        Correlation between Heart-type Fatty Acid-binding Protein Gene Polymorphism and mRNA Expression with Intramuscular Fat in Baicheng-oil Chicken

        This study aims to determine the polymorphism and mRNA expression pattern of the heart-type fatty acid-binding protein (H-FABP) gene and their association with intramuscular fat (IMF) content in the breast and leg muscles of Baicheng oil chicken (BOC). A total of 720 chickens, including 240 black Baicheng oil chicken (BBOC), 240 silky Baicheng oil chicken (SBOC), and 240 white Baicheng oil chicken (WBOC) were raised. Three genotypes of H-FABP gene second extron following AA, AB, and BB were detected by polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) strategy. The G939A site created AA genotype and G956A site created BB genotype. The content of IMF in AA genotype in breast muscle of BBOC was significantly higher than that of AB (p = 0.0176) and the genotype in leg muscle of WBOC was significantly higher than that of AB (p = 0.0145). The G939A site could be taken as genetic marker for higher IMF content selecting for breast muscle of BBOC and leg muscle of WBOC. The relative mRNA expression of H-FABP was measured by real-time PCR at 30, 60, 90, and 120 d. The IMF content significantly increased with age in both muscles. The mRNA expression level of H-FABP significantly decreased with age in both muscles of the three types of chickens. Moreover, a significant negative correlation between H-FABP abundance and IMF content in the leg muscles of WBOC (p = 0.035) was observed. The mRNA expression of H-FABP negatively correlated with the IMF content in both breast and leg muscles of BOC sat slaughter time.

      • The Research of Intelligent Irrigation Control System of Rice in Cold Region

        In traditional way of rice irrigation in the Heilongjiang region,there are many problems such as huge consumption of water, complex wiring and cold water harm. To improve these problems, the intelligent irrigation control system based on PLC and configuration software is designed. The data acquisition and monitoring of water level and water temperature and other parameters are carried through the wireless transmission with PLC as the core, to decide whether irrigation and drainage. In order to solve the problem of cold water harm of rice,the water of Sun pool is heated by solar heating system. The use of solar panels and batteries for power supply of each part, solve the problem of large power loss. The application proves that the system has a positive effect on water—saving and improving the quality and yield of rice, improving the intelligent of irrigation system.

        Protective effect of lncRNA CRNDE on myocardial cell apoptosis in heart failure by regulating HMGB1 cytoplasm translocation through PARP-1

        Long non-coding RNAs (lncRNAs) are boundup with the regulation of various diseases. Here, we probedinto the effect of lncRNA colorectal neoplasia differentiallyexpressed (CRNDE) on heart failure (HF). The pathologicalalterations and cell apoptosis of heart tissues were observedby hematoxylin–eosin and TUNEL staining. The viabilityor apoptosis of mouse myocardial cells HL-1 was tested byXTT or flow cytometry. The interaction between lncRNACRNDE and poly-ADP-ribose polymerase 1 (PARP-1) wasverified by RNA immunoprecipitation and RNA pull-down. The stability of the PARP-1 protein and the acetylation levelof high mobility group box-1 (HMGB1) were determined bycycloheximide-chase and immunoprecipitation, respectively. LncRNA CRNDE expression was decreased in HF micetissues and doxorubicin (Dox)-treated HL-1 cells, whereasPARP-1 and HMGB1 were increased. The overexpressionof lncRNA CRNDE restrained HL-1 cell apoptosis inducedby Dox. Moreover, the interaction between CRNDE andPARP-1 was corroborated, CRNDE negatively regulatedPARP-1 expression, and the overexpression of CRNDEreduced PARP-1 protein stability. In HL-1 cells, PARP-1positively regulated the acetylation level and cytoplasmtranslocation of HMGB1. CRNDE restrained Dox-inducedapoptosis in mouse myocardial cells via the PARP-1/HMGB1 pathway.

        Babeisa duncani infection alters gut microbiota profile in hamsters

        The genus Babesia includes parasites that can induce human and animal babesiosis, which are common in tropical and subtropical regions of the world. The gut microbiota has not been examined in hamsters infected by Babesia duncani. Red blood cells infected with B. duncani were injected into hamsters through intraperitoneal route. To evaluate the changes in gut microbiota, DNAs were extracted from small intestinal contents, acquired from hamsters during disease development. Then, the V4 region of the 16S rRNA gene of bacteria was sequenced using the Illumina sequencing platform. Gut microbiota alternation and composition were assessed according to the sequencing data, which were clustered with >97.0% sequence similarity to create amplicon sequence variants (ASVs). Bacteroidetes and Firmicutes were made up of the major components of the gut microbiota in all samples. The abundance of Bacteroidetes elevated after B. duncani infection than the B. duncani-free group, while Firmicutes and Desulfobacterota declined. Alpha diversity analysis demonstrated that the shown ASVs were substantially decreased in the highest parasitemia group than B. duncani-free and lower parasitemia groups. Potential biomarkers were discovered by Linear discriminant analysis Effect Size (LEfSe) analysis, which demonstrated that several bacterial families (including Muribaculaceae, Desulfovibrionaceae, Oscillospiraceae, Helicobacteraceae, Clostridia UGG014, Desulfovibrionaceae, and Lachnospiraceae) were potential biomarkers in B. duncani-infected hamsters. This research demonstrated that B. duncani infectious can modify the gut microbiota of hamsters.

        Programmed Cell Death-Related Gene Signature Associated with Prognosis and Immune Infiltration and the Roles of HMOX1 in the Proliferation and Apoptosis were Investigated in Uveal Melanoma

        Background Uveal melanoma (UVM) is the most common primary ocular malignancy, with a wide range of symptoms and outcomes. The programmed cell death (PCD) plays an important role in tumor development, diagnosis, and prognosis. There is still no research on the relationship between PCD-related genes and UVM. A novel PCD-associated prognostic model is urgently needed to improve treatment strategies. Objective We aim to screen PCD-related prognostic signature and investigate its proliferation ability and apoptosis in UVM cells. Methods The clinical information and RNA-seq data of the UVM patients were collected from the TCGA cohort. All the patients were classified using consensus clustering by the selected PCD-related genes. After univariate Cox regression and PPI network analysis, the prognostic PCD-related genes were then submitted to the LASSO regression analysis to build a prognostic model. The level of immune infiltration of 8-PCD signature in high- and low-risk patients was analyzed using xCell. The prediction on chemotherapy and immunotherapy response in UVM patients was assessed by GDSC and TIDE algorithm. CCK-8, western blot and Annexin V-FITC/PI staining were used to explore the roles of HMOX1 in UVM cells. Results A total of 8-PCD signature was constructed and the risk score of the PCD signature was negatively correlated with the overall survival, indicating strong predictive ability and independent prognostic value. The risk score was positively correlated with CD8 Tcm, CD8 Tem and Th2 cells. Immune cells in high-risk group had poorer overall survival. The drug sensitivity demonstrated that cisplatin might impact the progression of UVM and better immunotherapy responsiveness in the high-risk group. Finally, Overespression HMOX1 (OE-HMOX1) decreased the cell viability and induced apoptosis in UVM cells. Recuse experiment results showed that ferrostatin-1 (fer-1) protected MP65 cells from apoptosis and necrosis caused by OE-HMOX1. Conclusion The PCD signature may have a significant role in the tumor microenvironment, clinicopathological characteristics, prognosis and drug sensitivity. More importantly, HMOX1 depletion greatly induced tumor cell growth and inhibited cell apoptosis and fer-1 protected UVM cells from apoptosis and necrosis induced by OE-HMOX1. This work provides a foundation for effective therapeutic strategy in tumour treatment.         Cardanol with a Covalently Attached Organophosphate Moiety as a Halogen-Free, Intrinsically Flame-Retardant PVC Bio-Plasticizer

        Plasticizers that enable flexible polyvinyl chloride (PVC) are usually combustible, restricting the application ofPVC in fire-prone scenarios. In this context, intrinsically flame-retardant plasticizers displaying dual function continue to bethe focus of intensive research. Despite their efficiency, the majority of these dual-functional plasticizers previously reportedcontain halogen elements, which, once ignited, emanate toxic and potentially carcinogenic substances, along with toxic gasesand smoke, polluting the environment, damaging the biota, and threatening human health. Here, we report a strategy to obtaina halogen-free, intrinsically flame-retardant PVC bio-plasticizer that harnesses the phenolic hydroxyl of naturally occurringcardanol and covalent attachment of an organophosphate moiety. When combined with di-(2-ethylhexyl) phthalate (DOP),the organophosphate-containing cardanol is qualified as a co-plasticizer, while endowing the PVC materials with flameretardancy. Unlike inorganic flame-retardants, the engineered cardanol is compatible with PVC such that the mechanicalproperties of the PVC materials are not compromised. The rationale underlying the present effort may provide guidance fordeveloping sustainable alternatives to halogen-containing plasticizers to address the sustainability challenge now confrontingPVC industry.
