1-Kestose Blocks UVB-Induced Skin Inflammation and Promotes Type I Procollagen Synthesis via Regulating MAPK/AP-1, NF-κB and TGF-β/Smad Pathway

Jihye Baek,Jong-Hwa Kim,Jiwon Park,Do Hyun Kim,Soonok Sa,Jung-Sook Han,Wonyong Kim The Korean Society for Microbiology and Biotechnol 2024 Journal of microbiology and biotechnology Vol.34 No.4

Solar UVB irradiation cause skin photoaging by inducing the high expression of matrix metalloproteinase (MMPs) to inhibit the expression of Type1 procollagen synthesis. 1-Kestose, a natural trisaccharide, has been indicated to show a cytoprotective role in UVB radiation-induced-HaCaT cells. However, few studies have confirmed the anti-aging effects. In the present study, we evaluated the anti-photoaging and pathological mechanism of 1-kestose using Human keratinocytes (HaCaT) cells. The results found that 1-kestose pretreatment remarkably reduced UVB-generated reactive oxygen species (ROS) accumulation in HaCaT cells. 1-Kestose suppressed UVB radiation-induced MMPs expressions by blocking MAPK/AP-1 and NF-κB p65 translocation. 1-Kestose pretreatment increased Type 1 procollagen gene expression levels by activating TGF-β/Smad signaling pathway. Taken together, our results demonstrate that 1-kestose may serve as a potent natural trisaccharide for inflammation and photoaging prevention.

Potential Anti-Allergy and Immunomodulatory Properties of Lactococcus lactis LB 1022 Observed In Vitro and in an Atopic Dermatitis Mouse Model

2023 Journal of microbiology and biotechnology Vol.33 No.6

Lactococcus lactis is a lactic acid bacterium and used in the dairy food industry. The ameliorating effects of Lactobacillus species on atopic dermatitis (AD) have been extensively studied, but the specific effect of L. lactis strains has not yet been investigated. In this study, the efficacy of L. lactis LB 1022, isolated from natural cheese, was evaluated using RAW 264.7, HMC-1 and HaCaT cell lines and an ovalbumin-sensitized AD mouse model. L. lactis LB 1022 exhibited nitric oxide suppression and anti-allergy and anti-inflammatory activity in vitro. Oral administration of L. lactis LB 1022 to AD mice significantly reduced the levels of IgE, mast cells, and eosinophils, and a range of T cell-mediated T helper Th1, Th2, and Th17-type cytokines under interleukin (IL)-10, transforming growth factor-β (TGF-β), thymus and activation-regulated chemokine (TARC), and thymic stromal lymphopoietin (TSLP). In addition, L. lactis LB 1022 treatment increased the concentration of short-chain fatty acids. Overall, L. lactis LB 1022 significantly modulated AD-like symptoms by altering metabolites and the immune response, illustrating its potential as candidate for use in functional food supplements to alleviate AD.

Vol.28 No.-

Social isolation and loneliness, so widespread as to be called new pandemics, have emerged not only as individual health and life-satisfaction issues but also as social problems. Older adults, in particular, their chronic conditions deepening and their social networks thinning, are more likely to become socially isolated or lonely. The US has implemented multifaceted initiatives to address social isolation and loneliness among seniors. At the community level, older adults are encouraged to participate in “Village” social activities. When it comes to housing, the senior cohousing program is there to help older adults keep themselves from loneliness and foster a sense of belonging. Additionally, senior-friendly technology platforms and robotic systems are applied to assist older adults to connect with society and keep themselves from social isolation. The health sector has introduced an early screening system for older adults and provides and connects them with various services designed to alleviate their loneliness. This article examines some of the efforts ongoing in the US to reduce social isolation and loneliness among seniors and explores the implications they might have for Korea.

Autophagy-regulating small molecules and their therapeutic applications

Baek, Kyung-Hwa,Park, Jihye,Shin, Injae The Royal Society of Chemistry 2012 Chemical Society reviews Vol.41 No.8

Autophagy or self-eating is a complicated cellular process that is involved in protein and organelle digestion occurring via a lysosome-dependent pathway. This process is of great importance in maintaining normal cellular homeostasis. However, disruption of autophagy is closely associated with various human diseases such as cancer, neurodegenerative disorders, heart disease and pathogen infection. Therefore, small molecules that modulate autophagy can be employed to dissect this complex process and ultimately could have high potential for the treatment of a variety of diseases. This critical review discusses general aspects of autophagy, autophagy-associated diseases and autophagy regulators for biological research and therapeutic applications (207 references).

Graphic Abstract

This critical review discusses general aspects of autophagy, autophagy-associated diseases and autophagy regulators for biological research and therapeutic applications.

Vol.18 No.2

Popular music is a spce where intrinsic desires and values of young people are well exposed in its process of production and consumption. Previous discussions of Korean youth and youth culture have been proceeding mainly in the sense of their resistances to older generation. However, this is limited in that it neglects the inherent microscopic characteristics of youth culture. The purpose of this study is to investigate what the signs and discourses of youth generation make and share in popular music, and how it relates to social context through the ‘Mimesis’ theory. As a result, the songs of the 1990s remarkably showed the resistance to the old generation in its narrative structure and have signs which expresses repression and uniformity. On the other hand, the songs of the 2010s have many fragmented signs which has been reconstructed into individualized discourse structure. It is assumed that the songs have been able to gain a lot of empathy and popularity from the young generation by visualizing emotions and desires experienced by themselves in the social context. Through this study, it suggests to pay attention to the sensitivity, desire and creativity of young people in music in the social and cultural context of the time.

(Background and Purpose)As the ratio of aged population has increased dramatically, the support and consideration for the elderly have become an important challenge for everyone of us. This study was conducted to create new value of housing space for the socially and economically isolated class, or the single elders at the age of 65 of older who are residing in slice rooms, which are the basic living units, to contribute to improving their quality of life. (Method)This study strictly limited the subjects to the `single elders` residing in the `slice room town` in Seoul. First, literature such as preceding studies and publications and the periodicals of research institutes were searched, collected, and studied under the keywords of slice rooms, small-sized houses, single elders, and aging society. Second, the major research area in Changsin-1-dong, Jongno-gu was visited for a spatial analysis to identify the problems to resolve first to secure the substantial findings and subjectivity of research. Third, the single elders residing at the slice room town were visited with the cooperation of Dongdaemun Slice Room Counseling Center to examine their living situations. Single elders in the target area were followed through their daily schedule to be observed and interviewed to identify their complaints. (Results)As a result of analyzing the living environment of slice room residers, they spent much of their time in the slice rooms due to financial hardships and were living in narrow spaces with the living goods piled and unorganized in the rooms. Living inside the slice rooms primarily requires solutions for the basic clothing, food, and housing needs. The living goods should be easy to store and the environment should be sanitary and safe. The living space should provide minimal square footage, but they were residing and cooking in 1.06 py rooms that do not meed the minimum housing standards(14㎡). Therefore, this study examined the ways to arrange the essential living goods owned by the single elders residing in slice rooms within the minimum living space. It studied the ways to store the small/medium goods efficiently for the single elders. (Conclusions) A solution with a design may have limitations, but the following design solutions were suggested to improve their quality of life: first, designs for the socially isolated class should be emotionally touching for the users before providing aesthetics or functionality. Second, the design should be attainable and realistic than a design with viable performance. Third, the design should be built using environmentally-friendly designs without any harm for the body or the environment rather than pursuing low cost for the economical value. Fourth, the findings should not be limited to this study, but consider possible development into a sustainable design.
