        Kinematic response of industrial robot with uncertain-but-bounded parameters using interval analysis method

        The uncertain parameters of the deviation in link dimensions and joint clearances caused by manufacturing and assembling errors have a strong impact on the degree of accuracy of the industrial robot. Understanding how these uncertain parameters influence the kinematic response of the industrial robot is critical for design. In this paper, the interval method is used for analysing the kinematic response of the industrial robot with uncertain-but-bounded parameters. The interval used for describing the uncertain parameters is estimated by the grayscale theory. The Denavit-Hartenberg (D-H) method is used for establishing the kinematic model of a 6-degree-of-freedom industrial robot. The kinematic response of the industrial robot calculated by the interval method with uncertain parameters is compared with the results obtained by the probabilistic approach. The comparison results show that the upper or lower bound obtained by interval method is far away from the ideal kinematic response than that obtained by the probabilistic approach. That is a true portrayal of the essence of interval method and probabilistic approach. The research contributes to choose the tolerance zone of uncertain parameters, such as the link dimensions and joint clearances, during the design process of industrial robots.

        Investigate the Effect of Potassium on Nodule Symbiosis and Uncover an HAK/KUP/KT Member, GmHAK5, Strongly Responsive to Root Nodulation in Soybean

        Leguminous plants form root nodule organs with soil rhizobia bacteria, which can fix atmospheric nitrogen (N2) and supply ammonia to the host plant. It is established that the symbiotic N fixation efficiency is substantially influenced by plant nutrients, such as molybdenum (Mo), phosphorus (P), copper (Cu), and zinc (Zn). Potassium (K+) is the most abundant cation in plant cells; however, little evidence regarding the potential link between K nutrition and symbiotic N fixation efficiency is available to date. Here, we showed that K+ deficiency reduced the efficiency of symbiotic nitrogen fixation, and inoculated with rhizobia strain USDA110 could improve plant K+ acquisition. Furthermore, we identified a potassium transporter gene (GmHAK5) that was highly expressed in the root steles and in the vasculature cells of nodules. The GmHAK5 protein was localized at the plasma membrane and could rescue the growth phenotype of yeast K+ uptake-defective mutant. The results obtained from this study provides new insights for the understanding of the potential role of K+ nutrition in modulating symbiotic N fixation in soybean.

        TRIM24-Mediated Acetylation of STAT6 Suppresses Th2-Induced Allergic Rhinitis

        Purpose: Allergic rhinitis (AR) is a T helper type 2 (Th2)-mediated inflammatory disease. The E3 ligase tripartite motif-containing 24 (TRIM24) regulates the recruitment of acetyltransferase CREB-binding protein (CBP) to signal transducer and activator of transcription 6 (STAT6). CBP mediates the acetylation of STAT6 and decreases its activity. To date, the precise role of TRIM24 in AR has not been fully interpreted. Herein, our study aimed to explore the functions of TRIM24 in AR. Methods: The expression of TRIM24 in peripheral blood mononuclear cells (PBMCs) and CD4+ T cells from patients with AR was measured. TRIM24-conditional knockout mice with TRIM24 deficiency in CD4+ T cells were generated. Wide-type (WT) AR mice and TRIM24-conditional knockout AR mice were established. Then, AR symptoms and interleukin (IL)-4 levels were compared. Further, the proliferation, activation and polarization of CD4+ T cells from WT mice and TRIM24 knockout mice after stimulation were determined. The effects of TRIM24 deficiency on STAT6 activities were also evaluated. Results: Downregulated TRIM24 expression was detected in PBMCs and CD4+ T cells from patients with AR. TRIM24 conditional knockout mice had more sever AR symptoms with elevated IL-4 production. TRIM24-knockout CD4+ T cells had similar proliferation and activation when compared to WT CD4+ T cells, while they had enhanced Th2 polarization. TRIM24-knockout CD4+ T cells had decreased acetylation of STAT6 and enhanced STAT6 activities after IL-4 stimulation. The regulation of STAT6 activities by TRIM24 depended on TRIM24 N terminal RIGN domain and Lys383 acetylation site of STAT6. Conclusions: TRIM24 suppresses Th2-mediated AR by regulating the acetylation of STAT6.

        Growth and replication of ordered ZnO nanowire arrays on general flexible substrates

        <P>Vertically aligned and site controllable ZnO nanowire arrays have been synthesized and replicated <I>via</I> hydrothermal method on general flexible substrates. The replication was demonstrated for three generations. The morphology and density of the nanowire arrays could be optimized in the original generation by adjusting the chemical reaction parameters. The pattern of the original generation was inherited by the succeeding generations by a new transferring method. The growth mechanism of the replicated nanowire arrays was investigated with the help of inductively coupled plasma (ICP) etching and AFM tip scanning. The robustness of the ZnO nanowire arrays was obviously improved compared with the seedless ZnO nanowires grown on Au (111) surface, according to the excellent morphology preservation after ultrasonic wave treatment.</P> <P>Graphic Abstract</P><P>Vertically aligned and site controllable ZnO nanowire arrays have been synthesized and replicated <I>via</I> hydrothermal method on general flexible substrates.         Effects of stocking density on the homeostasis of uric acid and related liver and kidney functions in ducks

        Objective: Stocking density (SD) is an important issue in the poultry industry, which is related to the production performance, intestinal health and immune status. In the present study, the effects of SD on the metabolism and homeostasis of uric acid as well as the related functions of the liver and kidney in ducks were examined. Methods: A total of 360 healthy 56-day-old Shan-ma ducks were randomly divided into the low stocking density (n = 60, density = 5 birds/m2 ), medium stocking density (n = 120, density = 10 birds/m2 ) and high stocking density groups (HSD; n = 180, density = 15 birds/m2 ). Samples were collected in the 3rd, 6th, and 9th weeks of the experiment for analysis. Results: The serum levels of uric acid, lipopolysaccharide and inflammatory cytokines (interleukin-1β [IL-1β], IL-8, and tumor necrosis factor-α [TNF-α]) were increased significantly in the HSD group. Serious histopathological lesions could be seen in both the livers and kidneys in the HSD group in the 9th week. The mRNA expression levels of inflammatory cytokines (IL-8 and TNF-α) and related pathway components (toll-like receptor 4, myeloid differentiation primary response gene 88, and nuclear factor-κB) were increased significantly in both the livers and kidneys in the HSD group. The mRNA expression levels of enzymes (adenosine deaminase, xanthine oxidase, phosphoribosyl pyrophosphate amidotransferase, and phosphoribosyl pyrophosphate synthetase 1) related to the synthesis of uric acid increased significantly in the livers in the HSD group. However, the mRNA expression level of solute carrier family 2 member 9, which plays an important role in the excretion of uric acid by the kidney, was decreased significantly in the kidneys in the HSD group. Conclusion: These results indicated that a higher SD could cause tissue inflammatory lesions in the liver and kidney and subsequently affect the metabolism and homeostasis of uric acid, and is helpful for guiding decisions related to the breeding and production of ducks.

        Correlation between red blood cell distribution width/platelet count and prognosis of newly diagnosed diffuse large B-cell lymphoma

        Background Red blood cell distribution width/platelet count ratio (RPR) is a reliable prognostic assessment indicator for numerous diseases. However, no studies to date have examined the relationship between RPR and the prognosis of diffuse large B-cell lymphoma (DLBCL). Therefore, this study aimed to investigate the correlation between RPR and the clinical characteristics and prognosis of patients with diffuse large B-cell lymphoma. Methods We retrospectively studied 143 patients with newly diagnosed DLBCL and used the median value as the RPR threshold. We also investigated the correlation of pretreatment RPR level with clinical characteristics and its impact on DLBCL prognosis. Results Using the median value as the cut-off, patients with DLBCL were divided into a low RPR group (<0.0549) and a high RPR group (≥0.0549). Patients in the high RPR group were older, had a later Ann Arbor stage, were prone to bone marrow invasion, and had a higher National Comprehensive Cancer Network International Prognostic Index score (P < 0.05). A survival analysis showed that progression-free survival (PFS) (P =0.003) and overall survival (OS) (P <0.0001) were significantly shorter in the high versus low RPR group. A multifactorial Cox analysis showed that bone marrow invasion and elevated lactate dehydrogenase (LDH) were separate risk factors for PFS (P <0.05), while an RPR ≥0.0549 and elevated LDH were separate risk factors for OS (P <0.05). Conclusion A high RPR (≥0.0549) in patients with newly diagnosed DLBCL is an independent risk factor for a poor prognosis.

        CCL12 induces trabecular bone loss by stimulating RANKL production in BMSCs during acute lung injury

        In the last three years, the capacity of health care systems and the public health policies of governments worldwide were challenged by the spread of SARS-CoV-2. Mortality due to SARS-CoV-2 mainly resulted from the development of acute lung injury (ALI)/acute respiratory distress syndrome (ARDS). Moreover, millions of people who survived ALI/ARDS in SARS-CoV-2 infection suffer from multiple lung inflammation-induced complications that lead to disability and even death. The lung-bone axis refers to the relationship between lung inflammatory diseases (COPD, asthma, and cystic fibrosis) and bone diseases, including osteopenia/osteoporosis. Compared to chronic lung diseases, the influence of ALI on the skeleton has not been investigated until now. Therefore, we investigated the effect of ALI on bone phenotypes in mice to elucidate the underlying mechanisms. In vivo bone resorption enhancement and trabecular bone loss were observed in LPS-induced ALI mice. Moreover, chemokine (C-C motif) ligand 12 (CCL12) accumulated in the serum and bone marrow. In vivo global ablation of CCL12 or conditional ablation of CCR2 in bone marrow stromal cells (BMSCs) inhibited bone resorption and abrogated trabecular bone loss in ALI mice. Furthermore, we provided evidence that CCL12 promoted bone resorption by stimulating RANKL production in BMSCs, and the CCR2/Jak2/STAT4 axis played an essential role in this process. Our study provides information regarding the pathogenesis of ALI and lays the groundwork for future research to identify new targets to treat lung inflammation-induced bone loss.

        Bta-miR-365-3p-targeted FK506-binding protein 5 participates in the AMPK/mTOR signaling pathway in the regulation of preadipocyte differentiation in cattle

        Objective: MicroRNAs (miRNAs) are endogenous non-coding RNAs that can play a role in the post-transcriptional regulation of mammalian preadipocyte differentiation. However, the precise functional mechanism of its regulation of fat metabolism is not fully understood.Methods: We identified bta-miR-365-3p, which specifically targets the 3′ untranslated region (3′UTR) of the FK506-binding protein 5 (FKBP5), and verified its mechanisms for regulating expression and involvement in adipogenesis.Results: In this study, we found that the overexpression of bta-miR-365-3p significantly decreased the lipid accumulation and triglyceride content in the adipocytes. Compared to inhibiting bta-miR-36 5-3p group, overexpression of bta-miR-365-3p can inhibit the expression of adipocyte differentiation-related genes <i>C/EBPα</i> and <i>PPARγ</i>. The dualluciferase reporter system further validated the targeting relationship between bta-miR-365-3p and FKBP5. FKBP5 mRNA and protein expression were detected by quantitative real-time polymerase chain reaction and Western blot. Overexpression of bta-miR-365-3p significantly down-regulated FKBP5 expression, while inhibition of bta-miR-365-3p showed the opposite, indicating that bta-miR-365-3p negatively regulates FKBP5. Adenosine 5′-monophosphate (AMP)-activated protein kinase/mammalian target of rapamycin (AMPK/mTOR) signaling pathway is closely related to the regulation of cell growth and is involved in the development of bovine adipocytes. In this study, overexpression of bta-miR-365-3p significantly inhibited mRNA and protein expression of <i>AMPK, mTOR</i>, and <i>SREBP1</i> genes, while the inhibition of bta-miR-365-3p expression was contrary to these results. Overexpression of FKBP5 significantly upregulated <i>AMPK, mTOR</i>, and <i>SREBP1</i> gene expression, while inhibition of FKBP5 expression was contrary to the above experimental results.Conclusion: In conclusion, these results indicate that bta-miR-365-3p may be involved in the AMPK/mTOR signaling pathway in regulating Yanbian yellow cattle preadipocytes differentiation by targeting the <i>FKBP5</i> gene.
