      • MicroRNA Expression Profile Analysis Reveals Diagnostic Biomarker for Human Prostate Cancer

        Prostate cancer is a highly prevalent disease in older men of the western world. MicroRNAs (miRNAs) are small RNA molecules that regulate gene expression via posttranscriptional inhibition of protein synthesis. To identify the diagnostic potential of miRNAs in prostate cancer, we downloaded the miRNA expression profile of prostate cancer from the GEO database and analysed the differentially expressed miRNAs (DE-miRNAs) in prostate cancerous tissue compared to non-cancerous tissue. Then, the targets of these DE-miRNAs were extracted from the database and mapped to the STRING and KEGG databases for network construction and pathway enrichment analysis. We identified a total of 16 miRNAs that showed a significant differential expression in cancer samples. A total of 9 target genes corresponding to 3 DE-miRNAs were obtained. After network and pathway enrichment analysis, we finally demonstrated that miR-20 appears to play an important role in the regulation of prostate cancer onset. MiR-20 as single biomarker or in combination could be useful in the diagnosis of prostate cancer. We anticipate our study could provide the groundwork for further experiments.

        Phases and morphorlogy of CuFeS2 films prepared by electrodeposition

        This work reports one low-priced ways for production of CuFeS2 films on conductive glass surface. CuFeS2 films wereprepared by electrodeposition and post-sulfurization treatment at 220 oC for 30 h in N2 atmosphere for the first time. CuFeS2films were prepared by electrodeposition with sulfate, ascorbic acid, complexing agent sodium citrate as raw materials. Whenthe deposition potential is -1.0 V and the deposition time is 20 min, CuFeS2 thin films with good phrase formation can beobtained at pH 4 and 0.02M Na2S2O3·H2O. The phase of samples were characterized using X-ray diffraction (XRD). Themorphology of samples were characterized via scanning electron microscopy (SEM). The composition of samples werecharacterized by energy dispersive spectrometer (EDS). The crystallinity of CuFeS2 thin films prepared under these conditionsare relatively good. The microcosmic morphology of the samples is flaky crystal. It is shows that post-sulfurization method ishelpful for the phase formation of CuFeS2 films electrodeposited.

        The effects of paeoniflorin injection on soluble triggering receptor expressed on myeloid-1 (sTREM-1) levels in severe septic rats

        Paeoniflorin (PAE) is the most abundant compound in Xuebijing injection widely used to treat sepsis. We aimed to investigate effect of PAE on expression of soluble triggering receptor expressed on myeloid cells-1 (sTREM-1) in a rat model of sepsis. Wistar rats were divided into Normal, Model, and PAE groups (n=20 each). Endotoxin was administrated at 5 mg/ml/kg in Model and PAE rats to establish rat sepsis model. 1 h after endotoxin administration, PAE was administrated at 4 ml/kg in PAE group once per day for 3 days. Routine blood tests and biochemical indexes were assessed, including aspartate aminotransferase (AST) and creatine kinase-MB (CK-MB). The plasma sTREM-1 level was measured using quantitative ELISA. At the end of experiment, the small intestine, liver, kidney and lung were subjected to pathological examinations. A rat model of sepsis-induced multiple organ dysfunction syndrome (MODS) was established successfully with endotoxin administration (5 mg/ml/kg), evidenced by histo-pathological examinations, routine blood tests and biochemical indexes: platelet count decreased and white blood cell count increased (p<0.05), CK-MB and AST increased (p<0.05). PAE treatment significantly reduced the plasma levels of AST, CK-MB, and sTREM-1, compared to Model group (p<0.05). Meanwhile, sepsis-induced damages in the liver, lung, stomach and intestinal mucosa were also markedly ameliorated by PAE treatment. PAE demonstrated a significantly protective effect in a rat model of sepsis by decreasing plasma sTREM-1 level, reducing inflammation, preventing MODS and protecting organ functions.

        Supervised-learning-based algorithm for color image compression

        A correlation exists between luminance samples and chrominance samples of a color image. It is beneficial to exploit such interchannel redundancy for color image compression. We propose an algorithm that predicts chrominance components Cb and Cr from the luminance component Y. The prediction model is trained by supervised learning with Laplacian-regularized least squares to minimize the total prediction error. Kernel principal component analysis mapping, which reduces computational complexity, is implemented on the same point set at both the encoder and decoder to ensure that predictions are identical at both the ends without signaling extra location information. In addition, chrominance subsampling and entropy coding for model parameters are adopted to further reduce the bit rate. Finally, luminance information and model parameters are stored for image reconstruction. Experimental results show the performance superiority of the proposed algorithm over its predecessor and JPEG, and even over JPEG-XR. The compensation version with the chrominance difference of the proposed algorithm performs close to and even better than JPEG2000 in some cases.

        Adaptive polymorphism of tetrameric alpha-amylase inhibitors in wild emmer wheat

        α-Amylase inhibitors are attractive candidates for the control of seed weevils as these insects are highly dependent on starch as an energy source. Wheat tetrameric α-amylase inhibitor (WTAI) is a mixture (60 kDa) of 3 units: WTAI-CM2 plus 2 WTAI-CM3 plus WTAI-CM16, where none of the subunits is active on its own. A total of 334 gene sequences were obtained from 14 populations (131 accessions= genotypes) of wild emmer wheat. The frequencies of SNPs in WTAI-CM2,WTAI-CM3 and WTAI-CM16 were 1 out of 87.6, 101.4, and 108.0 bases, where 5, 5 and 4 SNPs were detected in the coding sequence, respectively. The nucleotide sequence of each unit of tetrameric α-amylase inhibitors were much more conserved than that of dimeric or monomeric inhibitors. The wild emmer wheat populations showed diversity on three WTAI loci,both between and within populations. It was revealed that WTAI were naturally selected for across populations by a ratio of dN/dS as expected. The results of purifying and positive selection hypothesis (p<0.05) also showed that the sequences of WTAI were contributed by natural selection, which ensures the protein function conservation as well as the inhibition diversity with insects amylase enzyme. Ecological factors, singly or in combination, explained a significant proportion of the variations in the SNPs. Ecological factors have an important evolutionary role in gene differentiation at these loci, and tetrameric α-amylase inhibitors are obviously adaptively selected under different environments.

      • Design of 3-electrode system for <i>in situ</i> monitoring direct methanol fuel cells during long-time running test at high temperature

        <P>To understand the effect mechanisms of long-time running and high operation temperature on performance of the direct methanol fuel cell (DMFC) more clearly and directly, in this paper, a new design of 3-electrode system with a solution-type salt bridge has been developed to distinguish the integral polarization into anodic and cathodic polarizations at various temperatures and explore the attenuation mechanism by in situ monitoring the potential of anode during long-time running process at 80 degrees C, for the first time. The results indicate that the optimized 3-electrode system consists of a standard calomel electrode (SCE) and a solution-type salt bridge placed in the anode hole filled by 0.5 mol L-1 H2SO4 solution. By utilization of the 3-electrode system, the effect mechanisms of the running temperature and time on electrochemical parameters of the DMFC have been found: (1) The increasing operation temperature improves cathodic performance more significantly than that of anode; (2) the attenuation of fuel cell performance mainly comes from that of anode during the 20-h running test at 80 degrees C, resulting from the sharp drop of electrochemical active surface area of anode. More important, the new 3-electrode system has simplified the detection equipment and reduced the operating difficulty in a practical application for DMFCs, resulting in its portability.         Study on proton irradiation effect of GaN optical and electrical properties

        In-situ spectra of GaN material during proton irradiation and the infuence of proton energy, fuence and temperature are obtained by the measurement of ion beam-induced luminescence (IBIL). The results show that the main luminescence peak comes from the radiation recombination from shallow donors to deep acceptors. The near-band emission was observed at low temperature, and the intensity presents a single decrease with fuence. When the temperature increases, the near-band emission appears red shift, while the yellow band appears blue shift. The depth distributions of protons in the material are diferent with diferent energies, resulting in variations of spectra due to the interface lattice mismatch and trace impurities. The capacitance–voltage curve and the output characteristic curve of GaN devices before and after irradiation were measured. It is found that the carrier density of the devices after 500 keV proton irradiation decreases more, and the degradation degree of low feld mobility is more serious.

        Population genetics reveals new introgression in the nucleus herd of min pigs

        Objective Min pigs are a unique genetic resource among local pig breeds in China. They have more excellent characteristics in cold and stress resistance, good meat quality, and a high reproductive rate. However, the genetic structure and driving factors remain unclear in the nucleus herd. In this study, the genetic diversity of Min pigs was studied to reveal the formation mechanism of its unique genetic structure. We hope to protect and develop the genetic resources of Min pigs. Methods We analyzed different types of genes to identify the genetic structure and gene introgression pattern of Min pigs. The nuclear DNA dataset includes information on 21 microsatellite loci and 6 Y-chromosome genes, and the mitochondrial D-loop gene is selected to represent maternal lineages. The above genes are all from the nucleus herd of Min pigs. Results The results of genetic structure identification and analysis of potential exogenous gene introgression patterns indicate that the nucleus herd of Min pigs maintains a high level of genetic diversity (polymorphism information content = 0.713, expected heterozygosity = 0.662, observed heterozygosity = 0.612). Compared with other Asian pig breeds, the formation of Min pig breeds is more special. Gene introgression from European pig breeds to Min pigs has occurred, which is characterized by complete introgression of paternal genes and incomplete introgression of maternal genes. Conclusion Gene introgression caused by cross-breeding is not the main factor leading to the formation of the current genetic structure of Min pigs, but this process has increased the level of genetic diversity in the nucleus herd. Compared with the influence of gene introgression, our research suggest that artificial selection and environmental adaptive evolution make Min pigs form unique genetic characteristics.
