcriptional activation in certain cell types. In the present study, we investigated the effect of sumoylation on TGF-${\beta}1$-induced, Smad3/4-mediated $GL{\alpha}$ transcription and IgA switching by mouse B cell line, CH12F3-2A. Overexpression of small ubiquitin-like modifier (SUMO)-1, SUMO-2 or SUMO-3 did not affect TGF-${\beta}1$-induced, Smad3/4-mediated $GL{\alpha}$ promoter activity, expression of endogenous $GL{\alpha}$ transcripts, surface IgA expression, and IgA production. Next, we tested the effect of the E3 ligase PIASy on TGF-${\beta}1$-induced, Smad3/4-mediated $GL{\alpha}$ promoter activity. We found that PIASy overexpression suppresses the $GL{\alpha}$ promoter activity in cooperation with histone deacetylase 1. Taken together, these results suggest that SUMO itself does not affect regulation of $GL{\alpha}$ transcription and IgA switching induced by TGF-${\beta}1$/Smad3/4, while PIASy acts as a repressor.

We examined the minimal amino acid sequence of bovine lactoferricin (Lfcin-B), a cationic peptide corresponding to residues 17-41 near the N-terminus of bovine lactoferrin, to induce apoptosis in THP-1 human monocytic leukemic cells using synthetic peptides. A synthetic peptide (Lfc-17/29, amino acid sequence; FKCRRWQWRMKKL) consist of 13 amino acids near the N-terminus of Lfcin-B induced cell death in THP-1 cells in a dose-dependent manner, showing apparent apoptotic changes such as hypodiploid forms of genomic DNA and apoptotic DNA fragmentation. However, another synthetic peptide (Lfc-30/41, amino acid sequence; GAPSITCVRRAF) consist of 12 amino acids near the C-terminus of Lfcin-B was inactive. The characteristics of Lfc-17/29-induced apoptosis was entirely identical with that of Lfcin-B-induced apoptosis, with regard to increased apoptosis with reduction of serum concentration, and inhibition of apoptosis by addition of Ca²⁺/Mg²⁺-dependent endonuclease inhibitor or antioxidants. In an analysis using various synthetic peptides having the partial sequences of Lfc-17/29, we found that a peptide consist of 10 amino acids (Lfc-17/26, FKCRRWQWRM), corresponding to residues 17-26 near the N-terminus of Lfcin-B, was the minimal sequence of Lfc-17/29 responsible for apoptosis induction in tumor cells.

This paper analyzes English and Korean binomials in the structure of X and Y by resorting to the iconicity of temporal sequence, the iconicity of directionality, and the iconicity of markedness. it is claimed that (ⅰ) X precedes Y if X is an event or a state that took place before what is represented by Y; (ⅱ) directionality iconicity is found in egocentric, vertical, and horizontal orientations where X stands for a word that refers to something that is closer, above, or in front of the speaker and Y stands for something that is farther away, below, or behind the speaker; (ⅲ) an unmarked or positive element occurs in the X slot and a marked and negative element occupies the Y slot, so that the unmarked-marked word order is observed. When due attention is paid to the order of substantial English and Korean data, the order of binomials appears to be iconic in terms of temporality, directionality, and markedness and applicable across languages.
Dectin-1 Stimulation Selectively Reinforces LPS-driven IgG1 Production by Mouse B Cells

Dectin-1, which specifically recognizes ${\beta}$-glucan of fungal cell walls, is a non-Toll-like receptor (TLR) pattern recognition receptor and a representative of C-type lectin receptors (CLRs). The importance of Dectin-1 in innate immune cells, such as dendritic cells and macrophages, has previously been well studied. However, the function of Dectin-1 in B cells is very poorly understood. To determine the role of Dectin-1 in B cell activation, we first investigated whether mouse B cells express Dectin-1 and then assessed the effect of Dectin-1 stimulation on B cell proliferation and antibody production. Mouse B cells express mRNAs encoding CLRs, including Dectin-1, and surface Dectin-1 was expressed in B cells of C57BL/6 rather than BALB/c strain. Dectin-1 agonists, heat-killed Candida albicans (HKCA) and heat-killed Saccharomyces cerevisiae (HKSC), alone induced B cell proliferation but not antibody production. Interestingly, HKSC, HKCA, and depleted zymosan (a selective Dectin-1 agonist) selectively enhanced LPS-driven IgG1 production. Taken together, these results suggest that, during fungal infection, ${\beta}$-glucan-stimulated Dectin-1 may cooperate with TLR4 to specifically enhance IgG1 production by mouse B cells.

Background: CBT is an established alternative donor transplantation for treatment of patients with advanced hematologic diseases. Methods: To assess clinical outcome after CBT and provide future recommendations for alternative donor, we conducted the first retrospective multicenter study of adult CBT in Korea. Results: Between June 2001 and April 2014, 48 patients (male, 19) were enrolled from 16 hospitals. The median age was 36 years (range, 16-65 years). Forty patients (83.3%) were acute leukemia: 33 (68.8%) in remission and 7 (14.6%) in persistence. The others were myelodysplastic syndrome (n=3, 6.3%), severe aplastic anemia (n=3, 6.3%), and lymphoma (n=2, 4.2%). Thirty two patients (66.7%) underwent double unit CBT. Twenty two patients (45.8%) underwent myeloablative conditioning and 26 patients (54.2%), reduced-intensity conditioning. Total body irradiation (TBI)- based conditioning regimens were used in 23 patients (47.9%). The median time to neutrophil >500/μL was 20 days among grafted, and 14 patients (29.2%) failed to recover. During follow-up, 8 patients died of disease and non-relapse mortality (NRM) developed in 23 patients. The 2-year overall survival (OS) was 29.6%. In multivariable analysis, ECOG performance status (PS) =2 (hazard ratio [HR] 6.42, p=0.001) and total nucleated cell dose <3.0x10^7/kg (HR 2.59, p=0.012) were poor prognostic factors for OS. Factors associated with graft failure were ECOG PS 2 (p=0.012) and non-TBIbased conditioning (p=0.026). CBT outcomes after January 2009 were significantly improved compared to those before January 2009 (p=0.012). Conclusions: Although in the past, the clinical outcome of adult CBT was not satisfactory in Korea because of high NRM, it has been improved in recent years. Our data suggest that the clinical outcome substantially depends on the cord blood cell dose and PS.
