        Characterization of Mutations in AlHK1 Gene from Alternaria Longipes: Implication of Limited Function of Two-Component Histidine Kinase on Conferring Dicarboximide Resistance

        Four series (S, M, R, and W) of Alternaria longipes isolates were obtained based on consecutive selection with Dimethachlon (Dim) and ultraviolet irradiation. These isolates were then characterized according to their tolerance to Dim, sensitivity to osmotic stress, and phenotypic properties. All the selected Dim-resistant isolates showed a higher osmosensitivity than the parental strains, and the last generation was more resistant than the first generation in the M, R, and W series. In addition, the changes in the Dim resistance and osmotic sensitivity were not found to be directly correlated, and no distinct morphologic characteristics were found among the resistant and sensitive isolates, with the exception of the resistant isolate K-11. Thus, to investigate the molecular basis of the fungicide resistance, a group III two-component histidine kinase (HK) gene, AIHK1, was cloned from nineteen A. longipes isolates. AIHK1p was found to be comprised of a six 92-amino-acid repeat domain (AARD), HK domain, and response regulator domain, similar to the Os-1p from Neurospora crassa. A comparison of the nucleotide sequences of the AIHK1 gene from the Dim-sensitive and -resistant isolates revealed that all the resistant isolates contained a single-point mutation in the AARD of AIHK1p, with the exception of isolate K-11, where the AIHK1p contained a deletion of 107 amino acids. Moreover, the AIHK1p mutations in the isolates of each respective series involved the same amino acid substitution at the same site, although the resistance levels differed significantly in each series. Therefore, these findings suggested that a mutation in the AARD of AIHK1p was not the sole factor responsible for A. longipes resistance to dicarboximide fungicides.

        Strategies for controlling the ghost ant, Tapinoma melanocephalum (Hymenoptera: Formicidae) with liquid bait

        The ghost ant, Tapinoma melanocephalum (F.), is a household pest and a considerable nuisance. The aim of this study is to demonstrate the toxicity and control efficacy of boric acid in liquid bait against queen and worker ghost ants. The LT50 values for workers fed with 0.5%–2.5% boric acid and 2% chicken peptone in 20% sucrose water solutions were 4.3–2.4 days. The lethal times (LT50=5.2–7.6 days) for queen ghost ants fed with various concentrations of a boric acid solution depended on the feeding behavior of the queens. The high boric acid (4%)content solutions were not repellent to the ghost antworkers. The liquid bait formulation of 1% boric acid,which caused a 100%worker, brood, and queen population reduction in 4, 4, and 5 weeks, respectively,was significantly more effective than the solid bait formulation containing the same concentration (p≦0.05). The simulation tests involved using chicken peptone and sucrose as the attractant, and colonies were provided an alternate food source (20% sucrose solution and dog food) to achieve a more accurate assessment of bait acceptability in screening for the efficacy of the liquid boric acid bait. The control efficiency attained 99.9% inweek 4. The results demonstrated that liquid bait, containing 2% chicken peptone, 20% sucrose as a food attractant, and 1% boric acid as the toxin, is efficient and highly recommended for ghost ant control.

      • Depletion of LINE1-ORF1P Causes GV Arrest in Mouse Oocytes In Vitro

        LINE-1 is an autonomous non-LTR retrotransposon in mammalian genomes and encodes ORF1P and ORF2P. ORF2P has been clearly identified as the enzyme supplier needed in LINE-1 retrotransposition. However, the role of ORF1P is not well explored and requires further elucidation. In this study, we depleted ORF1P to investigate its role in mouse oocyte meiotic maturation. The results showed that depletion of ORF1P caused oocyte arrest at the germinal vesicle (GV) stage as well as down-regulation of Cdc2 and Cyclin B1, components of the maturation promoting factor (MPF). Further analysis demonstrated a decreased expression of the P21 upstream factors Smad4 and Dcp1a after ORF1P depletion. However, SMAD4 and DCP1A became accumulated in the nucleus. This translocation would up-regulate the expression of P21. Furthermore, ORF1P knockdown also increased the expression of microRNA-494, which could decrease the expression of Cyclin B1 and Cdc2. Propidium Iodide (PI) staining after ORF1P depletion revealed abnormal chromatin configuration in the GV of mouse oocytes. Taken together, these results indicate that ORF1P is involved in the TGF-β pathway to modulate the GV breakdown (GVBD) during mouse oocyte meiotic maturation.

        Identification and Characterization of an Anti-fungi Fusarium oxysporum f. sp. cucumerium Protease from the Bacillus subtilis Strain N7

        A newly discovered alkaline antifungal protease named P6 from Bacillus subtilis N7 was purified and partially characterized. B. subtilis N7 culture filtrates were purified by 30–60% (NH4)2SO4 precipitation, anion-exchange chromatography and gel filtration chromatography. Sodium dodecyl sulfatepolyacrylamide gel electrophoresis (SDS-PAGE) revealed a single band of 41.38 kDa. Peptide sequence of protease P6 was determined using a 4800 Plus MALDI TOF/TOFTM Analyzer System. Self-Formed Adaptor PCR (SEFA-PCR) was used to amplify the 1,149 bp open read frame of P6. Dimensional structure prediction using Automatic Modeling Mode software showed that the protease P6 consisted of two β-barrel domains. Purified P6 strongly inhibited spore and mycelium growth of Fusarium oxysporum f. sp. cucumerium (FOC) by causing hypha lysis when the concentration was 25 μg/ml. Characterization of the purified protease indicated that it had substrate specificity for gelatin and was highly active at pH 8.0–10.6 and 70°C. The P6 protease was inhibited by EDTA (2 mmol/L), phenyl methyl sulfonyl fluoride (PMSF, 1 mmol/L), Na+, Fe3+, Cu2+, Mg2+ (5 mmol/L each) and H2O2 (2%, v/v). However, protease activity was activated by Ca2+, K+, Mn2+ (5 mmol/L each), mercaptoethanol (2%, v/v) and Tween 80 (1%, v/v). In additon, activity was also affected by organic solvents such as acetone, normal butanol and ethanol, but not hexane (25%, v/v each).

        <P>Polo-like kinase (PLK) 4 is a unique member of the PLK family that plays vital roles in centriole biogenesis during mitosis. The localization of PLK4 on centrioles must be precisely regulated during mitosis to ensure correct centriole duplication. However, little is known about the function of PLK4 in mammalian oocyte meiosis. We addressed this question by examining the expression and localization of PLK4 in mouse oocytes and using RNA interference and protein overexpression to investigate its function in meiosis. PLK4 expression peaked at the germinal vesicle breakdown (GVBD) stage, and the protein was localized in the cytoplasm throughout meiotic maturation. Depletion of PLK4 caused meiotic arrest at the GV stage and suppressed CYCLINB1 and CDC2 activities. Moreover, PLK4 depletion prevented the de-phosphorylation of CDC2-Tyr15 in nucleus and induced a decrease in the level of the CDC25C protein. PLK1 overexpression failed to rescue GV-stage arrest in PLK4-depleted oocytes, whereas overexpressing PLK4 resulted in normal GVBD in oocytes in which PLK1 activity was inhibited. In addition, PLK4 overexpression did not cause abnormal spindle formation or affect extrusion of the first polar body. These results illustrate the fact that PLK4 is essential for meiotic resumption but may not influence spindle formation in mouse oocytes during meiotic maturation.</P>

        Optimization of Medium Composition for Lipopeptide Production from Bacillus subtilis N7 using Response Surface Methodology

        The nutritional requirements for the maximum production of lipopeptides by Bacillus subtilis N7 (B. subtilis N7) were investigated and optimized using response surface methodology (RSM) under shake flask fermentation. A one-factor-at-a time experimental setup was used to screen carbon and nitrogen sources. A Plackett?Burman design (PBD) was employed to screen the most critical variables for lipopeptides production amongst ten nutritional elements. The central composite experimental design (CCD) was finally adopted to elucidate the composition of the fermentation medium. Statistical analyses (analysis of variance, ANOVA) of the results showed that KCl, MnSO4 and FeSO4·6H2O were important components and that their interactions were strong. Lipopeptide production was predicted to reach 709.87 mg/L after a 60 h incubation using an optimum fermentation medium composed of glucose 7.5 g/L, peanut oil 1.25 g/L, MgSO4 0.37 g/L, KH2PO4 0.75 g/L, monosodium glutamate 6.75 g/L, yeast extract and NH4Cl (5:3 w/w) 10 g/L, KCl 0.16 g/L, FeSO4·6H2O 0.24 mg/L, MnSO4 0.76 mg/L, and an initial pH of 7.0. Lipopeptide production (706.57 ± 3.70 mg/L) in the optimized medium confirmed the validity of the predicted model.

        What are the Individual`s Real Cares to Switch Personal Cloud Services?

        As the fast development of information technology (IT), abundant attractive alternatives have showed up and challenged to the traditional information technology (IT). Cloud services, the hottest representative among these alternatives, has attracted the attention from all walks of life. Considering the diversity of cloud services, this study attempts to find out the important factors affected on individual`s switching intention from incumbent IT to cloud services mainly based on two-factor theory (switching enablers and switching inhibitors) and explore the direct influences of these factors on individual`s switching intention. According to our findings, individual`s switching intention to cloud services is not only positively influenced by switching enablers but also negatively influenced by switching inhibitors. All the switching enablers in this study (perceived omnipresence, perceived collaboration support and perceived compatibility of cloud services) have positively significant influences on individual`s switching intention as well as the switching inhibitors (usage habit of incumbent IT and perceived risk of cloud services) have negatively significant influences on individual`s switching intention. It`s noteworthy that satisfaction of incumbent IT has insignificant influence on individual`s switching intention in this study. Moreover, as we expected, both social influence and personal innovativeness have significant influences on the generation of individual`s switching intention.
