        A Molecular Dynamics Simulation Study of the Gibbs-Tolman-Koening-Buff Equation

        The applicability of the Gibbs-Tolman-Koening-Buff (GTKB) equation has been a concern. A scheme is designed to test the GTKB equation. Molecular dynamics simulations are carried out for argon liquid of different systems containing different numbers of atoms with a Lennard-Jones (LJ) potential at a temperature of 78 K. The Tolman length and the surface tension are calculated. Based on the analysis, we conclude the GTKB equation is tenable for R s > 8.

        Anti-thrombotic activity of phenolic acids obtained from Salvia miltiorrhiza f. alba in TNF-a-stimulated endothelial cells via the NF-kB/JNK/p38 MAPK signaling pathway

        Over the past 100 years, Salvia miltiorrhiza f. alba (Lamiaceae) (RSMA) roots have been used to curethromboangiitis obliterans (TAO) in local clinics. This studyaimed to confi rm the anti-thrombotic effi cacy of 12 phenolicacids obtained from RSMA and to clarify the possibleunderlying mechanisms. The results of quantitative real-timepolymerase chain reaction (qRT-PCR) and enzyme-linkedimmunosorbent assay (ELISA) experiments demonstratedthat most of the phenolic acids markedly inhibited PAI-1protein and mRNA levels but increased t-PA protein andmRNA levels in TNF-α-induced EA.hy926 cells ( P < 0.05or 0.001), with lithospermic acid displaying the strongesteff ect. In vitro anticoagulation and antiplatelet aggregationassays showed that lithospermic acid and salvianolic acidB signifi cantly prolonged prothrombin time (PT), activatedpartial thromboplastin time (APTT), decreased fi brinogen concentration (FIB), and inhibited platelet aggregationinduced by adenosine diphosphate (ADP) in rat blood. Bothlithospermic acid and salvianolic acid B markedly down-regulatedthe expression of factor Xa and factor IIa on the externalsurface of EA.hy926 cells and demonstrated signifi cantanti-factor IIa and anti-factor Xa activity using chromogenicsubstrates in vitro. Western blot results revealed that bothlithospermic acid and salvianolic acid B also signifi cantlyinhibited the expression of TF, p-p65, p-p38, and pJNK proteinsinduced by TNF-α. These results indicated that all ofthe phenolic acids appeared to have some anti-thromboticactivity, with salvianolic acid B and lithospermic acid markedlydecreasing the chance of thrombosis by regulating theNF-κB/JNK/p38 MAPK signaling pathway in response toTNF-α.

        The mechanism of mesna in protection from cisplatin-induced ovarian damage in female rats

        Objective: Cisplatin is a widely used chemotherapeutic agent in the treatment of cancers in clinic; but it often induces adverse effects on ovarian functions such as reduced fertility and premature menopause. Mesna could attenuate the cisplatin-induced ovarian damages; however, the underlying mechanism is still unknown. This study aimed to figure out the underlying mechanism of the protection of mesna for ovaries against cisplatin therapy in cancers. Methods: We performed female adult Sprague-Dawley rats into normal saline control (NS), low-dose cisplatin (CL), high-dose cisplatin (CH), CL plus mesna (CL+M), and CH plus mesna (CH+M) groups and detected anti-Müllerian hormone (AMH)-positive follicle, oxidative stress status and anti-oxidative capability in ovaries. Results: AMH-positive follicles were significantly decreased after cisplatin administration, which was significantly reversed when mesna was co-administered with cisplatin. The end product of lipid peroxidation, malondialdehyde (MDA), was significantly increased, but the anti-oxidative enzymatic activity of superoxide dismutase (SOD) and glutathione (GSH) were significantly decreased in cisplatin groups when compared with NS group. In contrast, after co-administration of cisplatin with mesna, MDA was significantly decreased whereas the activity of SOD and the concentration of GSH were increased. Moreover, mesna did not decrease the anti-tumor property of cisplatin in HePG2 cell lines. Conclusion: Cisplatin damages the granulosa cells by oxidative stress to deplete the ovarian reserve and mesna could protect ovarian reserve through anti-oxidation. These results might highlight the mechanism of the protection of mesna for ovarian reserve and open an avenue for the application of mesna as a protective additive in cisplatin chemotherapy in clinical practise.

        Effects of Dietary Flaxseed Lignan Extract on Symptoms of Benign Prostatic Hyperplasia

        A flaxseed lignan extract containing 33% secoisolariciresinol diglucoside (SDG) was evaluated for its abilityto alleviate lower urinary tract symptoms (LUTS) in 87 subjects with benign prostatic hyperplasia (BPH). A randomized, dou-ble-blind, placebo-controlled clinical trial with repeated measurements was conducted over a 4-month period using treatmentdosages of 0 (placebo), 300, or 600 mg/day SDG. After 4 months of treatment, 78 of the 87 subjects completed the study.For the 0, 300, and 600 mg/day SDG groups, respectively, the International Prostate Symptom Score (IPSS) decreased . 3.67.1.56, . 7.33. 1.18, and . 6.88. 1.43 (mean. SE, P. .100,. .001, and. .001 compared to baseline), the Quality of Lifescore (QOL score) improved by . 0.71. 0.23, . 1.48. 0.24, and . 1.75. 0.25 (mean. SE, P. .163 and .012 comparedto placebo and P. .103,. .001, and. .001 compared to baseline), and the number of subjects whose LUTS grade changedfrom “moderate/severe” to “mild” increased by three, six, and 10 (P. .188, .032, and .012 compared to baseline). Maximumurinary flows insignificantly increased 0.43. 1.57, 1.86. 1.08, and 2.7. 1.93 mL/second (mean. SE, no statistical sig-nificance reached), and postvoiding urine volume decreased insignificantly by . 29.4. 20.46, . 19.2. 16.91, and . 55.62.36.45 mL (mean. SE, no statistical significance reached). Plasma concentrations of secoisolariciresinol (SECO), enterodiol(ED), and enterolactone (EL) were significantly raised after the supplementation. The observed decreases in IPSS and QOLscore were correlated with the concentrations of plasma total lignans, SECO, ED, and EL. In conclusion, dietary flaxseed lig-nan extract appreciably improves LUTS in BPH subjects, and the therapeutic efficacy appeared comparable to that of com-monly used intervention agents of .1A-adrenoceptor blockers and 5.-reductase inhibitors.
