      • Combined Effects of Curcumin and Triptolide on an Ovarian Cancer Cell Line

        Background: As natural medicines in Asia, curcumin and triptolide extracted from different drug plants have proven to possess anticancer potential and widely used for anti-cancer research. The present study attempted to clarify that curcumin and triptolide synergistically suppress ovarian cancer cell growth in vitro. Methods: To test synergic effects, cell viability and apoptosis were analyzed after curcumin and triptolide combination treatment on ovarian cancer cell lines. Synergistic effects on apoptosis induction were determined by lactate dehydrogenase (LDH) leakage assay, intracellular reactive oxygen species (ROS) assay, mitochondrial membrane potential (MMP) loss assay and flow cytometry analysis. Critical regulators of cell proliferation and apoptosis related were analyzed by qRT-PCR and Western blotting. Results: We showed that the combination of curcumin and triptolide could synergistically inhibit ovarian cancer cell growth, and induce apoptosis, which is accompanied by HSP27 and HSP70, indicating that HSP27 and HSP70 play the important role in the synergic effect. Conclusions: From the result present here, curcumin and triptolide combination with lower concentration have a synergistic anti-tumor effect on ovarian cancer and which will have a good potential in clinical applications.

        High alcohol-soluble MoOx gel for interfacial layer in organic solar cells

        Water-free solvent soluble, low-temperature processed metal oxides are important for preparing efficient and stable electronic devices, as well as the convenience in simplifying the device production process. Here we reported a facile approach with the features of low-temperature and solution-based process for the formation of a MoOx (s-MoOx) film as interface layer in polymer solar cells (PSCs). The absorbability, elementary composition, electronic property and surface microstructure of the s-MoOx are investigated in detail by ultravioletevisible spectrophotometer (UVevis), X-ray photoelectron spectrometry (XPS), ultraviolet photo-electron spectrometer (UPS) and atomic force microscopy (AFM). These investigations confirmed that such MoOx xerogel has high solubility in the organic alcohol solvents, such as ethanol and methanol. Meanwhile, this s-MoOx can be applied as the interfacial layer in organic solar cells via a low-temperature treatment (about 100 C) due to its proper physical properties, and a power conversion efficiency (PCE) over 3% was achieved. In addition, the devices with s-MoOx shows excellent air-stability, and the PCE efficiency can maintain about 84% of its initial value after 100 h exposure in air, which is dramatically enhanced comparing with the common devices with PEDOT:PSS layer.

        Comparison between Matrix Method, Equation Method and Full Air-Buoyancy Correction Method for Dissemination of Microgram Weights

        Over the past 9 years since 2007, there has been growing interest in the field of small weight measurement, like determination of sensitivity property of microbalance. This drives the requirement for more accurate small weight measurement down to microgram level. NMIs researched on the microgram from manufacture, material and shape, storage and holding method, measurement equipment and weighing scheme. Subdivision comparison is commonly adopted and matrix method (MM) is used to solve the measurement Eq.. In this paper, the density of test and standard weight rather than the volume were adopted in MM. Details on how to use this method based on Matlab software were presented. Then the comparison between MM, Eqution method (EM) and full air-buoyancy correction method (FACM) were made. Differences between these three methods were analyzed and evaluation results were given out. Experiment showed that EM and FACM were of the same relative uncertainty, but MM had lower measurement uncertainty than EM and FACM.

        Study on mechanism of macro failure and micro fracture of local nearly horizontal stratum in super-large section and deep buried tunnel

        The stability of surrounding rock will be poor when the tunnel is excavated through nearly horizontal stratum. In this paper, the instability mechanism of local nearly horizontal stratum in super-large section and deep buried tunnel is revealed by the analysis of the macro failure and micro fracture. A structural model is proposed to explain the mechanics of surrounding rock collapse under the action of stress redistribution and shed light on the macroscopic analytical approach of the stability of surrounding rock. Then, some highly effective formulas applied in the tunnel engineering are developed according to the theory of mixed-mode micro fracture. And well-documented field case is made to demonstrate the effectiveness and accuracy of the proposed analytical methods of mixed-mode fracture. Meanwhile, in order to make the more accurate judgment about yield failure of rock mass, a series of comprehensive failure criteria are formed. In addition, the relationship between the nonlinear failure criterion and $K_I$ and $K_{II}$ of micro fracture is established to make the surrounding rock failure criterion more comprehensive and accurate. Further, the influence of the parameters related to the tension-shear mixed-mode fracture and compression-shear mixed-mode fracture on the propagation of rock crack is analyzed. Results show that ${\sigma}_3$ changes linearly with the change of ${\sigma}_1$. And the change rate is related to ${\beta}$, angle between the cracks and ${\sigma}_1$. The proposed simple analytical approach is economical and efficient, and suitable for the analysis of local nearly horizontal stratum in super-large section and deep buried tunnel.

      • Expression of Cytoplasmic 8-oxo-Gsn and MTH1 Correlates with Pathological Grading in Human Gastric Cancer

        Background: Cancers have dysfunctional redox regulation resulting in production of reactive oxygen species (ROS), damaging DNA, RNA and free NTPs, and causing the accumulation of oxidative nucleic acids in cytoplasm. The major types are 8-oxo-7,8-dihydroguanine(8-oxoGsn) in RNA and 8-oxo-7,8-dihydro-2' deoxyguanosine(8-oxodGsn) in Mt-DNA. The MTH1 protein sanitizes oxidized nucleotide pools from NTPs to monophosphates, preventing the occurrence of transversion mutations. This study concerned cytoplasmic 8-oxodGsn/Gsn and MTH1 expression in gastric cancer and para-cancer tissues and elucidated roles of nucleic-acid oxidation and anti-oxidation. Materials and Methods: A polymer HRP detection system was used to detect 8-oxo-Gsn/dGsn and MTH1 expression in 51 gastric cancer and para-cancer tissue samples. Analyses of patient clinical and pathological data were also performed. Results: The expression of MTH1 and the 8-oxo-dGsn/Gsn ratio were significantly higher in cancer tissues than para-cancer tissues (P<0.05). Cytoplasmic 8-oxo-Gsn and MTH1 were both found to positively correlate (P<0.05) with tumor differentiation, while no significant associations were found with gender, age, invasion depth, lymph node metastasis and clinical stage (P>0.05). Conclusions: We found 8-oxo-dGsn/Gsn and MTH1 are both highly expressed in gastric cancer tissues, especially in well differentiated lesions. In addition, oxidated mtDNA is prevalently expressed in gastric cancers, while 8-oxo-Gsn expression in cytoplasmic RNA is a bit lower, but more selectively.

      • Clinicopathologic Characteristics and Prognostic Factors in Patients with Operable HER-2 Overexpressing Breast Cancer

        Objective: To study the relationship between clinical pathologic characteristics, treatment modalities and prognostic factors in HER-2 (Human Epidermal growth factor Receptor-2) overexpressed breast carcinoma. Materials and Methods: Major clinico-pathological factors including therapeutic modalities and survival status of 371 breast cancer patients with HER2 over-expression, teated at Yantai Yuhuangding Hospital from March of 2002 to December of 2010 were retrospectively studied, with special attention focused on survival-related factors. Results: The median age of the total 371 patients in this study was 48 years at time of diagnosis, among which, the leading pathological type was infiltrating ductal carcinoma (92.5%); 62.8% presented with a primary tomor larger than 2 cm in diameter at diagnosis, 51.0% had axillary lymph node (ALN) metastases; ER (Estrogen receptor)/PR (Progesterone receptor) double negative occured in 52.8% of cases, and PCNA (proliferation cell nuclear antigen) (+++) was found in 55.1%. HER-2 overexpressed patients were usually in advanced stage when the diagnosis was made (72.8% at stages IIA~IIIC). The prognosis and survival were assessed in 259 patients with complete follow-up data. 5-year DFS (disease-free survival) and OS (overall survival) rate was 68.0% and 78.0% respectively. Univariate analysis revealed that age, tumor size, ALN metastases, LVSI (lymph-vascular space involvement), PCNA status, hormonal therapy, chemotherapy cycles, and HER-2 overexpression, correlated closely with the prognosis. ALN metastases, LVSI, PCNA status and chemotherapy cycles were independent predictors of survival. Conclusions: HER-2 overexpressed breast cancer has special clinical and pathological characteristics, with advanced clinical stages and high rate of ER/PR double negative. Lymph node metastases, LVSI, PCNA and chemotherapy cycles are independent predictors of prognosis.

        An association study between IL1RAPL2 gene and non-specific mental retardation in Chinese children

        Non-specific mental retardation (NSMR) is one of common children psychiatric diseases with a high prevalence (1−3%). Here we investigated the association between the genetic variants of IL1RAPL2 gene and NSMR in the children of QinBa region of China. We chose five common SNPs of IL1RAPL2,examined their individual genotype frequencies using the conventional polymerase chain reaction single strand conformation polymorphism (PCR-SSCP) method, and evaluated the association between these genetic polymorphisms and NSMR with the suitable biostatistic software. The allele and genotype distributions of two SNPs (rs5962298 and rs9887672) showed significant differences between the control and NSMR groups (allele: p = 0.020 and 0.017; genotype: p =0.025 for rs9887672 respectively). The distribution differences became more significant in girls, but disappeared in boys, suggesting a gender effect. Taken together, we provide substantial evidence that IL1RAPL2 conferred a NSMR susceptibility to children of Qinba region in China. In future, further work should be carried out to scan mutations and to investigate the specific-gender effect in this gene.

        Evaluation of Liver Metastases Using Contrast-Enhanced Ultrasound: Enhancement Patterns and Influencing Factors

        Background/Aims: To evaluate the enhancement patterns of liver metastases and their influencing factors using dynamic contrast-enhanced ultrasound (CEUS). Methods: A total of 240 patients (139 male and 101 female; 58.5±11.2 years of age) diagnosed with liver metastases in our hospital were enrolled in this study to evaluate tumor characteristics using CEUS. A comparison of enhancement patterns with tumor size and primary tumor type was performed using the chi-square test. The differences between quantitative variables were evaluated with the independent-sample t-test and one-way analysis of variance. Results: The enhancement patterns of liver metastases on CEUS were categorized as diffuse homogeneous hyperenhancement (133/240, 55.4%), rim-like hyperenhancement (80/240, 33.3%), heterogeneous hyperenhancement (10/240, 4.2%), and isoenhancement (17/240, 7.1%). There were significant differences in the enhancement patterns during the arterial phase based on the nodule size (p=0.001). A total of 231 of the nodules showed complete washout during the portal phase, and 237 nodules were hypoenhanced during the delayed phase. The washout time was correlated with tumor vascularity, with a longer washout time observed in hypervascular metastases compared to hypovascular metastases (p=0.033). Conclusions: Diffuse homogeneous hyperenhancement followed by rapid washout was the most common enhancement pattern of liver metastases on CEUS and was affected by the nodule size and tumor vascularity. Small metastases were prone to show diffuse homogeneous hyperenhancement. Hypervascular metastases showed a significantly longer washout time compared to hypovascular metastases.
