        Effects of Dietary Supplementation with Hainanmycin on Protein Degradation and Populations of Ammonia-producing Bacteria In vitro

        An in vitro fermentation was conducted to determine the effects of hainanmycin on protein degradation and populations of ammonia-producing bacteria. The substrates (DM basis) for in vitro fermentation consisted of alfalfa hay (31.7%), Chinese wild rye grass hay (28.3%), ground corn grain (24.5%), soybean meal (15.5%) with a forage: concentrate of 60:40. Treatments were the control (no additive) and hainanmycin supplemented at 0.1 (H0.1), 1 (H1), 10 (H10), and 100 mg/kg (H100) of the substrates. After 24 h of fermentation, the highest addition level of hainanmycin decreased total VFA concentration and increased the final pH. The high addition level of hainanmycin (H1, H10, and H100) reduced (p<0.05) branched-chain VFA concentration, the molar proportion of acetate and butyrate, and ratio of acetate to propionate; and increased the molar proportion of propionate, except that for H1 the in molar proportion of acetate and isobutyrate was not changed (p>0.05). After 24 h of fermentation, H10 and H100 increased (p<0.05) concentrations of peptide nitrogen and AA nitrogen and proteinase activity, and decreased (p<0.05) $NH_3$-N concentration and deaminase activity compared with control. Peptidase activitives were not affected by hainanmycin. Hainanmycin supplementation only inhibited the growth of Butyrivibrio fibrisolvens, which is one of the species of low deaminative activity. Hainanmycin supplementation also decreased (p<0.05) relative population sizes of hyper-ammonia-producing species, except for H0.1 on Clostridium aminophilum. It was concluded that dietary supplementation with hainanmycin could improve ruminal fermentation and modify protein degradation by changing population size of ammonia-producing bacteria in vitro; and the addition level of 10 mg/kg appeared to achieve the best results.

        Fabrication and characterization of fluorohydroxyapatite nanocrystals/poly(d,l-lactide) composite scaffolds

        Poly(D,L-lactide) (PDLLA)/uorohydroxyapatite nanocrystals (nano-F-HA) porous scaolds were successfully fabricated through asolvent-casting and particulate-leaching technique. Nano-HA/PDLLA scaold and PDLLA scaold were prepared by using the sameprocess for comparison. The structure, phase and morphology of the nanocomposite scaolds were observed by SEM. The results indi-cated that F-HA nanocrystals were homogeneously dispersed in the PDLLA matrix. The porosity of the scaolds was up to 90%, andmacropores and micropores coexisted and interconnected throughout the scaolds. Cell culture experiments demonstrated that the nano-F-HA/PDLLA scaffold had the best adhesion tendency to chondrocytes among the scaffolds investigated.

        Global Histone H4 Acetylation of IGF1 and GH Genes in Lungs of Somatic Cell Cloned Calves

        Histone acetylation modification is one key mechanism in the regulation of gene activation. In this study, we investigated the global levels of histone H4 acetylation of insulin like growth factor I (IGF1) and growth hormone (GH) genes in the lungs of two somatic cell cloned calves. Data showed the levels of histone H4 acetylation of IGF1 and GH genes vary widely within different gene regions, and, in almost all regions of the two genes, acetylation levels are lower in the aberrant clone than in the normal clone. Thus we suggest that inefficient epigenetic reprogramming in the clone may affect the balance between acetylation and deacetylation, which will affect normal growth and development. These findings will also have implications for improvement of cloning success rates.

        Covalent attachment of chiral alkaloids onto silicon surfaces

        In this work, we report the covalent attachment of a chiral molecule onto a hydrogen-terminated Si(SiH) surface by using an extre-mely mild approach. Cinchonine, a chiral alkaloid was self-assembled onto a SiH surface via a photochemistry method at room tem-perature. The self-assembled monolayers (SAMs) obtained were characterized by means of ellipsometry, XPS and wettability studies.The lm formation kinetics was monitored by ellipsometry and XPS, which indicated that a full monolayer was formed within 15 h.

      • Pseudonocardia endophytica sp. nov., isolated from the pharmaceutical plant Lobelia clavata

        <P>An endophytic actinomycete strain, designated YIM 56035(T), was isolated from the inner tissue of the traditional Chinese medicinal plant Lobelia clavata. The strain stained Gram-positive, was aerobic and exhibited branching, white aerial mycelium and yellowish-brown substrate mycelium. The strain formed spore chains on aerial hyphae. The G+C content of the genomic DNA was 70.3 mol%. On the basis of morphological, physiological, chemotaxonomic and phylogenetic characteristics, strain YIM 56035(T) was assigned to the genus Pseudonocardia. Analysis of 16S rRNA gene sequences showed 98.5, 97.3, 97.3 and 97.1 % similarity to the closely related type strains Pseudonocardia kongjuensis LM 157(T), Pseudonocardia autotrophica IMSNU 20050(T), Pseudonocardia ammonioxydans H9(T) and Pseudonocardia compacta IMSNU 20111(T), respectively. The results of DNA-DNA hybridizations and comparison of some phenotypic characteristics revealed that the strain represents a novel species of the genus Pseudonocardia. The name Pseudonocardia endophytica sp. nov. is proposed, with the type strain YIM 56035(T) (=DSM 44969(T) =CCTCC AA 206026(T) =KCTC 19150(T)).</P>

        Preparation and characterization of glass-ceramics with α-cordierite as the main crystalline phase from bluestone tailings

        The bluestone tailings are quickly expanded in China with the continuous utilization and mining of bluestone resources. Theincremental recycling of bluestone tailings is important to solve the resources waste and environmental pollution. This studyaims to reuse the bluestone tailings as the main material to prepare α-cordierite glass-ceramics based on melting process. Theresults show that the nucleation temperature, crystallization peak temperature and the activation energy for crystallizationdecreased gradually with increasing the percentage of bluestone tailings. The complete preparation parameters of α-cordieriteglass-ceramics include a bluestone tailings percentage of 70%, a heating rate of 5 oC·min^-1, a crystallization duration of 1.5h, a crystallization temperature of 970 oC, a nucleation temperature of 830 oC, and a nucleation duration of 1.0 h. Theperformances analyses reveals that the α-cordierite glass-ceramics based on the optimized parameters exhibits high densityand Vickers hardness, low dielectric loss and water absorption, and strong chemical resistance.

        Taxonomy of fungal complex causing red-skin root of Panax ginseng in China

        Background: Red-skin root of Asian ginseng (Panax ginseng) significantly reduces the quality and limits theproduction of ginseng in China. The disease has long been thought to be a noninfectious physiologicaldisease, except one report that proved itwas an infectious disease. However, the causal agents have not beensuccessfully determined. In the present study, we were to reveal the pathogens that cause red-skin disease. Methods: Ginseng roots with red-skin root symptoms were collected from commercial fields in NortheastChina. Fungi were isolated from the lesion and identified based on morphological characters alongwith multilocus sequence analyses on internal transcription spacer, b-tubulin (tub2), histone H3 (his3),and translation elongation factor 1a (tef-1a). Pathogens were confirmed by inoculating the isolates inginseng roots. Results: A total of 230 isolates were obtained from 209 disease samples. These isolates were classifiedinto 12 species, including Dactylonectria sp., D. hordeicola, Fusarium acuminatum, F. avenaceum, F. solani,F. torulosum, Ilyonectria mors-panacis, I. robusta, Rhexocercosporidium panacis, and three novel speciesI. changbaiensis, I. communis, and I. qitaiheensis. Among them, I. communis, I. robusta, and F. solani had thehighest isolation frequencies, being 36.1%, 20.9%, and 23.9%, respectively. All these species isolated werepathogenic to ginseng roots and caused red-skin root disease under appropriate condition. Conclusion: Fungal complex is the causal agent of red-skin root in P. ginseng.

        Evaluation of Houttuynia cordata and Taraxacum officinale on Growth Performance, Nutrient Digestibility, Blood Characteristics, and Fecal Microbial Shedding in Diet for Weaning Pigs

        A total of 144 pigs ((Landrace${\times}$Yorkshire)${\times}$Duroc)] with an average initial BW of $8.45{\pm}0.57$ kg were used in a 5-wk growth trial. Pigs were randomly allocated to 4 treatments with 9 replications per pen in a randomized complex block design. Dietary treatments included: i) CON (basal diet), ii) ANT (CON+tylosin 1 g/kg), iii) H1 (CON+H. cordata 1 g/kg) and iv) T1 (CON+T. officinale 1 g/kg). In this study, pigs fed the ANT and T1 treatment had a higher (p<0.05) average daily gain (ADG) and gain:feed (G:F) ratio than those fed CON and H1 treatment. Dietary ANT and T1 treatment led to a higher energy digestibility than the CON group. No difference (p>0.05) was observed on the growth performance and apparent total tract digestibility with H1 supplementation compared with the CON treatment. The inclusion of ANT treatment led to a higher (p<0.05) lymphocyte concentration compared with the CON treatment. Dietary supplementation of herbs did not affect (p>0.05) the blood characteristics (white blood cell (WBC), red blood cell (RBC), IgG, lymphocyte). No difference was observed on (p<0.05) fecal microbial shedding (E. coli and lactobacillus) between ANT and CON groups. Treatments H1 and T1 reduced the fecal E. coli concentration compared with the CON treatment, whereas the fecal lactobacillus concentration was not affected by the herb supplementation (p>0.05). In conclusion, the inclusion of T. officinale (1 g/kg) increased growth performance, feed efficiency, energy digestibility similarly to the antibiotic treatment. Dietary supplementation of T. officinale and H. cordata (1 g/kg) reduced the fecal E. coli concentration in weaning pigs.

        Measurement of aerodynamic coefficients of tower components of Tsing Ma Bridge under yaw winds

        Tsing Ma Bridge in Hong Kong is the longest suspension bridge in the world carrying both highway and railway. It has two H-shape concrete towers, each of which is composed of two reinforced concrete legs and four deep transverse prestressed concrete beams. A series of wind tunnel tests have been performed to measure the aerodynamic coefficients of the tower legs and transverse beams in various arrangements. A 1:100 scaled 3D rigid model of the full bridge tower assembled from various tower components has been constructed for different test cases. The aerodynamic coefficients of the lower and upper segments of the windward and leeward tower legs and those of the transverse beams at different levels, with and without the dummy bridge deck model, were measured as a function of yaw wind angle. The effects of wind interference among the tower components and the influence of the bridge deck on the tower aerodynamic coefficients were also investigated. The results achieved can be used as the pertinent data for the comparison of the computed and field-measured fully coupled buffeting responses of the entire bridge under yaw winds.
