        Genetic diversity and population structure of the amylolytic yeast Saccharomycopsis fibuligera associated with Baijiu fermentation in China

        The amylolytic yeast Saccharomycopsis fibuligera is a predominant species in starters and the early fermentation stage of Chinese liquor (Baijiu). However, the genetic diversity of the species remains largely unknown. Here we sequenced the genomes of 97 S. fibuligera strains from different Chinese Baijiu companies. The genetic diversity and population structure of the strains were analyzed based on 1,133 orthologous genes and the whole genome single nucleotide polymorphisms (SNPs). Four main lineages were recognized. One lineage contains 60 Chinese strains which are exclusively homozygous with relatively small genome sizes (18.55–18.72 Mb) and low sequence diversity. The strains clustered in the other three lineages are heterozygous with larger genomes (21.85–23.72 Mb) and higher sequence diversity. The genomes of the homozygous strains showed nearly 100% coverage with the genome of the reference strain KPH12 and the sub-genome A of the hybrid strain KJJ81 at the above 98% sequence identity level. The genomes of the heterozygous strains showed nearly 80% coverage with both the sub-genome A and the whole genome of KJJ81, suggesting that the Chinese heterozygous strains are also hybrids with nearly 20% genomes from an unidentified source. Eighty-three genes were found to show significant copy number variation between different lineages. However, remarkable lineage specific variations in glucoamylase and α-amylase activities and growth profiles in different carbon sources and under different environmental conditions were not observed, though strains exhibiting relatively high glucoamylase activity were mainly found from the homozygous lineage.

        Effect of Lonicera edulis polysaccharide on reducing oral dyeing of lonicera edulis juice

        Fluorescence spectroscopy, particle size determination, and potential analysis were exploited to elucidate the effect of Lonicera edulis polysaccharide on polyphenol protein. The results revealed that Lonicera edulis polysaccharides mediated the binding of polyphenols and proteins through competition and formation of ternary complexes and were also able to enhance the stability of the polyphenol-protein complex solution system. A certain electrostatic effect was also present in the process simultaneously. As confirmed by the dyeing test, to improve oral dyeing, the optimum conditions of adding polysaccharide, pectin, and casein were as follows: the dosage of the polysaccharide group was 1.2 mg/mL, coloring time was 100 min, pH value was 4.0. Pectin group added 0.8 mg/mL with coloring time 80 min, pH 5.0. The addition of casein was 1.2 mg/mL; the coloring time was 100 min with pH 5.0. The sample juice substantiated a significant improvement in the dyeing of porcine tongue mucosa. Under the optimal conditions, microscopic observation validates that the mucosal color of the porcine tongue epidermis was closer to that of unstained porcine tongue epidermis, which significantly improved astringency and oral staining.       • 2,1,3-Benzothiadiazole-5,6-dicarboxylicimide-Based Polymer Semiconductors for Organic Thin-Film Transistors and Polymer Solar Cells

        <P>A series of polymer semiconductors incorporating 2,1,3-benzothiadiazole-5,6-dicarboxylicimide (BTZI) as strong electron-withdrawing unit and an alkoxy-functionalized head-to-head linkage containing bithiophene or bithiazole as highly electron-rich co-unit are designed and synthesized. Because of the strong intramolecular charge transfer characteristics, all three polymers BTZI-TRTOR (P1), BTZI-BTOR (P2), and BTZI-BTzOR (P3) exhibit narrow bandgaps of 1.13, 1.05, and 0.92 eV, respectively, resulting in a very broad absorption ranging from 350 to 1400 nm. The highly electron-deficient 2,1,3-benzothiadiazole-5,6-dicarboxylicimide and alkoxy-functionalized bithiophene (or thiazole) lead to polymers with low-lying lowest unoccupied molecular orbitals (-3.96 to -4.28 eV) and high-lying highest occupied molecular orbitals (-5.01 to -5.20 eV). Hence, P1 and P3 show substantial and balanced ambipolar transport with electron mobilities/hole mobilities of up to 0.86/0.51 and 0.95/0.50 cm(2) V-1 s(-1), respectively, and polymer P2 containing the strongest donor unit exhibited unipolar p-type performance with an average hole mobility of 0.40 cm(2) V-1 s(-1) in top-gate/bottom-contact thin-film transistors with gold as the source and drain electrodes. When incorporated into bulk heterojunction polymer solar cells, the narrow bandgap (1.13 eV) polymer P1 shows an encouraging power conversion efficiency of 4.15% with a relatively large open-circuit voltage of 0.69 V, which corresponds to a remarkably small energy loss of 0.44 eV. The power conversion efficiency of P1 is among the highest reported to date with such a small energy loss in polymer:fullerene solar cells.</P>

        Modified preparation of gangue-loess-based inorganic ceramic membrane and its oil-water separation performance test

        Purpose: Ceramic membranes are recognized for their high mechanical strength and satisfactory acid/alkaline resistance. The innovation of this paper is the preparation of ceramic membrane carriers using cost-effective raw materials gangue andloess as well as oak powder (pore forming agent) and carboxymethyl cellulose (CMC) (binder). These materials are processedthrough extrusion molding and solid particle sintering to create the supports. In addition, with tetraethyl orthosilicate (TEOS)as the precursor, SiO2 thin film was prepared via the sol–gel method, and the support and thin film were modified via thedip coating method to obtain the modified hydrophobic composite ceramic film. In this study, the oil removal rate of thebest ceramic membrane reached 94.1%, the oil–water flux was 1091.3 L/(m2·h·MPa), and the membrane flux recovery ratewas greater than 98%, which provided ideas for the resource utilization of industrial waste and the application of a ceramicmembrane in the field of water treatment.

        RNA sequencing reveals that Prx II gene knockout can down-regulate the allograft rejection of dermal mesenchymal stem cells

        In this study, we used RNA sequencing (RNA-seq) to analyze and compare bulk cell samples from wild-type (WT) dermal mesenchymal stem cells (DMSCs) (n = 3) and Prx II knockout DMSCs (n = 3). The purpose of the study was to elucidate the role of Prx II on allogeneic immune rejection of transplanted DMSCs. The results revealed differential expression of 472 genes (176 up-regulated and 296 down-regulated; p ≤ 0.05) between the PrxII+/+ (WT) and PrxII−/− sample groups. When highly regulated genes were categorized according to the Gene Ontology (GO) molecular function classification and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, the PrxII−/− samples showed a robust downward trend in allograft rejection. The study identified 43 all immunologically rejected differentially expressed genes, of which 41 showed lower expression in the PrxII−/− vs. PrxII+/+ (WT) samples. These findings suggest that Prx II gene knockout may down-regulate the allograft rejection that occurs during DMSCs transplantation and improve the survival rate of DMSCs in the host. This study provides a new perspective on the clinical treatment of stem cell transplantation.

      • Benchtop fabrication of three-dimensional reconfigurable microfluidic devices from paper-polymer composite.

        <P>We presented a benchtop technique that can fabricate reconfigurable, three-dimensional (3D) microfluidic devices made from a soft paper-polymer composite. This fabrication approach can produce microchannels at a minimal width of 100 μm and can be used to prototype 3D microfluidic devices by simple bending and stretching. The entire fabrication process can be finished in 2 hours on a laboratory bench without the need for special equipment involved in lithography. Various functional microfluidic devices (e.g., droplet generator and reconfigurable electronic circuit) were prepared using this paper-polymer hybrid microfluidic system. The developed method can be applied in a wide range of standard applications and emerging technologies such as liquid-phase electronics.</P>

      • Prognostic Significance of Interactions Between ER Alpha and ER Beta and Lymph Node Status in Breast Cancer Cases

        Objective: Both estrogen receptors, ER alpha ($ER{\alpha}$) and ER beta ($ER{\beta}$), are expressed in 50-70% of breast cancer cases. The role of $ER{\alpha}$ as a prognostic marker in breast cancer has been well established as its expression is negative correlated with tumor size and lymph node metastasis. $ER{\beta}$ is also a favorable prognostic predictor although this is less well documented than for $ER{\alpha}$. Materials and Methods: To explore whether ERs independently or together might influence clinical outcome in breast cancer, the correlation between the ERs with the clinicopathological features was analyzed in 84 patients. Results: $ER{\alpha}$ expression negatively correlated with tumor stage (r=-0.246, p=0.028) and tended to be negatively correlated with lymph node status (r=-0.156, p=0.168) and tumor size (r=-0.246, p=0.099). Also, $ER{\beta}$ was negatively correlated with nodal status (r=-0.243, p=0.028), as was coexpression of $ER{\alpha}$ and $ER{\beta}$ (p=0.043, OR=0.194, 95% CI= 0.040-0.953). Conclusion: Coexpression of ERs might serve as an indicator of good prognosis in breast cancer patients.

        《说文解字》刀部刑罚相关汉字考察

        刑罚是统治阶级惩戒罪犯、警醒世人的重要手段。据中国历史文献记载,夏朝已经具备一套成体系的刑罚制度。肉刑的行刑过程离不开各类刀具的使用,因此中国古代的刑罚与“刀”有着密不可分的关系。《说文解字》包含大量和刑罚有关的汉字,这些汉字对于明确古代刑罚种类和内容、探究古代文化生活有着重要的意义和价值。《说文解字》刀部字涉及刑罚的汉字共有8个字。其中“㓷”、“劓”代表“劓刑”,“刖”代表“剕刑”,“ ”、“刭”和“刎”则是“大辟”的一种。“刵”作为割耳刑罚,与意义相近的“聝”、“取”等字联系紧密。“㓷”和“劓”作为割鼻刑罚仅在汉代以前使用广泛。“刖”表示割腿刑罚,古代又有“剕”、“跀”等字表示相同意义,同时又与意义相近的“膑”有所差别。“ ”、“刭”和“刎”三字均表示砍头的刑罚。“罚”则表示对情节较轻犯罪行为的处罚,与表示对严重犯罪行为予以惩戒的“刑”字共同构成汉语中的“刑罚”一词。 Punishment is an important method for the ruling class to discipline criminals and alert people. According to historical records, a systematic punishment system was already existent during the Xia Dynasty. The “Ancient Five Punishments” recorded in Book of Documents occupies an important position in ancient Chinese punishments. Because they were all corporal punishment and the execution methods were extremely cruel so they were abolished in the Han Dynasty. The execution process of corporal punishment depends on the use of various knives. Therefore, punishment in ancient China is closely related to the “knife”. Shuo Wen Jie Zi contains a large number of Chinese characters related to punishment, and they are closely related to the “Ancient Five Punishments”. “刵(Er)”, as an ear-cutting penalty, can be traced back to the Zhou Dynasty or even earlier, and has similar meaning with the words “聝(Guo)” and “取(Qu)”. “㓷(Yi)” and “劓(Yi)” are variants of each other, and they were widely used as a penalty for cutting the nose only before the Han Dynasty. “刖(Yue)” means the punishment of cutting the legs, which has same meaning with the words “剕(Fei)” and “跀(Yue)”, but it was different from the word of “膑(Bin)” though they have similar meaning. The three characters “ (Xing)”, “刭(Jing)” and “刎(Wen)” can be explained by each other, and they all refer to the punishment of beheading. “罚(Fa)” means punishment for minor crimes. This word and the word of “刑(Xing)” meaning the punishment for serious crimes constitute the phrase of “刑罚(Xingfa)” in Chinese.

        MiR-34a, miR-21 and miR-23a as potential biomarkers for coronary artery disease: a pilot microarray study and confirmation in a 32 patient cohort

        The aim of this study was to investigate the expression of circulating microRNAs (miRNAs) in apolipoprotein E (apoE) knockout mice (apoE− / −) and to validate the role of these miRNAs in human coronary artery disease (CAD). Pooled plasma from10 apoE− / − mice and 10 healthy C57BL/6 (B6) mice was used to perform the microarray analysis. The results showed that miR-34a, miR-21, miR-23a, miR-30a and miR-106b were differentially expressed in apoE− / − mice, and these expression changes were confirmed by real-time quantitative reverse-transcription PCR. Then, miR-34a, miR-21, miR-23a, miR-30a and miR-106b were detected in the plasma of 32 patients with CAD and of 20 healthy controls. Only miR-34a, miR-21 and miR-23a were significantly differentially expressed in the plasma of CAD patients (all Po0.01). In conclusion, miR-34a, miR-21 and miR-23a were elevated in CAD patients, which means that these miRNAs might serve as biomarkers of CAD development and progression.

        Ortho-topolin riboside induces apoptosis in Acute myeloid leukemia HL-60 cells

        6-(2-hydroxybenzylamino)-9-D-ribofuranosylpurine (ortho-topolin riboside, oTR), a naturally occurring cytokinin and nucleoside analog has potential anticancer effects. However, the molecular mechanisms remain elusive. We found that oTR strongly inhibited Acute myeloid leukemia HL-60 cell proliferation, altered the cell cycle, induced cytochrome c release from mitochondria into the cytosol, and increased caspase-3 activity. Apoptosis was confirmed by DNA ladder formation following gel electrophoresis. These results indicated that oTR induced apoptosis through activation of the intrinsic mitochondrial pathway. Moreover, the apoptosis was significantly suppressed by the adenosine transporter inhibitor dipyridamole and adenosine kinase inhibitor A-134974. These data indicated that cellular uptake of oTR was an active process involving an adenosine transporter, and subsequently phosphorylated by an adenosine kinase. Taken together, Our study suggests that oTR is taken up by HL-60 cells, converted to the phosphorylated form, and induces apoptosis.
