Background/Aims: To investigate if BK virus (BKV)-specific T cell immunity measured by an interferon-γ enzyme-linked immunospot (ELISPOT) assay can predict the outcome of BK virus infection in kidney transplant recipients (KTRs). Methods: We included 68 KTRs with different viremia status (no viremia [n = 17], BK viremia [n = 27], and cleared viremia [n = 24]) and 44 healthy controls (HCs). The BK viremia group was divided into controller (< 3 months) and noncontroller ( > 3 months) according to sustained duration of BKV infection. We compared BKV-ELISPOT results against five BKV peptides (large tumor antigen [LT], St, VP1-3). Results: BKV-ELISPOT results were higher in three KTRs groups with different BKV infection status than the HCs group (p < 0.05). In KTR groups, they were higher in cleared viremia group than no viremia or BK viremia group. Within the BK viremia group, controller group had higher LT-ELISPOT results compared to noncontroller group (p = 0.032). Also, KTRs without BK virus-associated nephropathy (BKVN) had higher LT, St, VP1, and VP2-ELISPOT results than those with BKVN (p < 0.05). Conclusions: BKV-ELISPOT assay may be effective in predicting clinical outcomes of BKV infection in terms of clearance of BK virus and development of BKVN.

Acute respiratory syndrome coronavirus SARS-CoV-2 causing COVID-19 has been rapidly spreading worldwide since it occurred in December 2019. SARS-CoV-2 infection occurs as the receptor-binding domain (RBD) of the coronavirus envelope spike protein binds to the angiotensin-converting enzyme 2 (ACE2) receptor. ACE2 is a cell membrane protein activated when it is cleaved and released from the cell surface. The soluble ACE2 can act as a decoy receptor to neutralize the SARS-CoV-2 virus, which is a potential therapy for COVID-19. In this study, the recombinant ACE2 protein was produced in a transient Nicotiana benthamiana plant expression system. The ACE2 was cloned to a fragment crystallizable (Fc) tagged with the KDEL sequence, endoplasmic reticulum (ER) retention signal (ACE2-FcK) in pEAQ-HT, the Cowpea mosaic virus (CPMV)-based transient plant expression vector. To design the recombinant protein structure of ACE2-FcK, the transmembrane region of ACE2 was removed using a transmembrane region prediction program. Four N-glycosylation sites recognized were identified using the N-glycosylation prediction program. Finally, Agrobacterium (LBA4404) carrying pEAQ-HT ACE2-FcK vector was applied for transient expression of ACE2-FcK in N. benthamiana. The proteins (100kDa) were successfully expressed and purified from leaves using protein A affinity chromatography. Additionally, suggested that the plant-derived ACE2-FcK has bioactivities as a therapeutic agent for coronavirus. Taken together, the ACE2-FcK protein can be transiently produced with an optimized expression time and leaf position post-infiltration in plant.
Bilateral medial iliac lymph node excision by a ventral laparoscopic approach: technique description

<P>The aim of this study was to describe a ventral laparoscopic technique for bilateral medial iliac lymphadenectomy in dogs. Twelve intact male purpose-bred research dogs, weighing less than 15 kg, were positioned in dorsal recumbency, and a 3-portal technique was used. Bilateral dissection was performed with vessel-sealing devices while tilting the surgical table by up to 30° towards the contralateral side of the target medial iliac lymph node (MILN) without changing the surgeon’s position. Using a ventral laparoscopic approach, bilateral MILNs were identified and excised in all dogs. The mean times for unilateral and bilateral MILN dissections were 9.7 ± 3.8 and 21.0 ± 6.0 min, respectively. The mean times for the right and left MILN dissections were 10.8 ± 4.3 and 9.8 ± 2.5 min, respectively. The mean total surgery time was 43.7 ± 7.7 min. In total, 26 MILNs were dissected. Several complications, including mild to moderate capillary hemorrhage from perinodal fat and vessels (controlled laparoscopically), mild spleen trauma caused by the first trocar insertion and capsular damage of MILNs, were observed. However, there were no other major complications. All MILN samples were evaluated and deemed suitable for histopathologic diagnosis. Laparoscopic excision of MILNs is a useful method of excisional biopsy for histopathologic diagnosis. Using this ventral laparoscopic approach with the 3-portal technique, bilateral MILN dissection suitable for obtaining histopathologic samples could be achieved in a short time in dogs weighing less than 15 kg.</P>
Kinetin Improves Barrier Function of the Skin by Modulating Keratinocyte Differentiation Markers

Background: Kinetin is a plant hormone that regulates growth and differentiation. Keratinocytes, the basic building blocks of the epidermis, function in maintaining the skin barrier. Objective: We examined whether kinetin induces skin barrier functions in vitro and in vivo. Methods: To evaluate the efficacy of kinetin at the cellular level, expression of keratinocyte differentiation markers was assessed. Moreover, we examined the clinical efficacy of kinetin by evaluating skin moisture, transepidermal water loss (TEWL), and skin surface roughness in patients who used kinetin-containing cream. We performed quantitative real-time polymerase chain reaction to measure the expression of keratinocyte differentiation markers in HaCaT cells following treatment. A clinical trial was performed to assess skin moisture, TEWL, and evenness of skin texture in subjects who used kinetin- containing cream for 4 weeks. Results: Kinetin increased involucrin, and keratin 1 mRNA in HaCaT cells. Moreover, use of a kinetin-containing cream improved skin moisture and TEWL while decreasing roughness of skin texture. Conclusion: Kinetin induced the expression of keratinocyte differentiation markers, suggesting that it may affect differentiation to improve skin moisture content, TEWL, and other signs of skin aging. Therefore, kinetin is a potential new component for use in cosmetics as an anti-aging agent that improves the barrier function of skin. (Ann Dermatol 29(1) 6∼12, 2017)
