        손상 척수의 Lipid Peroxidation과 유수신경섬유의 미세구조에 미치는 Naloxone의 영향

        Spinal Cord contusions in cats were produced experimentally by impact injuries to the surgically exposed cord at the second lumbar vertebral level. As a step in the investigation of the possible effect of spinal cord trauma on biochemical and ultrastructural changes in the injured cord, activities of lipid peroxidation were measured in the frozen-dried sample of the spinal cord and the fine structure of the myelinated nerve fiber in the white matter were observed before and after the cord injury. An increase of lipid peroxidatin level was found as early as 30 minutes after the injury and the highest concentration was reached at 4 hours of injury. Fine structures of the myelinated nerve fibers were changed progressively with the lapse of time after the injury. By 1 hour after the contusion the myelinated nerve fibers showed moderately enlarged periaxonal space, attenuation and disarray of the myelin sheaths. By 4 hour after the contusion, there appeared disarray of the myelin sheaths, greatly enlarged periaxonal space and irregulary contoured axons. The effect of naloxone of the lipid peroxidation and fine ultrastructure of the myelinated nerve fibers were studied, and the results of this study suggested that naloxone have a beneficial effect on the change in lipid peroxidation and the preservation of the myelinated nerve fibers after injury.

        The erythrocytes agglutination test was applied to the common korean plants for lectin activity screening by using human blood. During the four years, 108 species from 46 families of floras were collected, identified and subjected to the test after dividing into several different parts. Only 13 species demonstrated strong lectin activities. Meanwhile 66 species did not shown any agglutination. All others were observed as low activity or as having hemolytic constituents.

      • 별불가사리(Asteriaa pectinifera) 렉틴의 사이토카인 생성 양상

        The purpose of this study is to define whether Asterina pectinifera Lectin (APL) is effective on the cytokine production. Isolated mRNA from hPBMC (human peripheral blood mononuclear cells) stimulated with APL for various reaction times (1 to 96 hours) was detected by RT-PCR. The intensity of band for IL-1 and IFNα mRNA was markedly increased at 1 hour, and IL-2 mRNA was strongly expressedat 4 hours. The mRNA band of APL-induced IL-2 and TFNα was weaker than that of IL-1, IL-6 and TNFα. The mRNA expression of 4 cytokines (IL-1, IL-2, IFN?? and TNFα) was detected up to 48 hours, and that of IL-6 was detected until 72 hours. ELISA was used to look protein secretion of the cytokine gene with IL-1, IL-2 and TNFα expressed strongly in RT-PCR. The highest protein secretion was at 4 hours with IL-1, at 8 hours with IL-2 and at 4 hours with TNFα. These results suggest that APL can induce the pro-duction of some cytokines and the immune response from PBMC was done within the first few hours of stimulation with APL.

        대퇴골절 수술 후 회복실에서 나타난 지방색전증후군

        The fat embolism syndrome is a serious and potentially life threatening complication of long bone trauma, blunt trauma and intramedullary manipulation. A 26-year-old woman underwent an intramedullary nailing of the femur under general anesthesia 45 hours after a traffic accident. The operation ended uneventfully and the patient was extubated in the operatirg room. About one hour after the patient arrived at the recovery room, she progressively developed tachycardia, tachypnea, cyanosis and drowsiness. The fat embolism syndrome was suspected because of the above clinical signs, and because pulmonary edema appeared on a simple chest A-P. During six days of intensive treatment in response to the fat embolism syndrome, the patient,s vital signs and ventilatory status progressively improved. The patient was uneventfully discharged from the hospital 19 days after her operation. (Korean J Anesthesiol 1999; 37: 731∼736)

      • 병원성 원생동물의 병이와 약물내성 : Leishmania major의 클로로퀸 내성 Chloroquine Resistance in Leishmania major

        A comparative study was peformed on two different strains of Leishmania major, chloroquine sensitive strains (Chl^(S)) and its mutant chloroquine resistant strains (Chl^(R)). Chl^(R) strains were obtained at 5×10^(-4) M chloroquine. Remarkable differences were observed at the initial chloroquine uptake in Chl^(R) and Chl^(S), i.e., the rate of uptake was very reduced in Chl^(R) (Km values were 70 nM and 125 nM, respectively). Influx and accumulation of chloroquine were also compared between wild type and mutant. An increasing tendency in both influx and accumulation of chloroquine was shown in Chl^(S), but Chl^(R) demonstrated a rapid release after a little uptake (influx) at the early stage. This result is thought to be basis of their resistance for Chl^(R) strains.

        Observations of Infection Structures on the Leaves of Cucumber PlantsPre-treated with Arbuscular Mycorrhiza Glomus intraradices after Challenge Inoculation with Colletotrichum orbiculare

        Resistance inductions on the leaves of cucumber plant by an arbuscular mycorrhiza Glomus intraradices were investigated. In addition, the infection structures were observed at the penetration sites on the leaves of plant inoculated with Colletotrichum orbiculare using a fluorescence microscope. The severity of anthracnose disease caused by Colletotrichum orbiculare was significantly decreased on the leaves of cucumber plant colonized with G. intraradices compared with those of non-treated control plants. As a positive control, pretreatment with DL-3-aminobutyric acid (BABA) caused a remarkable reduction of the disease severity on the pathogen-inoculated leaves. There were no significant differences in the frequency of either germination or appressorium formation of the plant pathogen between mycorrhiza colonized and non-treated plants. It was also the same on the BABA pre-treated plants. However,the frequency of callose formation was significantly high on the leaves of G. intraradices colonized plants compared to those of non-treated control plants at 5 days after challenge inoculation. On the leaves of BABA treated plants callose formation was not significantly high than those of non-treated, although the disease severity was more strongly suppressed. It was suggested that the resistance induced by colonization with G.intraradices might be related to the enhancement of callose formation at the penetrate sites on the leaves invaded by the pathogen, whereas resistance by BABA did not

      • IMMUNOMODULATING LECTINS FROM MARINE NATURAL PRODUCTS

        The lectins, a very particular bioactive substance, are isolated and purified from natural products and tried to develop as not only medical uses but also biochemical reagents mainly in advanced country. Since these are macro molecules, obtaining through chemical synthesis is not accomplished yet. For this reason, lectins are extremely expensive. For example, according to the Sigma Chemical Co 1995 catalogue, plant lectins Abrin A costs 59.20$/㎎, Con A type V is 158.20$/g PHA-L is 42.00$/㎎ and a marine animal lectin Limulin costs 125.10$/㎎. This research is designed based on our many years of lectins research experiences throughout the world. We have collected over 100 marine natural products (mainly animals). For these marine natural products, our research team has been tried a series of intensive studies accompanying by modem biochemical techniques. These are lectins screening, extraction, separation, purification of the lectin molecules, purity identification, carbohydrate specificity, heat and pH effects on the activity, determination of molecular size, identification of pI, amino acids assay, carbohydrate assay, immunochemical studies, lymphocytes mitogenic activity lymphokine secretion, cancer cell agglutinability, etc. The four marine animals (Asterina pectinifera, Lunella coronata coreenisis, Meretrix lusoria, and Neptunea intersculpta) turned out as important lectin sources through our recent researches. And continuing further investigations are strongly recommended for the development of new drugs as immunomodulators or as cancer treatment drugs.

      • 천연물자원과 향기(방향)요법

        색깔과 향기는 생명현상의 표현 그 자체중의 하나라 할 수 있다. 모든 생명체는 각각 감지되는 정도의 차이는 있을지언정 모두가 독특한 색과 냄새를 지닌 채 태어나고, 자라고, 노화하고, 죽음에 이르는 물질대사의 현상을 나타낸다. 그리고 사람들은 누구나 아름다운 색깔과 신선하고 감미로운 냄새를 좋아한다. 이렇게 좋은 냄새로 더 좋은 환경을 만들어 정신과 육체가 더 건강해 지고 그러므로 생활의 질이 향상되도록 하는 방법, 이것이 바로 향기(방향)요법(aromatherapy)이다. 여기에 이용되는 향의 원료는 천연물중 대부분이 식물의 생식기관에 해당되는 꽃, 열매, 과일, 종자 등에서 얻어진다. 이와 같은 현상은 고대에서부터 현대 문명에 이르기까지 그러했듯이, 미래에도 변함없이 인류는 자연의 산물중 하나인 방향성 식물을 이용할 것이다. 특히 앞으로는 과학기술의 발달과 더불어 열대 다우(多雨)지역 원시림 등 아직 사람들이 접근하지 못한 미개척지에서의 신품종 발견, 환경조절이 가능한 phytotron의 개발 이용, 새로운 생명공학기법의 도입 응용 등으로 더 많고 좋은 새로운 천연자원을 개발하게 될 것이다. 이런 꾸준하고 다양한 노력의 결과로 우리 인류의 생활의 질은 지속적으로 향상되리라 믿는다. 자연과학의 연구목표는 우리가 익혀 터득한 과학기술을 통해 인류를 더 행복하게 하려는데 있다. 이와 같은 자연과학의 바탕에는 문화예술이 있어야 그 과학은 더욱 건실하고 그런 사회와 국가는 번영함을 우리는 알아야한다. 꽃의 색과 향기를 찾아내고 이용하는 방안은 유사이래 문화예술임과 동시에 과학이었음을 강조하며, 다음과 같은 내용에 대하여 언급하고자 한다.

        Extra-Axial Medulloblastoma in the Cerebellar Hemisphere

        Extra-axial medulloblastoma is a rare phenomenon. We report a case in a 5-year-old boy who presented with nausea, vomiting, and gait disturbance. He was treated with total removal of the tumor. This is the first case of an extra-axially located medulloblastoma occurring in the cerebellar hemisphere posteriolateral to the cerebellopontine angle in Korea. Although the extra-axial occurrence of medulloblastoma is rare, it should be considered in the differential diagnosis of extra-axial lesions of the posterior fossa in children.

        The objective of this research is to find new biopotential natural substances from marine invertebrates. 16 species from 11 families of marine invertebrates were collected and studied extensively for their lectin components. Among these, three species, Styela plicata, Halocynthia roretzi, Asterina pectinifera, contained lectins in relatively high amounts.
