        Autophagy May Mediate Cellular Senescence by Nicotine Stimulation in Gingival Fibroblasts

        Background: When cells are damaged by nicotine, cellular senescence due to oxidative stress accelerates. In addition, stress-induced inflammatory response and cellular senescence cause the accumulation of damaged organelles in cells, and autophagy appears to remove them. Conversely, when autophagy is reduced, harmful cell components accumulate, and aging is accelerated. This study aimed to determine the association between nicotine-induced cellular senescence and autophagy expression patterns in human gingival fibroblasts. Methods: Cells were treated with various concentrations of nicotine (0, 0.1, 0.5, 1, 2, and 5 mM) and 10 nM rapamycin was added to 1 mM nicotine to investigate the relationship between autophagy and cellular senescence. Cell viability was confirmed using WST-8 and the degree of cellular senescence was measured by SA-β-gal staining. The expression of the inflammatory proteins (COX-2 and iNOS) and autophagy markers (LC3-II, p62, and Beclin-1) was analyzed by western blotting. Results: The cell viability tended to decrease in a concentration-dependent manner. COX-2 showed no concentration-dependent expression and iNOS increased in the 0.5 mM nicotine treated group. The degree of cellular senescence was the highest in the 1 mM nicotine treatment group. In the group treated with rapamycin and nicotine, the conversion ratio of LC3-II to LC3-I was the highest, that of p62 was the lowest, and the level of Beclin-1 proteins was significantly increased. Furthermore, the degree of cellular senescence was reduced in the group in which rapamycin was added to nicotine compared to that in the group treated with nicotine alone. Conclusion: This study provides evidence that autophagy activated in an aging environment reduces cellular senescence to a certain some extent.

        Biocompatibility and Bioactivity of Four Different Root Canal Sealers in Osteoblastic Cell Line MC3T3-El

        Background: Endodontic sealers or their toxic components may become inflamed and lead to delayed wound healing when in direct contact with periapical tissues over an extended period. Moreover, an overfilled sealer can directly interact with adjacent tissues and may cause immediate necrosis or further resorption. Therefore, the treatment outcome conceivably depends on the endodontic sealer’s biocompatibility and osteogenic potential. This study aimed to evaluate the cell viability and osteogenic effects of four different sealers in osteoblastic cells. Methods: AH Plus (resin-based sealer), Pulp Canal Sealer EWT (zinc oxide-eugenol sealer), BioRoot RCS (calcium silicate-based sealer), and Well-Root ST (MTA-based calcium silicate sealer) were mixed strictly according to the manufacturer’s instructions, and dilutions of sealer extracts (1/2, 1/5 and 1/10) were determined. Cell viability was measured using the water-soluble tetrazolium-8 (WST-8) assay. Differentiation was assessed by alkaline phosphatase (ALP) activity and mineralized nodule formation by Alizarin Red S staining. Results: The cell viability of the extracts derived from the sealers excluding Well-Root ST was concentration dependent, with sealer extracts having the least viability at a 1/2 dilution. At sealer extract dilution of 1/10, the test groups showed the same survival rate as that control group, with the exception of BioRoot RCS. Among all experimental groups, BioRoot RCS showed the highest cell viability after 48 hours. The ALP activity was significantly higher in a concentration-dependent manner. Furthemore, all four materials promoted ALP activity and mineralized nodule formation compared to the control at 1/10 dilutions. Conclusion: This is the first study to highlight the differences in biological activity of these four materials. These results suggest that the composition of root canal sealers appears to alter the form of biocompatibility and osteoblastic differentiation.

        신생아 중환자에 대한 i-STATTM 휴대용 분석기의 유용성에 대한 연구

        연구목적:POCT(Point of Care Test)는 환자 곁 검사(bedside laboratory)라고 번역되며, 의료 선진국에서는 몇 년 전부터 많이 사용되고 있는 검사법이다. 주로 중환자실에서 신속하게 결과를 얻을 수 있다는 장점으로 많이 사용된다. 본 연구에서는 신생아중환자실에 입원한 만삭아나 미숙아를 대상으로 적은 혈액 샘플로 신속하고 정확한 결과를 얻을 수 있다고 알려진 POC검사법 중 i-STATTM검사법을 시행하여 기존 검사에 대비한 i-STATTM검사결과의 상관관계를 알아보고자 하였다. 방법:2000년 6월부터 2001년 6월까지 12개월간 서울아산병원 신생아중환자실에 입원하였던 환아중 체중이 1.0kg 이상이면서 임상적으로 동맥혈 가스검사와 전해질, 헤모글로빈, 헤마토크리트 검사가 필요하여 동맥혈 채취를 하게 되었던 환아 60명을 대상으로 하였다. 이들 대상 환아에게서 채취한 혈액으로 기존의 검사를 검사실로 보내고, 잉여혈액을 i-STATTM분석기를 사용하여 검사하였다. 분석대상은 동맥혈가스검사의 pH, 이산화탄소분압, 산소분압, HCO3와 나트륨, 칼륨, 헤모글로빈, 헤마토크리트로 하였다. 결과:총 60명의 환아에게서 동맥혈가스검사는 245건, 나트륨, 칼륨, 헤모글로빈, 헤마토크리트는 195건이 실시되었다. 실시한 결과를 상관분석법을 이용해서 분석했을 때 상관계수는 동맥혈가스검사의 pH 0.954, 이산화탄소분압 0.944, 산소분압 0.941, HCO3 0.880이며 나트륨 0.713, 칼륨 0.860, 헤모글로빈 0.864, 헤마토크리트 0.880으로 통계학적으로 유의한 상관관계를 보였다(p<0.01). 결론:본 연구 결과 i-STATTM 검사의 결과는 기존 검사 결과에 대비하여 유의한 상관관계를 보임을 알 수 있었고, 신생아중환자를 치료하는데 유용하게 사용될 수 있을 것으로 사료된다. Objectives:POCT(Point-of-Care Test), often translated as "Bedside laboratory", is a testing method used in most developed countries to conduct medical research. It is known to extract rapid results that can be applied in the intensive care unit. This study was conducted to investigate the correlation of the i-STATTM POCT analyzer with the traditional test in the neonatal intensive care units. Methods:60 babies(birth weight≥1.0kg) who were admitted to neonatal intensive care unit from June, 2000 to June, 2001 at Asan Medical Center requiring blood samples for the testing of arterial blood gas analysis(ABGA), electrolytes, hemoglobin(Hb), hematocrit(Hct), were included in the study. Blood samples were taken simultaneously to be tested by the traditional laboratory method and by i-STATTM(i-STAT Co. USA) POCT analyzer. The data used for comparison analyzation included pH, pCO2, pO2, HCO3, Na, K, Hb, and Hct. Results:245 measurements of ABGA and 195 measurements of Na, K, Hb, Hct from 60 babies were used for comparison. Good correlation of data was found between i-STATTM POCT analyzer and traditional laboratory method obtained from the central laboratory(correlation coefficient:pH 0.954, pCO2 0.944, pO2 0.941, HCO3 0.880, Na 0.713, K 0.860, Hb 0.864, Hct 0.880). Conclusion:This study showed that i-STATTM POCT analyzer provided accurate analytic results when compared with traditional laboratory method used in the neonatal intensive care units.

        맥문동 물 추출물의 선천면역 활성과 염증억제 효과

        Objectives: This study was designed to examine the anti-cancer activity by innate immunomodulating and anti-inflammatory effects of liriopis tuber water extract (LPE). Methods: Cell cytotoxicity was tested with 4T1 mouse mammary carcinoma cells, spleen cells, macrophage, and RAW264.7 cells. To investigate innate immunomodulating effects of LPE on macrophage, we measured tumor necrosis factor-alpha (TNF-α), interleukin-12 (IL-12), and interleukin-10 (IL-10). To investigate innate immunomodulating effects of LPE on RAW264.7 cell, we measured TNF-α, interleukin-6 (IL-6). In addition, TNF-α and nitric oxide (NO) induced by lipopolysaccharide (LPS) were measured after treating with LPE to observe innate immunomodulating effect of LPE on RAW264.7 cell. Also, mitogen-activated protein kinase (MAPK) and nuclear factor κB (NF-κB) were examined by western blot analysis. Results: In an in vitro cytotoxicity analysis, LPE affected tumor cell growth above specific concentration. As compared with the control group, the production of TNF-α, IL-12, and IL-10 were increased in macrophage. As compared with the control group, the production of TNF-α and IL-6 were increased in RAW 264.7 cell. The expression of TNF-α and NO induced by LPS after treating LPE was decreased. In addition, treatment of RAW 264.7 cell with LPE increased the phosphorylation levels of p-extracellular signal-regulated kinase (p-ERK), p-Jun N-terminal kinase (p-JNK), and p-p38. Conclusions: LPE might have impact on the anti-cancer effect by activation of innate immune system and inflammation control.

        대학생의 인터넷 중독과 우울과의 관련성

        Objectives: To investigate the relationship of internet addiction and depression, and evaluate other possible related factors. Methods: Data was based on the questionnaires from 300 college students in Seoul. Gender, age, major, family income, academic performance, people they live with, location, and main use of internet were included. We used Young's Internet Addiction Test and Beck Depression Inventory(BDI) to measure the depression and addiction degree. Results: In logistic regression analysis, it was found that gender, depression degree and main use of internet were significant variables. There were 136 males, 144 females in this study, and the risk of being addicted to internet were 10.9 times(CI:2.67~44.68) higher in male than female. People with moderate depression (Beck's Depression Inventory(BDI) Score>20) also had higher risk(11.4 times, CI:2.03~63.95) to be addicted in internet than normal group. Lastly, in case of internet use, people who use internet for games and chatting were 6.38(CI:1.38~29.63) and 4.84 times(CI:1.30~18.06) risky compare to the group with web surfing. Conclusion: The result of this study suggests that there is noticeable relationship of internet addiction and depression. However we do oat have evidence of exact relation between them, therefore more studies should be done.

        ISAG-recommended Microsatellite Marker Analysis Among Five Korean Native Chicken Lines

        The objective of this study was to determine genetic variation of five Korean native chicken lines using 30 microsatellite(MS) markers, which were previously recommended by ISAG(International Society for Animal Genetics). The initial study indicated that two microsatellite markers, MCW0284 and LEI0192, were not amplified in these lines and excluded for further analysis. Twenty eight microsatellite markers were investigated in 83 birds from five Korean native chicken lines. The identified mean number of alleles was 4.57. Also, the expected, observed heterozygosity(He, Ho) and polymorphism information content(PIC) values were estimated in these markers and they ranged from 0.31~0.868, 0.145~0.699, and 0.268~0.847, respectively. The results were used for the discrimination of five chicken lines using genetic distance values and also neighbor-joining phylogenetic tree was constructed. Based on the He and PIC values, eighteen markers are enough for the discrimination of these Korean native chicken lines for the expected probability of identity values among genotypes of random individuals(PI), random half sibs(PIhalf-sibs) and random sibs(PIsibs). Taken together, these results will help the decision of conservation strategies and establishment of traceability system in this native chicken breed. Also, the use of ISAG-recommended microsatellite markers may indicate that the global comparison with other chicken breeds is possible.

        RESEARCH : Open Access ; Discrimination of the commercial Korean native chicken population using microsatellite markers

        Background: Korean native chicken (KNC) is a well-known breed due to its superior meat taste. This breed, however, owing to a low growth rate, has a high market price. In order to overcome this disadvantage, the National Institute of Animal Science (NIAS) in Korea developed a commercial KNC breed, named Woorimatdag version 2 (WM2), an upgraded version of the Woorimatdag (WM1) breed and the WM2 was created by crossing the KNC with meat type breeds. This study aims to discriminate between WM2 and other chicken breeds using microsatellite (MS) markers. Methods: A total of 302 individuals from eight Korean chicken populations were examined. The genetic diversity and population structure analysis were investigated using Cervus, API-CALC, STRUCTURE, PowerMarker programs. Results: Based on heterozygosity and polymorphic information content (PIC) values, 30 MS markers were initially selected from 150 markers. The identified average number of alleles (Na), expected heterozygosity, and PIC values for the WM2 samples were 7.17, 0.741, and 0.682, respectively. Additionally, the paternity of individuals was assigned with a success rate of greater than 99% using 12 markers, the best minimum number of markers. The 12 selected markers contained heterozygosity and PIC values above 0.7 and probability of identity values around zero. Using these markers, the determined probability of identity (PI), PIhalf-sibs, and PIsibs values were 3.23E-33, 5.03E-22, and 8.61E-08, respectively. Conclusions: WM2 is well differentiated with respect to other chicken breeds based on estimated genetic distances. The results presented here will contribute to the identification of commercial WM2 chicken in the market.
