Effect of the worn status of wheel/rail profiles on wheel wear over curved tracks

The worn status of wheel/rail profiles could change the wheel-rail contact and influence the wear rates of the wheel and rail. Worn wheel profiles of a Chinese intercity train and the worn rail profile of curved tracks were taken into account in this work. According to the dynamics model of the vehicle, teh wheel-rail three-dimensional steady-state rolling contact model, and the Archard wear model, a new fast calculation method of wheel wear rate was proposed. Simulation results show that obviously severe wear would distribute in the flange when the wheel and rail were worn. The side wear of rail gauge corner will seriously aggravate the wear of new wheel flange. The wear of the wheel is serious for teh worn wheel in the initial phase of service. Therefore, the re-profiling of wheels and rails should be scheduled cooperatively, and the wear of flange in the initial phase of service should addressed.

The purpose of this study was to develop paclitaxel-loaded poly(lactide-co-glycolide) (PLGA) nanoparticles coated with cationic SM5-1 single-chain antibody (scFv) containing a polylysine (SMFv-polylys). SM5-1 scFv (SMFv) is derived from SM5-1 monoclonal antibody, which binds to a 230 kDa membrane protein specifically expressed on melanoma, hepatocellular carcinoma and breast cancer cells. SMFv-polylys was expressed in Escherichia coli and purified by cation-exchange chromatography. Purified SMFv-polylys was fixed to paclitaxel-loaded PLGA nanoparticles to form paclitaxel-loaded PLGA nanoparticles coated with SMFv-polylys (Ptx-NP-S). Ptx-NP-S was shown to retain the specific antigen-binding affinity of SMFv-polylys to SM5-1 binding protein-positive Ch-hep-3 cells. Finally, the cytotoxicity of Ptx-NP-S was evaluated by a non-radioactive cell proliferation assay. It was demonstrated that Ptx-NP-S had significantly enhanced in vitro cytotoxicity against Ch-hep-3 cells as compared with non-targeted paclitaxel-loaded PLGA nanoparticles. In conclusion, our results suggest that cationic SMFv-polylys has been successfully generated and may be used as targeted ligand for preparing cancer-targeted nanoparticles.

Tuberculosis (TB) remains a serious health issue around the word. Adenovirus (Ad)-based vaccine and modified vaccinia virus Ankara (MVA)-based vaccine have emerged as two of the most promising immunization candidates over the past few years. However, the performance of the homologous and heterologous prime-boost immunization regimens of these two viral vector-based vaccines remains unclear. In the present study, we constructed recombinant Ad and MVA expressing an Ag85B-TB10.4 fusion protein (AdH4 and MVAH4) and evaluated the impact of their different immunization regimens on the humoral and cellular immune responses. We found that the viral vector-based vaccines could generate significantly higher levels of antigen-specific antibodies, IFN-γ-producing splenocytes, CD69<sup>+</sup>CD8<sup>+</sup> T cells, and IFN-γ secretion when compared with bacillus Calmette-Guerin (BCG) in a mouse model. AdH4-containing immunization regimens (AdH4-AdH4, AdH4-MVAH4, and MVAH4-AdH4) induced significantly stronger antibody responses, much more IFN-γ-producing splenocytes and CD69<sup>+</sup>CD8<sup>+</sup> T cells, and higher levels of IFN-γ secretion when compared with the MVAH4-MVAH4 immunization regimen. The number of IFN-γ-producing splenocytes sensitive to CD8<sup>+</sup> T-cell restricted peptides of Ag85B (9-1p and 9-2p) and Th1-related cytokines (IFN-γ and TNF-α) in the AdH4-MVAH4 heterologous prime-boost regimen immunization group was significantly higher than that in the other viral vector-based vaccine- and BCG-immunized groups, respectively. These results indicate that an immunization regimen involving AdH4 may have a higher capacity to induce humoral and cellular immune responses against TB in mice than that by regimens containing BCG or MVAH4 alone, and the AdH4-MVAH4 prime-boost regimen may generate an ideal protective effect.

This study explored an integrated strategy for extraction and preparative separation of four anthocyaninsfrom Ribes nigrum L. using DES-based microwave assisted extraction andflash chromatography. Theextraction yield of anthocyanins reached 2.03 mg/g after optimization. An efficient preparativeseparation process of four main anthocyanins by macroporous resin andflash chromatography wasestablished afterwards. The yields of D3G, D3R, C3G and C3R reached 0.32 mg/g, 0.47 mg/g, 0.18 mg/g and0.43 mg/g with the purities of more than 95%, respectively. The developed procedure represents anefficient and convenient alternative for the systematic utilization of natural products from plantmaterials.
Parametric study of pyrolysis and steam gasification of rice straw in presence of K2CO3

A parametric study of pyrolysis and steam gasification of rice straw (RS) was performed to investigate the effect of the presence of K2CO3 on the behavior of gas evolution, gas component distribution, pyrolysis/gasification reactivity, the quality and volume of synthetic gas. During pyrolysis, with the increase in K2CO3 content in RS (i) the instantaneous CO2 concentration was increased while CO concentration was relatively stable; (ii) the yield of CO2 and H2 increased on the cost of CH4. During steam gasification of RS, with the increase in K2CO3 content in RS (i) the instantaneous concentration of CO2 and H2 increased while instantaneous concentration of CO and CH4 decreased; (ii) the yield of CO2 and H2 production and total yield increased; and (iii) yield of CO and CH4 production followed the order: 9% K2CO3 RS<6% K2CO3 RS<raw RS<3% K2CO3 RS<water-leached RS. Water-leached RS showed the highest pyrolysis reactivity, while stream gasification reactivity was proportional to K2CO3 content in RS. The results of this study reveal that the presence of K2CO3 during pyrolysis and steam gasification of RS effectively improves production of H2 rich gas.

Integrated and environmentally friendly methods for extraction and enrichment of natural products havegained increasing interests. The proposed preparation process provides the possibility that extractionand pre-concentration for astragalosides from Radix Astragali roots can be conducted with the surfactantaqueous system in one single procedure. The formulated surfactant system composing of 2% TX-114 assurfactant and 0.03% (w/v) of Gemini 16-5-16 as co-surfactant showed higher extraction efficiency withultrasonic-assisted method and four astragalosides could be enriched by the cloud point phenomenon ofsurfactant. This work represents a valuable alternative for the eco-friendly extraction and enrichment ofsecondary metabolites from plants.
The rationale and potential for using Lactobacillus in the management of periodontitis

Periodontitis refers to a wide range of the inflammatory conditions of supporting dental structures. For some patients with periodontitis, antibacterial agents are needed as an adjuvant to mechanical debridement treatments and oral hygiene maintenance. However, the widespread use of broad-spectrum antibiotics for the prophylaxis and treatment of periodontal infections results in the emergence of resistant pathogens. Therefore, probiotics have become markedly interesting to researchers as a potentially safe alternative to periodontal treatment and maintenance. Probiotics have been used in medicine for decades and extensively applied to the treatment of inflammatory diseases through the modulation of microbial synergy and other mechanisms. A growing amount of evidence has shown that using Lactobacillus strains for oral cavity maintenance could improve periodontal health. In this study, we reviewed studies showing proof of the inhibitory effects of Lactobacillus species on periodontal inflammation. We also explored the rationale and potential for using Lactobacillus species in the management of periodontitis.
