        Novel splice isoforms of pig myoneurin and their diverse mRNA expression patterns

        Objective: The aim of this study was to clone alternative splicing isoforms of pig myoneurin (MYNN), predict the structure and function of coding protein, and study temporal and spatial expression characteristics of each transcript. Methods: Alternative splice isoforms of MYNN were identified using RNA sequencing (RNA-seq) and cloning techniques. Quantitative real-time polymerase chain reaction (qPCR) was employed to detect expression patterns in 11 tissues of Large White (LW) and Mashen (MS) pigs, and to study developmental expression patterns in cerebellum (CE), stomach (ST), and longissimus dorsi (LD). Results: The results showed that MYNN had two alternatively spliced isoforms, MYNN-1 (GenBank accession number: KY470829) and MYNN-2 (GenBank accession number: KY670835). MYNN-1 coding sequence (CDS) is composed of 1,830 bp encoding 609 AA, whereas MYNN-2 CDS is composed of 1,746 bp encoding 581 AA. MYNN-2 was 84 bp less than MYNN-1 and lacked the sixth exon. MYNN-2 was found to have one $C_2H_2$ type zinc finger protein domain less than MYNN-1. Two variants were ubiquitously expressed in all pig tissues, and there were significant differences in expression of different tissues (p<0.05; p<0.01). The expression of MYNN-1 was significantly higher than that of MYNN-2 in almost tissues (p<0.05; p<0.01), which testified that MYNN-1 is the main variant. The expression of two isoforms decreased gradually with increase of age in ST and CE of MS pig, whereas increased gradually in LW pig. In LD, the expression of two isoforms increased first and then decreased with increase of age in MS pig, and decreased gradually in LW pig. Conclusion: Two transcripts of pig MYNN were successfully cloned and MYNN-1 was main variant. MYNN was highly expressed in ST, CE, and LD, and their expression was regular. We speculated that MYNN plays important roles in digestion/absorption and skeletal muscle growth, whereas the specific mechanisms require further elucidation.

      • The Design of Anti-aliasing Analog Filter for Data Acquisition in the Surface Measurement

        In the surface measurement system, the data acquisition is key part and the anti-aliasing analog filter is necessary. This paper deals with the design of the anti-aliasing analog filter. Based on the principle of anti-aliasing filtering, the parameters of filter are computed, the filter circuit is designed, and the frequency characteristic is drawn. Combined with digital filter, the filtering can maintain the low frequency components very well and suppress effectively the high frequency signals in the original surface profile, which reduces the distortion caused by noise and makes the filtering effect better. This method has been applied in the surface measurement system and the actually measured data verified the performance of the filter.

        In this short paper, the SAIST method recently proposed for image denoising is improved by exploiting the fixed rank and sparsity constraints. Experimental results show that the improved SAIST obtains better performance than the original SAIST and BM3D.

        Adjustment of Hydrophilic-Lipophilic Balance of Hexamethyldisilazane-Modified Nanosilica for Enhanced Oil Recovery

        Different dosages of hexamethyldisilazane (denoted as HMDS), a silane coupling agent, were adopted to modify nanosilica (denoted as NS) to afford a series of HMDS-NS nanoparticles with different hydrophilic-lipophilic balance governed by the amount of surface hydroxyl. The amounts of the hydrophilic hydroxyl of the as-prepared HMDS-NS nanoparticles and their water contact angles were measured, and their dispersing behavior in water and oil was examined in relation to their transfer behavior therein. Moreover, the effects of the as-prepared HMDS-NS nanofluids on the oil–water interfacial tension as well as the oil recovery were investigated based on interfacial tension measurements and simulated rock core flooding tests. Findings indicate that the hydrophilic-lipophilic balance of HMDS-NS nanoparticles highly depends on the amount of the surface hydroxyl, and the surface hydroxyl amount can be well adjusted by properly selecting the dosage of HMDS modifier. Besides, the transfer behavior of HMDS-NS nanoparticles in oil and water is closely related to their hydrophilic-lipophilic balance, and they can greatly reduce the oil–water interfacial tension and increase the oil recovery by 7.7–11.1% as compared with conventional water flooding. This is because the surface grafting of the hydrophobic segments of HMDS leads to a significant increase in the hydrophobicity of nanosilica, thereby changing the wettability of oil on the sand surface and favoring the stripping of oil droplets. Particularly, the HMDS-NS nanofluid obtained with 2wt.% of HMDS modifier has a water contact angle of 83.6 ° and can dramatically reduce the oil–water interfacial tension from 20.22mN/m to 0.28mN/m, showing desired hydrophilic-lipophilic balance and potential for enhanced oil recovery associated with chemical flooding.

        Green and Efficient Synthesis of an Adsorbent Fiber by Plasma-induced Grafting of Glycidyl Methacrylate and Its Cd(II) Adsorption Performance

        A new chelating adsorbent for the removal of Cd(II) from aqueous solution, PP-g-GMA-DETA fibers, is prepared by plasma induced grafting of glycidyl methacrylate (GMA) onto the surface of polypropylene (PP) fibers, followed by modification with diethylenetriamine (DETA). The effects of grafting parameters on the grafting degree are investigated. Fourier transform infrared spectroscopy, scanning electron microscopy, and X-ray photoelectron spectroscopy prove the successful grafting of GMA onto the surfaces of PP fibers and the subsequent conversion of epoxy groups of grafted GMA to amine groups, on reaction with DETA. The adsorption performances of Cd(II) by the chelating fibers are evaluated in detail. Kinetic and isothermal parameters are also evaluated. The data fit well with the pseudo-second order and Langmuir isotherm model, respectively. The maximum adsorption capacity of Cd(II) is 46.47 mg g-1 and the prepared fibers show selective adsorbability towards target Cd(II) in presence of competing Mg(II) ions.

        Appraisal of Guidelines for the Management of Blood Pressure in Patients with Diabetes Mellitus: The Consensuses, Controversies and Gaps

        Background: Currently available guidelines contain conflicting recommendations on the management of blood pressure (BP) in patients with diabetes mellitus (DM). Therefore, it is necessary to appraise the guidelines and summarize the agreements and differences among recommendations.Methods: Four databases and the websites of guideline organizations were searched for guidelines regarding BP targets and thresholds for pharmacologic therapy in DM patients, and the included guidelines were appraised with the Appraisal of Guidelines for Research and Evaluation (AGREE) II instrument.Results: In 6,498 records identified, 20 guidelines met our inclusion criteria with 64.0% AGREE II scores (interquartile range, 48.5% to 72.0%). The scores of the European and American guidelines were superior to those of the Asian guidelines (both adjusted P<0.001). Most of the guidelines advocated systolic BP targets <130 mm Hg (12 guidelines, 60%) and diastolic BP targets <80 mm Hg (14 guidelines, 70%) in DM patients. Approximately half of the guidelines supported systolic BP thresholds >140 mm Hg (10 guidelines, 50%) and diastolic BP thresholds >90 mm Hg (nine guidelines, 45%). The tiny minority of the guidelines provided the relevant recommendations regarding the lower limit of official BP targets and the ambulatory BP monitoring (ABPM)/home BP monitoring (HBPM) targets and thresholds in DM patients.Conclusion: The lower official BP targets (<130/80 mm Hg) in patients with DM are advocated by most of the guidelines, but they contain conflicting recommendations on the official BP thresholds. Moreover, the gaps regarding the lower limit of official BP targets and the ABPM/HBPM targets and thresholds need to be considered by future study.

        A Method for k Nearest Neighbor Query of Line Segment in Obstructed Spaces

        In order to make up the deficiencies of the existing research results which cannot effectively deal with the nearest neighbor query based on the line segments in obstacle space, the k nearest neighbor query method of line segment in obstacle space is proposed and the STA_OLkNN algorithm under the circumstance of static obstacle data set is put forward. The query process is divided into two stages, including the filtering process and refining process. In the filtration process, according to the properties of the line segment Voronoi diagram, the corresponding pruning rules are proposed and the filtering algorithm is presented. In the refining process, according to the relationship of the position between the line segments, the corresponding distance expression method is put forward and the final result is obtained by comparing the distance. Theoretical research and experimental results show that the proposed algorithm can effectively deal with the problem of k nearest neighbor query of the line segment in the obstacle environment.

        Fractional Order Sliding Mode Control for Permanent Magnet Synchronous Motor Speed Servo System via an Improved Disturbance Observer

        A fractional order sliding mode control (FOSMC) method is developed in this paper to deal with the control problem of permanent magnet synchronous motor (PMSM) speed servo system subject to multiple disturbances including model uncertainties, unknown constant disturbances and harmonic disturbances. The lumped exogenous disturbances and uncertainties of the PMSM speed servo are estimated by an improved disturbance observer (DO) and an extended state observer (ESO), respectively. Then, a novel FOSMC law is developed by incorporating the feedforward compensation and a fractional order switching law. The stability of the closed-loop system is established based on Lyapunov stability approach. Under the FOSMC scheme, the tracking performance and robustness of the PMSM servo system are improved simultaneously in the presence of mismatched disturbance torques and measurement noise. The effectiveness and advantages of the proposed method are demonstrated by the PMSM speed regulation experiments and the comparisons with some existing methods.

        Glutamic-oxaloacetic transaminase 1 regulates adipocyte differentiation by altering nicotinamide adenine dinucleotide phosphate content

        Objective: This study was performed to examine whether the porcine glutamic-oxaloacetic transaminase 1 (GOT1) gene has important functions in regulating adipocyte differentiation. Methods: Porcine GOT1 knockout and overexpression vectors were constructed and transfected into the mouse adipogenic 3T3-L1 cells. Lipid droplets levels were measured after 8 days of differentiation. The mechanisms through which GOT1 participated in lipid deposition were examined by measuring the expression of malate dehydrogenase 1 (MDH1) and malic enzyme (ME1) and the cellular nicotinamide adenine dinucleotide phosphate (NADPH) content. Results: GOT1 knockout significantly decreased lipid deposition in the 3T3-L1 cells (p< 0.01), whereas GOT1 overexpression significantly increased lipid accumulation (p<0.01). At the same time, GOT1 knockout significantly decreased the NADPH content and the expression of MDH1 and ME1 in the 3T3-L1 cells. Overexpression of GOT1 significantly increased the NADPH content and the expression of MDH1 and ME1, suggesting that GOT1 regulated adipocyte differentiation by altering the NADPH content. Conclusion: The results preliminarily revealed the effector mechanisms of GOT1 in regulating adipose differentiation. Thus, a theoretical basis is provided for improving the quality of pork and studies on diseases associated with lipid metabolism.         Selection of candidate genes affecting meat quality and preliminary exploration of related molecular mechanisms in the Mashen pig

        Objective: The aim of this study was to select the candidate genes affecting meat quality and preliminarily explore the related molecular mechanisms in the Mashen pig. Methods: The present study explored genetic factors affecting meat quality in the Mashen pig using RNA sequencing (RNA-Seq). We sequenced the transcriptomes of 180-day-old Mashen and Large White pigs using longissimus dorsi to select differentially expressed genes (DEGs). Results: The results indicated that a total of 425 genes were differentially expressed between Mashen and Large White pigs. A gene ontology enrichment analysis revealed that DEGs were mainly enriched for biological processes associated with metabolism and muscle development, while a Kyoto encyclopedia of genes and genomes analysis showed that DEGs mainly participated in signaling pathways associated with amino acid metabolism, fatty acid metabolism, and skeletal muscle differentiation. A MCODE analysis of the protein-protein interaction network indicated that the four identified subsets of genes were mainly associated with translational initiation, skeletal muscle differentiation, amino acid metabolism, and oxidative phosphorylation pathways. Conclusion: Based on the analysis results, we selected glutamic-oxaloacetic transaminase 1, malate dehydrogenase 1, pyruvate dehydrogenase 1, pyruvate dehydrogenase kinase 4, and activator protein-1 as candidate genes affecting meat quality in pigs. A discussion of the related molecular mechanisms is provided to offer a theoretical basis for future studies on the improvement of meat quality in pigs.
