        Screening potential reference genes for reverse transcription-quantitative polymerase chain reaction normalization in Atrijuglans hetaohei (Lepidoptera: Gelechioidea)

        Atrijuglans hetaohei Yang (Lepidoptera: Gelechioidea), is one of the major pests that seriously damage the walnut fruits. Although the morphology and physiology of A. hetaohei have been widely studied, suitable reference genes for normalizing target gene expression have not been identified. In this study, the expressions of eight candidate reference genes including GAPDH, β-Tubulin, TATA-binding protein (TBP), Histone, Ubiquitin-conjugating enzyme 9 (UBC9), arginine kinase (AK), 28S and elongation factor 1 alpha (EF1α) in different developmental stages and various larval tissues of A. hetaohei were determined using reverse transcription-quantitative polymerase chain reaction. Moreover, the expression stability of the selected reference genes was evaluated by ΔCt method, BestKeeper, NormFinder, geNorm and ReFinder, a comprehensive software platform. These results demonstrated that the best reference genes were GAPDH and 28S at different developmental stages of A. hetaohei; and the optimum references in various larval tissues of A. hetaohei were 28S, Histone and TBP. Our study should be useful to analyze the expression profiles of target genes and form a solid foundation for future research on the understanding of the genes’ biological functions in A. hetaohei.

        Identification, characterization, and expression of two novel P450 genes CYP6BQ21 and CYP6BQ22 from Dastarcus helophoroides (Coleoptera: Bothrideridae)

        Chemical pesticides often applied to effectively control the long-horned beetles to protect the forests are reported to affect the non-target organisms adversely. Dastarcus helophoroides is an active natural enemy of long-horned beetles. Studying the molecular mechanism of P450 genes will help to elucidate the metabolic mechanism of pesticides in D. helophoroides to better coordinate the use of chemical and biological controls. In this study, two novel genes, CYP6BQ21, and CYP6BQ22 were successfully cloned from D. helophoroides using the rapid ampli fication of cDNA ends technique. The sequence and homology analyses indicated that CYP6BQ21 was highly similar to CYP6BQ1 from Tribolium castaneum, while CYP6BQ22 was closely related to CYP6BQ13 from T. castaneum. Gene expression patterns showed that CYP6BQ21 and CYP6BQ22 were specifically expressed in the adult stage of D. helophoroides. In addition, CYP6BQ21 and CYP6BQ22 were significantly expressed under the treatment of a high concentration of cypermethrin. Based on these findings, we proposed that CYP6BQ21 and CYP6BQ22 played possible roles in the development and pesticide stress of D. helophoroides. Our findings are an important first step in identifying and characterizing CYP6BQ21 and CYP6BQ22 from D. helophoroides, and lay the groundwork for future research into the role of these novel CYP6s in the regulation of pesticide resistance in D. helophoroides.

        Transcriptome Analysis Reveals Molecular Mechanisms Underlying Methyl Jasmonate-mediated Biosynthesis of Protopanaxadiol-type Saponins in Panax notoginseng Leaves

        Methyl jasmonate (MeJA) has been widely used to improve the biosynthesis of secondary metabolites such as triterpenoid saponins in medicinal plants. However, the underlying molecular mechanisms remain poorly understood. Differing from roots that accumulate protopanaxatriol-type saponins, Panax notoginseng leaves with a lower biomass mainly contain protopanaxadiol (PPD)-type saponins. Therefore, it is interesting to explore whether MeJA can activate the biosynthesis of PPD-type saponins in P. notoginseng leaves. In this study, we found MeJA could effectively induce the accumulation of PPD-type saponins, including ginsenoside Rb1, Rc, Rb2, Rb3 and notoginsenoside Fa, Fe in P. notoginseng leaves based on a newly established high-performance liquid chromatography method. Transcriptome analysis showed that differentially expressed genes (DEGs) induced by MeJA were mainly enriched in “terpenoid backbone biosynthesis”, “biosynthesis of unsaturated fatty acids”, “sesquiterpenoid and triterpenoid biosynthesis”, “fatty acid metabolism”, and “phenylpropanoid biosynthesis”. Furthermore, the expression profile and quantitative real-time PCR analysis of DEGs showed that MeJA could positively induce the molecular response of endogenous jasmonic acid (JA) signaling pathway, and increased PPD-type saponins mediated by MeJA in P. notoginseng leaves may be related to the high expression of FPS, SS, SE, DS and UGTs, and the low expression of CYP716A53v2 and β-AS. The results provide a molecular understanding for MeJA-elicited biosynthesis of triterpenoid saponins and facilitate the further characterization of the genes responsible for biosynthesis of PPD-type saponins in P. notoginseng leaves.

        2-(2-Cyanopropyl) Dithiobenzoate-mediated Grafting Polymerization of Methyl Methacrylate from Vinyl Modified Silica

        Grafting polymerization of methyl methacrylate (MMA) from vinyl modified silica mediated by 2-(2-cyanopropyl) dithiobenzoate was first conducted. Surface radicals generated by the addition reaction of “free” polymeric radicals with surface vinyl could initiate reversible addition-fragmentation chain transfer (RAFT) grafting polymerization of MMA from silica. The RAFT grafting polymerization of MMA from silica exhibited a living character, evident from the linear relationship of grafting ratio with monomer conversion. Grafting polymerization rate appeared dependent on target molecular weight and initiator concentration because of diffusion-controlled RAFT process on silica. The PMMA-grafted silica allowed for a second-step grafting polymerization due to the existence of dithioester group on the chain ends of grafted PMMA. In this case, surface radicals were generated by the reaction of “free” polymeric radicals with the surface RAFT agents, which differed from the RAFT process of grafting polymerization from vinyl modified silica. This method could be extended to prepare a variety of block copolymers from silica.

        Online Cutting Temperature Measurement System Based Thermocouple

        In order to measure cutter temperature, the on-line thermocouple temperature measurement system was proposed. In this system, temperature sensing element thermocouple was welded in the flank face close to the cutting edge with double bare wires welding way. With this method, this system has advantages of a single point temperature measurement and no effect on cutter temperature distribution. For a higher sampling rate, two AD were used to sample alternately; and for the convenient signal transmission when the cutter is rotating at high speed, wireless transmission was adopted to transmit thermocouple output signal and cold side temperature signal to the host computer. After temperature compensation to the cold side, the compensated temperature will be real-time displayed, stored and played back. System measurement range is 0 ~ 1300 ° C, and the measurement uncertainty is 0.58%. For 8000rpm rotational speed of XH714D machine, experimental results show that the system can measure cutter temperature online, and has advantages of low cost and great practicability.

        Characterisation of Shear Creep and Stress Relaxation of Serrate Discontinuity in Rock Mass under Different Loading Paths

        In engineering applications, rocks rarely exist in their monolithic form and discontinuities always occur in the rocks. Intuitively there is a considerable difference in rheological properties between monolithic rocks and rocks with discontinuities. The present paper aims to characterise the shear creep and stress relaxation of serrate discontinuity in rock mass under different loading paths. A comprehensive experimental program, including creep tests, stress relaxation tests and unloading tests, on serrate discontinuities, is developed with focus on the effect of the slope angle of the serrated surface of the discontinuity on its creep and stress relaxation. Empirical models for shear creep and stress relaxation are also developed. We found in the paper that the larger the slope angle of a discontinuity is the larger both the creep and stress relaxation are, and the stress-strain relation of discontinuities in rock masses can be different under different loading paths. The significance of the research is that it investigates the simultaneous shear creep and stress relaxation of a serrate discontinuity under various loading paths. The paper concludes that the empirical models for creep and stress relaxation developed in the paper can help geotechnical engineers to analyse and assess the behaviour of rock masses with discontinuities.

        Adaptive Complex Interpolator for Channel Estimation in Pilot-Aided OFDM System

        In an orthogonal frequency division multiplexing system,conventional interpolation techniques cannot correctly balanceperformance and overhead when estimating dynamic longdelaychannels in single frequency networks (SFNs). In this study,classical filter analysis and design methods are employed to derivea complex interpolator for maximizing the resistible echo delay in achannel estimator on the basis of the correlation between frequencydomain interpolating and time domain windowing. The coefficientcomputation of the complex interpolator requires a key parameter,i.e., channel length, which is obtained in the frequency domain witha tentative estimation scheme having low implementation complexity. The proposed complex adaptive interpolator is verified in asimulated digital video broadcasting for terrestrial/handheld receiver. The simulation results indicate that the designed channelestimator can not only handle SFN echoes with more than 200 µsdelay but also achieve a bit-error rate performance close to the optimumminimum mean square error method, which significantlyoutperforms conventional channel estimation methods, while preservinga low implementation cost in a short-delay channel.

        Optimistic Fault Diagnosis in Discrete Event Systems by Labeled Petri Nets and Basis Markings

        This paper deals with the fault diagnosis problem of discrete event systems modeled with labeled Petri nets. Its main contributions are threefold. First, depending on whether a diagnosis function examines the fault transitions that possibly occur after the last observed event, we formally divide the diagnosis functions into two types: optimistic and pessimistic, which aims to facilitate the exploration of different diagnosis approaches. Second, a framework is proposed, which extends a given diagnosis approach for Petri nets to the case of labeled Petri nets. The main idea of the framework is to compute and combine the diagnosis results of observable transition sequences corresponding to an observed word. Third, we convert a basis-marking-based approach that is originally pessimistic to the optimistic case and prove the correctness of this conversion.

        The plate-type fuel assembly adopted in nuclear research reactor suffers from complicated effect induced by non-uniform irradiation, which might affect stress conditions, mechanical behaviors and thermal-hydraulic performance of the fuel assembly. This paper is the Part II work of a two-part study devoted to analyzing the complex unique mechanical deformation and thermal-hydraulic characteristics for the typical plate-type fuel assembly under irradiation effect, which is on the basis of developed and verified numerical thermal-fluid-structure coupling methodology under irradiation in Part I of this work. The mechanical deformation, thermal-hydraulic performance and Mises stress have been analyzed for the typical plate-type fuel assembly consisting of support plates under non-uniform irradiation. It was interesting to observe that: the plate-type fuel assembly including the fuel plates and support plates tended to bend towards the location with maximum fission rate; the hot spots in the fuel foil appeared at the location with maximum thickness increment; the maximum Mises stress of fuel foil was located at the adjacent location with the maximum plate thickness increment et al.

        The plate-type fuel assembly adopted in nuclear research reactor suffers from complicated effect induced by non-uniform irradiation, which might affect its stress conditions, mechanical behavior and thermal-hydraulic performance. A reliable numerical method is of great importance to reveal the complex evolution of mechanical deformation, flow redistribution and temperature field for the plate-type fuel assembly under non-uniform irradiation. This paper is the first part of a two-part study developing the numerical methodology for the thermal-fluid-structure coupling behaviors of plate-type fuel assembly under irradiation. In this paper, the thermal-fluid-structure coupling methodology has been developed for plate-type fuel assembly under non-uniform irradiation condition by exchanging thermal-hydraulic and mechanical deformation parameters between Finite Element Model (FEM) software and Computational Fluid Dynamic (CFD) software with Mesh-based parallel Code Coupling Interface (MpCCI), which has been validated with experimental results. Based on the established methodology, the effects of non-uniform irradiation and fluid were discussed, which demonstrated that the maximum mechanical deformation with irradiation was dozens of times larger than that without irradiation and the hydraulic load on fuel plates due to differential pressure played a dominant role in the mechanical deformation.
