        Carbon disulfide induces mitochondria-mediated apoptosis in Sertoli-germ cells coculture

        Carbon disulfide (CS2), a common organic solvent, induces a variety of adverse effects in the male reproductive system. In this study, we investigated the cytotoxicity, ultrastructural changes, and potential apoptotic induction mechanisms of CS2 in mixed cultures of Sertoli and germ cells. Sertoli and germ cells were cocultured and treated with CS2 for 24 h. Growth rates were noted, and apoptotic cells were identified by Hoechst 33258 staining. Ultrastructure changes were observed via transmission electron microscopy (TEM). Mitochondrial membrane potential and expressions of apoptosis-related factors (cytochrome c, Bax, Bcl- 2, caspase-3 and caspase-9) were examined by JC-1 staining, western blot, and real-time PCR. The results showed that CS2 treatment was associated with reduced growth rates of Sertoli-germ cells. Ultrastructure changes in Sertoli-germ cells treated with CS2 were typical of apoptosis. In addition, CS2 treatment depolarized mitochondrial membrane potential, upregulated Bax levels and downregulated Bcl-2 levels, released cytochrome c from the mitochondrial intermembrane space to the cytosol, and triggered mitochondria-mediated apoptosis. Subsequently, caspase-9 and caspase-3 were activated, resulting in Sertoli-germ cells apoptosis. The above data suggest that CS2 has adverse effect on the viability of Sertoli-germ cells and induces apoptosis through mitochondrial pathway.

        Genome Sequencing Highlights the Plant Cell Wall Degrading Capacity of Edible Mushroom Stropharia rugosoannulata

        The basidiomycetous edible mushroom Stropharia rugosoannulata has excellent nutrition, medicine, bioremediation, and biocontrol properties. S. rugosoannulata has been widely and easily cultivated using agricultural by-products showing strong lignocellulose degradation capacity. However, the unavailable high-quality genome information has hindered the research on gene function and molecular breeding of S. rugosoannulata. This study provided a high-quality genome assembly and annotation from S. rugosoannulata monokaryotic strain QGU27 based on combined Illumina-Nanopore data. The genome size was about 47.97 Mb and consisted of 20 scaffolds, with an N50 of 3.73 Mb and a GC content of 47.9%. The repetitive sequences accounted for 17.41% of the genome, mostly long terminal repeats (LTRs). A total of 15,726 coding gene sequences were putatively identified with the BUSCO score of 98.7%. There are 142 genes encoding plant cell wall degrading enzymes (PCWDEs) in the genome, and 52, 39, 30, 11, 8, and 2 genes related to lignin, cellulose, hemicellulose, pectin, chitin, and cutin degradation, respectively. Comparative genomic analysis revealed that S. rugosoannulata is superior in utilizing aldehyde-containing lignins and is possible to utilize algae during the cultivation.

        Mechanical behavior of rock-coal-rock specimens with different coal thicknesses

        To explore the influence of coal thickness on the mechanical behavior and the failure characteristics of rock-coal-rock (RCR) mass, the experimental investigation of uniaxial compressive tests was conducted first and then a systematic numerical simulation by particle flow code (PFC2D) was performed to deeply analyze the failure mechanical behavior of RCR specimens with different coal thicknesses in conventional compression tests. The overall elastic modulus and peak stress of RCR specimens lie between the rock and the coal. Inter-particle properties were calibrated to match the physical sample strength and the stiffness response. Numerical simulation results show that the deformation and strength behaviors of RCR specimens depend not only on the coal thickness, but also on the confining pressure. Under low confining pressures, the overall failure mechanism of RCR specimen is the serious damage of coal section when the coal thickness is smaller than 30 mm, but it is shear failure of coal section when the coal thickness is larger than 30 mm. Whereas under high confining pressures, obvious shear bands exist in both the coal section and the rock section when the coal thickness is larger than 30 mm, but when the coal thickness is smaller than 30mm, the failure mechanism is serious damage of coal section and shear failure of rock section.

        CT-Guided Microcoil Localization of Small Peripheral Pulmonary Nodules to Direct Video-Assisted Thoracoscopic Resection without the Aid of Intraoperative Fluoroscopy

        Objective: To evaluate the feasibility, safety, and effectiveness of CT-guided microcoil localization of solitary pulmonary nodules (SPNs) for guiding video-assisted thoracoscopic surgery (VATS). Materials and Methods: Between June 2016 and October 2019, 454 consecutive patients with 501 SPNs who received CTguided microcoil localization before VATS in our institution were enrolled. The diameter of the nodules was 0.93 ± 0.49 cm, and the shortest distance from the nodules to the pleura was 1.41 ± 0.95 cm. The distal end of the microcoil was placed less than 1 cm away from the nodule, and the proximal end was placed outside the visceral pleura. VATS was performed under the guidance of implanted microcoils without the aid of intraoperative fluoroscopy. Results: All 501 nodules were marked with microcoils. The time required for microcoil localization was 12.8 ± 5.2 minutes. Microcoil localization-related complications occurred in 179 cases (39.4%). None of the complications required treatment. A total of 463 nodules were successfully resected under the guidance of implanted microcoils. VATS revealed 38 patients with dislocated microcoils, of which 28 underwent wedge resection (21 cases under the guidance of the bleeding points of pleural puncture, 7 cases through palpation), 5 underwent direct lobectomy, and the remaining 5 underwent a conversion to thoracotomy. In 4 cases, a portion of the microcoil remained in the lung parenchyma. Conclusion: CT-guided microcoil localization of SPNs is safe and reliable. Marking the nodule and pleura simultaneously with microcoils can effectively guide the resection of SPNs using VATS without the aid of intraoperative fluoroscopy.

      • Effect of Portal Vein Chemotherapy on Liver Metastasis after Surgical Resection of Colorectal Cancer

        Objective: To explore the effect of portal vein chemotherapy on liver metastasis after surgical resection of colorectal cancer. Methods: Patients fulfilling the eligibility criteria were assigned to receive either surgery plus 1-week continuous infusion of 5-FU (study group) or surgery alone (observational group). Patients in the study group received portal vein chemotherapy, whereby 5-FU (1000 mg/d) and heparin (5000 IU/d) infusion was initiated from the day of surgery and lasted for 7 consecutive days. Liver metastasis was monitored during five years follow-up postoperatively. Results: Sixty four patients were recruited and assigned to the study group (12 with colon and 20 with rectal cancer) or the control group (10 with colon and 22 with rectal cancer). Liver metastasis rate was 12.5% in study and 25.0% in observational group, the difference being significant (P<0.01). Conclusion: Portal vein chemotherapy could be an effective treatment in preventing liver metastasis after surgical resection of colorectal cancer.

        Effects of Cryogenic Deformation on Second-Phase Al2Cu Particles and Mechanical Properties of 2219 Al–Cu Alloy Rings

        2219 Al–Cu alloy transition rings are widely used in launch vehicles. However, the coarse and agglomerated second-phaseAl2Cuparticles significantly deteriorate the mechanical properties and ductility of 2219 Al–Cu alloy rings manufactured bytraditional thermal deformation processes. In this study, cryogenic deformation (− 190 °C) is applied for the manufacturingof 2219 Al–Cu alloy rings to alleviate this problem. The effects on the evolution of second-phase Al2Cuparticles and themechanical properties of the T8-aged samples were examined in comparison with the results of room-temperature (25 °C)and conventional thermal deformation at 480 °C. The results indicate that cryogenic deformation can effectively producehigh-density dislocations and strongly crush coarse particles, promoting the dissolution of Al2Cuparticles and improving theirdistribution in the Al matrix when combined with subsequent solution treatment and rolling processes. As the deformationtemperature was decreased from 480 to −190 °C, the area fraction of the coarse particles was decreased from 1.55 to 0.47%,while their mean size was decreased from 11.8 to 8.3 μm. Correspondingly, the uniformity and density of the precipitatesafter T8 aging were improved. Thus, the mechanical properties of the T8-aged samples were improved with decreasingdeformation temperatures; the average ultimate tensile strength, yield strength, and elongation were increased by 20 MPa,22 MPa, and 3.1% at room temperature.

        Characterization of sacbrood virus and its infection in Apis cerana in the Qiandao Lake region in Zhejiang Province, China

        Chinese sacbrood virus (CSBV) is a disastrous virus that fatally threatens honeybees (Apis cerana) in China. Here, we identified the complete genome of CSBV using the rapid amplification of cDNA ends (RACE) method and compared the obtained sequences with those of other SBVs through multiple sequence alignment. The results showed that the full-length of CSBV genome from Zhejiang Province (CSBV-ZJ) is 8824 bp and contains a single, large open reading frame (ORF) that encodes 2842 amino acids flanked by 190 nucleotides (nts) of the 5′-untranslated region (UTR) and 108 nts of the 3′-UTR. The multiple genome comparison showed that SBVs have five conserved domains and that CSBV-ZJ belong to SBV type II. The phylogenetic analysis demonstrated that the CSBV-ZJ strain was similar to the CSBV from Fuzhou (CSBV-FZ) strain. Colonies collected from most of the sampling sites in the area surrounding Qiandao Lake were carriers of CSBV. The haplotypes of the VP1 gene formed a network with radiated and reticular structures. In conclusion, this characterization of the CSBV-ZJ genome will help us understand the SBV prevalence mechanism and aid the protection of honeybees.

        Effect of Heat Treatments on Magnetic Properties and Thermal Expansion Behavior in Two Distinct Types of Fe-Ni Invar Alloys

        The casting process of Fe-Ni Invar alloy could drastically affect magnetic properties and thermal expansion. We have observed analogous trends of saturation magnetization and thermal expansion coefficient in two distinct types of casted samples, as we perform post-annealing at different temperatures. X-ray diffraction measurements show that all the alloys remain in the same face-centered cubic structure after different heat treatments without phase transformation. With the help of X-ray diffraction and differential thermal analysis, we have inferred that these changes in properties might be induced by magnetic lattice transitions and analyzed the possible reason for property differences in samples. This work delivers a perspective on the relationships between thermal expansion, magnetic properties, and heat treatments, which could help to improve the industrial assembly line design.
