This article conducted an pilot study on Lee Gyu-Jun(李圭晙)’s life and thoughts. He championed, and is mainly based on, old annotations of ancient Confucian bibles古文 that were made in the Han漢 and Tang唐 era in Chinese history and consequently took additionally into account Zuxi朱熹’s annotations on Confucian bibles. This cause big trouble to him in conservative Gyeong Shang Province(嶺南). He participated in the Confucian Religion Movement孔敎運動 and proposed constitutional monarchy as a new polity fit for changing Korean history. He closely interacted with many resistants to the Japanese rule of Korea and had a mixed perspective on Western culture, science and technology, and social system, positive or negative, contingent on cases. He made great footprints in the history of both modern Korean medicine and philosophy. Thus there is an urgent need for the overall study on Lee Gyu-Jun in every aspect.

<P>Mesenchymal stem cells (MSC) are multipotent in nature and believed to facilitate the engraftment of hematopoietic stem cells (HSC) when transplanted simultaneously in animal studies and even in human trials. In this study, we transfected culture-expanded MSC with granulocyte macrophage-colony stimulating factor (GM-CSF) and stem cell factor (SCF) cytokine genes and then cotransplanted with mononuclear cells (MNC) to further promote HSC engraftment. MNC were harvested from cord blood and seeded in long-term culture for ex vivo MSC expansion. A total of 1×10<SUP>7</SUP> MNC plus MSC/µL were introduced to the tail vein of nonobese diabetic/severe combined immunodeficiency mice. After 6-8 weeks later, homing and engraftment of human cells were determined by flow cytometry and fluorescence in situ hybridization studies. The total nucleated cell count and the engraftment of CD45+/CD34+ cells and XX or XY positive human cells were significantly increased in cotransplanted mice and even higher with the cytokine gene-transfected MSC (GM-CSF>SCF, <I>p</I><0.05) than in transplantation of MNC alone. These results suggest that MSC transfected with hematopoietic growth factor genes are capable of enhancing the hematopoietic engraftment. Delivering genes involved in homing and cell adhesions, CXCR4 or VLA, would further increase the efficiency of stem cell transplantation in the future.</P>

<P>We have proposed and experimentally demonstrated a bidirectional hybrid wavelength-division-multiplexed/subcarrier-multiplexed (SCM)-passive optical network based on a noise-suppressed Fabry-Peacuterot laser diode (FP-LD) and a reflective semiconductor optical amplifier (RSOA). For downlink data transmission, an FP-LD with multiple wavelength characteristics was used, and the mode partition noise of the FP-LD was reduced using a gain-saturated SOA with high-pass filter characteristics. For uplink data transmission, the downlink optical source was reused and remodulated by an SCM method, using an RSOA. A bidirectional SCM link with radio frequencies (RFs) of 2.4 GHz in the downlink and 1.0 GHz in the uplink was implemented. To confirm the validity of the proposed configuration, a 16-quadrature amplitude modulation transmission experiment was performed on a 10-km bidirectional optical access link. The transmission performance was investigated by means of the error vector magnitude and RF spectrum</P>
Evaluation of the Anti-Tumor Effects of Paclitaxel-Encapsulated pH-Sensitive Micelles

We evaluated the efficacy of pH-sensitive micelles, formed by methoxy poly(ethylene glycol)-b-poly($\beta$)-amino ester) (PEG-PAE), as carriers for paclitaxel (PIX), a drug currently used to treat various cancers. PTX was successful encapsulated by a film hydration method. Micelles encapsulated more than 70% of the PTX and the size of the PTX-encapsulated micelles (PTX-PM) was less than 150 nm. In vitro experiments indicated that the micelles were unstable below pH 6.5. After encapsulation of PTX within the micelles, dynamic light scattering (DLS) studies indicated that low pH had a similar demicellization effect. An in vitro release study indicated that PTX was slowly released at pH 7.4 (normal body conditions) but rapidly released under weakly acidic conditions (pH 6.0). We demonstrated the safety of micelles from in vitro cytotoxicity tests on HeLa cells and the in vivo anti-tumor activity of PTX-PM in B16F 10 tumor-bearing mice. We concluded that these pH-sensitive micelles have potential as carriers for anti-cancer drugs.
Effects of Codon Usage Changes on the Expression of Human Interleukin - 2 - Gene in E . coli

Interleukin-2(IL-2) exerts many immunological responses. To study in in vivo as well as in vitro action of IL-2, we have produced recombinant IL-2 in E. coli previously. The IL-2 was produced in the form of insoluble inclusion bodies in E. coli. Production of IL-2 in E. coli as a soluble protein would increase the specific activity of purified IL-2. Since the codon usage of human IL-2 gene was very different from the preferred codons of E. coli, we attempted to change the IL-2 gene using the E. coli codons. A synthetic gene coding for IL-2 was constructed using the oligonucleotides synthesized by an automatic DNA synthesizer. The nucleotide sequence of the synthetic gene was chosen considering the preferred codons of E. coli by not changing the amino acid sequence of IL-2 polypeptide. The levels of IL-2 expression from the synthetic and the natural gene were compared using the expression vectors with similar constructs. Details will be discussed.
Anticoagulation and Anticancer Constituents from Eugenia caryophyllata Thunb

From the EtOAc fraction of Eugenia caryophyllata, four compounds were isolated through activity-guided silica gel column chromatography, From the result of spectroscopic data including NMR, MS and IR, the chemical structures of the compounds were determined as 1-allyl-4-hydroxy-3-methoxybezene acetate (eugenol acetate, 1), 1-allyl-4-hydroxy-3-methoxybezene (eugenol, 2), $3{\beta}-hydroxyolean-12-en-28-oic$ acid (oleanolic acid, 3) and $2{\alpha}$, $3{\beta}-dihydroxyolean-12-en-28-oic$ acid (maslinic acid, 4). Compounds 3 and 4 were isolated for the first time from this plant. Also, compounds 1, 2 and 3 exhibited relatively high platelet aggregation inhibitory activity with the $IC_{50}$ values of 0.24, 0.09 and 0.07 mM, respectively. Compound 2 significantly prolonged activated partial thromboplastin time (aPTT) with the value of $124{\pm}11.2$ seconds as compared to the control with the value of $37.5{\pm}2.2$ seconds at the concentration of 50 ${\mu}g/ml$. Compounds 1 and 3 revealed inhibitory activity on farnesyl protein transferase (FPTase) with the $IC_{50}$ values of 0.49 and 0.24 mM and compounds 1 and 2 highly inhibited the growth of rat-H-ras cells with the $Gl_{50}$ values of 6.63 and 5.70 ${\mu}M$, respectively.

Metagenomic library was constructed from compost made with pig manure and mushroom cultural waste using fosmid vector. Composting was carried out using moving roller system with intermittent mixing and forced aeration, and samples were taken from $7^{th}$- and $14^{th}$-day spots, which represent the mid and late stages of the process, respectively. DNAs of about 40 kb were obtained by preparative electrophoresis on 0.4% low-melting agarose gel to fractionate DNA fragments and to remove humic substances. Total 12,380 fosmid clones were obtained. Restriction analysis of randomly selected clones showed that most clones had different inserts, and average size of inserts was about 35 kb. Two cellulase-positive and five xylanase-positive clones were selected from metagenomic library. Cellulase of clone C1 showed maximal activity at $50^{\circ}C$ and pH 6.0, and retained its original activity after 30 min of heat treatment at $60^{\circ}C$. Optimum temperature for xylanases of clones X1, X2, X3, and X4 was $50^{\circ}C$, and that of clone X5 was $55^{\circ}C$. Thermostabilities of xylanases were in the order of X4>X5>X1, X2, and X3. Optimum pH of xylanases of X1, X2, and X3 was 6.0, that of X4 was 5.5, and that of X5 was 5.5-8.0. Xylanasepositive clones could be divided into three groups, X1/X2/X3, X4, and X5, based on influences of temperature and pH on enzyme activity. Sequence analysis of positive subclone of clone C1 showed cellulase, Cel6H, had the highest similarity of 64% to that of Cellulomonas fimi (P07984), suggesting cellulase and xylanase from metagenomic library are novel enzymes.
Analysis of Ginsenoside Composition of Woods-grown Ginseng Roots

The objective of this research is to provide basic information necessary to differentiate between ginseng (Panax ginseng) grown in woods environments and cultivated ginseng. The ginseng saponin (ginsenoside) contents of Korean woods-grown, 4 year-old cultivated, and 6 year-old cultivated ginsengs were determined via HPLC analysis. The total saponins in the woods-grown ginseng (0.648%) were approximately twice that of the 4 year-old cultivated (0.270%) and the 6 year-old cultivated ginsengs (0.280%). The protopanaxadiols (PD)/protopanaxatriols (PT) ratio of the woods-grown ginseng (3.258%) was higher than that of the 4 year-old cultivated (2.456%) and the 6 year-old cultivated ginsengs (2.183%). The $Rb_1/Rg_1$ ratio of the woods-grown ginseng (10.225%) was also higher than those of the 4 year-old cultivated (3.514%) and the 6 year-old cultivated ginsengs (4.865%).
